Parasitoids introduce various virulence factors when parasitism occurs, and some taxa generate teratocytes to manipulate the host immune system and metabolic homeostasis for the survival and development of their progeny. Host-parasitoid interactions are extremely diverse and complex, yet the evolutionary dynamics are still poorly understood. A category of serpin genes, named CvT-serpin s, was discovered to be specifically expressed and secreted by the teratocytes of Cotesia vestalis , an endoparasitoid of the diamondback moth Plutella xylostella. Genomic and phylogenetic analysis indicated that the C. vestalis serpin genes are duplicated and most of them are clustered into 1 monophyletic clade. Intense positive selection was detected at the residues around the P1–P1′ cleavage sites of the Cv-serpin reactive center loop domain. Functional analyses revealed that, in addition to the conserved function of melanization inhibition (CvT-serpins 1, 16, 18, and 21), CvT-serpins exhibited novel functions, i.e. bacteriostasis (CvT-serpins 3 and 5) and nutrient metabolism regulation (CvT-serpins 8 and 10). When the host-parasitoid system is challenged with foreign bacteria, CvT-serpins act as an immune regulator to reprogram the host immune system through sustained inhibition of host melanization while simultaneously functioning as immune effectors to compensate for this suppression. In addition, we provided evidence that CvT-serpin8 and 10 participate in the regulation of host trehalose and lipid levels by affecting genes involved in these metabolic pathways. These findings illustrate an exquisite tactic by which parasitoids win out in the parasite–host evolutionary arms race by manipulating host immune and nutrition homeostasis via adaptive gene evolution and neofunctionalization. Many endoparasitoids adopt several parasitic factors, such as venom, polydnavirus and teratocytes, to suppress the immune response of their associated hosts including melanization for successful parasitism. A teratocyte‐specific expressed serpin gene, designated as CvT‐serpin6, was identified from the parasitoid Cotesia vestalis. The immunoblot result suggested that CvT‐serpin6 was secreted into extracellular space. qPCR results showed that CvT‐serpin6 was mainly transcribed at later stages of parasitism, and the transcriptional abundance of CvT‐serpin6 in teratocytes was significantly increased in response to the challenge of bacteria. Inhibitory assay indicated that recombinant CvT‐serpin6 (rCvT‐serpin6) could inhibit the activation of Plutella xylostella prophenoloxidase and ultimately resulted in the inhibition of melanization in P. xylostella haemolymph. Furthermore, we confirmed that rCvT‐serpin6 could form SDS‐stable complexes with activated PxPAP1 and PxPAP3 in a dose‐dependent manner. BACKGROUND: Parasitic wasps are an important group of entomophagous insects for pest control. As parasitic wasps often lay eggs on or into their associated hosts, parasitoids evolve to utilize several factors including venom, polydnavirus (PDV) to alter host physiology for successful parasitism. Some taxa of endoparasitoids produce teratocytes, which are a type of cell that is released into host insects when wasp eggs hatch. Teratocytes display multifunction in parasitism such as host nutritional exploration, immune and developmental regulation, by secreting plenty of proteins into host hemocoel. RESULTS: A serpin (CvT‐serpin15) secreted by teratocytes was characterized. QPCR results showed the expressional level of CvT‐serpin15 was upregulated following bacterial challenges. Enzyme activity experiment indicated the recombinant CvT‐serpin15 protein could interfere with the growth of Gram‐positive bacteria Staphylococcus aureus. The survival rate assay demonstrated CvT‐serpin15 increased survival rate of Plutella xylostella infected by S. aureus. CONCLUSION: CvT‐serpin15 secreted by teratocytes would boost the host immune system when pathogens invade host hemocoel during parasitism, and ultimately protect the development of wasp larva from bacterial infection. © 2021 The Authors. The molecular bases of the host-parasitoid interactions in the biological system Acyrthosiphon pisum (Harris) (Homoptera, Aphididae) and Aphidius ervi (Haliday) (Hymenoptera, Braconidae) have been elucidated allowing the identification of a gamma-glutamyl transpeptidase, the active component of maternal venom secretion, and