Opisuju se sami bioaktivni peptidi, te njihova uloga i pozitivni učinci na zdravlje ljudskog organizama. Navedeno je koji se to bioaktivni peptidi nalaze u samom pršutu, koje su njihove funkcije i od kakvog su značaja. Als Alternative zu konventionellen Batch-Prozessen können enzymatische Hydrolysen auch kontinuierlich durchgeführt werden. Um solche Prozesse durchführen zu können, wurde ein kontinuierliches Keramikkapillarmembran-Reaktorsystem entwickelt, das mit unterschiedlichsten hydrolytischen Enzymen betrieben werden kann. Letztere sind auf den Kapillarmembranen immobilisiert. Dieses System bietet zahlreiche Vorteile hinsichtlich Stabilität, Reproduzierbarkeit und Kontrollierbarkeit gegenüber Batch-Prozessen und kann einfach an die optimalen Reaktionsbedingungen angepasst werden.

Ici, nous avons comparé la dégradation des protéines myofibrillaires musculaires et l'autophagie, l'ubiquitine-protéasome et le Ca 2+-protéolyse dépendante par rapport au statut énergétique et redox chez les souris de type sauvage (WT) et les souris knock-out pour la myostatine (KO) au cours du stockage post-mortem précoce. Les muscles KO ont montré une dégradation plus élevée des protéines myofibrillaires dans les 24 premières heures après la mort, associée à un statut antioxydant préservé, par rapport aux muscles WT. L'analyse des principaux marqueurs de l'autophagie et du système ubiquitine-protéasome a indiqué que ces deux voies n'étaient pas régulées à la hausse dans le muscle post-mortem (les deux génotypes), mais que le flux autophagique basal et la teneur en ATP étaient inférieurs dans les muscles KO. Les activités du protéasome et de la caspase n'étaient pas différentes entre les souris WT et KO. À l'inverse, l'activité de la calpaïne était plus élevée dans les muscles KO, en même temps qu'une dégradation plus élevée de la troponine T et de la desmine. International audience; Rapeseed meal proteins (RP) are enzymatically hydrolyzed using three individual proteases (Alcalase, Flavourzyme, and Prolyve) and the enzymatic mechanism is studied. Rapeseed hydrolysates are produced under controlled conditions and the Prolyve hydrolysate is separated by membrane filtration. Their capacity to reduce free radicals (by transfer of hydrogen or electron) or transition metals (by electron transfer) in the absence of an oxidizable substrate, their metal chelating capacity as well as the antioxidant performances in model (conjugated autoxidizable triene assay) are investigated. All hydrolysates show a reduction capacity (by transfer of hydrogen or electron) and antioxidant activities, in a dose-dependent manner, which are however not significantly increased in comparison to the native proteins. A noteworthy metal chelating activity of the peptides produced with Prolyve is highlighted. These results indicate the potential of valorization of RP as a source of high metal chelating peptides to counteract lipid oxidation in foods. Practical applications: Over the last decade, the antioxidative potential of peptides from plant biomass has been evidenced by much research. Considering the myriad of possible sources and the diversity of technology and means to obtain peptides from protein materials, it is reasonable to expect more applications. Concomitantly, preventing lipid oxidation, especially with the polyunsaturated fat-based products, is a major concern in sectors such as agri-food and cosmetic. Although the efficacy of synthetic antioxidants is recognized, both consumers and manufacturers are looking for more innovative, healthy, environmental-friendly processes and quality products. In this context, a controlled proteolysis of proteins from plant by-products can be used as a sustainable strategy to produce antioxidant peptides. Among them, new peptides released from rapeseed proteins with Prolyve can provide interesting metal chelators to counteract lipid oxidation in foods.

