Purpose: Quantitative sensory testing commonly utilizes the unaffected, contralateral side as a control to detect somatosensory dysfunction. There is scant evidence that somatosensory function for the volar dominant and non-dominant hands is equivalent, therefore intra-patient comparisons are unwarranted. This study aimed to identify dominance-related differences in palmar hand somatosensation, thereby determining if the unaffected contralateral hand is a valid comparator in clinical populations. Participants and Methods: With ethical approval (IREC_13_1_10) and informed consent, 110 healthy adult volunteers' participated in this clinical measurement study. Somatosensory function was assessed with the German Research Network on Neuropathic Pain (DFNS) quantitative sensory testing (QST) protocol. Half of the participants were tested on the dominant hand. Thirteen parameters of thermal and mechanical detection and pain threshold were evaluated at both the dorsal and volar hand (distal middle finger). Tests were performed in the same order and instructions were read from a standardized script. Results for dorsal hand tests were compared to DFNS normative data to confirm participants met study inclusion criteria. Between-group differences for age and sex were investigated with the independent samples t-test and Chi-square test of independence, respectively. Group differences for dominant and non-dominant hands for all 13 continuous QST parameters were investigated with the Mann–Whitney U-test. Results: Data for 106 participants were included in statistical analysis. Fifty percent of participants were tested on the dominant hand [n=53]; there were no differences for age or sex between groups (dominant or non-dominant hand test group). The dominant volar hand was significantly more sensitive to vibration detection threshold than the non-dominant hand (P=0.001). There were no significant differences related to dominance for other DFNS QST measures. Conclusion: For quantitative sensory testing with the DFNS protocol in healthy cohorts, the contralateral, unaffected hand is a valid control, with the exception of vibration detection threshold. Many haematophagous insects use the heat emitted by warm‐blooded animals as a cue for locating suitable hosts. Blood‐feeding stable flies, Stomoxys calcitrans (L.) (Diptera: Muscidae), are known to respond to visual and olfactory host cues. However, the effects of thermal host cues on the foraging behaviour of these flies remain largely unknown. Here we tested the hypothesis that host‐foraging stable flies preferentially land on objects with host‐like temperature, and on objects with both visual and thermal host‐like cues. In laboratory bioassays, stable flies were offered a choice between paired temperature‐controlled copper discs. Flies preferentially landed on the disc with a host‐like temperature (40 °C), discriminating against discs that were cooler (26 or 35 °C) or warmer (50 or 60 °C) than vertebrate hosts. Flies that were well fed and thus not in foraging mode, or host‐foraging flies that were offered infrared radiation but not the conductive and convective heat of different temperature discs, failed to discriminate between the stimuli. In greenhouse experiments, when flies were offered a choice between paired barrels as surrogate hosts, flies preferentially landed on barrels that were both thermally and visually appealing (38–39 °C, black), discriminating against barrels that were cold (10 °C), white, or both cold and white. Thermal cues augmented the overall landing responses of flies but their initial (mid‐range) attraction to barrels was mediated by visual cues. Overall, the data suggest that thermal host cues affect the host‐foraging behaviour of stable flies primarily at close range, prompting landing on a host. [ABSTRACT FROM AUTHOR]

Background. The study of the mechanisms of sensation, primarily visual, emergence under the influence of a light stimulus on a person is a classic task for psychological science. At the same time, little attention has been paid to the sensations arising from light exposure of the skin surface, despite the surprising results obtained in a well-known series of experiments by Russian psychologist A.N. Leontiev. The task of studying skin-optical sensitivity with the modern element base of experimental equipment is relevant from the point of view of the development of the general psychological theory of activity and understanding sensory-perceptual processes of sensation formation due to light exposure. Objective of the study was to investigate the ability of subjects to recognize the effect of light on their skin surface. Methods. A specially designed experimental setup was used for the purpose of the study, which included a LED light source and a control programme. The experimental procedure involved the psychophysical method of two-alternative forced choice. The main experiment consisted of three series of cycles for each participant: first, 10 cycles were presented to fix the initial state; 100 cycles followed to train the subject; then, 10 cycles were used to fix the final state. Sample. 12 participants took part in the experiment. Results. showed that the number of correct answers when determining the cycle phase corresponding to the LED switching on, is significantly more than 50% (t = 10,992, df = 1414, p < 0.001). Conclusion. The conclusions of A.N. Leontiev and N.B. Poznanskaya about the possibility of skin sensitivity to light exposure were confirmed. A biophysical hypothesis is proposed to explain the result.

