Lectins are omnipresent proteins in living beings, they bind specifically to carbohydrates, this interaction is important for several biological processes and makes lectins valuable tools for applications in biomedical sciences. The red marine alga lectin Solieria filiformis (SfL) presents biological effects such as antidepressant, antinociceptive, antitumor and anti-inflammatory. Since this lectin has this ability to modulate the inflammatory response, it is promising for the control of disorders of immunoinflammatory origin, such as periodontitis. Periodontitis is a disease characterized by damage to the supporting periodontitis due to an immunoinflammatory disorder triggered by the accumulation of dental biofilm. In view of the above, this study aimed at investigating the efficacy of the SfL in an experimental model of periodontitis in rats. For this, male Wistar rats (n=6) were ligature-induced by nylon and under daily use of LSf (1, 3 or 9 mg/kg) intraperitoneally (i.p.) for 11 days. As controls, groups of animals without periodontitis and without treatment (naive) were used, and rats with periodontitis and under the use of sterile saline (i.p.). For the evaluation of this effect, morphometric study and histopathological analysis of the maxilla, interleukin (IL)-1β and 6 and tumor necrosis factor-α (TNF-α) were performed. Finally, systemic parameters were evaluated using serum biochemical dosages, hemogram, body mass variation, weight changes and organ histopathology. The use of LSf during 11 days in animals submitted to periodontitis proved to be efficient in reducing the evolution of periodontitis, as shown in the study of macroscopic analysis of the alveolar bone, reduced 60, 57 and 51%, with the respective doses 1, 3 and 9 mg/kg and in the analysis of the histology of the periodontitis there was a significant improvement in the parameters evaluated, which were inflammatory infiltrate, cemento and bone. There was modulation of pro-inflammatory cytokines, IL-1β, IL- 6 and TNF-α, suggesting that the reduction in the evolution of periodontitis under the use of LSf is involved in the ability to modulate the inflammation of this lectin, moreover, LSf proved to be non-toxic, because it does not alter the systemic parameters evaluated. Thus, this lectin proved promising as an adjuvant in periodontitis. This is the first report in the literature of an algal lectin with the effect of reducing the evolution of periodontitis. Lectinas são proteínas onipresente nos seres vivos, se ligam especificamente a carboidratos, esta interação é importante para diversos processos biológicos e faz das lectinas valiosas ferramentas para aplicações nas ciências biomédicas. A lectina da alga marinha vermelha Solieria filiformis (LSf) apresenta efeitos biológicos, tais como antidepressivo, antinociceptivo, antitumoral e anti-inflamatório. Uma vez que a referida lectina possui essa capacidade de modular a resposta inflamatória, a mesma mostra-se promissora para o controle de desordens de origem imunoinflamatória, tal como a periodontite. A periodontite é uma doença caracterizada por danos no periodonto de sustentação, devido a desordem imunoinflamatório desencadeado pelo acúmulo de biofilme dental. Diante do exposto, este trabalho teve como objetivo investigar a eficácia da LSf em modelo experimental de periodontite em ratos. Para isso, ratos Wistar machos (n=6) foram submetidos à indução da periodontite por ligadura de fio de náilon e sob o uso diário de LSf (1, 3 ou 9 mg/kg) por via intraperitoneal (i.p.), durante 11 dias. Como controles, foram utilizados grupos de animais sem periodontite e sem tratamento (naïve), e ratos com periodontite e sob o uso de salina estéril (i.p.). Para a avaliação do referido efeito, foram realizados, estudo morfométrico e análise histopatológica das maxilas, e dosagem das interleucinas (IL)-1β e 6 e do fator de necrose tumoral-α (TNF-α). Por fim, foram avaliados parâmetros sistêmicos, através de dosagens bioquímicas séricas, hemograma, variação de massa corpórea, alterações no peso e histopatologia de órgãos. O uso de LSf durante 11 dias em animais submetidos a periodontite, mostrou-se eficiente na redução da evolução da periodontite, como mostrado no estudo da análise macroscópica do osso alveolar, reduziu 60, 57 e 51%, com as respectivas doses 1, 3 e 9 mg/kg e nas análises da histologia do periodonto houve uma melhora significativa dos parâmetros avaliados, que foram infiltrado inflamatório, cemento e osso. Houve modulação de citocinas pró-inflamatórias, IL-1β, IL- 6 e TNF-α, sugerindo que a redução da evolução da periodontite sob o uso da LSf, esteja envolvido na capacidade de modular a inflamação desta lectina, além disso, LSf mostrou-se atóxica, por não alterar os parâmetros sistêmicos avaliados. Assim, essa lectina mostrou-se promissora como adjuvante na periodontite. Esse é o primeiro relato na literatura de uma lectina algal com efeito de redução da evolução da periodontite.