teratocytes, the embryonic parasitic factors responsible for host physiology regulation after parasitization. Teratocytes, cells deriving from the dissociation of the serosa, the parasitoid embryonic membrane, are responsible for extra-oral digestion of host tissues in order to provide a suitable nutritional environment for the development of parasitoid larvae. Teratocytes rapidly grow in size without undergoing any cell division, synthesize, and release in the host hemolymph two proteins: a fatty acid binding protein (Ae-FABP) and an enolase (Ae-ENO). Ae-FABP is involved in transport of fatty acids deriving from host tissues to the parasitoid larva. Ae-ENO is an extracellular glycolytic enzyme that functions as a plasminogen like receptor inducing its activation to plasmin. Both Ae-FABP and Ae-ENO lack their signal peptides, and they are released in the extracellular environment through an unknown secretion pathway. Here, we investigated the unconventional mechanism by which teratocytes release Ae-FABP and Ae-ENO in the extracellular space. Our results, obtained using immunogold staining coupled with TEM and western blot analyses, show that these two proteins are localized in vesicles released by teratocytes. The specific dimension of these vesicles and the immunodetection of ALIX and HSP70, two exosome markers, strongly support the hypothesis that these vesicles are exosomes.

Insect parasitic factors of both maternal and embryonic origin allow the endoparasitoid progeny to elude the humoral and cellular immune system responses of their host. Endoparasitoid wasps of lepidopteran larval stages inject, along with venom and ovarian proteins, Polydnavirus particles acting in synergy with all other factors in host regulation for parasitism success. To date, the molecular mechanisms used by endoparasitoid to circumvent the host immune system are little known. Neverthless, several of these strategies are conserved through the wasp parasitoid species. Heliothis virescens is a noctuid moth, host of the endophagous parasitoid Toxoneuron nigriceps. The first observed effect of parasitism is immune system suppression, as direct consequence of the array of host regulation factors, both of embryonic and maternal origin. This review describes the contribution of all the parasitic components during alterations of the host immune response observed after oviposition by T. nigriceps.

Background Information While enolase is a ubiquitous metalloenzyme involved in the glycolytic pathway, it is also known as a multifunctional protein, since enolases anchored on the outer surface of the plasma membrane are involved in tissue invasion. Results We have identified an extracellular enolase ( Ae-ENO) produced by the teratocytes, embryonic cells of the insect parasitoid Aphidius ervi. We demonstrate that Ae-ENO, although lacking a signal peptide, accumulates in cytoplasmic vesicles oriented towards the cell membrane. Ae-ENO binds to and activates a plasminogen-like molecule inducing digestion of the host tissue and thereby ensuring successful parasitism. Conclusions These results support the hypothesis that plasminogen-like proteins exist in invertebrates. Interestingly the activation of a plasminogen-like protein is mediated by a mechanisms involving the surface enolase/fibrinolytic system considered, until now, exclusive of vertebrates, and that instead is conserved across species. Significance To our knowledge, this is the first example of enolase mediated Plg-like binding and activation in insect cells, demonstrating the existence of an ECM degradation process via a Plg-like protein in invertebrates. Abstract: The larval endoparasitoid Toxoneuron nigriceps (Viereck) (Hymenoptera: Braconidae) has a toolbox of biological weapons to secure for host colonization and the successful parasitization of its host Heliothis virescens (F.) (Lepidoptera: Noctuidae). The cDNA of a putative chitinase has been previously isolated and initially characterized from teratocytes of this parasitoid among the plethora of molecules available in the venom and calyx fluids injected by females, oral and/or anal secretions released by the parasitoid larvae and/or produced by the expression of genes of the symbiotic associated polydnavirus. This putative chitinase has been initially associated with the host cuticle digestion to allow for parasitoid egression and with the asepsis of the host