Several hypotheses tried to explain the advantages of zebra stripes. According to the most recent explanation, since the borderlines of sunlit white and black stripes can hamper thermal vessel detection by blood-seeking female horseflies, striped host animals are unattractive to these parasites which prefer hosts with a homogeneous coat, on which the temperature gradients above blood vessels can be detected more easily. This hypothesis has been tested in a field experiment with horseflies walking on a grey barrel with thin black stripes which were slightly warmer than their grey surroundings in sunshine, while in shade both areas had practically the same temperature. To eliminate the multiple (optical and thermal) cues of this test target, we repeated this experiment with improved test surfaces: we attracted horseflies by water- or host-imitating homogeneous black test surfaces, beneath which a heatable wire ran. When heated, this invisible and mechanically impalpable wire imitated thermally the slightly warmer subsurface blood vessels, otherwise it was thermally imperceptible. We measured the times spent by landed and walking horseflies on the test surface parts with and without underlying heated or unheated wire. We found that walking female and male horseflies had no preference for any (wired or wireless) area of the water-imitating horizontal plane test surface on the ground, independent of the temperature (heated or unheated) of the underlying wire. These horseflies looked for water, rather than a host. On the other hand, in the case of host-imitating test surfaces, female horseflies preferred the thin surface regions above the wire only if it was heated and thus warmer than its surroundings. This behaviour can be explained exclusively with the higher temperature of the wire given the lack of other sensorial cues. Our results prove the thermal vessel recognition of female horseflies and support the idea that sunlit zebra stripes impede the thermal detection of a host's vessels by blood-seeking horseflies, the consequence of which is the visual (non-thermal) unattractiveness of zebras to horseflies. Solitary bees play a crucial role in ecological systems, contributing to the pollination of crops and wild plants. All females are reproductive, and their habitat requirements include nesting sites, food resources and nesting materials. Although these activities require the ability to detect biotic and abiotic stimuli in the environment, the sensory system of these species is poorly studied. In this study, the antennal sensilla of five solitary bee species belonging to three Apoidea families were investigated using scanning electron microscopy. These included two species of stem-nesting bees, Ceratina cucurbitina (Rossi, 1792) (Apidae) and Osmia scutellaris (Morawitz, 1868) (Megachilidae), and three species of ground-nesting bees, Lasioglossum brevicorne (Schenck, 1870), Lasioglossum leucozonium (Schrank, 1781), and Lasioglossum villosulum (Kirby, 1802) (Halictidae). Thirteen different types of antennal sensilla were identified in females based on their morphological characteristics: sensilla trichodea (subtypes STI, II, III), chaetica (subtypes SchI, II), basiconica (subtypes SBI, II, III, IV), placodea, campaniformia, coeloconica, and ampullacea. Their functional role was discussed and morphology was compared among the species and within the antennal segments in each species. The results provide a baseline for further physiological and behavioural studies to determine the role of antennal sensilla in habitat selection, food search and nesting site selection. • Antennal sensilla are described in females of five species of Apoidea. • Seven types of sensilla and related morphological subtypes are identified using scanning electron microscopy. • Differences in types and distribution are found among ground and stem nesting species. Temperature is a critical factor for living organisms. Many microorganisms migrate toward preferable temperatures, and this behavior is called thermotaxis. In this study, the molecular and physiological bases for thermotaxis are examined in Chlamydomonas reinhardtii. A mutant with knockout of a transient receptor potential (TRP) channel, trp2-3, showed defective thermotaxis. The swimming velocity and ciliary beat frequency of wild-type Chlamydomonas increase with temperature; however, this temperature-dependent enhancement of motility was almost absent in the trp2-3 mutant. Wild-type Chlamydomonas showed negative thermotaxis, but mutants deficient in the outer or inner dynein arm showed positive thermotaxis and a defect in temperature-dependent increase in swimming velocity, suggesting involvement of both dynein arms in thermotaxis., (© 2024 The Authors. Introduction: Capsaicin, a chili pepper extract, can stimulate increased skin blood flow (SkBF) with a perceived warming sensation on application areas. Larger surface area application may exert a more systemic thermoregulatory response. Capsaicin could assist with maintaining heat transport to the distal extremities, minimizing cold weather injury risk. However, the thermoregulatory and perceptual impact of topical capsaicin cream application prior to exercise in the cold is unknown., Methods: Following application of either a 0.1% capsaicin or control cream to the upper and lower extremities (10 g total, ∼40-50% body surface area), 11 participants in shorts and a t-shirt were exposed to 30 min of cold (0 °C, 40% relative humidity). Exposures comprised of 5 min seated rest, 20 min walking (1.6 m·s -1 , 5% grade), and 5 min seated rest. Temperature (skin, core), SkBF, skin conductivity, heart rate, thermal sensation, and thermal comfort were measured throughout., Results: The capsaicin treatment did not differ from the control treatment in skin temperature (treatment mean: 30.0 ± 2.5, 30.1 ± 2.4 °C, respectively, p  = 0.655), core temperature (treatment mean: 37.3 ± 0.5, 37.4 ± 0.4 °C, respectively, p  = 0.113), SkBF (treatment mean: -8.4 ± 10.0, -11.1 ± 10.7 A.U., respectively, p  = 0.492), skin conductivity (treatment mean: -0.7 ± 5.1, 0.4 ± 6.4 µS, respectively, p  = 0.651), or heart rate (treatment mean: 83 ± 29, 85 ± 28 beats·minute -1 , respectively, p  = 0.234). The capsaicin and control treatments also did not differ in thermal sensation ( p  = 0.521) and thermal comfort ( p  = 0.982), with perceptual outcomes corresponding with feeling "cool" and "just uncomfortable," respectively., Conclusions: 0.1% topical capsaicin application to exposed limbs prior to walking in a cold environment does not alter whole-body thermoregulation or thermal perception.