Recent researches have shown that substances produced by seaweeds have an amazing biotechnological potential with several industrial uses. However, the natural pigments stand out, but they have not been intensively explored yet, especially when obtained from macroalgae. Because of the need to use natural substances, this work intends to extract and purify the pigments of the Hypnea musciformis red sea macroalgae, as known as phycobiliproteins, and characterize the red pigment R-phycoerythrin (R-FE). The pigments were extracted from the fresh macroalgae in potassium phosphate buffer 25 mM (pH 6.5), followed by fractionation by ammonium sulfate 0-30% and 30-45% saturated for posterior application in chromatographic columns in DEAE-Sephacel and Sephacryl S-300 matrix. The spectrophotometric analysis of the obtained fractions showed pigments related to the said macroalgae species, R-phycocyanin (λ = 550 and 610 nm), and R-FE, with absorption peaks at 495, 540 and 564 nm and fluorescence emission at 575 nm. It was obtained pure R-FE with 15.75% recovery, 8.56 μg/g productivity (seaweed dry weight) and 4.17 purity index. By electrophoresis (PAGE-SDS) in reducing conditions, the R-FE showed three protein bands with 18, 20, and 37 kDa apparent molecular mass, relative to the α, β and γ subunits. The R-FE showed 95.35% of its secondary structure in α-helix, maintaining this structure stability until 60 °C. Starting from 70 °C, it was observed changes in the absorption specter, occurring reductions in absorption (28.26%) and fluorescence emission (49.01%). The R-FE absorption specter, when stored for 20 days, remained stable when in refrigerated conditions. However, when stored at 25 °C, the maximum absorption peaks at 495, 540 and 564 nm showed an 89.7, 89.7 and 92.8% decrease, respectively. Concerning its fluorescence emission, it was observed the loss of 38.8, 73.7 and 100% under the temperature of -20, 4 and 25 °C respectively within the first five days. It was possible to extract phycobiliproteins and purify specifically the R-FE fluorescent pigment with a high purity rate from Hypnea musciformis, showing thermostable α-helix structure predominance, highly stable spectroscopic properties and especially in proper storage conditions. Pesquisas recentes têm mostrado que compostos produzidos por algas marinhas têm um incrível potencial biotecnológico com inúmeras aplicações industriais. Entretanto, os pigmentos naturais se destacam, porém os mesmos ainda não foram explorados de forma intensiva, principalmente quando obtidos a partir de macroalgas. Diante da necessidade do uso de compostos de origem natural, o presente trabalho teve por objetivo extrair e purificar os pigmentos da macroalga marinha vermelha Hypnea musciformis, também conhecidos como ficobiliproteínas, e caracterizar o pigmento vermelho R-Ficoeritrina (R-FE). Os pigmentos foram extraídos a partir da macroalga fresca em tampão fosfato de potássio 25 mM (pH 6,5), seguidos por fracionamento por sulfato de amônio a 0-30% e 30-45% de saturação para posterior aplicação em colunas cromatográficas em matriz de DEAE-Sephacel e Sephacryl S-300. As análises espectrofotométricas das frações obtidas mostraram pigmentos relativos à referida espécie de macroalga, R-Ficocianina (λ = 550 e 610 nm), e R-FE, com picos de absorção a 495, 540 e 564 nm e emissão de fluorescência a 575 nm. Obteve-se a R-FE com recuperação de 15,75%, rendimento de 8,56 µg/g (peso seco da alga) e índice de pureza de 4,17. Por eletroforese (PAGE-SDS) em condições redutoras, a R-FE apresentou três bandas proteicas com massas moleculares aparentes de 18, 20 e 37 kDa, referente as subunidades α, β e γ. A R-FE apresentou 95,35% da estrutura secundária em α-hélix, mantendo a estabilidade dessa estrutura até 60 ºC e, a partir de 70 °C, foi observado alterações no espectro de absorção, ocorrendo reduções na absorção de 28,26% e emissão de fluorescência de 49,01%. O espectro de absorção da R-FE, quando armazenada por 20 dias, se manteve estável sob condições refrigeradas (-20 º e 4 ºC), entretanto quando armazenada a 25 °C, os picos máximos de absorção a 495, 540 e 564 nm apresentaram um decréscimo de 89,7, 89,7 e 92,8% respectivamente. Quanto a sua emissão de fluorescência, foi observado a perda de 38,8, 73,7 e 100% sob as temperaturas de – 20, 4 e 25ºC, respectivamente, nos primeiros cinco dias. Foi possível extrair ficobiliproteínas e purificar especificamente o pigmento fluorescente R-FE com alto grau de pureza a partir da macroalga Hypnea musciformis, apresentando predominância de estrutura em α-hélix termoestável, propriedades espectroscópicas de elevada estabilidade e sobretudo em condições adequadas de armazenamento.