The valorisation of agro-industrial by-products and the search for alternative sources of protein to produce peptides are of great importance. This work proposes the development of a process enabling the production of fractions rich in bioactive peptides from the by-product from brewing called “spent brewer’s yeast”. The motivation of this subject of research is based on the increasing demand to reuse agro-industrial by-products such as spent yeasts and on the production bioactive peptides using clean and efficient technologies. Spent brewer’s yeast slurry was collected after maturation and was submitted to cell wall disruption methods. Autolysis, glass bead milling and enzymatic hydrolysis using Brauzyn® were compared, and enzymatic hydrolysis presented a higher protein recovery and improved antioxidant activity. A simultaneous cell wall disruption and peptide production was proposed using a mixture design employing Brauzyn®, Protamex™ and Alcalase™ (pH 7.0, 50 °C, 2000 U g-1 for 2 h), being able to reduce steps during the processing of spent brewer’s yeast. Protein hydrolysates characteristics varied with the proportion of enzymes used, changing the extent of the release of hydrophobic residues, the degree of hydrolysis, antioxidant properties, browning extent and yield of solids and peptides. Membrane separation of the complex protein hydrolysate was studied firstly using polymeric membranes of regenerated cellulose and polyethersulfone and then in ceramic ones. A smaller susceptibility to fouling was observed for more hydrophilic surfaces, and at higher feed pH values. Results confirmed that the main foulants during ultrafiltration of spent brewer’s yeast protein hydrolysate are peptides that adsorb easily onto the membrane surface. Fractionation using ceramic membranes of 50-1 kg mol-1 of molecular weight cut-off was able to separate multi-active peptides from total sugars and ribonucleic acids. Spent brewer’s yeast peptides presented antioxidant activity by different mechanisms, in vitro anti-diabetic activity (inhibition of α-glucosidase and α-amylase) and anti-Alzheimer activity (inhibition of acetylcholinesterase). Sequential processing of spent brewer’s yeast using enzymatic and membrane separation technologies was able to recover peptides with multiple bioactivities from an underused by-product from brewing. Fractions produced represent an alternative as peptide-rich ingredients in the food and pharmaceutical industries., La valorisation des sous-produits agro-industriels et la recherche de sources alternatives de protéines pour la production de peptides sont d’une grande importance. Ce travail propose le développement d’un procédé capable d’obtenir des fractions riches en peptides bioactifs à partir du sous-produit de brasserie appelé « levure résiduelle de bière ». Ces travaux de recherche sont motivés par la nécessité de valorisation des sous-produits agro-industriels et la demande croissante de peptides bioactifs produits par des technologies vertes et efficaces. La suspension de levure résiduelle de bière a été recueillie après maturation puis soumise à différents traitements de rupture de la paroi cellulaire. L’autolyse, le broyage par billes de verre et l’hydrolyse enzymatique utilisant Brauzyn® ont été comparés, et l’hydrolyse enzymatique a permis une récupération plus importante des protéines avec une activité antioxydante plus élevée. Une rupture de la paroi cellulaire simultanée à la production de peptides a été proposée grâce à un plan de mélanges en utilisant Brauzyn®, Protamex™ et Alcalase™ (pH 7,0, 50 °C, 2000 U g-1 pendant 2 h). Cette procédure a permis de réduire le nombre d’étapes requises pour le traitement des levures résiduelles de bière. Il a été montré que les caractéristiques des hydrolysats obtenus (la quantité de résidus hydrophobes libérés, le degré d’hydrolyse, les propriétés antioxydantes, l’évolution du brunissement et le rendement en solides) est fonction de la proportion en enzymes utilisée. Le fractionnement par techniques membranaires de l’hydrolysat protéique a d’abord été étudié en utilisant des membranes polymères en cellulose régénérée et en polyethersulfone, et ensuite avec des membranes céramiques. Les phénomènes de colmatage sont moins importants lorsque les surfaces sont hydrophiles et lorsque la valeur du pH de l’alimentation est élevée. Les résultats ont confirmé que les principaux éléments colmatants de l’hydrolysat de protéines de levure résiduelle de bière sont les peptides qui s’absorbent facilement à la surface des membranes. Le fractionnement en cascade avec des membranes céramiques de 50 à 1 kg mol-1 de seuil de coupure a permis de séparer des peptides multi-bioactifs des sucres totaux et des acides ribonucléiques. Les peptides de levure résiduelle de bière ont présenté des activités antioxydantes impliquant différents mécanismes d’action, une activité anti-diabétique (inhibition de l’α-glucosidase et de l’α-amylase) et une activité anti-Alzheimer (inhibition de l’acétylcholinestérase). Le traitement séquentiel de la levure résiduelle de bière couplant des technologies d’hydrolyse enzymatique et les techniques de séparation par membranes a permis de récupérer des peptides ayant de multiples bio-activités à partir d’un résidu sous-utilisé du brassage. Les fractions produites représentent une alternative en tant qu’ingrédients riches en peptides pour des applications en industries alimentaires et pharmaceutiques.

Der co-translationale Proteintransport von Membranproteinen ist ein entscheidender Prozess zur Aufrechterhaltung der Membranintegrität, um somit die Lebensfähigkeit der Zelle zu gewährleisten. In \(\textit {Escherichia coli}\) wird der co-translationale Proteintransport durch den konservierten Signalerkennungspartikel (SRP-Komplex) vermittelt. Der SRP-Komplex ist ein Ribonukleoprotein, welches aus dem Ffh-Protein und der 4,5S RNA besteht. In der vorliegenden Arbeit wurde die Lon-abhängige Proteolyse von Ffh in \(\textit {E. coli}\) untersucht. Freies Ffh, welches nicht an die 4,5S RNA gebunden ist, wird der Proteolyse zugeführt. Die Proteolyse erfolgt solange, bis eine minimale Ffh-Konzentration in der Zelle erreicht ist. Für die Erkennung von Ffh durch Lon, scheint die RNA-Bindefähigkeit des Ffh-Proteins ausschlaggebend zu sein. Durch die Proteolyse von Ffh ist die AAA\(^{+}\)-Protease Lon in die Aufrechterhaltung des co-translationalen Proteintransports als auch der Membranintegrität involviert.

National audience; Introduction et but de l’étude Le processus de fabrication des préparations pour nourrissons (PPNs) implique des traitements thermiques, modifiant les propriétés physico-chimiques des protéines et éventuellement leur cinétique de protéolyse. L'objectif de l'étude consiste en l’analyse des effets de la composition et des structures protéiques thermo-induites des PPNs sur la cinétique de protéolyse. Matériel et méthodes Deux PPNs modèles ont été formulées à 1,3% de protéines. Elles se distinguent par la qualité des protéines du lactosérum (α-lactalbumine, β-lactoglobuline et lactoferrine) : la PPN « standard » mime la composition protéique de la majorité des PPNs 1er âge commercialisées et la PPN « LF+ ALA+ » celle du lait maternel. Les PPNs ont été traitées thermiquement à 67,5°C ou 80°C avec un taux de dénaturation des protéines du lactosérum identique de 65%. Les PPNs sans traitement thermique ont servies de référence. Les structures protéiques thermo-induites ont été caractérisées par électrophorèse, fractionnement d’écoulement de champ couplé à la diffusion de lumière multi-angle et microscopie électronique. D’autre part, les PPNs ont été soumises à la digestion gastro-intestinale en condition statique in vitro au stade nouveau-né à terme (n = 3). Le degré d’hydrolyse a été mesuré par dosage des amines primaires, la cinétique de protéolyse suivie par électrophorèse et l’évolution de la structure au cours de la digestion par granulométrie à diffraction laser. Résultats et analyse statistique A taux de dénaturation protéique équivalent, les caractéristiques morphologiques des structures protéiques thermo-induites diffèrent selon la composition protéique des PPNs et la température de traitement thermique. La formation d’agrégats solubles a été observée uniquement pour la PPN « standard », tandis que la fixation de protéines dénaturées sur les micelles de caséines a été constatée pour les deux PPNs. Les agrégats formés à 67,5°C ont une plus petite masse pour la PPN « standard » et un plus petit diamètre pour la PPN « LF+ ALA+ » que ceux formés à 80°C. Au cours de la phase gastrique, le traitement thermique accélère la cinétique de protéolyse des caséines de la PPN « standard » contrairement à la PPN « LF+ ALA+ ». La cinétique de protéolyse de l’α-lactalbumine dépend de la composition protéique des PPNs et de la température de chauffage de la PPN « LF+ ALA+ ». La protéolyse de la lactoferrine est fortement affectée par le traitement thermique. Les entités protéiques formées par agrégation en fin de phase gastrique sont affectées par la composition protéique des PPNs et le traitement thermique. Conclusion Notre étude a permis de mettre en évidence les effets de la composition protéique des PPNs et des structures protéiques thermo-induites sur les cinétiques de protéolyse. Ces deux paramètres sont donc à prendre en compte pour l’optimisation des PPNs en termes de stratégie nutritionnelle.