Tabanid flies (Diptera: Tabanidae) are attracted to shiny black targets, prefer warmer hosts against colder ones and generally attack them in sunshine. Horizontally polarised light reflected from surfaces means water for water-seeking male and female tabanids. A shiny black target above the ground, reflecting light with high degrees and various directions of linear polarisation is recognised as a host animal by female tabanids seeking for blood. Since the body of host animals has differently oriented surface parts, the following question arises: How does the attractiveness of a tilted shiny black surface to male and female tabanids depend on the tilt angle δ? Another question relates to the reaction of horseflies to horizontal black test surfaces with respect to their surface temperature. Solar panels, for example, can induce horizontally polarised light and can reach temperatures above 55 °C. How long times would horseflies stay on such hot solar panels? The answer of these questions is important not only in tabanid control, but also in the reduction of polarised light pollution caused by solar panels. To study these questions, we performed field experiments in Hungary in the summer of 2019 with horseflies and black sticky and dry test surfaces. We found that the total number of trapped (male and female) tabanids is highest if the surface is horizontal (δ = 0°), and it is minimal at δ = 75°. The number of trapped males decreases monotonously to zero with increasing δ, while the female catch has a primary maximum and minimum at δ = 0° and δ = 75°, respectively, and a further secondary peak at δ = 90°. Both sexes are strongly attracted to nearly horizontal (0° ≤ δ ≤ 15°) surfaces, and the vertical surface is also very attractive but only for females. The numbers of touchdowns and landings of tabanids are practically independent of the surface temperature T. The time period of tabanids spent on the shiny black horizontal surface decreases with increasing T so that above 58 °C tabanids spent no longer than 1 s on the surface. The horizontally polarised light reflected from solar panels attracts aquatic insects. This attraction is adverse, if the lured insects lay their eggs onto the black surface and/or cannot escape from the polarised signal and perish due to dehydration. Using polarotactic horseflies as indicator insects in our field experiment, we determined the magnitude of polarised light pollution (being proportional to the visual attractiveness to tabanids) of smooth black oblique surfaces as functions of δ and T. Honeybee foragers are exposed to thermal stimuli when collecting food outside and receiving food rewards inside the nest. In both contexts, there is an opportunity for foragers to associate warmth with food rewards. However, honeybee thermal learning is poorly understood. Using an associative learning paradigm (the proboscis extension reflex), we show that honeybees can learn to associate a nectar reward with a heated stimulus applied to the antenna to mimic natural contact with a warm flower or nectar-offering forager. Conditioning with longer inter-trial intervals (ITI) significantly improved learning acquisition. We also trained bees to discriminate between temperatures above (warm) and below (cold) ambient air temperature. Learning acquisition improved by 38% per 10 degrees C increase in absolute stimulus intensity (difference between the rewarded temperature and unrewarded ambient air temperature). However, bees learned positive temperature (warm) significantly better than negative temperature (cold) differences, approximately twice as well for 10 degrees C as compared with a -10 degrees C difference. Thus, thermosensation, a sensory modality that is relatively unexplored in honeybees, could play a role in the acquisition of information from nestmates (social learning) and in foraging decisions influenced by associations between floral temperature and nectar rewards.

From a large distance tabanid flies may find their host animal by means of its shape, size, motion, odour, radiance and degree of polarization of host-reflected light. After alighting on the host, tabanids may use their mechano-, thermo-, hygro- and chemoreceptors to sense the substrate characteristics. Female tabanids prefer to attack sunlit against shady dark host animals, or dark against bright hosts for a blood meal, the exact reasons for which are unknown. Since sunlit darker surfaces are warmer than shady ones or sunlit/shady brighter surfaces, the differences in surface temperatures of dark and bright as well as sunlit and shady hosts may partly explain their different attractiveness to tabanids. We tested this observed warmth preference in field experiments, where we compared the attractiveness to tabanids (Tabanus tergestinus) of a warm and a cold shiny black barrel imitating dark hosts with the same optical characteristics. Using imaging polarimetry, thermography and Schlieren imaging, we measured the optical and thermal characteristics of both barrels and their small-scale models. We recorded the number of landings on these targets and measured the time periods spent on them. Our study revealed that T. tergestinus tabanid flies prefer sunlit warm shiny black targets against sunlit or shady cold ones with the same optical characteristics. These results support our new hypothesis that a blood-seeking female tabanid prefers elevated temperatures, partly because her wing muscles are more rapid and her nervous system functions better (due to faster conduction velocities and synaptic transmission of signals) in a warmer microclimate, and thus, she can avoid the parasite-repelling reactions of host animals by a prompt take-off.

Thermal stimulation of various parts of the skin in Hirudo medicinalis increases the frequency of spontaneous rhythmic excitation of Retzius neurons in leech ganglia. It was shown that the frequency of spontaneous rhythmic excitation of Retzius cells in the segmental ganglion increases only in response to thermal stimulation and returns to initial values upon cooling. This effect was also detected in neurons that are not directly connected by nerve fibers with the particular skin area. Changes in the frequency of spontaneous rhythmic excitation of Retzius cells in the segmental ganglion were observed during thermal stimulation of not only leech body, but also of the head and caudal suckers. These changes in spontaneous rhythmic excitation of Retzius cells in the segmental ganglion during thermal stimulation were observed in Hirudo medicinalis, but not in Macrobdella decora. In nature, living organisms, such as octopuses, cabrito, and frogs, have already evolved admirable adhesive abilities for better movement and predation in response to the surroundings. Inspired by biological structures, researchers have made enormous efforts in developing actuators that can respond to external stimuli, while such adhesive property is very desired, yet there is still limited research in responsive hydrogel actuators. Here, a bilayer actuator with high stretchability and robust interface bonding is presented, which has a smart adhesion and thermoreception function. The system consists of an adhesive passive layer copolymerized of amphoteric ([2-(methacryloyloxy) ethyl] dimethyl-(3-sulfopropyl), SBMA) and acrylic acid (AA), and an active layer hydrogel composed of poly( N -isopropylacrylamide) (PNIPAm) containing polydopamine-modified MXene (P-MXene) and calcium chloride (CaCl 2 ). The coordination of carboxylate and Ca 2+ at the interface of the two layers enhances the interfacial bonding from 14 to 30 N m -1 , which facilitates withstanding large strain and preventing stratification. The resulting hydrogel actuator can bend approximately 360° in a mere 10 s, exhibiting excellent photothermal effect, a large angle bending deformation, and ultrafast photoresponsive ability. As a proof of concept, the photothermal actuators are programmed to present various shapes and grab objects. Importantly, the hydrogel actuator exhibits remarkable adhesion capabilities toward diverse substrates, with a maximum peel force of up to 280 N m -1 . Relying on their own adhesion and the photoresponse properties, these flexible adhesion actuators show outstanding gripping capability, enabling them to grip and release objects of different shapes and weights. More interestingly, the hydrogel exhibits a smart adjustable adhesion capability at different temperatures, which enables it as a gripper to recognize temperature signals through real-time different feedback actions based on its own adhesion. This study presents innovative insights into biomimetic hydrogel actuators, providing new opportunities for developing intelligent soft robots with multiple functions.