environment, acting as an antimicrobial. As chitinases are commonly expressed in plants against plant pathogens, the chitinase derived from the teratocytes of T. nigriceps is a potential tool for the development of insect pest control methods based on the disruption of the perithrophic membrane of herbivores. Therefore, we aimed to characterize the activity of the putative chitinase from teratocytes of T. nigriceps (Tnchi) produced using the Escherichia coli expression system and its potential to control H. virescens larvae when expressed into transgenic tobacco plants. The purified E. coli-produced Tnchi protein showed no chitinolitic activity, but was active in binding with colloidal and crystalline chitins in water and with colloidal chitin in buffered solution (pH=6.74). Transgenic tobacco plants showed no enhanced chitinolitic activity relative to control plants, but survival of three-day old larvae of H. virescens was severely affected when directly fed on transgenic tobacco leaves expressing the recombinant Tnchi protein. Abstract: Defining host ranges in parasitoid insects is important both from a theoretical and an applied point of view. Based on the literature, some species seem able to use a wide range of hosts, while field studies indicate possible local host specialization. In koinobiont endoparasitoid species, such specialization could involve physiological processes. We tested the ability of two strains of the cosmopolitan and polyphagous parasitoid Diaeretiella rapae, to develop in three of its recorded aphid host species. Both strains produced high parasitism rates on the cabbage aphid Brevicoryne brassicae and the green peach aphid Myzus persicae but almost no progeny on the cherry-oat aphid Rhopalosiphum padi. This last species was less attacked by female parasitoids. Moreover, parasitoid eggs and larvae were smaller than in the two other host aphid species and their development was delayed. This abnormal development appeared to be due to an incomplete host regulation process, probably related to the low number and the size of teratocytes produced by D. rapae in R. padi individuals. Such a failure as far as gaining control of the host''s metabolism is concerned could play an important role in shaping the host range of parasitoid insects, leading to local variation of the host spectrum in populations from various geographical areas. Abstract: Glyptapanteles liparidis is a gregarious, polydnavirus (PDV)-carrying braconid wasp that parasitizes larval stages of Lymantria dispar. In previous studies we showed that parasitized hosts dramatically increase juvenile hormone (JH) titers, whereas JH degradation is significantly inhibited in the hemolymph. Here we (i) quantified the effects of parasitism on JH esterase (JHE) activity in hemolymph and fat body of penultimate and final instars of L. dispar hosts and (ii) assessed the relative contribution of individual and combined wasp factors (PDV/venom, teratocytes, and wasp larvae) to the inhibition of host JHE activity. The effects of PDV/venom was investigated through the use of gamma-irradiated wasps, which lay non-viable eggs (leading to pseudoparasitization), while the effects of teratocytes and wasp larvae were examined by injection or insertion of these two components in either control or pseudoparasitized L. dispar larvae. Parasitism strongly suppressed host JHE activity in both hemolymph and fat body irrespective of whether the host was parasitized early (premolt-third instar) or late (mid-fourth instar). Down-regulation of JHE activity is primarily due to the injection of PDV/venom at the time of oviposition, with only very small additive effects of teratocytes and wasp larvae under certain experimental conditions. We compare the results with those reported earlier for L. dispar larvae parasitized by G. liparidis and discuss the possible role of JH alterations in host development disruption. In some parasitoid species, the serosa membrane breaks apart at hatching and produces teratocyte cells that assume various functions (immunossupression, secretion and nutrition) mediating host–parasitoid relationships. Teratocyte growth pattern may thus reflect the host suitability for a parasitoid. The teratocyte growth pattern (increase in size and number of teratocytes as a function of time) is studied and used as an indirect measure of fitness to compare the development of the endoparasitoid