Proteasomes are large ubiquitous macromolecular assemblies composed of a 20S catalytic complex and a regulatory particle containing an AAA-ATPase module. This cellular machine is responsible for selectively degradation of intracellular proteins in order to allow proteome renewal, elimination of defective proteins and control of many biological functions. The objective of this thesis was to reveal the mechanisms by which the AAA-ATPase complexes selectively unfold substrate proteins and translocate them into the 20S particle, where they are destroyed. To this end, an innovative approach was used by combining time-resolved small-angle neutron scattering (TR-SANS) combined with fluorescence spectroscopy to monitor biochemical activity. The proteasome of the hyperthermophilic archaea Methanocaldococcus jannaschii was used as a model system. It is composed of the 20S protease of the regulatory particle AAA-ATPase PAN. A variant of the fluorescent protein GFP was used as a substrate protein.The data obtained show that PAN unfolding activity generates denatured species of GFP forming aggregates. The association with the 20S particle prevents the formation of these species and indicates that once the unfolding of a substrate by PAN is engaged, translocation into the 20S complex and degradation processes are closely coupled. Analysis of the neutron scattering spectra of the GFP substrate reveal that the native GFP population is rapidly disappearing in favor of peptides generated by the 20S protease, as confirmed by mass spectrometric analysis. This demonstrates the highly processive nature of the proteasome. Finally, two modes of action of PAN have been identified depending on the amount of proteins to be degraded compared to the PAN-20S proteasome. This work allows to experimentally validate one of the proteasome function models previously proposed and emphases the importance of controlling the association of proteasomes in vivo. This study also highlights the interest of TR-SANS technique to study the functional dynamics of large cellular machines., Les protéasomes sont de grands assemblages macromoléculaires ubiquitaires composés d’un complexe catalytique 20S et d’une particule régulatrice comprenant un module AAA-ATPase. Cette machine cellulaire est chargée de dégrader sélectivement les protéines intracellulaires pour permettre le renouvellement du protéome, éliminer les protéines défectueuses et contrôler de nombreuses fonctions biologiques. Le travail de cette thèse avait pour objectif de mettre à jour les mécanismes qui permettent aux complexes AAA-ATPase de déplier sélectivement les protéines substrat et de les transférer à la particule 20S, dans laquelle elles sont détruites. Pour cela, une approche novatrice a été utilisée en combinant la diffusion de neutrons aux petits angles résolue en temps (TR-SANS) avec la spectroscopie de fluorescence permettant de suivre l’activité biochimique. Le protéasome de l’archée hyperthermophile Methanocaldococcus jannaschii, a été utilisé comme système modèle. Il est composé de la protéase 20S et de la particule régulatrice AAA-ATPase PAN. Un variant de la protéine fluorescente GFP a été utilisé comme protéine substrat.Les données obtenues montrent que l’activité de dépliement de PAN génère des formes de GFP dénaturée formant des agrégats. En revanche, l’association avec la particule 20S prévient la formation de ces espèces et indique qu’une fois le dépliement d’une protéine par PAN engagé, les processus de transfert dans le complexe 20S et de dégradation sont étroitement couplés. L’analyse des spectres de diffusion neutronique du substrat GFP montrent que la population de GFP native disparait rapidement au profit des peptides générés par la protéase 20S, comme confirmé par une analyse en spectrométrie de masse. Cela démontre le caractère hautement processif du protéasome. Enfin, deux modes d'action de PAN ont été mis en évidence selon la quantité de protéines à dégrader par rapport au protéasome PAN-20S. Ces travaux permettent de valider expérimentalement un des modèles de fonctionnement du protéasome préalablement proposés et soulignent l’importance d’un contrôle de l’association des protéasomes in vivo. Cette étude met également en valeur l’intérêt de la technique TR-SANS pour étudier la dynamique fonctionnelle de grandes machines cellulaires.