To elucidate the nociceptive system of the brown treesnake, Boiga irregularis, we exposed isolated brown treesnake trigeminal neurons to thermal and chemical stimulation. We measured responses as changes in intracellular calcium using ratiometric fluorescent calcium imaging. Responses to aversive thermal and chemical identified several classes of putative nociceptors. Compounds that were aversive excited many trigeminal neurons, putative chemonociceptors. Identification as nociceptors was further supported by lack of activation by compounds that were not aversive. Brown treesnake neurons had thermal thresholds ranging from 32 to 49 °C. The distribution was discontinuous, with a population of thresholds from 32 to 45 °C and a population with thresholds > 48 °C. Thermal stimulation of 48 °C has been shown to be strongly aversive to brown treesnakes, is lethal, and suggests the presence of thermonociceptors. Thermal sensitivity of brown treesnake trigeminal neurons greatly overlaps with chemical sensitivity; only 1.1% of neurons were sensitive to only thermal stimulation. 50% of brown treesnake trigeminal neurons tested with both > 48 °C and cinnamaldehyde responded to both stimuli, identifying putative polymodal nociceptors. Although a previous study found brown treesnakes insensitive to capsicum extract containing capsaicin, brown treesnake trigeminal neurons responded to capsaicin. Determine if topical capsaicin, a transient receptor potential vanilloid heat thermoreceptor activator, alters thermoregulation and perception when applied topically prior to thermal exercise. Twelve subjects completed 2 treatments. Subjects walked (1.6 m s−1, 5% grade) for 30 min in the heat (38 °C, 60% relative humidity) with either a capsaicin (0.025% capsaicin) or control cream applied to the upper (shoulder to wrist) and lower (mid-thigh to ankle) limbs covering ∼50% body surface area. Skin blood flow (SkBF), sweat (rate, composition), heart rate, temperature (skin, core), and perceived thermal sensation were measured prior to and during exercise. The relative change in SkBF was not different between treatments at any time point (p = 0.284). There were no differences in sweat rate between the capsaicin (1.23 ± 0.37 L h−1) and control (1.43 ± 0.43 L h−1, p = 0.122). There were no differences in heart rate between the capsaicin (122 ± 38 beats·min−1) and control (125 ± 39 beats·min−1, p = 0.431). There were also no differences in weighted surface (p = 0.976) or body temperatures (p = 0.855) between the capsaicin (36.0 ± 1.7 °C, 37.0 ± 0.8 °C, respectively) and control (36.0 ± 1.6 °C, 36.9 ± 0.8 °C, respectively). The capsaicin treatment was not perceived as hotter than the control treatment until minute 30 of exercise (2.8 ± 0.4, 2.5 ± 0.5, respectively, p = 0.038) Topical capsaicin application does not alter whole-body thermoregulation during acute exercise in the heat despite perceiving the treatment as hotter late in exercise. • Capsaicin application prior to thermal exercise induces greater perceived heat. • Despite greater perceived heat there were no thermoregulation alterations. • Use of topical capsaicin for thermoregulatory purposes may not be warranted. The dynamical mechanisms underlying thermoreception in the nematode C. elegans are studied with a mathematical model for the amphid finger-like ciliated (AFD) neurons. The equations, equipped with Arrhenius temperature factors, account for the worm's thermotaxis when seeking environments at its cultivation temperature, and for the AFD's calcium dynamics when exposed to both linearly ramping and oscillatory temperature stimuli. Calculations of the peak time for calcium responses during simulations of pulse-like temperature inputs are consistent with known behavioral time scales of C. elegans. The recent development of the olfactory conditioning of the sting extension response (SER) has provided new insights into the mechanisms of aversive learning in honeybees. However, until now, very little information has been obtained concerning US detection and perception in this aversive conditioning. In the initial version of SER conditioning, bees learned to associate an odor CS with an electric shock US. Recently, we proposed a modified version of SER conditioning, in which thermal stimulation with a heated probe is used as US. This procedure has the advantage of allowing topical US applications virtually everywhere on the honeybee body. In this study, we made use of this possibility and mapped thermal responsiveness on the honeybee body, by measuring workers' SER after applying heat on 41 different structures. We then show that bees can learn the CS-US association even when the heat US is applied on body structures that are not prominent sensory organs, here the vertex (back of the head) and the ventral abdomen. Next, we used a neuropharmalogical approach to evaluate the potential role of a recently described Transient Receptor Potential (TRP) channel, HsTRPA, on peripheral heat detection by bees. I argue for sensory pluralism. This is the view that there are many forms of sensory interaction and unity, and no single category that classifies them all. In other words, sensory interactions do not form a single natural kind. This view suggests that how we classify sensory systems (and the experiences they generate) partly depends on our explanatory purposes. I begin with a detailed discussion of the issue as it arises for our understanding of thermal perception, followed by a general account and defense of sensory pluralism.

