ABSTRACT: This study aims to identify candidate genes related to ovarian development after ovarian tissue transplantation through transcriptome sequencing (RNA-seq) and expression network analyses, as well as to provide a reference for determining the molecular mechanism of improving ovarian development following ovarian tissue transplantation. We collected ovarian tissues from 15 thirty-day-old ducks and split each ovary into 4 equal portions of comparable sizes before orthotopically transplanting them into 2-day-old ducks. Samples were collected on days 0 (untransplanted), 3, 6, and 9. The samples were paraffin sectioned and then subjected to Hematoxylin-Eosin (HE) staining and follicular counting. We extracted RNA from ovarian samples via the Trizol method to construct a transcriptome library, which was then sequenced by the Illumina Novaseq 6000 sequencing platform. The sequencing results were examined for differentially expressed genes (DEG) through gene ontology (GO) function and the Kyoto encyclopedia of genes and genomes (KEGG) pathway analyses, gene set enrichment analysis (GSEA), weighted correlation network analysis (WGCNA), and protein-protein interaction (PPI) networks. Some of the candidate genes were selected for verification using real-time fluorescence quantitative PCR (qRT-PCR). Histological analysis revealed a significant reduction in the number of morphologically normal follicles at 3, 6, and 9 d after ovarian transplantation, along with significantly higher abnormality rates (P < 0.05). The transcriptome analysis results revealed 2,114, 2,224, and 2,257 upregulated DEGs and 2,647, 2,883, and 2,665 downregulated DEGs at 3, 6, and 9 d after ovarian transplantation, respectively. Enrichment analysis revealed the involvement multiple pathways in inflammatory signaling, signal transduction, and cellular processes. Furthermore, WGCNA yielded 13 modules, with 10, 4, and 6 candidate genes mined at 3, 6 and 9 d after ovarian transplantation, respectively. Transcription factor (TF) prediction showed that STAT1 was the most important TF. Finally, the qRT-PCR verification results revealed that 12 candidate genes exhibited an expression trend consistent with sequencing data. In summary, significant differences were observed in the number of follicles in duck ovaries following ovarian transplantation. Candidate genes involved in ovarian vascular remodeling and proliferation were screened using RNA-Seq and WGCNA.

Human pluripotent stem cells (hPSCs) are a promising resource for the replacement of degenerated ventral midbrain dopaminergic (vmDA) neurons in Parkinson's disease. Despite recent advances in protocols for the in vitro generation of vmDA neurons, the asynchronous and heterogeneous nature of the differentiations results in transplants of surprisingly low vmDA neuron purity. As the field advances toward the clinic, it will be optimal, if not essential, to remove poorly specified and potentially proliferative cells from donor preparations to ensure safety and predictable efficacy. Here, we use two novel hPSC knock-in reporter lines expressing GFP under the LMX1A and PITX3 promoters, to selectively isolate vm progenitors and DA precursors, respectively. For each cell line, unsorted, GFP+, and GFP- cells were transplanted into male or female Parkinsonian rodents. Only rats receiving unsorted cells, LMX1A-eGFP+, or PITX3-eGFP- cell grafts showed improved motor function over 6 months. Postmortem analysis revealed small grafts from PITX3-eGFP+ cells, suggesting that these DA precursors were not compatible with cell survival and integration. In contrast, LMX1A-eGFP+ grafts were highly enriched for vmDA neurons, and importantly excluded expansive proliferative populations and serotonergic neurons. These LMX1A-eGFP+ progenitor grafts accelerated behavioral recovery and innervated developmentally appropriate forebrain targets, whereas LMX1A-eGFP- cell grafts failed to restore motor deficits, supported by increased fiber growth into nondopaminergic target nuclei. This is the first study to use an hPSC-derived reporter line to purify vm progenitors, resulting in improved safety, predictability of the graft composition, and enhanced motor function. Les patients présentant une maladie rénale chronique ont un risque infectieux accru qui peut être diminué par la vaccination. Cette susceptibilité aux infections peut être secondaire à la néphropathie comme, par exemple, dans le cadre du syndrome néphrotique, aux traitements utilisés, notamment immunosuppresseurs ou au traitement de suppléance. De plus, l'altération de la fonction rénale entraîne une diminution de la réponse vaccinale pouvant nécessiter une adaptation des schémas vaccinaux. Les traitements immunosuppresseurs couramment employés en néphrologie contre-indiquent l'utilisation de certains vaccins et peuvent impliquer la réalisation de sérologies préalables. Le but de cette synthèse est de proposer des pistes de calendrier vaccinal selon les caractéristiques des patients rencontrés en néphrologie et de leurs pathologies. This increased susceptibility to infection can be related to the nephropathy itself as observed in nephrotic syndrome, to the treatment especially in situations requiring immunosuppressive drugs or related to dialysis. Despite a less effective response to vaccination, some data emphasize similar benefits from immunization among people with CKD to the general population. However, some situations encountered in nephrology require adaptation of immunization practices. Objective： To explore the effect bone marrow mononuclear cells （BM-MNCs） transplantion on the ulcerative colitis in rats. Methods： BM-MNCs labeled with DAPI were transplanted into UC rat （model group） through caudal vein, the rats injected with the same amount of PBS were used as the control group. The pathological changes of colon tissues was observed by light microscope; and the planting and distributed conditions of the BM-MNCs labeled DAPI in colon tissues were detected by fluorescence microscope, the expressions of CK19 and CD34 in BM-MNCs were detected by immunofluorescence. Results： The neogenetic mucosal epithelium and gland submucous neogenetic and ripe granulation tissue were observed in the transplanting group, which were obviously better than those of the control group; DAPI labeled BM-MNCs cells can express CD34 and CK19. Conclusions： BM-MNCs can transfer and plant into the iniured colon tissues and differentiate into the vascular endothelial cell and mucosal epithelial cell. [ABSTRACT FROM AUTHOR]