Pulmonary inflammatory diseases such as cystic fibrosis and COPD, have in common a high influx of neutrophils that secrete proteases responsible for tissue injury and the progressive loss of lung function. To date there is no method able to assess lung enzyme activity in vivo. Such a method would allow an early diagnostic of any protease/inhibitor imbalance long before the detection of pulmonary lesions by anatomical imaging methods. Lungs function could thus be preserved with protease inhibitors. MRI enhanced by dynamic nuclear polarization (DNP) is an approach that reveal proteolytic activity through specific nitroxide. However, lung imaging by MRI is demanding, because of intrinsic properties. In order to obtain signal in the lung by MRI, a radial sequence imaging is often favorable. Indeed, radial trajectories benefit from various advantages like their low sensitivity to motion artefacts as well as the opportunity to highly undersample acquisitions. Thus the aim of this thesis was to develop a 3D Ultra Short Echo Time sequence on a new low field MRI system (0.2T). This sequence has been compared to a robust spin echo sequence. Both methods have been implemented in the system to obtain 3D images in a limited acquisition time. After their in vitro evaluation, their in vivo use in mice gives access to the biodistribution of nitroxides in the lung, like substrate of neutrophil elastase. Enhanced MRI allowed in vitro monitoring of the enzymatic activities of serine proteases inthe case of infected lungs.; Les maladies inflammatoires pulmonaires tels que la mucoviscidose et le BPCO, ont pour point commun un influx important de neutrophiles qui libèrent des protéases, responsables des lésions tissulaires et de la perte progressive de fonction. À ce jour, il n'existe aucune méthode permettant d'évaluer l'activité des protéases dans le poumon in vivo. Une telle méthode permettrait un diagnostic précoce de tout déséquilibre protéase / inhibiteur bien avant la détection des lésions pulmonaires par des méthodes d'imagerie anatomique ainsi que l'évaluation de thérapies avec des inhibiteurs de protéases. L'IRM rehaussée par Polarisation dynamique nucléaire (PDN) est une approche possible pour révéler de telles activités enzymatiques en utilisant des nitroxydes spécifiques. Cependant, l’imagerie du poumon par IRM est encore aujourd’hui exigeante, en raison des propriétés intrinsèques à l'organe. Dans la nécessité d'obtenir du signal dans le poumon en IRM, une séquence radiale est souvent privilégiée. En effet, les trajectoires radiales bénéficient de nombreuses avantages comme leur faible sensibilité aux artefacts de mouvements ainsi que la possibilité de sous-échantillonner les acquisitions. Ainsi, l’objectif de cette thèse est de développer une séquence 3D à Temps d'Echo Ultra court sur un nouveau système IRM bas champ (0.2T) et de la comparer à une séquence robuste utilisant l'écho de spin. Les deux méthodes ont été implémentés au système afin d'obtenir des images en 3D en un temps d’acquisition limité. Après leur évaluation in vitro, leur utilisation in vivo chez la souris donnent accès à la biodistribution de nitroxydes dans le poumon, dont un substrat de l'élastase du neutrophile. L'IRM rehaussée a par ailleurs permis d'évaluer in vitro les activités enzymatiques de protéases à sérine dans le cas de poumons infectés.

Infant formulas (IF) are the only source of nutrition for infants when breastfeeding is not possible. Usually, IF are spray-dried to a powder form and include cow milk proteins as well as whey protein concentrate. In the future, plant proteins could be an interesting alternative to the use of animal proteins for nutritional, functional and sustainable concerns. The objective of this work was to study the influence of the partial substitution of whey protein concentrate by plant proteins on the functional properties and the digestibility of IF. For this purpose, proteins from pea, faba bean, rice and potato replaced half of the proteins in plant-based IF and where compared to a reference IF based on whey proteins. The digestibility of these IF, manufactured at pilot scale then at semi-industrial scale, was assessed through in vitro digestion simulating infant physiological conditions using static and dynamic models. Although all IF showed a balanced nutritional profile compared to the infant needs, it was demonstrated that the protein source had a great impact on IF properties. Indeed, solubility and viscosity limits observed for rice and potato IF affected their manufacturing as well as their digestibility. On the contrary, pea and faba bean IF showed physicochemical properties and protein nutritional quality very close to the reference. These promising results highlight future innovation possibilities in infant nutrition field. However, they need to be validated at the real industrial scale and extended through in vivo studies.; Les formules infantiles (FI) sont la seule source nutritionnelle pour les nourrissons lorsque l’allaitement est impossible. Les FI en poudre sont obtenues par séchage par atomisation et sont composées de protéines de lait de vache, majoritairement des protéines du lactosérum. A l’avenir, les protéines végétales pourraient constituer une alternative nutritionnelle, fonctionnelle et durable à l’utilisation de protéines animales. L’objectif de ce travail était d’étudier l’influence de la substitution partielle des protéines laitières par des protéines végétales sur les propriétés fonctionnelles et la digestibilité de FI innovantes. A cet effet, des protéines de pois, féverole, riz et pomme de terre ont remplacé la moitié des protéines dans des FI végétalisées et ont été comparées à une FI de référence composée de protéines du lactosérum. La digestibilité de ces FI, fabriquées à échelle pilote puis semi-industrielle, a été étudiée en digestions in vitro statique puis dynamique simulant les conditions physiologiques du nourrisson. La source de protéine influence les propriétés des FI, qui étaient cependant toutes nutritionnellement adaptées aux besoins du nourrisson. Les limites de solubilité et de viscosité observées pour les FI à base de riz et de pomme de terre affectent leur fabrication et leur digestibilité. A l’inverse, les FI à base de pois et de féverole présentent des propriétés physicochimiques et nutritionnelles très proches de la référence. Ces résultats prometteurs éclairent les possibilités d’innovation en nutrition infantile. Ils devront toutefois être confirmés à échelle industrielle et complétés par des études in vivo.

Serratia liquefaciens is a psychrotrophic species, frequently found in raw milk, which secretes Ser2, a heat-resistant protease. Involvement of this species in UHT milk destabilisation was investigated in the present study. Microfiltered milk was inoculated independently with strains S. liquefaciens L53 or L64. Then, UHT treatment was performed and stability of the corresponding UHT milk was investigated during three months of storage. The residual proteolytic activity of strain L53 led to destabilisation of UHT milk, with sedimentation and formation of aggregates. Hydrolysis of casein micelles was confirmed by the increase in the content of non-casein nitrogen and the identification of numerous peptides coming from the four caseins using mass spectrometry. For strain L64, no visual and biochemical alteration were found. This study showed that Ser2 resists UHT treatment and could be a cause of UHT milk destabilisation; however, this destabilisation by S. liquefaciens was strain-dependent.