Synthetic genes from the red marine algae Hypnea musciformis (HML) and Bryothamnion triquetrum (rBTL) were cloned into differents vectors and transformed into several bacterial expression strains. The recombinant lectins were obtained from the soluble fraction of bacterial cultures using Escherichia coli Rosetta-gami 2 (DE3) strain for rHML and E. coli BL21 (DE3) strain for rBTL. Haemagglutination tests showed that rHML and rBTL are able to agglutinate rabbit erythrocytes with strong haemagglutination activity only after treatment with papain and trysine indicating that their ligands are not directly accessible at the cell surface. The haemagglutinating properties of rHML and rBTL confirm the correct folding and functional state of the proteins. A study of the specificity of these lectins by glycan array was conducted. HML, BTL and rBTL showed a restricted specificity for complex N-glycans with core (α1-6) fucose. A more detailed analysis of the specificity of these lectins showed a preference for non bisecting N-glycans, bi- and tri-antennary branching sugars with short chains. Addition of Sialic acid at the non-reducing end of N-glycans favors their recognition by the lectins. This is the first characterization of lectins from red algae by glycan array. An interaction between BTL and a core (α1-6) fucosylated octasaccharides was also observed by STD-NMR. The toxic activity of wild and recombinant lectins were evaluated against Artemia sp. and the human lung adenocarcinoma cell line (A549). In cytotoxicity assays, HML, rHML, BTL and rBTL showed no toxicity against Artemia sp. Only HML and rHML showed a low cytotoxic activity against cell line (A549). The first crystal of rBTL was obtained in micro-scale level using a robot and diffracted at 15 Å. Os genes sintéticos das lectinas das algas marinhas vermelhas Hypnea musciformis (HML) e Bryothamnion triquetrum (BTL) foram clonados em diferentes vetores e transformados em diferentes células bacterianas de expressão. As lectinas recombinantes foram obtidas a partir da fração solúvel das culturas bacterianas de Escherichia coli Rosetta-gami 2 (DE3) para rHML e BL21 (DE3) para rBTL. Os testes de hemaglutinação mostraram que rHML e rBTL são capazes de aglutinar eritrócitos de coelho tratados com diferentes enzimas proteolíticas. As propriedades hemaglutinantes de rHML e de rBTL confirmam o enovelamento correto e o estado funcional das proteínas. A caracterização da especificidade de ligação a carboidratos da HML, BTL e da rBTL por glycan array mostrou uma especificidade restrita por oligossacarídeos complexos contendo o núcleo de fucosilação (α1-6), com uma preferência particular por N-glicanos não bisectados, bi e tri-antenados de cadeia curta. A presença de ácido siálico na extreminada não-redutora dos glicanos favorece o reconhecimento. Essa foi a primeira caracterização de lectinas de algas vermelhas por glycan array. Experimentos de STD-RMN com a BTL mostraram uma interação com um octassacarídeo contendo o núcleo de fucosilação (α1-6). A atividade tóxica das lectinas selvagens e recombinantes foi avaliada contra Artemia sp. e contra células de adenocarcinoma de pulmão (A549). Nos ensaios de citotoxicidade, HML, rHML, BTL e rBTL não mostraram nenhuma toxicidade contra Artemia sp. e somente HML e rHML mostraram uma baixa toxicidade contra células de adenocarcinoma de pulmão (A549). O primeiro cristal de rBTL foi obtido a um nível de microescala com a ajuda de um robôt de cristalização e difratou a 15 Å de resolução.

La introducción de especies exóticas marinas y su posible actividad invasiva se considera hoy en día la 3ª gran amenaza global para la biodiversidad. Para evitar estos daños es necesario mantener controles que permitan una certera y rápida identificación de las especies que están invadiendo o siendo introducidas. Los métodos morfológicos muchas veces no son capaces de discernir especies cripticas por lo que hoy en día se usan métodos de identificación genética, con especial relevancia el uso de barcoding. El caso de las algas rojas foliosas es muy característico en Europa, ya que debido a que su simple morfología han ocurrido introducciones cripticas, como el caso de G. turuturu que se confundía con G. doryphora. La presencia de esta en Galicia nos pone en antecedentes, de que esto sucede en costas cantábricas. Por lo que nosotros proponemos un estudio para la identificación de especies cripticas introducidas en Asturias, mediante el uso de técnicas de barcoding usando a COI como marcador principal.