The simultaneous application of innocuous cutaneous warm and cold stimuli with a thermal grill can induce both paradoxical pain and paradoxical warmth (heat). The goal of this study was to investigate further the relationships between these paradoxical sensations. Stimuli were applied to the palms of the right hands of 21 volunteers with a thermode consisting of 6 bars, the temperature of which was controlled by Peltier elements. We assessed the quality and intensity of the sensations evoked by series of stimuli consisting of progressively colder temperatures combined with a series of given warm temperatures. We applied a total of 116 series of stimuli, corresponding to 785 combinations of warm and cold temperatures. The 2 paradoxical phenomena were reported for most of the series of stimuli (n = 66). In each of these series, the 2 phenomena occurred in the same order: paradoxical warmth followed by paradoxical pain. The difference between the cold–warm temperatures eliciting paradoxical warmth was significantly smaller than that producing paradoxical pain. The intensities of the warmth and unpleasantness evoked by the stimuli were directly related to the magnitude of the warm–cold differential. Our results suggest that there is a continuum between the painful and nonpainful paradoxical sensations evoked by the thermal grill that may share pathophysiological mechanisms. These data also confirm the existence of strong relationships between the thermoreceptive and nociceptive systems and the utility of the thermal grill for investigating these relationships. In the second segment of the antennae of haematophagous reduviids an unusual cave-like organ is found the function os which was investigated in Triatoma infestans. the morphology of the organ makes it difficult to ascribe it to a mechno- or chemoreceptive function, but shows some characteristics shared with thermoreceptors of other animals. The electrical activity of sense cells was recorded in the presence of stimuli that evoke behavioural responses in this species, i.e. warm, CO2, lactic and butyric acids at different concentrations. The three compounds tested failed to evoke a response at all concentrations assayed. Only thermal stimulation evinced a clear modification in the electrical activity of the sense cells.Both the morphological and electrophysiological findings support a thermoreceptive finding, habitat selection and circadian synchronization.

Background: The great majority of afferent neurons of insect legs project into their segmental ganglion. Intersegmental projections are rare and are only formed by sense organs associated with the basal joints of the legs. Such intersegmental projections never ascend as far as the brain and they form extensive ramifications within thoracic ganglia. A few afferents of chordotonal organs of the subcoxal joints ascend as far as the suboesophageal ganglion. Results: We describe novel afferent neurons in distal segments of locust legs that project directly into the brain without forming ramifications in other ganglia. In the brain, the fibres terminate with characteristic terminals in a small neuropile previously named the superficial ventral inferior protocerebrum. The somata of these neurons are located in the tibiae and tarsi of all legs and they are located within branches of peripheral nerves, or closely associated with such branches. They are not associated with any accessory structures such as tendons or connective tissue strands as typical for insect internal mechanoreceptors such as chordotonal organs or stretch receptors. Morphologically they show great similarity to certain insect infrared receptors. We could not observe projections into the superficial ventral inferior protocerebrum after staining mandibular or labial nerves, but we confirm previous studies that showed projections into the same brain neuropile after staining maxillary and antennal nerves, indicating that most likely similar neurons are present in these appendages also. Conclusion: Because of their location deep within the lumen of appendages the function of these neurons as infrared receptors is unlikely. Their projection pattern and other morphological features indicate that the neurons convey information about an internal physiological parameter directly into a special brain neuropile. Background: Tsetse flies are obligatory blood feeders, accessing capillaries by piercing the skin of their hosts with the haustellum to suck blood. However, this behaviour presents a considerable risk as landing flies are exposed to predators as well as the host's own defense reactions such as tail flicking. Achieving a successful blood meal within the shortest time span is therefore at a premium in tsetse, so feeding until replete normally lasts less than a minute. Biting in blood sucking insects is a multi-sensory response involving a range of physical and chemical stimuli. Here we investigated the role of heat and humidity emitted from host skin on the biting responses of Glossina pallidipes, which to our knowledge has not been fully studied in tsetse before. Methods: The onset and duration of the biting response of G. pallidipes was recorded by filming movements of its haustellum in response to rapid increases in temperature and/or relative humidity (RH) following exposure of the fly to two airflows. The electrophysiological responses of hygroreceptor cells in wall-pore sensilla on the palps of G. pallidipes to drops in RH were recorded using tungsten electrodes and the ultra-structure of these sensory cells was studied by scanning and transmission electron microscopy. Results: Both latency and proportion of tsetse biting are closely correlated to RH when accompanied by an increase of 13.1°C above ambient temperature but not for an increase of just 0.2°C. Biting persistence, as measured by the number of bites and the time spent biting, also increases with increasing RH accompanied by a 13.1°C increase in air temperature. Neurones in wall-pore sensilla on the palps respond to shifts in RH. Conclusions: Our results show that temperature acts synergistically with humidity to increase the rapidity and frequency of the biting response in tsetse above the levels induced by increasing temperature or humidity separately. The interconnection between vision and somatosensation is already well-established and is further supplemented by the evolutionary link between eyes and photoreceptors, and the functional connection between photosensation and thermoreception. However, our analysis shows that the relation between vision and somatosensation is much much deeper and suggests that somatosensation may possibly be the basis of vision. The purpose of this study was to determine whether ingestion of a small bolus of ice slurry (1.25 g kg(-1)) could attenuate the reduction in maximal voluntary isometric