Human milk feeding is an important recommendation for preterm newborns considering their vulnerability and digestive immaturity. Holder pasteurization (62.5 C, 30 min) applied in milk banks modifies its biological quality and its microstructure. We investigated the impact of pasteurization of preterm human milk on its gastrointestinal kinetics of lipolysis, proteolysis and structural disintegration. An in vitro dynamic system was set up to simulate the gastrointestinal digestion of preterm newborns. A pool of preterm human milk was digested as raw or after Holder pasteurization. Pasteurization impacted the microstructure of undigested human milk, its gastrointestinal disintegration and tended to limit the intestinal lipolysis. Furthermore, the gastrointestinal bioaccessibility of some fatty acids was decreased by pasteurization, while the intestinal bioaccessibility of some amino acids was selectively modulated. The impact of pasteurization on the digestion of human milk may have nutritional relevance in vivo and potentially modulates preterm development and growth.

IntroductionPlant proteins have recently gained attention in research and industrial areas by their relatively modestenvironmental, ethical and economic impact.ObjectiveFood structure associated to food processing are known to generally impact sensitivity of proteinsto digestive enzymes.MethodologyFour plant-based foods produced from different protein sources (soy, pea or wheat) and offeringdifferent structure and their corresponding concentrate or isolate were digested following the INFOGESTprotocol. Since tofu and seitan cannot be stored without structure degradation, the impact offreeze drying on protein digestion was also studied. Proteolysis progress was followed by monitoringthe release of free amino-groups and proteolysis degree was assessed. Multi-scale characterizationof foods was performed to allow data interpretation.Main findingsPea emulsion exhibited similar droplet size and lipid content than soymilk, only the protein sourcechanged. Final proteolysis degree was higher with pea (60 %) than soy (45 %). When comparingthe same source, protein digestion of tofu was more extensive than that of soymilk. This could beexplained by differences in macrostructure since tofu is obtained through soy protein coagulation,but also by differences in the origin of beans. Indeed, proteolysis degrees at the end of the intestinalphase were different between protein isolates created from beans used for tofu vs soymilk (15% vs 40 %). Proteins of tofu isolate were less hydrolyzed than tofu (15 % vs 26 %), that could beexplained by different protein denaturation levels. Comparing a simple solubilization of commercialpea isolate with its emulsion with soy lipids, proteolysis was twice higher for the emulsion (60 % vs30 %).ConclusionUltimately, freeze-drying effect of protein digestion was highlighted for seitan, increasing final proteolysisrate from 9 % to 22 %, but not for tofu explained through enzymatic accessibility improvedafter freeze drying in the case of seitan.

Receptor cell-surface levels can be regulated by internalization, proteolytic cleavage of the membrane-bound receptor or synthesis of new receptors. Following internalization, the receptors are either recycled to the cell-surface or degraded in the lysosome. I could show that the IL-6R and gp130 are internalized in a clathrin- and dynamin-dependent manner which was not influenced by IL-6. Furthermore, the ICD of the IL-6R is dispensable for IL-6R internalization, however, I could show that gp130 influences IL-6R internalization to some extent. Following internalization, both receptors are recycled back to the cell surface via Rab11-positive recycling endosomes but are also degraded in the lysosome. Stimulation with IL-6 led to an increase in recycling of both receptors, but only degradation of gp130 in the lysosome was enhanced. Additionally, it was revealed that STAT3 signaling was only activated upon internalization of the IL-6/IL-6R/gp130 signaling complex, indicating that signaling is initiated at endosomes. Only little is known about endogenous stimuli leading to the release of sIL-6R in vivo. I could show that, in humans, activation of TLR2 resulted in an increase in sIL-6R release mainly from monocytes which was generated by proteolytic cleavage of the membrane-bound IL-6R mediated by ADAM10 and 17. Furthermore, TLR2 activation led to the activation of the ERK signaling pathway which was found to differentially regulate the release of IL-6 and sIL-6R. The sIL-6R is also released constitutively but the responsible protease has not been clearly identified so far. Using a screening procedure, cathepsin S was identified as a new protease that can proteolytically cleave the IL-6R. However, cathepsin S uses a different cleavage site in the IL-6R in vitro as has been determined for the sIL-6R in vivo, indicating that it is rather not involved in the generation of the constitutively released sIL-6R. Nach der Internalisierung von Rezeptoren können diese entweder recycled oder im Lysosom abgebaut werden. Ich konnte zeigen, dass IL-6R und gp130 über einen Clathrin- und Dynamin-abhängigen Mechanismus internalisiert werden, wobei IL-6 die Internalisierung nicht beeinflusst. Der intrazelluläre Teil des IL-6R wird für die IL-6R-Internalisierung nicht benötigt, jedoch konnte ich zeigen, dass gp130 die Internalisierung des IL-6R zum Teil beeinflusst. Anschließend an die Internalisierung werden der IL-6R und gp130 sowohl über Rab11-positive Recycling Endosomen an die Zelloberfläche zurück transportiert, als auch im Lysosom abgebaut. Stimulation mit IL-6 führte zu einem erhöhten Recycling beider Rezeptoren, jedoch wurde nur gp130 vermehrt im Lysosom abgebaut. Des Weiteren zeigte sich, dass die STAT3-Signaltransduktion erst nach Internalisierung des IL-6/IL-6R/gp130-Komplexes aktiviert wird, was darauf schließen lässt, dass diese an Endosomen stattfindet. Bislang ist wenig über endogene Aktivatoren der sIL-6R Generierung in vivo bekannt. Ich konnte zeigen, dass im Menschen die Aktivierung von TLR2 zur Generierung des sIL-6R durch proteolytische Spaltung des membrangebundenen IL-6R durch ADAM10 und ADAM17 von Monozyten führt. TLR2-Aktivierung resultierte außerdem in der Aktivierung des ERK-Signalweges, welcher die Freisetzung von sIL-6R und IL-6 gegensätzlich reguliert. Der sIL-6R wird auch konstitutiv gebildet, jedoch ist die hierfür verantwortliche Protease noch nicht eindeutig nachgewiesen. Durch ein Screeningverfahren wurde Cathepsin S als eine neue Protease identifiziert, die den IL-6R proteolytisch spalten kann. Jedoch zeigte sich, dass Cathepsin S in vitro eine andere Schnittstelle im IL-6R verwendet als in vivo nachgewiesen wurde und daher vermutlich keine Rolle in der Generierung des konstitutiv gebildeten sIL-6R spielt.