Objectives The aim of this study was to evaluate the anti-inflammatory effect of a sulphated polysaccharide fraction (PLS) extracted from the alga Hypnea musciformis and investigate the possible involvement of the nitric oxide (NO) pathway in this effect. Methods The anti-inflammatory activity of PLS was evaluated using inflammatory agents (carrageenan and dextran) to induce paw oedema and peritonitis in Swiss mice. Samples of paw tissue and peritoneal fluid were removed to determine myeloperoxidase (MPO) activity, NO3/NO2 levels, and interleukin-1b (IL-1b) level. The involvement of NO in the modulation of neutrophil migration in carrageenan-induced paw oedema or peritonitis was also investigated. Key findings Compared with vehicle-treated mice, mice pretreated with PLS (10 mg/kg) inhibited carrageenan-induced and dextran-induced oedema; it also inhibited total and differential peritoneal leucocyte counts in a model of peritonitis. These PLS effects were reversed by l-arginine treatment and recovered with the administration of a NO synthase blocker (aminoguanidine). Furthermore, PLS reduced the MPO activity, decreased IL-1b levels, and increased NO3/NO2 levels in the peritoneal cavity. Conclusions PLS reduced the inflammatory response by modulating neutrophil migration, which appeared to be dependent on the NO pathway.

Entre septiembre 2006 y junio 2007 se estudió la composición y abundancia de poliquetos asociados a algas rojas intermareales, en 7 estaciones de la franja costera cordobesa del Caribe colombiano. Para la recolección de los especímenes, se delimitó un cuadrante de 625 cm², con 5 réplicas dispuestas al azar en cada punto de muestreo. Todos los frondes de algas rojas examinados evidenciaron la presencia de poliquetos, con un registro de 288 individuos pertenecientes a 19 especies de 7 familias. La especie más abundante fue Platynereis dumerilii (Nereididae) con 188 individuos, seguida de Pseudonereis gallapagensis (Nereididae) con 23 individuos y Syllis corallicola (Syllidae) con 12 individuos. También se colectaron los poliquetos sedentarios, Hydroides sanctaecrusis (Serpulidae) con 4 individuos y Pista palmata (Terebellidae) con 5 individuos. La abundancia más alta de poliquetos se registró en diciembre (T2) y marzo (T3) con 79 y 103 individuos respectivamente, y la menor ocurrió en septiembre (T1) y junio (T4) con 47 y 59 individuos. Con relación a la prueba ANOSIM, no se registró una agrupación significativamente diferente de la otra (R = 0,09, P = 0,52), debido a que P. dumerilii mostró el más alto porcentaje de contribución (> 77%) en cada agrupación, lo que indica que fue la especie más importante, en términos de abundancia, durante los meses de muestreo. Finalmente, las especies colectadas en los frondes algales son especies características de sustratos de fondos blandos y rocosos, sugiriendo una adaptación no específica de estas especies a los sustratos algales.

Seaweeds are classified in the kingdom protist, predominantly aquatic and with great adaptive capacity. Found in all regions of the planet, algae play an important role ecological and economic, representing a great potential for bioresources. The red seaweed Gracilaria birdiae, collected during twelves months, was submitted to aqueous extraction of polysaccharides at 100ºC, showed yield exceeding 50 %, except in February, September and November 2007 (49,5%, 41,8% and 49,4% respectively). The sulfate content was obtained by microanalysis and it not vary significantly over the months examined averaging 1,35%. The sulfate degree was 0.15. The protein content in the samples also showed no considerable variation, with average of 0,5%, howevwr in April, the protein content increased about 3 times the average found. The twelve samples were chemically characterized by Gel Permeation Chromatography (GPC), Infrared (FT-IR) and Nuclear Magnetic Resonance (NMR) spectroscopy. The GPC chromatograms of the samples showed similarity and typical characteristics of polysaccharides from red algae. In general, we observed a main peak, at approximately 9,2 mL volume of eluition, with the exception of August, October and November 2007, which showed chromatograms with main peak at approximately 8,7 mL volume of eluition, detected by a refractive index. The polysaccharides investigated showed hight molecular weight, ranging from 0.11 x 104 and 8.51 x 105 g/mol. The infrared spectra of the twelve samples of polysaccharides presented similar profiles, with bands characteristics of agarocolloids, more specifically agaran-like, with evidence of protein. NMR were employed to the structural characterization of the samples throughout the year and revealed that the structure of polysaccharides is composed by (1→3)-β-D-galactopyranosyl linkage to (1→4)-α-L-anhydrogalactopyranosyl segments. The study of seasonal variation in proximate composition, in general, showed no marked variation. Regarding the amount of soluble protein in the crude extract, the study of seasonal variation showed a small variation in the content of soluble protein, with maximum value (0,94 mgP/mL) in November/06 and minimum (0,39 mgP/mL) in December/06. The hemagglutinating activity during the period of one year, except in December/06. Algas marinhas são seres