contraction (MVC) torque output during a 2-min sustained task following exercise-induced hyperthermia. On two separate occasions, 10 males (age: 24 ± 3 years, .VO(2peak): 49.8 ± 4.7 ml kg(-1) min(-1)) ran to exhaustion at their first ventilatory threshold in a hot environment (34.1 ± 0.1°C, 49.5 ± 3.6% RH). Prior to and after exercise, subjects performed a 2-min sustained MVC of the right elbow flexors in a thermoneutral environment (24.6 ± 0.8°C, 37.2 ± 4.5% RH). The post exercise MVC was performed immediately following the ingestion of either 1.25 g kg(-1) of ice slurry (-1°C; ICE) or warm fluid (40°C; CON), in a counterbalanced and randomised order. Run time to exhaustion (42.4 ± 9.5 vs. 41.7 ± 8.7 min; p = 0.530), and rectal (39.08 ± 0.30 vs. 39.08 ± 0.30°C; p = 0.934) and skin temperatures (35.26 ± 0.65 vs. 35.28 ± 0.67°C; p = 0.922) and heart rate (189 ± 5 vs. 189 ± 6 beats min(-1); p = 0.830) at the end of the run were similar between trials. Torque output during the post-exercise 2-min sustained MVC was significantly higher (p = 0.001) following ICE (30.75 ± 16.40 Nm) compared with CON (28.69 ± 14.88 Nm). These results suggest that ice slurry ingestion attenuated the effects of exercise-induced hyperthermia on MVC, possibly via internal thermoreceptive and/or temperature-related sensory mechanisms. This is a review of recent advances in our understanding of the sensory modalities of touch, temperature sensitivity, and pain. Most of the research described is psychophysical or perceptual in nature, but physiological and imaging studies are included when they sharpen issues or reveal underlying mechanisms. Coverage of touch research comprises the subjects of acuity, vibrotaction and texture perception, perception of location and movement, tactile attention, and cross-modal phenomena. For pain, the covered topics are central sensitization, pain-touch and pain-pain interactions, placebo effects, the role of attention and emotion in pain, and the genetics of pain. For touch, the topics are arranged roughly in order of increasing cognitive involvement, but such an ordering is not feasible for pain, where attitudes and expectations can substantially affect even the most "sensory" of judgments. Abstract: The evolution of triatomine bugs towards haematophagy has demanded different types of adaptations, i.e., morphological, physiological and behavioural. In fact, haematophagy evolved as a secondary adaptation facilitated by frequent vertebrate contact. As derived from other Heteroptera, probably from an entomophagous group, some main morphological pre-adaptations, as piercing mouthparts and sucking pumps were already present. Thus, the most important novel acquisitions of triatomines include physiological and behavioural traits to obtain and handle the blood meal. In this review, we discuss how the sensory system and the behaviour of triatomines have been shaped by natural selection to accomplish the tasks of finding a vertebrate host and getting access to its blood. The feeding behaviour of triatomines is presented in its spatial and temporal context. Finally, some methods to study these topics are described. [Copyright &y& Elsevier]

Cold sensing in mammals is heterogeneous and more than one type of receptor molecule is likely to be involved in the transduction process. Most features of innocuous cold receptors have been explained by TRPM8, the cold and menthol receptor, but their fast adaptation to cooling has not yet been reproduced in cellular systems. In this study we have used a newly developed system for applying fast thermal stimuli to dissociated dorsal root ganglia (DRG) neurons from young rats (150–200 g) in primary culture. We describe a novel type of cold-sensitive rat DRG neuron with rapid adaptation to cooling. These cells (4.3% of the total DRG population) do not express either TRPM8 or the other cold-activated TRP channel, TRPA1, and the epithelial sodium channel (ENaC) is not involved in their transduction. Increases in intracellular calcium induced by cooling in rapidly adapting neurons are caused by calcium entry. These neurons express a large and rapidly adapting cold-induced inward current with a time constant of adaptation in the seconds range, and may correspond to the rapidly adapting cold receptors described in vivo. The thermal dependence of biochemical reaction rates means that many animals regulate their body temperature so that fluctuations in body temperature are small compared to environmental temperature fluctuations. Thermoregulation is a complex process that involves sensing of the environment, and subsequent processing of the environmental information. We suggest that the physiological mechanisms that facilitate thermoregulation transcend phylogenetic boundaries. Reptiles are primarily used as model organisms for ecological and evolutionary research and, unlike in mammals, the physiological basis of many aspects in thermoregulation remains obscure. Here, we review recent research on regulation of body temperature, thermoreception, body temperature set-points, and cardiovascular control of heating and cooling in reptiles. The aim of this review is to place physiological thermoregulation of reptiles in a wider phylogenetic context. Future research on reptilian thermoregulation should focus on the pathways that connect peripheral sensing to central processing which will ultimately lead to the thermoregulatory response. The distribution in the thalamus of terminal projections from lamina I neurons of the trigeminal, cervical, and lumbosacral dorsal horn was investigated with the anterograde tracer Phaseolus vulgaris leucoagglutinin (PHA-L) in the cat. Iontophoretic injections were guided by single- and multi-unit physiological recordings. The injections in particular cases were essentially restricted to lamina I, whereas in others they spread across laminae I-III or laminae I-V. The trigemino- and spinothalamic (TSTT) terminations were identified immunohistochemically. In all cases, regardless of the level of the injections, terminal fibers were consistently distributed in three main locations: the submedial nucleus; the ventral aspect of the basal ventral medial nucleus and ventral posterior nuclei; and, the dorsomedial aspect of the ventral posterior medial nucleus. The terminal fields in the submedial nucleus and the ventral aspect of the ventral posterior group were topographically organized. Terminations along the ventral aspect of the ventral posterior group extended posterolaterally into the caudal part of the posterior nucleus and anteromedially into the ventromedial part of the ventral lateral nucleus. In several cases with trigeminal lamina I injections, a terminal labeling patch was observed within the core of the ventral posterior medial nucleus. In cases with spinal lamina I injections, terminations were also