This study tested the hypothesis that neuropeptide Y (NPY) and NPY receptors 1 (Y1) and 2 (Y2) participate in lung allograft rejection. Inflammation in grafts may include interaction between blood leukocytes and graft endothelial cells and marked accumulation of intravascular blood leukocytes. Fewer leukocytes accumulate in lung than in kidney allografts. Lung transplantion was performed in the Dark Agouti to Lewis rat strain combination. Intravascular and intraalveolar leukocytes were isolated from the grafts, and we evaluated the mRNA expression of NPY, Y1, and Y2 by real-time RT-PCR as well as the peptide expression of NPY by radioimmunoassay and immunohistochemistry. NPY and Y1 were expressed by pulmonary intravascular and intraalveolar leukocytes. Y1 was up-regulated by pulmonary intravascular and intraalveolar leukocytes during allograft rejection while Y2 could not be detected. Higher NPY expression levels in intravascular leukocytes were observed in lung compared to kidney allografts, which were investigated previously. Our findings suggest that an increased leukocytic expression of NPY in lung compared to kidney allografts results in a reduced accumulation of leukocytes in allograft vessels. [ABSTRACT FROM AUTHOR]

Objective To investigate the clinical effects of bone marrow mesenchymal stem cell ( BMSC) transplantation via different approaches in the treatment of liver cirrhosis in mice. Methods A total of 46 BALB/c mice were randomly divided into normal control group with 5 mice and liver cirrhosis model group with 41 mice. Subcutaneously injected carbon tetrachloride olive oil was used to establish the mouse model of liver cirrhosis. A total of 36 mice with liver cirrhosis were randomly divided into control group, caudal vein BMSC transplantation group, and spleen BMSC trailsplantation group, with 12 mice in each group. Whole bone marrow adherent culture was performed to obtain the third - generation BMSCs, and flow cytometry was used for cell surface identification. BMSCs were injected into the mice through the caudal vein or spleen. Blood samples were collected at 4 weeks after transplantation to measure liver function. HE and Masson staining and a - smooth muscle actin (a -SMA) immunohistochemistry were performed for liver sections. Liver injury and fibrosis in mice were examined. A one - way analysis of variance was used for comparison between groups. Results At 8 weeks after the establishment of the model, the mice in the model group had sparse and dark yellow hair, reduced food consumption and activity, and a reduction in body weight. After transplantation, compared with the model control group, the caudal vein BMSC transplantation group and spleen BMSC transplantation group showed a significant increase in albumin and significant reductions in alanine aminotransferase and aspartate aminotransferase (all P < 0.01 ). There were no significant differences between the two transplantation approaches (P >0. 05 ). After transplantation, there were significant changes in diseased tissue, alleviated liver cirrhosis, reduced collagen fiber and necrotic area, and a good structure. Immunohistochemistry showed both transplantation groups showed significant reductions in the number of cells with positive a - SMA. Conclusion BMSC transplantation via the tail vein or spleen can improve liver function and diseased tissue in mice with liver cirrhosis and these two approaches have comparable clinical effects. Estudo exploratório descritivo, com abordagem qualitativa cujo objetivo foi: conhecer as percepções dos pacientes em lista de espera para transplante renal. O local da pesquisa foi a Unidade de Hemodiálise de um hospital universitário de Porto Alegre. Os participantes foram os pacientes ativos na lista de espera para fazer o transplante renal, sendo a amostra constituída de 09 portadores de IRC, que realizam hemodiálise. Como resultados identificou-se as seguintes percepções: esperança, ansiedade, liberdade, ambivalência, medo, culpa e fé. A dependência da diálise para sobreviver remete o paciente ao confronto com a morte e a descrença, ao mesmo tempo em que busca força e fé para lutar e manter-se na espera por um doador.Estudio exploratorio descriptivo, con abordaje cualitativo cuyo objetivo fue: conocer las percepciones de los pacientes en lista de espera para transplante renal. La dependencia de la diálisis para supervivir remete al paciente al enfrentamiento con la muerte y la descreencia, al mismo tiempo en que busca fuerza y fe para luchar y mantenerse en la espera por un donador.A descriptive and exploratory study with a qualitative approach whose aim was to get to know the feelings of patients on the waiting list for a kidney transplant. The research was conducted at the Hemodialysis Unit of a university hospital of Porto Alegre. The participants were the active patients on the waiting list for a kidney transplant. The sample comprised 9 patients affected with chronic renal insufficiency who undergo hemodialysis. The following feelings have been identified: hope, anxiety, freedom, ambivalence, fear, guilt, and faith. Dependence on dialysis for survival causes patients to face death and disbelief at the same time that they search for strength and faith in order to fight and wait for a donor.