Dinocampus coccinellae in a marginal host, the coccinellid Harmonia axyridis, and in a suitable host, Coleomegilla maculata. Indirect measures of fitness recorded in both host species confirm that C. maculata is a suitable host for D. coccinellae contrary to the marginal host H. axyridis. According to regression analysis, teratocyte numbers decrease linearly whereas teratocyte size increases linearly with time in the suitable host C. maculata (larvae or adults). In the marginal host, parasitism occurs only in the larval stage where a delay in the parasitoid larval development is observed. Increase in teratocyte size is also highly variable. The teratocyte growth pattern of the parasitoid in the marginal host does not follow the linear model found in the suitable host. Abstract: The braconid wasp Toxoneuron nigriceps induced qualitative and quantitative changes in the protein composition of the moth Heliothis virescens host hemolymph. Total protein concentration was found to be higher in parasitized host 4 days after parasitism as compared to control hosts, mainly due to changes in a particular group of proteins. Host proteins with a molecular mass of 173 and 72 kDa were found in higher levels in the hemolymph of parasitized larvae as control hosts approached pupation, while an 80 kDa peptide was found in reduced concentration in the hemolymph of parasitized hosts. Levels of these three peptides were maintained throughout parasitoid development, while two of them (173 and 72 kDa) were cleared from the host hemolymph close to pupation. Besides the regulation of host proteins, three parasitism-specific proteins (PSPs) were released into the host hemolymph. Two of them (PSP1-MW=116 kDa, pI =6.3; PSP2-MW=114 kDa, pI =6.2) first appeared in the hemolymph of parasitized hosts soon after pupation of control host and increased in concentration as the parasitoid developed. The third PSP (PSP3-MW=56 kDa, pI =5.8) was produced towards the end of parasitoid larval development, close to parasitoid egression. Database searches based on the amino acid composition and amino terminal sequence of PSP1 and PSP2 did not produce any significant matches, while PSP3 was identified as a putative chitinase. Incubation of host derived tissues, parasitoid larvae and teratocytes in 35S conditioned media suggested PSPs were a product of teratocytes. Abstract: Teratocytes derived from the embryonic membrane (serosa) of parasitoids are released into the host hemocoel when the parasitoid eggs hatch, where they perform several functions during the post-embryonic stage. A full-length cDNA encoding a putative carboxylesterase was isolated from the teratocytes of Dinocampus coccinellae and was designated as teratocyte-specific carboxylesterase (TSC). It contained an open reading frame of 2571bp coding for a protein of 857 amino acids with a calculated molecular mass of 89kDa. The deduced amino acid sequence had many structural features that are highly conserved among serine hydrolases including Ser, Glu and His as a catalytic triad, carboxylesterase type-B (FGGNPNSVTLLGYSAG)/ lipase-serine (VTLLGYSAGA) active sites, and six N-glycosylation sites. Interestingly, the mRNA encoding the TSC gene was expressed exclusively in teratocytes but not in the parasitoid larva or in the non-parasitized host. Most notably, the TSC protein was distinguished by an insertion of 294 amino acids towards the N-terminal region and was flanked by carboxylesterase domains. Furthermore, sequence alignment and homology search revealed these additional amino acids to be unique to TSC and the insertion contributed significantly to its molecular mass resulting in a larger protein than other esterases. In addition to sequence analysis, the possible role of TSC in relation to the host (Coccinella septempunctata) and parasitoid (D. coccinellae) system is discussed. [Copyright &y& Elsevier]