classificados no reino protista, predominantemente aquáticos e com grande capacidade adaptativa. Encontradas em todas as regiões do planeta, as algas desempenham importante papel ecológico e econômico, representando um grande potencial de biorrecursos. A alga marinha vermelha Gracilaria birdiae, coletada durante doze meses, foi submetida à extração aquosa a 100 ºC de polissacarídeos e apresentou rendimento superior a 50 %, com exceção nos meses de fevereiro, setembro e novembro de 2007 ( 49,5%, 41,8% e 49,4% respectativamente). O teor de sulfato foi obtido por microanálise e não variou significativamente ao longo dos meses analisados, apresentando valor médio de 1,35%. O grau de sulfatação médio encontrado foi de 0,15. O conteúdo protéico nas amostras também não apresentou variação considerável, tendo média de 0,5%, entretanto, no mês de abril o conteúdo protéico aumentou cerca de 3 vezes do valor médio encontrado. As doze amostras foram caracterizadas quimicamente por cromatografia de permeação em gel (GPC), espectroscopia na região do infravermelho (FT-IR) e por ressonância magnética nuclear (RMN). Os cromatogramas de GPC das amostras apresentaram semelhanças e características típicas de polissacarídeos das algas vermelhas. Em geral, foram observados um pico principal, em aproximadamente 7,58 mL e um ombro em aproximadamente 9,2 mL de volume de eluição, com exceção dos meses de agosto, outubro e novembro de 2007, que apresentaram cromatogramas com pico principal em aproximadamente 8,27 mL de volume de eluição, detectados pelo índice de refração. Os polissacarídeos investigados apresentaram altas massas molares , variando de 0,11 x 104 a 8,51 x 105 g/mol. Os espectros de infravermelho das doze amostras de polissacarídeos apresentaram perfis semelhantes, com as bandas características de agarocolóides, mais especificamente, do tipo agarana. Espectros de RMN foram empregados para a caracterização estrutural das amostras ao longo do ano e, revelaram que a estrutura do polissacarídeo é composta por segmentos de (1→ 3)-β-D-galactopiranosil ligada a (1→4)-α-L-anidrogalactopiranosil. O estudo da variação sazonal da composição centesimal, em geral, não apresentou variação acentuada. Com relação à quantidade de proteínas solúveis e a atividade hemaglutinante no extrato protéico, o estudo da variação sazonal revelou uma pequena variação no teor de proteínas solúveis, com valor máximo (0,94 mgP/mL) em novembro/06 e mínimo no mês de dezembro/06 (0,39 mgP/mL). Em geral, a atividade hemaglutinante observada em Gracilaria birdiae é fraca, porém, ela esteve presente em concentrações variáveis ao longo de todo o período estudado, com exceção no mês de dezembro/06, independente do conteúdo protéico total e solúvel encontrado.

Seaweed consists in a vital resource to the economy of many countries and it has been explored all over the world thanks to its capacity of producing colloids such as agar, carrageen and sodium alginate as well as the useful aspect either in food industry, fertilizers or medicine area. Studies on induction, cultivation and callus reorganization have been shown an important issue on tissue culture technique and its usage. In different tissue experience in vitro the usage of regulating growth is so fundamental to establish the competence and necessary conditions to callus formation and regeneration. In this present study has been tested the auxin and citocinin effects in different dosages of agar in ASP 12-NTA. The indolacetic acid effect and 6- benzilalaminopurine have been tested as well in seaweed in the species Gracilaria birdiae with two separated doses, 0,5mg/L and 0,8mg/L and together in concentration of 1:5mg/L and 5;1mg/L. Regarding to agar concentration, it has been tested two concentrations with 0,5% and 0,8% in all treatments. As to sterilization process has been done with fungicide (nistatina) and antibiotic (ciprofloxacin) besides iodopovidona and the sodium hypochlorite. The complete experiment had been lead in a germination chamber with 25±2ºC and photoperiod of more or less 16 hs of light and 08hs dark and 30,2‰ saltiness, pH around 7,6 within 60 days approximately. In more or less 50 days of cultivation had been noticed callus formation and regeneration process therein. The results had been underwent by statistic analyze treatments of contingent through (x2) where many meaningful different observations had been checked in callus and regeneration and the agar levels studied, that is to say the agar level treatments in 0,8% presented a major number of regeneration Macroalgas marinhas constituem um recurso vital para a economia de diversos países e tem sido exploradas ao redor do globo devido a sua capacidade de produção de fico colóides como o ágar, carragenina e alginatos, bem como pelo seu espectro de utilização, seja na indústria de alimentos, de fertilizantes e na medicina. Estudos sobre a indução, cultivo e reorganização de calos, tem representado um importante papel nas técnicas de cultura de tecidos e suas aplicações. Em diferentes tecidos cultivados in vitro, a utilização de reguladores de crescimento é de importância primordial para o estabelecimento da competência e determinação, condições necessárias para a formação de calos e regenerações. No presente estudo