Two protein tanning methods were evaluated to contribute to the withdrawal of formaldehyde as a tanning agent of meals for feeding ruminants. The experimental materials were two fractions of rapeseed and sunflower meals collected at the positive electrode of an electrostatic separator, presenting high contents in proteins and phenolic compounds. The objective was to make phenolics and proteins interact without addition of exogenous tannins. Treatment CH incubated a meal fraction:water mixture (1:2, w:w) for 48 h at 50 degrees C. Treatment FR incubated a meal fraction:water mixture (1:10, w:w) at pH 9.0 for 48 h at 4 degrees C. Microbial proteolysis on meal fractions were quantified during 24 h rumen batch fermentations with cellulose and starch as nitrogen-free energy sources. The net production of ammonia tended to be reduced by treatment FR mostly on rapeseed, corresponding to an 8% saving of rapeseed meal proteins degradable in the rumen. When untreated, the sunflower fraction decreased methane production by 50%, while treatments restored the fermentation pattern. Cold alkaline treatment could be considered to protect meal proteins from degradation by rumen micro-organisms., Deux méthodes de tannage des protéines ont été évaluées pour contribuer au remplacement du formaldéhyde comme agent tannant des tourteaux destinés à l’alimentation des ruminants. Les matériaux expérimentaux étaient deux fractions de tourteaux de colza et tournesol collectées à l’électrode positive d’un séparateur électrostatique, présentant des teneurs élevées en protéines et en composés phénoliques. Le but était de faire interagir les composés phénoliques et les protéines sans addition de tanins exogènes. Le traitement CH a consisté à incuber un mélange tourteau/eau (1/2, poids/poids) pendant 48 h à 50 °C. Le traitement FR a consisté à incuber un mélange tourteau/eau (1/10, poids/poids) à pH 9,0 pendant 48 h à 4 °C. La protéolyse des fractions de tourteau par les microbes du rumen a été quantifiée lors de fermentations in vitro de 24 h avec de la cellulose et de l’amidon comme sources d’énergie sans azote. Le traitement FR a eu tendance à réduire la production nette d’ammoniac, principalement avec le colza, équivalant à la protection de 8 % des protéines de tourteau de colza dégradables dans le rumen. La fraction de tournesol non traitée a diminué la production de méthane de 50 %, cependant les traitements ont restauré le profil fermentaire. Le traitement alcalin à froid pourrait être envisagé afin de protéger les protéines du tourteau de la dégradation par les micro-organismes du rumen

Introduction Propionibacterium freudenreichii (PF) is a Swiss-type cheeses starter and has versatile probiotic properties due to various beneficial metabolites. PF recently emerged as a suitable candidate to develop fermented functional foods. Delivery of viable bacteria to the colon is crucial for in situ beneficial metabolites production. The delivery vehicle was shown to influence P. freudenreichii tolerance towards digestive stresses. It is also suggested to influence PF adaptation to the colon environment. In addition, during digestion, the delivery vehicle may prevent or allow proteolysis of surface proteins. These surface proteins are generally thought to contribute to bacteria tolerance towards environmental stresses. Aim This study aimed at evaluating PF (strain CIRM-BIA 129) adaptation to the caecal environment, according to the delivery vehicle. Besides, we evaluated PF survival in the caecal environment after removal of bacterial surface proteins by enzymatic proteolysis. We address the plausible role of S-layer protein SlpB in PF adaptation and survival. Methods Caecal medium was prepared from caecal contents recovered from piglets. PF pellets were harvested from cultures in Yeast Extract-Lactate (YEL), in Milk ultrafiltrate, or from cheese. Wild-type and SlpB KO mutant PF were further compared with respect to survival in caecal medium. Bacterial pellets were also submitted to surface proteins proteolysis to mimic digestion. Cultures were incubated 50 h at 37°C in anaerobiosis. Survival was monitored by plate counting, optical density and by live/dead fluorescence labeling. Results PF survival decreased in the caecal medium with a similar rate, whatever the growth medium tested. However, optical density was stable and the epifluorecence images did not show dead bacteria. Reduced survival rate was not due to significant cell death.PF seemed rather to enter a viable but nonculturable state in the caecal medium. Surprisingly, proteolytic removal of surface layer proteins enhanced PF culturable state in caecal medium. In addition, the mutation of SlpB did not change PF culturable state but surface proteins proteolysis enhanced also PF KO SlpB s culturable state. Conclusion These results indicate that surface proteins proteolysis during digestion may enhance PF survival and metabolic activity within the caecal medium. A comparative proteomic analysis will address the molecular mechanisms involved in enhanced survival.

