Abstract

Equine Chorionic Gonadotropin (eCG) previously named Pregnant Mare Serum Gonadotropin (PMSG) has been widely used since the 40s in animal reproduction control. It is extracted from the blood of pregnant mares between days 40 and 120 of gestation. Animal welfare organizations have voiced concerns against mares bleeding conditions. There is currently no effective substitute for the natural PMSG. In this review, we summarize the basic knowledge of the structure and biology of eCG, and the research on recombinant eCG production in the past five years.

18. Establishment of an in-house one-step real-time RT-PCR assay for detection of Zaire ebolavirus

Abstract

Ebola virus is a deadly causative agent with a high mortality rate of up to 90%, therefore it has been classified by the Center for Disease Control and Prevention (CDC) as a category A biological agent. The World Health Organization (WHO) recommended using RT-PCR based assays to rapidly detect the virus. In the present study, we established an in-house assay for detection of Zaire ebolavirus via real-time RT-PCR. The nucleotide sequence of the Zaire ebolavirus nucleoprotein (NP) gene was retrieved from the Genbank for designing primer pairs and probes using Primer Express 3.0 software. The RNA positive control was generated by in vitro RNA transcript synthesis. The optimal components in the 20 μl final volume of the real-time RT-PCR assay were 10 μl 2X QuantiTect Probe RT-PCR master mix, 0,6 μM of each primer, 0,1 μM of the probe, 0,2 μl RT mix and 5 μl of RNA template. The thermal cycle conditions were as follows: 50°C for 30 min, 95°C for 15 min, then 45 cycles of 15 s at 94°C, 60s at 60°C. The limit of detection of the assay was 100 copies/reaction and 1414 FFU/ml with the positive RNA panel and sample panel of RNA extracted from cell culture supernatants of cells infected with Zaire ebolavirus 2014/Gueckedou-C05, respectively. The specificity of this assay was 100% when tested with the positive RNA panel of Ebola virus and other haemorrhagic fever viruses. In conclusion, we successfully established an in-house real-time RT-PCR assay for detection of Zaire ebolavirus in Vietnam with a limit of detection of 1414 FFU/ml and specificity of 100%.

Abstract

Objective: this study aims to develop a methodology for detecting mutations in the K13 gene and determine mutation frequencies in clinical samples. Method: total DNAs were extracted blood samples collected from 50 patients in failure with artemisinin treatmentin the Central Highlands region. A fragment of the K13 gene was amplified, purified, and sequenced by the Sanger method. The K13 sequences were analysed by using Bioedit and aligned with the reference sequence to determine K13 mutations. Results: successfully amplified the K13 gene segment with size 799 bp in P. falciparum. After sequencing, there was a nucleotide substitution of A>G at position 1740, leading to changes in amino acids C>Y at the respective position 580. In the 50 patient samples, 41/50 (82%) samples showed the C580Y mutation, 9/50 (18%) of the samples had no mutation, and there was no other mutation. Conclusion: the authors have successfully developed and optimised a procedure for detecting the mutation C580Y in K13 in P. falciparum and determined the mutation frequency in the K13 gene.

Abstract

We did not record any complications intra-operation and post-operation, including: co-twin demise, bleeding in the amniotic cavity, PPROM within 24 hours, maternal infection, anesthetic complications, or maternal blood loss requiring transfusion. Firstly, because of the difference in gestational age of the surgical procedure, we performed surgery in the second trimester differently from some studies performed in the third trimester, increasing the risk of premature birth following the procedure. Abstract

Background Hepatitis E virus (HEV) infection can occur through consumption of undercooked pork meat or exposure to animal feces. Because there are scarce data only in developing countries, we assessed whether pigs might be a potential source of human HEV infections in Vietnam. In addition, we determined anti-HEV seroprevalences in the general population and in individuals professionally exposed to pigs and pork meat. Methods The study took place in Hanoi, Vietnam. Liver tissues from domestic pigs (n = 210) and serum samples obtained from individuals occupationally exposed to pigs and pork meat (n = 283) and from unexposed healthy controls (n = 168) were screened for HEV-ribonucleic acid (RNA) by reverse-transcription polymerase chain reaction. The exposed group was divided into pork meat vendors (n = 81), pig farmers (n = 96), and slaughterers (n = 106). Serum samples were subjected to HEV immunoglobulin (Ig)G and IgM enzyme-linked immunosorbent assays. The HEV genotypes were assessed by direct sequencing, followed by phylogenetic analyses. Results Hepatitis E virus seroprevalence was higher among persons occupationally exposed to pigs/pork meat compared with unexposed individuals (anti-HEV IgM 11% vs 6%, P = .07; anti-HEV IgG 53% vs 31%, P < .0001). Positivity of anti-HEV IgG among slaughterhouse staff was 66%, followed by 51% in pig-farmers and 38% in pork meat vendors (P = .00073). A similar trend was observed for IgM positivity. Of the pig liver tissues, 26 of 210 (12.4%) were positive for HEV-RNA and assessed to be HEV genotype 3. Conclusions Hepatitis E virus circulates in domestic pigs in Hanoi and constitutes a permanent zoonotic disease risk. The high HEV seroprevalence among occupationally exposed individuals indicates an associated risk of HEV infection.

Abstract

Hepatocellular carcinoma (HCC) is one of the most common malignant tumors, which is also often fatal. An early and accurate diagnosis is a decisive step towards the survival of the patients. Molecular biology improved significantly the prognosis of liver cancers through learned use of tumor markers like proteantigens, cytokines, enzymes, isoenzymes, circulating RNAs, gene mutations and methylations. Nevertheless, much improvement is still achievable and needed in this area, which is crucial in order to make an early diagnosis and monitor the progression of the disease. We present in this review what we believe to be the most relevant data regarding tissue and serum biomarkers related to HCC.

The complement receptor of the immunoglobulin superfamily (CRIg, encoded by the VSIG4 gene) is a macrophage receptor involved in the clearance of immune complexes and autologous cells. Abstract

Abstract Background Ficolin-2 coded by FCN2 gene is a soluble serum protein and an innate immune recognition element of the complement system. FCN2 gene polymorphisms reveal distinct geographical patterns and are documented to alter serum ficolin levels and modulate disease susceptibility. Methods We employed a real-time PCR based on Fluorescence Resonance Energy Transfer (FRET) method to genotype four functional SNPs including -986 G > A (#rs3124952), -602 G > A (#rs3124953), -4A > G (#rs17514136) and +6424 G > T (#rs7851696) in the ficolin-2 (FCN2) gene. We characterized the FCN2 variants in individuals representing Brazilian (n = 176), Nigerian (n = 180), Vietnamese (n = 172) and European Caucasian ethnicity (n = 165). Results We observed that the genotype distribution of three functional SNP variants (−986 G > A, -602 G > A and -4A > G) differ significantly between the populations investigated (p p Conclusions The observed distribution of the FCN2 functional SNP variants may likely contribute to altered serum ficolin levels and this may depend on the different disease settings in world populations.
