Supramolecular salicylic acid has the keratolytic and anti‐inflammatory properties, and zinc pyrithione is an antifungal drug delivered as a microparticle to skin epithelia.This study aimed to assess the efficacy and safety of 2% supramolecular salicylic acid (SSA) combined with 0.8% zinc pyrithione conditioner (SSA‐ZP) in treating scalp SD focusing on key outcome measures including sebum levels and scalp barrier function.Five patients with mild to moderate scalp SD were included in the 2‐week self‐controlled prospective study, receiving daily SSA‐ZP treatment. Evaluation on days 0, 7, and 14 included dermoscopy, the adherent scalp flaking scale (ASFS), sebum level, transepidermal water loss (TEWL), pH, and stratum corneum hydration. We also performed the fungal count across specific scalp regions, such as the left and right sides of the forehead, the top of the head, and the occiput.Five patients with mild to moderate scalp SD participated in this study. After 2 weeks of SSA‐ZP treatment, significant reductions in ASFS scores, sebum levels, and fungal count were observed, alongside improvements in TEWL and pH values across multiple scalp regions. Moreover, there was no difference in the hydration of stratum corneum.SSA‐ZP demonstrated efficacy in treating scalp SD without adverse effects, suggesting its potential as a first‐line treatment option. In order to explore the composition, pH and concentration changes of the antifouling agent zinc pyrithione (ZPT) during the service process of zinc pyrithione-containing self-polishing antifouling coating (ZPT-SPC), and clarify the antifouling mechanism of ZPT-SPC during service, SEM/EDS were employed to explore the changes in cross-sectional micromorphology and composition of coatings before and after polishing. Moreover, the open circuit potential-pH experiment was used to measure the changes in the micro-potential-pH information of paint scratches. The changes in ZPT composition and content on the paint surface during service were explored through Raman spectroscopy. SEM/EDS results showed that the surface became denser, and the Zn element content decreased after polishing. Notably, the Zn content was much greater than the Cu content, which indicated that he ZPT content was much greater than the Cu2O content. In addition, the open circuit potential-pH measurement results showed that the pH value of the ZPT-SPC scratch interface generally exhibited a trend of first decreasing and then increasing, and eventually approached the original pH value. Raman results showed that the ZPT slowly diffused to the outside, which reduced the internal concentration, and the ZPT component transfer occurred inside the antifouling coating, resulting in a more uniform distribution of components. Furthermore, this process could be characterized macroscopically by antifouling agent concentration-color gray correlation model.

Newly emerging roles of LRRC8 volume-regulated anion channels (VRAC) raise important questions about the therapeutic potential of VRAC in the treatment of epilepsy, type 2 diabetes, and other human diseases. A critical barrier to evaluating whether VRAC represents a viable drug target is the lack of potent and specific small-molecule inhibitors and activators of the channel. Here we review recent progress in developing the molecular pharmacology of VRAC made by screening a library of FDA-approved drugs for novel channel modulators. We discuss the discovery and characterization of cysteinyl leukotriene receptor antagonists Pranlukast and Zafirlukast as novel VRAC inhibitors, and zinc pyrithione (ZPT), which apparently activates VRAC through a reactive oxygen species (ROS)-dependent mechanism. These ongoing efforts set the stage for developing a pharmacological toolkit for probing the integrative physiology, molecular pharmacology, and therapeutic potential of VRAC.

Nowadays, regarding the important interest to hygienic conditions and human comfort and health, in this study, we were particularly focused on developing functional cotton for safety shoes insoles worn as personal protective equipment (PPE). In fact, numerous persons are exposed to several shoes problems such as bad odors caused by bacteria and many diseases due to foot infections. For this purpose, an AgNPs free antibacterial ecofriendly treatment respecting Oekotex standard 100 and Detox Campaign using zinc pyrithione compound was realized based on an experimental design to optimize simultaneously the zinc fixation rate under an exhaustion process and the antibacterial efficiency. The fixed zinc pyrithione content was analyzed by ICP-MS spectrometry as a reference method and a spectrophotometric method as an alternative, more straightforward, and cost-effective method. A good correlation between the two methods was established. Then, the impact of zinc pyrithione and zinc content on antibacterial effects was investigated. As a result, 173 mg/kg of fixed zinc obtained by applying only 0.14% of pure zinc pyrithione with the optimized exhaustion process showed a significant antibacterial effect. Furthermore, 0.28% of pure zinc pyrithione ensured an excellent fastness to abrasion and sweat of the antibacterial treated cotton fabric tested against bacteria Escherichia coli, Staphylococcus aureus and Staphylococcus Epidermidis.