As the main energy producer of the eukaryotic cell, mitochondria are also a major source of reactive oxygen species and a prime target for oxidative damage to proteins. To cope with these mechanisms involved in ageing and a large number of pathologies, the mitochondrion has protein maintenance systems among which the Lon protease. The objective of this thesis was to clarify the role of this protease localized in the mitochondrial matrix and particularly involved in mitochondrial proteins homeostasis and the elimination of oxidized proteins. This study was conducted on transformed HeLa cells so that Lon expression could be significantly reduced. Our results show that lack of Lon affects protein homeostasis by elevating carbonyl protein levels. We have also identified a number of proteins with varying levels of expression and oxidative changes in the absence of Lon. The mitochondrial function is also disturbed by Lon down expression as this is accompanied by a decrease in the activity of the respiratory chain. In addition, slightly more than a third of the proteins affected by Lon failure are mitochondrial and about half of them are involved in two areas: energy metabolism and protein quality control. Mitochondrial network fragmentation and potential mitochondrial DNA integrity damages noted in Lon-free cells are further arguments in favor of mitochondrial dysfunction. Finally, the impact of Lon deficiency on protein homeostasis and mitochondrial function can be modulated by the carbon source available to the cells since it varies according to whether the culture medium is supplemented with glucose or galactose.; Principal producteur d’énergie de la cellule eucaryote, la mitochondrie est aussi une source majeure d’espèces réactives de l’oxygène et une cible privilégiée des dommages oxydatifs causés aux protéines. Face à ces mécanismes impliqués dans le vieillissement et un nombre important de pathologies, la mitochondrie dispose de systèmes de maintenance des protéines parmi lesquels la protéase Lon. L’objectif de cette thèse a été de préciser le rôle de cette protéase de la matrice mitochondriale impliquée dans l’homéostasie des protéines mitochondriales et l’élimination des protéines oxydées. Cette étude menée sur des cellules HeLa transformées de façon à pouvoir significativement restreindre l’expression de Lon montre qu'une déficience en Lon affecte l’homéostasie protéique en élevant les niveaux de protéines carbonylées. Nous avons aussi identifié un certain nombre de protéines présentant une variation des niveaux d’expression et des modifications oxydatives en absence de Lon. La fonction mitochondriale est également perturbée en absence de Lon puisque celle-ci s’accompagne d’une baisse de l’activité de la chaîne respiratoire. De plus, plus du tiers des protéines affectées par une défaillance en Lon sont mitochondriales et environ la moitié d’entre elles interviennent dans deux domaines : le métabolisme énergétique et le contrôle qualité des protéines. La fragmentation du réseau mitochondrial et de potentielles atteintes à l’intégrité de l’ADN mitochondrial notées dans les cellules sans Lon sont d’autres arguments en faveur d’une dysfonction mitochondriale. Enfin l’impact d’une déficience en Lon varie selon que le milieu de culture est supplémenté en glucose ou en galactose.

The αs1-casein hydrolysis has been shown to greatly influence the texture and techno-functionalities of hard cooked cheeses. This hydrolysis can be performed by the proteinases of the thermophilic lactobacilli since rennet, also implied, is denatured during the cooking step. However, it is unclear whether these changes are only due to the proteolytic activity of lactobacilli, the pH decrease in cheese or both. Small scale Swiss-cheeses were made from microfiltered milk using various combinations of starters including or not lactobacilli: i) Streptoccocus thermophilus alone (STH cheeses), ii), S. thermophilus in enriched lactose cheesemilk (STH-LACT cheeses) to reach pH level similar to cheese with lactobacilli and iii) S. thermophilus and lactobacilli either L helveticus (STH/LH cheeses), or L. delbrueckii subsp lactis (STH/LL cheeses). As S. thermophilus does not metabolize the galactose moiety of lactose, the pH at day 1 of these STH-LACT cheeses was similar to those made with lactobacilli (pH 5.15-5.25) while pH values of the STH cheeses were higher (pH 5.40). Higher firmness and chewiness of the melted ripened STH cheeses were correlated with a higher pH at day 1 and with a lower proteolysis (twofold amount of remaining intact αs1-casein) compared to the STH-LACT cheeses as well as the cheeses with lactobacilli. Conversely, the stretchability showed different behaviors depending both on the quantitative and qualitative proteolysis and on the pH value. The lower pH and content in small peptides led to higher stretchability of the STH-LACT and STH/LH cheeses in contrast to the cheeses STH (high pH and few small peptides) and STH/LL (low pH and high amount of small peptides). In conclusion, the decrease in pH due to the use of lactobacilli plays a key role in the texture of raw and melted Swiss-cheese. Beside the pH, the proteolytic activity of lactobacilli is also a determinant factor for stretchability.