Laboratory studies investigated the development of teratocytes derived from the eggs of the parasitoidMeteous gyrator(Thun.) in its host, the tomato mothLacanobia oleracea(L.). At hatching, each parasitoid egg produced an average of approximately 1000 teratocytes, but this number declined to approximately 400 during the course of parasitism. The teratocytes increased in size markedly, such that 7 days after egg hatch their mean diameter was approximately four times that of the cells immediately after dissociation. The haemolymph of parasitized hosts had reduced phenoloxidase activity, and teratocytes inhibited phenoloxidase activity when coincubated with plasma from nonparasitized hosts. The injection of teratocytes into nonparasitized fifth-instarL. oleracealarvae suppressed growth and induced a supernumerary moult in some larvae. A number of parasitism-specific proteins were detected in the haemolymph of parasitized hosts, and incubation of teratocytes in culture media indicated that these cells were a source of at least two of these proteins. In several species of hymenopteran parasitoids of the superfamilies of Ichneumonoidea and Platygastroidea, the membrane enveloping the parasitoid embryo dissociates at hatching into a number of cells, called teratocytes, which autonomously develop in the host haemolymph. In this work we report for Encarsia berlesei and Encarsia citrina (Hymenoptera: Chalcidoidea), the dissociation of the extraembryonic membrane into cells whose morphological and embryological features correspond to those of teratocytes. In E. berlesei the membrane dissociated at hatching into 4–9 larger cells (100 μm diameter) and about 10 smaller cells (60 μm), which scarcely doubled their size during maturation. In E. citrina the membrane dissociated into five large cells (250 μm) which did not grow appreciably. Ultrastructural investigation of the dissociated cells in E. berlesei revealed that their surface was covered by microvilli, whose density and length increased from the egg stage to the 12 h following hatching. During the same period, rough endoplasmic reticulum evolved from a parallel profile to that of the cisternal type, while abundant vesicles represented the dominant cytological feature. The ploidy level of these cells ranged between 8c and 140c at hatching, but increased to 40c–350c at maturation. Abstract After parasitization, some wasps induce hosts prematurely to initiate metamorphic development that is then suspended in a postwandering, prepupal state. Following egression of the parasite larva, the host remains in this developmentally arrested state until death. Teratocytes, cells released at egg hatch from extra-embryonic serosal membranes of some wasp parasites, inhibit growth and development when injected into host larvae independent of other parasite factors (e.g. venom, polydnavirus). Synthesis of some developmentally regulated, abundantly expressed Heliothis virescens host proteins is inhibited in hosts parasitized by Microplitis croceipes and by teratocyte injection. A cDNA encoding a 13.9 kDa protein (TSP14) that inhibited protein synthesis, growth and development was isolated from a protein fraction secreted by teratocytes. TSP14 appears to be responsible, in part, for the teratocyte-mediated inhibition of host growth and development. Interestingly, this cDNA encoded a cysteine-rich amino acid motif similar to that described from Campoletis sonorensis polydnavirus, a mutualistic virus that enables wasp parasitization of lepidopteran larvae. Moreover, TSP14 inhibited protein synthesis in a dose-dependent manner in rabbit reticulocyte lysate and wheat germ extract translation systems. Abstract Embryo production in aphids is absolutely dependent on the function of symbiotic bacteria, mainly Buchnera , and the growth and development of koinobiont parasitoids in aphids requires the diversion of nutrients from aphid embryo production to the parasitoid. The implication that the bacterial symbiosis may be promoted in parasitized aphids to support the growing parasitoid was explored by analysis of the number and biomass of mycetocytes, and the aphid cells bearing Buchnera , in the pea aphid Acyrthosiphon pisum Harris (Hemiptera: Aphididae) parasitized by the wasp Aphidius ervi Haliday (Hymenoptera: Braconidae). Aphids hosting a young larval parasitoid bore more mycetocytes of greater total biomass, and embryos of lower biomass than unparasitized aphids. Furthermore, one of the three aphid clones tested, which limited teratocyte growth (giant cells of parasitoid origin having a trophic role), bore smaller mycetocytes and larger embryos, than one or both of the two aphid clones with greater susceptibility to the parasitoid. These data suggest that susceptibility of the aphid-Buchnera symbiosis to parasitoid-mediated manipulation may, directly or indirectly, contribute to aphid susceptibility to parasitoid exploitation. Microplitis croceipes wasps overcome