consistently found in the lateral habenula, the parafascicular nucleus, and the nucleus reuniens. Isolated terminal fibers were occasionally seen in the zona incerta, the dorsomedial hypothalamus, and other locations. These anatomical observations extend prior studies of TSTT projections and identify lamina I projection targets that are important for nociceptive, thermoreceptive, and homeostatic processing in the cat. The findings are consistent with evidence from physiological (single-unit and antidromic mapping) and behavioral studies. Summary 1. Why do larger predators generally consume larger prey items? Endemic pit-vipers (Gloydius shedaoensis ) on a small island in north-eastern China ambush passerine birds, usually from the branches of small trees. Both minimum and maximum prey sizes increase with predator size. 2. To clarify the reasons for this ontogenetic shift, 251 snakes in ambush postures were approached and offered potential prey items (dead birds, or models covered in feathers) to clarify the cues that trigger a foraging strike. 3. The snakes struck at the prey item in 101 of these trials, and this ‘decision’ was influenced by the size, movement and temperature of the prey item. A pit-viper's body size influenced its prey-size selectivity, with larger snakes refusing to strike at smaller prey items. Larger snakes also scavenge dead birds too large for smaller snakes to ingest, but do not ignore live birds: even the largest snakes use prey movement and prey temperature as cues to elicit feeding strikes. 4. The ontogenetic shift in prey size thus reflects a combination of processes. The absence of large prey from the diet of small snakes is due to gape-limitation (these snakes strike and attempt to swallow much larger prey). The absence of small prey items from the diet of larger snakes is due to active refusal to strike at small prey, as well as a behavioural shift to scavenging and to terrestrial rather than arboreal ambush-sites (and thus, higher rates of encounter with large prey items) by larger pit-vipers. At older ages, the circadian rhythm of body temperature shows a decreased amplitude, an advanced phase, and decreased stability. The present review evaluates to what extent these changes may result from age-related deficiencies at several levels of the thermoregulatory system, including thermoreception, thermogenesis and conservation, heat loss, and central regulation. Whereas some changes are related to the aging process per se, others appear to be secondary to other factors, for which the risk increases with aging, notably a decreased level of fitness and physical activity. Moreover, functional implications of the body temperature rhythm are discussed. For example, the relation between circadian rhythm and thermoregulation has hardly been investigated, while evidence showed that sleep quality is dependent on both aspects. It is proposed that the circadian rhythm in temperature in homeotherms should not be regarded as a leftover of ectothermy in early evolution, but appears to be of functional significance for physiology from the level of molecules to cognition. A new view on the functional significance of the circadian rhythm in peripheral vasodilation and the consequent out-of-phase rhythms in skin and core temperature is presented. It is unlikely that the strong, daily occurring, peripheral vasodilation primarily represents heat loss in response to a lowering of set point, since behavioral measures are simultaneously taken in order to prevent heat loss. Several indications rather point towards a supportive role in immunological host defense mechanisms. Given the functional significance of the temperature rhythm, research should focus on the feasibility and effectiveness of methods that can in principle be applied in order to enhance the weakened circadian temperature rhythm in the elderly. A noxious cold stimulus can evoke multiple sensations each occurring with a different time course. We have performed psychophysical studies to identify the time course of five sensations evoked by a noxious cold stimulus applied to the hand. Subjects continuously rated either pain, ache, cold, heat or prickle sensations throughout repeated presentations of a noxious cold stimulus (3°C) from a neutral (32°C) baseline. Separate runs were used to assess each of the five types of sensation. Cold was reported throughout the period of cooling. The time course of pain and ache sensations were similar. However, prickle and heat sensations had time courses that could be distinguished from each other, and from ache and pain. Identification of these temporal profiles could provide clues to their underlying mechanisms. The temporal dissociation of these sensations will also enable neuroimaging studies of the cortical mechanisms associated with these sensations. Thus our results constitute a first step toward identifying the distinct modes of neural activity associated with different types of pain sensation. [Copyright &y& Elsevier]

The calbindin-immunoreactivity of spinothalamic (STT) lamina I neurons and their ascending axons was examined in two experiments. In the first experiment, lamina I STT neurons in macaque monkeys were double-labeled for calbindin and for retrogradely transported WGA&ast;HRP following large (n=2) or small (n=1) injections that included the posterior thalamus. Most, but not all (78%) of the contralateral retrogradely labeled lamina I STT cells were positive for calbindin. Calbindin-immunoreactivity was not selectively associated with any particular anatomical type of lamina I STT cell; 82% of the fusiform cells, 78% of the pyramidal cells and 67% of the multipolar cells were double-labeled. In the second experiment, oblique transverse sections from upper cervical spinal segments of three macaque monkeys, one squirrel monkey and five humans were stained for calbindin-immunoreactivity. In each case, a distinct bundle of fibers was densely stained in the middle of the lateral funiculus. This matches the location of anterogradely labeled ascending lamina I axons observed in prior work in cats and monkeys, and it matches the location of the classically described ‘lateral spinothalamic tract’ in humans. This bundle had variable shape across cases, an observation that might have clinical significance. These findings support the view that lamina I STT neurons are involved in spinal cordotomies that reduce pain, temperature and itch sensations. [Copyright &y& Elsevier]

The article discusses a study of the possible involvement of G proteins in cold-sensory transduction in Paramecium multimicronucleatum (PM). Cells of PM were voltage-clamped and guanine nucleotide analogs were introduced into them. The results support the view that potassium channels, whose activities are regulated by calcium, are involved in the response to the cooling stimulus.