Background: Tinea versicolor is a common superficial fungal infection of the skin with various clinical manifestations. This review aims to familiarize physicians with the clinical features, diagnosis and management of tinea versicolor. Methods: A search was conducted in July 2022 in PubMed Clinical Queries using the key terms “tinea versicolor” OR “pityriasis versicolor”. The search strategy included all clinical trials, observational studies and reviews published within the past 10 years. Results: Tinea versicolor is caused by Malassezia species, notably M. globosa, M. furfur and M. sympodialis. The condition is characterized by scaly hypopigmented or hyperpigmented macules/patches, primarily located on the upper trunk, neck and upper arms. The diagnosis is usually based on characteristic clinical features. If necessary, a potassium hydroxide preparation test can be performed to reveal numerous short, stubby hyphae intermixed with clusters of spores. Most patients with tinea versicolor respond to topical antifungal therapy, which has a better safety profile (fewer adverse events, fewer drug interactions) and lower cost compared to systemic treatment and is therefore the treatment of choice. Oral antifungal therapy is typically reserved for patients with extensive disease, frequent recurrences or disease that is refractory to topical therapy. Advantages of oral antifungal therapy include increased patient compliance, shorter duration of treatment, increased convenience, less time involved with therapy and reduced recurrence rates. On the other hand, oral antifungal therapy is associated with higher cost, greater adverse events and potential drug–drug interactions and is therefore not the first-line treatment for tinea versicolor. Long-term intermittent prophylactic therapy should be considered for patients with frequent recurrence of the disease. Conclusion: Selection of antifungal agents depends on several factors, including efficacy, safety, local availability, ease of administration, likelihood of compliance and potential drug interactions of the antifungal agent.

[Display omitted] Seborrheic dermatitis (SD) is a common dermatological disorder with symptoms that include skin flaking, erythema and pruritus. This review discusses the topical products available for treating SD, which target several aspects of disease pathobiology, including cutaneous microbial dysbiosis (driven by Malassezia yeast), inflammation, sebum production and skin barrier disruption. Among the various treatments available, zinc pyrithione (ZnPT) based products that exhibit anti-fungal action are the market leaders. A skin compartment approach is presented here for combining ZnPT exposure information with threshold levels for anti-fungal efficacy and toxicity, overall providing a comprehensive picture of ZnPT therapeutics and safety. While Malassezia yeast on the surface are effectively targeted, yeast residing beyond the superficial follicle may not receive adequate ZnPT for anti-fungal effect forming the basis for skin re-colonisation. Levels entering systemic circulation from topical delivery are well below toxic thresholds, however the elevated zinc levels within the viable epidermis warrants further investigation. Strategies to improve formulation design can be broadly classified as influencing 1) topical delivery, 2) therapeutic bioactivity, 3) skin mildness, and 4) sensory attributes. Background. Atopic dermatitis is a multifactorial di­sease in which the microbial products of Malassezia yeast may play a certain role in triggering the disease. Topical application of zinc pyrithione has an antifungal effect and can improve the course of disease. The aim of this study was to evaluate the effectiveness of zinc pyrithione in the treatment of atopic dermatitis in children by conducting a randomized clinical trial using zinc pyrithione as an adjunct therapy. Materials and methods. Sixty-two patients with atopic dermatitis were included in the study and randomly divided into two groups: 31 people received only traditional therapy, 31 — additional topical zinc pyrithione in the form of 0.2% cream. Disease severity was evaluated using the dynamics of SCORAD (SCORing Atopic Dermatitis) indicators. Specific IgE to Malassezia was analyzed in 31 patients of the main group. Results. The improvement on the SCORAD was 45 % (T-W=496.0, p < 0.001) in the group of children who received zinc pyrithione in addition to traditional therapy, in the control group — 35.2 % (T-W = 496.0, p < 0.001). Children receiving zinc pyrithione locally had statistically significantly better dynamics on the SCORAD than controls: the average value of ΔSCORAD in the main group was 17.9, in the control group — 12.6 (W = 782.5, p = 0.006). Specific IgE to Ma­lassezia mixture (m227) was evaluated in 31 children of the main group, among them 38.7 % had a positive result: 3 patients had class 1 sensitization; 1 — class 2; 4 — class 3; 4 children — class 5 of sensitization. Sensitization to Malassezia had not any impact on the effectiveness of treatment (W = 179.0, p = 0.597). Conclusions. This study demonstrated the effectiveness of a 3-week course of using zinc pyrithione in the treatment of atopic dermatitis as an adjunct therapy. Since the pathogenetic mechanism and interaction between Malassezia spp. and atopic dermatitis remain partially unclear, there is still a need for further research to understand the exact role of these organisms in the disease course and the role of antifungal drugs in the treatment of atopic dermatitis.