foram testados os efeitos de uma auxina e de uma citocinina, em diferentes concentrações de ágar em meio ASP 12-NTA. O efeito do ácido indolacético (AIA) e da 6-benzilaminopurina (BAP) foram testados em explantes de macroalgas de espécie Gracilaria birdiae, em separado, com duas concentrações de 0,5 mg/L e 0,8 mg/L e em conjunto, nas concentrações de 1:5 mg/L e 5:1 mg/L. Com relação a concentração de ágar, foram testadas duas concentrações, com 0,5% e 0,8% para todos os tratamentos. Para o processo de esterilização foram aplicados um fungicida (nistatina) e um antibiótico (ciprofloxacina), além de iodopovidona e hipoclorito de sódio. Todo o experimento foi conduzido em câmara de germinação, com temperatura de 25 ± 2oC e fotoperíodo de 16hs de luz e 08hs escuro, salinidade de 30 ± 2‰ e pH em torno de 7,6, durante aproximadamente 60 dias. Ao final de 50 dias de cultivo, foi observada a formação de calos e de processos de regeneração indireta dos mesmos. Os resultados foram submetidos a tratamentos estatísticos de análise de contigência através do Quiquadrado (χ2 ), onde foram observadas diferenças significativas entre a incidência de calos e regenerações e os níveis de ágar estudados. Ou seja, nos tratamentos onde foram utilizados níveis de ágar de 0,8%, apresentaram o maior número de regenerações

Cancer is a desease that occurs in a larger number of people each year. The therapies used to treat it are still not satisfatory. Natural products have been studied to select new compounds capable of reducing tumours and with fewer side effects. The sulfated polysaccharide isolated from the seaweed C. feldmannii (Cf-PLS) was investigated for its antitumor and immunostimulating properties and also for the toxicological aspects related to Cf-PLS treatment. The Cf-PLS did not show any significant in vitro cytotoxic effect, but showed a strong in vivo antitumor effect. The inhibition rates of sarcoma 180 tumor development were 48.16 % and 48.62% at the doses of 10 and 25 mg/kg, respectively, and 68.32% when treated together with 5-fluorouracil (5-FU). The histopathological analysis of liver and kidney showed that the kidneys were affected by Cf-PLS-treatment, presenting some focal areas of acute tubular necrosis at the higher dose. Cf-PLS caused considerable changes in renal physiology, as shown by an increase in parameters such as perfusion pressure, renal vascular resistance, glomerular filtration rate, urinary flow and sodium, chloride and potassium excretion. Neither enzymatic activity of alanine aminotransferase, urea nor creatinin levels were significantly altered. In hematological analyses, leucopeny was observed after 5-FU treatment, but this effect was prevented when the treatment was associated with the Cf-PLS. It was also demonstrated that Cf-PLS acts as an immunomodulatory agent, raising the production of specific antibodies, and also increasing the production of OVA-specific antibodies. In addition, it induced a discreet increase of the white pulp and nest of megakaryocytic in spleen of treated mice and the neutrophil migration to the intraperitoneal cavity of mice. In conclusion, Cf-PLS has some interesting antitumour activity that could be associated with its immunostimulanting properties. O câncer é uma doença que afeta cada vez mais um número maior de pessoas em todo o mundo. As terapias atuais utilizadas para o tratamento do câncer são ainda insatisfatórias. Produtos naturais têm sido avaliados para seleção de compostos ativos, capazes de reduzir os tumores malignos de uma forma mais eficiente e com menos efeitos indesejáveis. O polissacarídeo sulfatado extraído da alga Champia feldmannii (Cf-PLS) foi testado para avaliação do seu potencial antitumoral e imunoestimulante. Além disso, foram feitos testes de toxicidade do fígado, rins e baço após o tratamento com Cf-PLS. Os resultados demonstraram que Cf-PLS não apresenta citotoxicidade in vitro, mas é capaz de reduzir o tumor Sarcoma 180 implantado em camundongos em 48,16% e 48,62% nas doses de 10 e 25mg/kg, respectivamente, e reduz 68,32% quando adiministrado juntamente com 5-Fluorouracil (5-FU). As análises do fígado e rins revelaram que houve certa toxicidade no rim, com áreas de necrose tubular aguda na maior dose. Os testes da perfusão renal revelaram que Cf-PLS causa aumento da pressão de perfusão, resistência vascular renal, ritmo de filtração glomerular e fluxo urinário, além da excreção de sódio, cloreto e potássio. Não houve alterações nos percentuais de transporte totais ou tubular proximais de sódio, cloreto ou potássio. Houve discreta deposição protéica nos glomérulos e túbulos renais. Não houve alterações nas análises bioquímicas e os testes hematológicos revelaram uma leucopenia causada por 5-FU, entretanto esta foi revertida pelo tratamento com Cf-PLS. Também foi demonstrado que Cf-PLS age como agente imunoestimulante e imunomodulador, aumentando a produção de anticorpos totais e específicos contra Cf-PLS e OVA, aumentando também a polpa branca e o número de megacariócitos nos baços dos animais tratados e induzindo a migração de neutrófilos para a cavidade peritoneal de camundongos. Pode-se concluir que Cf-PLS apresenta atividade antitumoral e que isso pode estar relacionado com suas propriedades imunoestimulantes.