host immunity by inducing changes in host physiology using factors derived from the embryo and/or larva. Teratocytes of some parasitic wasps circulate in the host hemolymph after egg hatch and synthesize proteins (TSPs), some of which are secreted to alter host physiology in support of endoparasitoid development. TSPs appear to alter host physiology, at least in part, by inhibiting synthesis of certain proteins. M. croceipes teratocytes synthesize a 13.9 kDa protein (TSP14), which inhibits synthesis of host proteins that are linked to larval growth and development. A cDNA encoding TSP14 was generated by RT-PCR from teratocyte RNA, and cloned into yeast expression vectors to produce sufficient recombinant protein for functional analyses. RecTSP14 was produced using the yeast expression system at a concentration of more than 300 μg/L. The recTSP14 inhibited in vitro translation of larval Heliothis virescens RNA, with the activity sensitive to boiling, protein concentration, incubation time, and storage temperatures. Although recTSP14 inhibited translation of some cellular RNAs in vitro, the in vivo incorporation of [35S]-methionine into proteins of selected insect and mammalian cell lines was not inhibited. The physiological mechanism of resistance to the endophagous braconid Aphidius ervi Haliday (Hymenoptera, Braconidae) by a pink clone (PC) of Acyrthosiphon pisum (Harris) (Homoptera, Aphididae) has been investigated. Comparative data on parasitoid development and associated host biochemical changes in the resistant PC aphids and in a susceptible green clone (GC) of A. pisum are reported. When the PC aphids were attacked as early 4th instars, the developing parasitoid larvae showed a strongly reduced increase in size, compared to those synchronously developing in GC aphids, and were unable to produce a regular mummy. In contrast, parasitism of 2nd instar PC aphids, allowed completion of parasitoid development, but adults had a prolonged developmental time, due to a longer duration of parasitoid’s final (3rd) instar. In all cases, teratocytes, cells deriving from the A. ervi serosal membrane, and the proteins abundantly synthesised by them, were never found in the haemolymph of parasitised PC aphids. Host castration, as demonstrated by total protein incorporation into reproductive tissues, was total in the majority of early (2nd instar) parasitised host aphids, while it was limited when later instars (4th) of PC aphids were parasitised. This is partly due to the absence of the cytolytic activity of teratocytes on host embryos, which, through their persistence, may compete for nutritional resources with the developing parasitoid larvae. In parasitised PC aphids, this competitive effect is further aggravated for the parasitoid by the absence of the regulated amino acid titre increase in the host haemolymph, which is regularly observed in GC aphids. Failure of teratocyte development in the PC clone of the pea aphid is, then, the major functional constraint accounting for the reduction/inhibition of A. ervi larval growth. The reported results allow to assess in vivo the role of teratocytes in the host physiological redirection and nutritional exploitation by the parasitoid, and to integrate and validate the proposed physiological model of host-parasitoid interactions in the system A. pisum-A.ervi. Teratocytes, cells which originate from the serosal membrane of some Braconidae and Scelionidae, can be found in the hemocoel of permissive hosts during part or all of the developmental time of the parasitoid larva. Teratocytes from Microplitis croceipes are known to secrete biologically active proteins, which contribute to developmental arrest and failure to pupate of Heliothis virescens larvae. One such protein, which has a molecular weight of approximately 14 kDa is called TSP14. The presence of parasitoid larvae is essential to maintain teratocytes under in vitro conditions with protein-free EX-CELL 400. The teratocyte viability was maintained in vitro for at least 12 days in the presence of larvae when medium was exchanged every three days. Western blots show that TSP14 was secreted during the entire period of exchanges. In the absence of parasitoid larvae, teratocyte viability was only 30% by day 6 and no TSP14 had been secreted. In the absence of parasitoid larvae, teratocytes maintained in vitro in EX-CELL 400 medium supplemented with 10% FBS remained viable for at least nine days and secreted TSP14 for at least six days. This suggests that parasitoid larval secretions are sufficient but not uniquely essential to maintain teratocyte viability. Summary 1. In solitary parasitoid Hymenoptera, normally only one offspring per host survives. Parasitism with more than one egg by the same female results in siblicide and hence is generally maladaptive. 