Aims: Although zinc pyrithione (ZPT) has been studied as topical antimicrobial and cosmetic consumer products, little is known about its pharmacological actions in gastrointestinal (GI) health and inflammation. Our aims were to investigate the effects of ZPT on transient receptor potential (TRP) channels and Ca 2+ signaling in intestinal epithelial cells (IECs) and its therapeutic potential for colitis., Main Methods: Digital Ca 2+ imaging and patch-clamp electrophysiology were performed on human colonic epithelial cells (HCoEpiC) and rat small intestinal epithelial cells (IEC-6). The transcription levels of proinflammatory cytokines such as IL-1β were detected by RTq-PCR. Dextran sulfate sodium (DSS) was used to induce colitis in mice., Key Findings: ZPT dose-dependently induced Ca 2+ signaling and membrane currents in IECs, which were attenuated by selective blockers of transient receptor potential ankyrin 1 (TRPA1) and transient receptor potential vanilloid 4 (TRPV4) channels, respectively. Interestingly, Ca 2+ entry via TRPA1 channels inhibited the activity of TRPV4 channels in HCoEpiC, but not vice versa. ZPT significantly promoted migration of IECs by activating TRPA1 and TRPV4 channels. ZPT reversed lipopolysaccharides (LPS)-induced changes in mRNA expression of TRPA1 and TRPV4. Moreover, ZPT decreased mRNA levels of pro-inflammatory factors promoted by LPS in HCoEpiC, which were restored by selective TRPA1 blocker. In whole animal studies in vivo, ZPT significantly ameliorated DSS-induced body weight loss, colon shortening and increases in stool score, serum calprotectin and lactic acid (LD) in mouse model of colitis., Significance: ZPT exerts anti-colitic action likely by anti-inflammation and pro-mucosal healing through TRP channels in IECs. Background: Seborrheic dermatitis is a common scalp condition affecting the quality of life of individuals across all age groups. The uninhibited proliferation of Malassezia spp. and enhanced sebaceous gland activity often leads to scalp flaking, mild erythema, and itching, thereby worsening the situation. Here, we aimed to study the efficacy of an antiseptic liquid as a pre-shampoo rinse followed by a non-anti-dandruff shampoo as a dandruff care strategy.  Methods: The anti-dandruff efficacy of a chlorhexidine-cetrimide-based antiseptic liquid (ASL) as a pre-shampoo scalp rinse, followed by a regular non-anti-dandruff shampoo, was compared with a commercial Zinc pyrithione (ZnPTO) based anti-dandruff shampoo following a half-head paired treatment design in a randomized, placebo-controlled, double-blind trial. The study was conducted on 50 healthy human adults of both genders with moderate to severe dandruff. During the 12 weeks of this study, the product's safety and efficacy were evaluated based on the dermatologist's visual assessment, the subjects' self-assessment, the loose flake density score, and hair fall., Results: ASL as a pre-shampoo scalp rinse demonstrated significant reduction (p<0.001 for n=47) in dandruff and itching after the treatment and the regression phase, performing at par with a marketed anti-dandruff shampoo. Moreover, ASL was established to be safe, non-irritant, and well-tolerated. Introduction: Dandruff is a common chronic condition that has a negative impact on the quality of life. Many years of experience with zinc pyrithione (ZPT) as an active ingredient led to an improved technology for administration to the scalp. Methods: The severity of dandruff was measured using the Adherent Scalp Flaking Score on a scale of 0 to 80 units by qualified assessors. Bacterial skin diseases of livestock could be a serious global threat, especially in association with overcoming bacterial resistance. Combinatory action of antimicrobial agents proves to be an effective strategy to overcome the problem of increasing antibiotic resistance of microorganisms. In this study, the in vitro combined effect of zinc pyrithione with gentamicin against bacterial skin pathogens of livestock (Staphylococcus aureus, Streptococcus agalactiae, and Streptococcus dysgalactiae) was evaluated according to the sum of fractional inhibitory concentration indices (FICI) obtained by checkerboard method. The results showed that a combination of zinc pyrithione with gentamicin produced a strong synergistic effect (p < 0.001) against all tested streptococcal strains (with FICI values ranging from 0.20 to 0.42). Compared to that, only three out of eight S. aureus strains were highly susceptible to the combination of antimicrobial agents at single concentration (0.25 µg/mL) of zinc pyrithione with range of FICI 0.35–0.43. These findings suggest that interference between agents tested in this study can be used for the development of future veterinary pharmaceutical preparations for the treatment of bacterial skin infections of livestock. Seed coating is receiving increased relevance in sustainable agriculture, to reduce the usage of pesticides and protect the natural environment. In this study, a new formulation based on the combination of two active constituents (zinc pyrithione and triclosan) and an inactive component (carboxymethyl cellulose polymer) was optimized for seed coating to control seed-borne pathogens. According to the in vitro antimicrobial tests, the combination of zinc pyrithione and triclosan showed a broader spectrum of phytopathogenic bacteria and fungi control. Coated seeds from different plant species with 2% of zinc pyrithione, 0.05% of triclosan and, 0.5% of carboxymethyl cellulose did not show any microorganisms development on culture media, contrarily to the noncoated ones. Germination quality and seedling growth of treated groups were enhanced in the presence of natural seed-borne pathogens and not affected in pathogen-free seeds compared to the nontreated groups. Thus, this novel seed coating formulation is a promising antimicrobial agent that could be used in the prevention of seed-borne diseases without any side effect on the seedling establishment. Zinc pyrithione (ZnPT) is one of the components used in antifouling paints and can be an alternative to classical toxic chemicals such as organotin. However, there is still remarkable concern about the environmental safeness of ZnPT due to rapid transchelation and degradation into several metabolites that have their own toxicity. The effect after acute exposure of ZnPT is investigated on Mediterranean mussels exposed to 20 and 40 μg/L concentrations for 48 and 96 h and antioxidant responses [superoxide dismutase (SOD), and reduced glutathione (GSH)], genotoxicity [micronuclei (MN) frequency], apoptotic and histological changes were determined. Severe histological changes in hepatopancreas and gill tissues of mussels were observed in ZnPT exposed groups due to dose-dependent increase. ZnPT also caused a dose-dependent increase of TUNEL-positive cell count in the mussel tissues, especially in the hepatopancreas. Increasing in SOD activities and decreasing in GSH levels in both ZnPT concentrations compared to the control were observed. MN and binuclei numbers in all exposure groups were significantly increased. The results of the present study demonstrate that acute exposure to ZnPT could cause an adverse effect on mussel tissues at especially higher concentrations. [ABSTRACT FROM AUTHOR]