To present dissertation it analyzes the effect of the brightness in the growth of the alga sea Gracilaria birdiae through the rate of specific growth (TCE) and biomass increase in different depths. A structure of the type long-line was used with main rope measuring 25m and 10 sec ondary strings of 2,5m installation to 200m of the coast with depth between 5m and 6m (tide evaporates). 12 seedlings were fastened by secondary rope with spacing of 20cm composing 12 depth lines. With 30 days it accomplished the measure of the weights of the seedlings of the structure identified by secondary rope and depth line. With 60 and 90 days the measure of the weights was just in the depth lines that presented seedlings in better physical conditions. In the tested depths from 0,2 to 2,4m, it was observed a better acting of the growth as well as better appearance and physical condition of the fastened seedlings from 0,2m to 1,2m, while fastened them from 1,4m to 1,8m or they had inferior growth or practically they didn't grow , and fastened them in the inferior limit from 2,0 to 2,4m reduced growth of the seedlings it was verified. The obtained medium weights presented significant statistical difference (F = 187,104; p = 0,05). TCE demonstrated negative correlation with the dept h (r = - 0,95; p = 0,05; n = 120). In the closest depths of the surface from 0,2 to 0,6m favored the growth of the algae reaching weights from 167,5 to 147,0g and TCE 2,92 and 2,69% day-1, among 0,8m to 1,2m weights from 139,0 to 124,0g and TCE fr om 2,29 to 1,90% day-1, and starting from 1,4m until 2,4m weights from 94,5 to 59,5g and TCE 0,9 to 0,53% day-1. The reduction in the weights below 1,2 m was quite accentuated evidencing that the algae had detriment in your growth with the reduction of the intensity brightness presenting reduced growth of some you change fastened in the extremities of the strings. The parameters of the water in the area of the cultivation temperature, salinity and degree of transparency of the water presented averages of 29,6 ± 2,41ºC, 35,6 ± 1,49‰ and 1,20 ± 0,76m. In the end of the experimental period (90 days) the seedlings reached final weight of 937,0g, ideal for crop. For Gracilaria birdiae, the brightness was the environmental factor that it affected your TCE and biomass production indeed, presenting in the conditions of the cultivation, satisfactory growth until the depth of 1,2m where the seedlings reached weight of 124,0 ± 9,94 g and TCE of 1,90 %dia-1. A presente dissertação analisa o efeito da luminosidade no crescimento da alga marinha Gracilaria birdiae através da taxa de crescimento específico (TCE) e aumento de biomassa em diferentes profundidades. Foi utilizada estrutura do tipo long-line com corda principal medindo 25m e 10 cordas secundárias de 2,5m instalação a 200m da costa com profundidade entre 5 e 6 m (maré seca). Foram fixadas 12 mudas por corda secundária com espaçamento de 20cm compondo 12 linhas de profundidade. Com 30 dias realizou-se a pesagem de todas as mudas da estrutura identificadas por corda secundária e linha de profundidade. Com 60 e 90 dias a pesagem foi apenas nas linhas de profundidade que apresentavam mudas em melhores condições físicas. Nas profundidades testadas de 0,2 a 2,4m, observou-se um melhor desempenho do crescimento como também melhor aparência e condição física das mudas fixadas de 0,2 a 1,2m, enquanto as fixadas entre 1,4 a 1,8m ou tiveram crescimento inferior ou praticamente não cresceram, e as fixadas no limite inferior entre 2,0 e 2,4m verificou-se atrofiamento das mudas. Os pesos médios obtidos apresentaram diferença estatística significativa (F = 187,104; p = 0,05). A TCE demonstrou correlação negativa com a profundidade (r = - 0,95; p = 0,05; n = 120). Nas profundidades mais próximas da superfície de 0,2 a 0,6m favoreceram o crescimento das algas alcançando pesos de 167,5 a 147,0g e TCE de 2,92 e 2,69% dia-1, entre 0,8 a 1,2m pesos de 139,0 a 124,0g e TCE de 2,29 a 1,90% dia-1, e a partir de 1,4m até 2,4m pesos de 94,5 a 59,5g e TCE de 0,9 a 0,53% dia-1. A redução nos pesos abaixo de 1,2m foi bastante acentuada evidenciando que as algas tiveram detrimento em seu crescimento com a redução da intensidade luminosidade apresentando atrofiamento de algumas mudas fixadas nas extremidades das cordas. Os parâmetros da água na área do cultivo temperatura, salinidade e grau de transparência da água apresentaram médias de 29,6 ± 2,41 ºC, 35,6 ± 1,49 ‰ e 1,20 ± 0,76 m. No final do período experimental (90 dias) as mudas atingiram peso final de 937,0g, ideal para colheita. Para Gracilaria birdiae, a luminosidade foi o fator ambiental que afetou efetivamente sua TCE e produção de biomassa, apresentando nas condições do cultivo, crescimento satisfatório até a profundidade de 1,2m onde as mudas atingiram peso médio de 124,0 ± 9,94 g e TCE de 1,90 %dia-1.