Antennal styloconic thermo-hygro sensilla of Antheraea were studied with DC-coupled transepithelial recordings. - The transepithelial voltage changed by about 2 mV · °C. The spike frequency of the cold cell reached 300 Hz at the onset of negative temperature steps, but only 30 Hz at static temperatures (as with metal electrodes). The cold cell spikes showed a brief afterhyperpolarization that increased with temperature. The spikes of the cold- and warm-stimulated cells facilitated each other at low temperature. Mechanical stimuli (push against the sensillum, hydrostatic pressure of < ± 50 kPa, ultrasonic vibrations ≈ 120 kHz) modified the responses of the cold- and the warm-stimulated cells. Latency of cold cell responses to ultrasonic stimulation was occasionally less than 3 ms. - The impulse frequencies of the warm and the cold cells depend on the temperature and the magnitude of temperature change. When the firing rate is high enough by either or both of these parameters, it can be forced still higher by application of clamp current (outside positive). The higher the firing rate prior to clamping, the greater the effect of the current. - By analogy with sensilla for other modalities, this relationship between frequency and clamp current strongly suggests that stimulus-dependent changes in the conductance of dendritic membranes control the excitation of the warm and cold cells. The sensillum coelocapitulum, a hygro- and thermoreceptive sensillum of the honey bee, Apis mellifera, was investigated by electron microscopy. The cuticular apparatus of the sensillum is a mushroomshaped protrusion, devoid of pores, set in a narrow cylindrical pit positioned centrally within a cuticular, shallow depression. There may be three or four receptor cells. Three receptor cells have unbranched sensory cilia, containing densely packed microtubules, which extend distally into the cuticular apparatus and completely fill its cavity. These connecting cilia are of the usual 9+0 type. The fourth receptor, if present, has a thin sensory cilium which terminates beneath the cuticular apparatus. Its connecting cilium has armed outer doublets. The outer cavity is formed by two enveloping cells and is completely sealed off. Lipid deposits are present within the cavity and the tormogen cell. The thecogen cell has scolopale rod-like structures around the inner cavity. Features common to the insect hygro- and thermoreceptive sensilla are discussed in comparison with those of other insects. Antennal sensilla of Leucophaea maderae were investigated electrophysiologically, labeled and then examined with the scanning or transmission electron microscope. The sensilla can be classified into morphological types according to their external shape and the structure of their hair wall. Sensilla showing similar reaction spectra of their cells can be cathegorized into physiological groups. The morphological classification corresponds to the physiological grouping: one morphological type of sensillum comprises one or several groups of physiologically similar sensilla. In many of these groups constant combinations of physiologically different cells occur. The possible functional significance of the relationships found between the structural features of the sensilla and the physiological properties of their sensory cells is discussed. Coeloconic sensilla on the antenna of Locusta migratoria were investigated electrophysiologically, labeled and then examined under the scanning or transmission electron microscope. They can be categorized into two main morphological types: 1) sensilla with wall pores and two concentric cuticular walls (wp-dw sensilla), and 2) sensilla lacking wall pores (np-sensilla). Hygroreceptors were observed only in np-sensilla. Olfactory receptors were observed in wp-dw sensilla, but not in np-sensilla. Cold-sensitive units were found in both types. Wp-dw sensilla with a cold-sensitive unit also manifest a fatty acid olfactory receptor. This cold-sensitive unit appears to be less sensitive to drops in temperature than the cold receptor combined with hygroreceptors in np-sensilla. Recordings from both types revealed up to three or sometimes four units that could be distinguished on the basis of impulse amplitude. The number of np-sensilla on each antennal segment ranges from 0 to 6. These sensilla are pegs 4-5 μm long with a terminal swelling. The dendritic outer segments of two sensory cells fit tightly into these pegs. The dendrite of the third sensory cell ends at its base. All three are up to 1 μm thick, unbranched, and densely filled with microtubules. The cuticular wall and socket of the peg are characterized by (1) an electron-dense inner layer surrounding the dendrites and continuous with electron dense clusters at the base of the peg, and (2) clefts in the cuticle at the insertion of the peg. The present observations favor the hypothesis that insect sensilla combining the triad of moist air, dry air and cold receptors generally lack pores. Specifical, though still alternative roles in stimulus transduction can be suggested for substructures demonstrated in the present and previous electron microscopic investigations. A sensillum in a narrow pit with a broad cuticular collar, located in a sensillum field on the 12th segment of the antennae of Carausius morosus, was investigated electrophysiologically. After marking, it was also examined with the transmission and the scanning electron microscopes. The number of sensory cells within the sensillum varies between three and four. One cell, present in half of the sensilla studied, exhibits a simple cilium of the 9×2+0 type as outer dendritic segment. The outer segment of a second unit is noteworthy in that it divides near its ciliary base into two branches. These flatten to form lamellae, then fold and wrap around each other. The remaining two sensory cells bear unbranched or bifurcate outer segments which contain densely packed microtubules. Only these outer segments extend into the cuticular peg; the others end beneath its base. The cuticular peg is devoid of pore systems. Electrophysiological recording yielded evidence that a cold, a dry and a moist air receptor are present. The fourth unit did not respond clearly to stimulation. [ABSTRACT FROM AUTHOR]

On the antennae of Periplaneta americana, 25 chemo-, hygro- or thermosensitive sensilla were investigated electrophysiologically and, after marking, by transmission and scanning electron microscopy. A clear-cut relationship of functional types to structural types was observed. Two different stimulus conducting structures were observed: a) pore tubules which are found only in smooth, single-walled sensory pegs and b) secretion-filled canals which occur only in grooved double-walled sensilla. Temperature and humidity-sensitive receptors occur only in double-walled sensilla with secretion material as the stimulus conducting system. Olfactory sensory cells were found in both types, however, those with a specific sensitivity for short-chain n-alcohols are restricted to single-walled pegs with pore tubules, while those which are most sensitive to short-chain n-acids and amines are found in double-walled sensilla, sometimes together with thermosensitive units. The stimulus conducting systems may control the access of odorous substances to the dendritic membranes and thus contribute to the discriminatory properties of the sensilla. [ABSTRACT FROM AUTHOR]

Dynamic properties of thermoresponsive units at the caudal trigeminal nucleus are not modified by heat-rearing. The modification of the thermoregulatory system by heat-rearing is unlikely to be a consequence of altered thermoreceptive function at this level. Rats were reared at ambient temperatures of 30°C (heat-reared) and 20°C (controls). Extracellular recordings of thermoresponsive units at the caudal trigeminal nucleus were made while the facial receptive fields of these thermoresponsive units were stimulated with 6 different rates of temperature change (+0.5, +0.1, +0.02, −0.02, −0.1 and −0.5°C/sec). All thermoresponsive units encountered were cold-sensitive. The static maxima were clustered around 13.5°C and did not differ between treatment groups. Dynamic maxima occurred at temperatures around 29-30°C and were not different in thermoresponsive neurones from the two treatment groups. There was no difference between the two treatment groups in the dynamic activity of trigeminal cold-responsive neurones at any of the 42 combinations of temperature and rate of temperature change examined.