Zinc Pyrithione (ZPT), a Food and Drug Administration (FDA) approved chemical, is widely used for topical antimicrobials and cosmetic consumer products, including anti-dandruff shampoos. ZPT and its degraded byproducts have detected in large quantities in the environment, and identified to pose healthy risks on aquatic organisms and human. However, so far, knowledge about ZPT effects on female reproduction, particularly oocyte maturation and quality, is limited. Herein, we investigated the adverse impact of ZPT on mouse oocyte maturation and quality in vitro and found exposure to ZPT significantly compromises oocyte maturation. The results revealed that ZPT disturbed the meiotic cell cycle by impairing cytoskeletal dynamics, kinetochore-microtubule attachment (K-MT), and causing spindle assembly checkpoints (SAC) continuous activation. Further, we observed the microtubule-organizing centers (MTOCs) associated proteins p-MAPK and Aurora-A were disrupted in ZPT-treated oocytes, signified by decreased expression and abnormal localization, responsible for the severe cytoskeletal defects. In addition, ZPT exposure induced a significant increase in the levels of H3K9me2, H3K9me3, H3K27me1, and H3K27me3, suggesting the alterations of epigenetic modifications. Moreover, the accumulation of zinc ions (Zn2+) was observed in ZPT-treated oocytes, which was detrimental because overmuch intracellular Zn2+ disrupted oocyte meiosis. Finally, these above alterations impaired spindle organization and chromosome alignment in metaphase-II (MII) oocytes, indicative of damaged oocytes quality. In conclusion, ZPT exposure influenced oocyte maturation and quality via involvement in MTOCs-associated proteins mediated spindle defects, altered epigenetic modifications and zinc accumulation.