Der sekundäre Botenstoff c-di-GMP kommt in vielen Bakterienspezies vor und fördert dort die Bildung von Biofilmen. Für Abbau und Synthese von c-di-GMP in der Zelle sorgen Diguanylatzyklasen (DGCs) und Phosphodiesterasen (PDEs), an deren N-Terminus häufig Sensordomänen die enzymatische Aktivität steuern. In dieser Arbeit wurde die Regulation und Wirkungsweise einer Gruppe von PDEs mit einer neuartigen Sensordomäne, der CSS-Domäne, genauer charakterisiert. Die CSS-Domäne ist im Periplasma lokalisiert und besitzt zwei hochkonservierte Cysteine, von denen eines in dem namensgebenden CSS-Motiv arrangiert ist. Die Integration in die innere Membran erfolgt durch zwei Transmembrandomänen (TM1 und TM2), wobei TM2 die Verbindung zu der C-terminal gelagerten EAL-Domäne mit PDE-Aktivität herstellt. Die Analyse von PdeC als eine von fünf CSS-PDE in Escherichia coli K-12 zeigte, dass die Bildung einer Disulfidbrücke zwischen den konservierten Cysteinen vom oxidierende DsbA/DsbB-System katalysiert wird und zu einer Verminderung der enzymatischen Aktivität der EAL-Domäne führt. Im Gegensatz dazu führt die reduzierte freie Thiol-Form der CSS-Domäne zu einer stark erhöhten PDE-Aktivität verbunden mit der Dimerisierung über die TM2. Die Reduktion der CSS-Domäne führt außerdem zur Prozessierung durch die HtrA-Proteasen DegP und DegQ in ein membranständiges Fragment aus TM2+EAL-Domäne mit hoher enzymatischer Aktivität. Der Abbau durch DegP und DegQ im Periplasma ist sehr effizient. Auf der cytoplasmatischen Seite hingegen erfolgt die weitere Degradierung durch noch unbekannte Proteasen eher langsam, wodurch es unter bestimmten Bedingungen zur Akkumulierung der hochaktiven TM2+EAL-Form kommt. Durch das Zusammenspiel von redox-abhängiger Aktivitätskontrolle und Proteolyse der c-di-GMP-spezifischen PDE PdeC wird die Produktion der amyloiden Curli-Fasern und Cellulose reguliert, welche Hauptbestandteil der extrazellulären Matrix von Biofilmen sind. The second messenger c-di-GMP promotes biofilm formation in many bacterial species. Phosphodiesterases (PDEs) and diguanylatcyclases (DGCs) - often controlled by various N-terminal sensor domains - degrade and synthesize c-di-GMP. The aim of this study was to characterize the mode of activation and physiological function of a new class of sensor domains, the CSS domain, which is coupled to an EAL domain with PDE activity and therefore potentially controlling c-di-GMP degradation. The CSS sensor domain contains two highly conserved cysteins in the periplasmic loop, of which one is arranged in the characteristic CSS motif. Integration into the inner membrane is ensured by two flanking transmembrane domains (TM1 and TM2), with TM2 providing a connection to the C-terminal EAL domain. Analysis of PdeC as one of 5 CSS-PDEs in Escherichia coli K-12 revealed a close linkage to the disulfide bond (DSB) system as well as important periplasmic proteases. Thus, formation of a DSB between the two conserved cysteins is promoted by the oxidizing DsbA/DsbB-system and reduces enzymatic activitiy of the EAL domain. In contrast, the free thiol form increases PDE activity and dimerizes via the TM2 domain. Moreover, reduction of the CSS domain results in degradation by the periplasmic HtrA proteases DegP and DegQ. These redundantly process PdeC to a shorter fragment containing the TM2 and EAL domain only, which shows dimerization as well and has high PDE activity. Degradation in the periplasm mediated by DegP and DegQ is very efficient. In contrast, further proteolysis in the cytoplasm by yet unidentified proteases is rather slow, allowing the accumulation of the highly active TM2+EAL form. Finally, the interplay of redox dependent activity control and proteolysis of the c-di-GMP specific PDE PdeC regulates production of curli and cellulose as major matrix components of the bacterial biofilm.

Tannine gehören zu den phenolhaltigen Verbindungen und galten in der Tierernährung bisher als sekundäre unerwünschte Pflanzeninhaltsstoffe. Sie werden in der Regel in zwei Hauptgruppen eingeteilt: kondensierte und hydrolisierbare Tannine. Im Mittelpunkt der vorliegenden Untersuchungen stand die Quantifizierung der Reduzierung der Proteolyse während der Silierung in Folge der Zulage von tanninhaltigen Pflanzenextrakten unterschiedlicher Herkunft zum Siliergut. Zu diesem Zweck wurden zunächst in Silierversuchen zwei tanninhaltige Pflanzenextrakte unterschiedlicher botanischer Herkunft (Mimosa und Quebracho) alleine sowie in Kombination mit Silierzusätzen dem Ausgangsmaterial am Beispiel Weidelgras (Lolium perenne L.) zugesetzt. Folgend wurden die Effekte unterschiedlicher Konzentrationen der beiden tanninhaltigen Pflanzenextrakte sowie die Effekte bei unterschiedlichen TM-Gehalten des Siliergutes auf den Umfang der Proteolyse am Beispiel der Luzerne (Medicago sativa L.) geprüft. In den Versuchen konnte gezeigt werden, dass die Tannine einen deutlichen Einfluss auf die Proteolyse während der Silierung insbesondere in den ersten Tagen hatten. Die Beurteilung der Effekte der Zulage von tanninhaltigen Pflanzenextrakten basierte unter anderem auf Veränderungen der Parameter der Rohproteinfraktionierung nach dem Cornell Net Carbohydrate and Protein System (CNCPS). Durch den Zusatz von tanninhaltigen Pflanzenextrakten konnte der Abbau von schwer- und mittellöslichen Rohproteinfraktionen (Fraktionen B1-B3) zur leichtlöslichen Rohproteinfraktion (Fraktion A) signifikant minimiert werden, parallel stieg der Gehalt der pansenstabilen Rohproteinfraktionen. Die Effekte unterschiedlicher TM-Gehalte in den Silierversuchen mit Luzerne (Medicago sativa L.) zeigten, dass die eingesetzten tanninhaltigen Pflanzenextrakte unabhängig von ihrer botanischen Zusammensetzung in ihrer Wirkung umso höher waren je geringer der TM-Gehalt war. Tannins belong to phenol compounds and have traditionally been classified as antinutritive substances in animal nutrition. Tannins are usually divided into two groups: hydrolyzed and condensed tannins. The present study focused on reducing proteolysis during ensilage by supplementing tannin extracts from different botanical sources. Ensilage studies were carried out with Lolium perenne dominated forage and two different tanniniferous extracts (Mimosa and Quebracho) alone as well as in combination with silage additives. In a second study, the effects of different tannin concentrations and two different dry matter levels on proteolysis during ensilage were tested with alfalfa (Medicago sativa L.). The results show that tannin extracts definitely reduce proteolysis during the first days of ensilage. The protein fractionation of the Cornell Net Carbohydrate and Protein System (CNCPS) was used as indicator. Degradations from slightly soluble (B1 – B3) to easily soluble (A) fractions were significantly reduced by the addition of tannin extracts. At the same time, the level of rumen undegradable true protein was increased. The alfalfa study used different dry matter levels showing that by supplementing tannin extracts the wetter the silage was, the clearer the effect on reducing proteolysis.