Many substances have been shown to possess anticoagulant and immunostimulant activities, including sulfated polysaccharides (SP) of marine algae. They also exhibit antithrombotic and antiviral activities. It is important to describe new substances capable of preventing known adverse effects of heparin such as hemorrhages and thrombocytopenia. Moreover, the use of immunostimulants can also lessen cultivated aquatic organism stress. The aim of this work was to evaluate both the anticoagulant activity of sulfated galactans present in two species of red marine seaweed belonging to genus Halymenia and their immunostimulant effects on white Pacific shrimp Litopenaeus vannamei. SP were extracted from marine seaweed collected in Praia de Fleixeiras-CE. Following the extraction, crude extracts were fractioned in DEAEcellulose column and fractions were eluted with different concentrations of NaCl solution. Fraction purification grade was evaluated by electrophoresis. Anticoagulant activity was analyzed by aPTT’s test using rabbit plasma and the clotting time was measured in a coagulometer. Three different amount of SP (0.5 and 1.5 μg.L-1 and 1.0 mg.L-1) were used twice a day (9 and 17h) to evaluate immunostimulant activity on L. vannamei through immersion baths, after changing tank water and before feeding the animals. The control group was maintained without SP addition. The experiment lasted 20 days. Shrimps were treated by immersion in SP for the first 15 days, and in the last 5 days they were submitted to stress when no water was renewed. There was no significant difference on mortality among the treatments. After the stress, the mortality of shrimps treated with 1.0 mg.L-1 was lower than the others (control group, 0.5 and 1.5 μg.L-1). Anticoagulant activity was detected in fractions obtained from H. pseudofloresia, the highest being in that eluted with 1.25 M NaCl from the first extraction procedure. Thus, it was possible to obtain SP from red marine seaweed H. pseudofloresia which showed higher anticoagulant activity than heparin and was capable of lowering mortality of L. vannamei under stress conditions. Muitos compostos têm sido descritos como anticoagulantese imunoestimulantes, dentre essas os polissacarídeos sulfatados (PS) de algas marinhas que também são conhecidos por apresentar outras atividades biológicas, tais como antitrombótica e antiviral. A descoberta de novas drogas anticoagulantes é importante para prevenir os efeitos adversos do uso da heparina, como hemorragias e trombocitopenia. Além disso, o uso de imunoestimulantes também pode reduzir o impacto do estresse no cultivo intensivo de organismos aquáticos. O objetivo deste trabalho foi avaliar a atividade anticoagulante de galactanas sulfatadas de duas espécies de algas marinhas vermelhas do gênero Halymenia e o seu efeito imunoestimulante no camarão branco Litopenaeus vannamei. Os PS foram extraídos, através de digestão enzimática, a partir de algas marinhas coletadas na Praia de Fleixeiras/CE. Em seguida, os extratos brutos foram fracionados em coluna de DEAE-celulose, sendo as frações eluídas com soluções de diferentes concentrações de NaCl e o grau de purificação analisado por eletroforese. A atividade anticoagulante das frações foi avaliada pelo teste do aPTT, utilizando plasma de coelho citratado, sendo o tempo de coagulação registrado em um coagulômetro. Para avaliar efeito imunoestimulante, três doses diferentes de PS foram administradas, através de banhos de imersão, no camarão L. vannamei. Os tratamentos, com três repetições cada, foram 0,5 μg.L-1; 1,5 μg.L-1 e 1,0 mg.L-1, e um controle, sem polissacarídeos. O experimento teve duração de 20 dias, dos quais 15 dias foram para a aplicação das soluções de PS e 5 dias de indução de estresse, através da ausência de renovação de água do cultivo. A administração das doses foi realizada, duas vezes por dia (09:00 e 17:00 h), após as trocas de água e antes da alimentação doa animais. Depois de 15 dias de imersão, não houve diferenças significativas entre as taxas de mortalidade dos tratamentos. Entretanto, depois do período do estresse, o tratamento com 1,0 mg.L-1 apresentou uma mortalidade significativamente menor do que as observadas nos tratamentos s/PS e 0,5 μg.L-1. Com relação à atividadeanticoagulante, as frações obtidas da espécie H. pseudofloresia apresentaram uma maior atividade, sendo a mais ativa aquela eluída com 1,25 Mde NaCl da coluna de troca iônica, obtida da 1ª extração. Assim, com a realização deste trabalho, foi possível obter PS da alga marinha vermelha H. pseudofloresia com atividade anticoagulante superior à heparina e capaz de reduzir a mortalidade do camarão L. vannamei submetidos à condições de estresse.