2. The braconid wasp Monoctonus paulensis Ashmead is a solitary parasitoid of aphids, including the Pea Aphid. Although all but one offspring are eliminated by competition between first-instar larvae, females often produce a multiple-egg clutch, even when searching alone. The hypothesis that the size and, presumably, the fitness of the surviving offspring in a clutch are greater than that of singly developing counterparts was tested in the laboratory. Adult size was measured as dry mass in parasitoids developing in Pea Aphids that differed in size (instar) and growth potential. 3. Parasitoid size increased with host size, and females were larger than males. Parasitoids developing in aphids reared under crowded conditions (which had low growth potential) were smaller relative to initial host size. As predicted, the male offspring of virgin mothers developing in a clutch were 4–5% larger than counterparts developing alone. Clutch size had no consistent effect on the size of male or female offspring of mated mothers, however. 4. A female of M. paulensis laying a multiple-egg clutch can gain in fitness in terms of increased offspring size. The surviving offspring has access to greater nutritional resources because superparasitized aphids feed more than single-parasitized aphids; the former also contain proportionately more teratocytes, which are egg-derived cells aiding in parasitoid nutrition. 5. A method for in vitro cultivation of teratocytes from the egg parasitoid Telenomus heliothidis (Hymenoptera: Scelionidae) is described. Parasitoid eggs, from which teratocytes were derived, were collected from 24-h-old Heliothis virescens (Lepidoptera: Noctuidae) eggs previously parasitized by T. heliothidis females. Optimal culture conditions, including species and concentration of serum, were determined experimentally. Thirty percent Manduca sexta hemolymph or 10% chicken serum in Hink's TNH-FH medium were found to generate the most satisfactory number of teratocytes per parasitoid larva. Teratocytes cultivated in vitro showed similar development and morphology to those produced in vivo. However, cultured teratocytes lived approximately 10 times longer than teratocytes in natural hosts and were not dependent upon the presence of the parasitoid larva for normal development. Biological control using entomopathogens and natural enemies is an ecofriendly method for pest management in agriculture. Biological control agents often can be simultaneously employed and compatibility between agents may improve pest suppression. We investigated the influence of the entomopathogen Bacillus thuringiensis (Bt) on the immune system of the sugarcane borer Diatraea saccharalis (Fabricius, 1794) (Lepidoptera: Crambidae) to determine if such changes impact parasitization by Cotesia flavipes Cameron, 1891 (Hymenoptera: Braconidae). The immune response of surviving D. saccharalis larvae fed with an LC 25 dosage of a Bt-based biopesticide (Dipel®) was analyzed (total hemocyte count, hemocyte adhesion, and activities of phenoloxidase and lysozyme). Furthermore, the suitability of surviving Bt-fed larvae as hosts for C. flavipes was assessed by measuring parasitoid attributes such as number and size of teratocytes, weight of pupae, length of adult female tibia and number of emerged adults. Total hemocyte count, but not hemocyte adhesion, total protein content and phenoloxidase activity increased in the hemolymph of non-parasitized Bt-fed larvae (Bt-NP) compared to control larvae (NBt-NP). Lysozyme activity increased only after parasitization without Bt exposure (NBt-P). After parasitization, the immunological parameters activated in Bt-NP larvae decreased to levels at or below those observed in control larvae, showing that C. flavipes can regulate the activated immune response of Bt-fed larvae. The development of C. flavipes was not impaired in Bt-fed larval hosts (Bt-P); no changes were observed for teratocyte size, weight of pupal mass, length of hind tibia and number of adults emerged.