The American experience in organ transplantation illuminates the attitudes and practices that underlie the ongoing failure to establish a national health insurance system. In both spheres, the public makes a sharp divide between the well-being of intimates and the well-being of strangers. Private and public duties remain distinct; obligations recognized on a personal level do not expand public commitments. As a result, individuals must shift for themselves, whether they require an organ donor or a doctor. Girls and young women who require ovariectomy or cancer therapy may consider having their own eggs, embryos or ovarian tissue stored (cryopreserved) for their own future use. Ovarian tissue is simple to collect and contains large numbers of germ cells. Transplantation of fresh and frozen- thawed ovarian tissue in healthy sheep and mice has resulted in normal live young. Similar techniques may be effective in the human but it is unclear whether ovarian tissue cryopreservation and grafting is suitable for ovaries from individuals with cancer or infections. If cancer cells were present in an ovary at the time of collection and survived cryopreservation and grafting they could establish cancer in the recipient. We therefore performed ovarian cryopreservation and transplantation trials using a mouse lymphoma model. This established that the lymphoma was transmitted by grafts of both fresh and frozen ovarian tissue. The normal healthy recipient mice died 9–43 days after receiving a small piece (1 mm of ovarian tissue from a donor with lymphoma. We conclude that ovarian tissue which is collected, cryopreserved and grafted while it contains cancer cells has the potential to spread the cancer to the graft recipient. [ABSTRACT FROM PUBLISHER]

Objective: To observe the inhibitory effect of the Chinese herbal compound Guizhu capsule (CGZC) on lung cancer and explore the possible mechanism underlying its actions by evaluating its effects on the expression of phosphate and tension homology deleted on chromsome ten (PTEN) and murine double mimute 2 (MDM2) in lung cancer model mice., Methods: A mouse model of transplanted lung cancer was established by subcutaneous inoculation of cancer cells into the axillae of mice. Twenty-four hours later, the mice were weighed and randomly divided into the model control group, cisplatin group (DDP group), and high, moderate and low dosage CGZC groups, with ten mice in each group. The control mice received an equal volume of distilled water. DDP was intraperitoneally injected at 1 mg/kg in the DDP group, once a day for 3 days. CGZC diluted with distilled water was administered at 20 g/kg (10 times the clinical adult dosage) in the high-dose group, 10 g/kg (5 times the clinical adult dosage) in the moderate-dose group and 5 g/kg (2.5 times the clinical adult dosage) in the low-dose group once a day for 10 days. On the 11th day, the mice were weighed and killed. The tumor tissues were weighed and the tumor inhibition rate was calculated. PTEN and MDM2 protein expression were detected by immunohistochemical analysis of tumor tissues., Results: The CGZC high- and moderate-dose groups showed a significant inhibitory effect on tumor growth (P < 0.05); there was no significant difference between the high dose group and the DDP group (P > 0.05). The CGZC high-dose group also showed enhanced expression of PTEN protein (P < 0.01) and decreased expression of MDM2 protein (P < 0.01) in lung cancer cells of the mice. There was no significant difference between the high dose group and the DDP group., Conclusion: CGZC has a significant inhibitory effect on transplanted lung cancer in mice. The mechanism may involve reducing expression of PTEN and decreasing the expression of MDM2 in the lung cancer tissues of the mice.