Psoriasis is a chronic immune-mediated inflammatory disease with predominant skin lesions. External therapy is an important area of treatment for such patients. Non-steroidal anti-inflammatory drugs based on zinc pyrithione do not have side effects inherent in the glucocorticosteroid drug and can be an alternative to them. Evaluation of the effectiveness of drugs is not always convenient in humans. The use of laboratory models allows us to study changes in the clinical, histological and immunohistochemical characteristics of the skin during topical therapy. Goal. On laboratory models of psoriasis, conduct a comparative assessment of the anti-inflammatory effectiveness of Skin-cap in comparison with other drugs. Materials and methods. In the experiment, 50 Balb / c inbred mice were used, randomized to 5 groups. The formation of a model of psoriasis by listening to them, after which I perform euthanasia with a skin biopsy in mice. Group II mice are treated with activated zinc pyrithione cream (Skin-cap), group III - zinc pyrithione cream (Zinocap), group IV betamethasone dipropionate cream (Akriderm) 2 p/d 14 days. Group V was monitored without therapy. The Clinical PASI Index (mPASI) score is given daily. On the 29 day of studies, group II-V mice undergo euthanasia and skin biopsies, which were examined histologically (epidermal thickness, severe inflammatory infiltrate) and immunohistochemical methods (Ki-67, CD3). The differences were considered significant at p0.05. Results. The initial mPASI index value was 7.2 [5.29.3]. The mPASI index in the Skin-cap group was 1.8 [1.13.2], zinc pyrithione 2.9 [2.14.2] and betamethasone dipropionate 1.3 [1.0] 3.1], which is significantly lower than in the group without therapy 5.8 [5.06.8] (p 0.05). A histological examination in mice receiving zinc pyrithione-activated showed a more pronounced decrease in the thickness of the epidermis (2.7 times) than with pyrithione zinc (2 times) and, immunohistochemical analysis showed the most significant decrease in the proliferative activity of the epidermis and the number of T-lymphocytes when using betamethasone and zinc pyrithione activated. Conclusions: it was found that the use of the drug leads to the suppression of the inflammatory process in the skin, comparable in effectiveness with a strong topical glucocorticosteroid. Skin-cap has no atrophogenic effect, which allows us to recommend this drug for the treatment of psoriasis patients with any localization of rashes.

Zinc pyrithione (ZPT) is widely used as an antimicrobial. Zinc is a necessary trace element of the human whose homeostasis associated with several cancers. However, the anticancer effect of increased Zinc in ovarian cancer is still unclear. This study focussed on the anti-tumour effects of ZPT combined with Zinc in SKOV3 and SKOV3/DDP cells. The cell viability, apoptosis, migration, and invasion assays were detected by CCK-8, flow cytometry, wound healing and transwell assay, respectively. The distribution of Zinc in cells was monitored by staining of Zinc fluorescent dye and lysosome tracker. The changes in lysosomal membrane stability were reflected by acridine orange fluorescence and cathepsin D reposition. Expression of the proteins about invasion and apoptosis was evaluated by western blot. The results indicated that ZPT combined with Zinc could notably reduce cell viability, inhibit migration and invasion in SKOV3 and SKOV3/DDP cells. Besides, ZPT performed as a Zinc carrier targeted lysosomes, caused the increase of its membrane permeability and the release of cathepsin D accompanied by mitochondrial apoptosis in SKOV3/DDP cells. In conclusion, our work suggests that ZPT combined with Zinc could inhibit proliferation, migration, invasion, and promote apoptosis by trigger the lysosome-mitochondrial apoptosis pathway in ovarian carcinoma.

G. mellonella larvae were inoculated with M. globosa on abraded cuticular surface. Then, integument cell viability, histological changes, and fungal burden were evaluated. Results. Larvae inoculated with M. globosa showed higher lesion melanization and tissue damage. In addition, M. globosa population showed to increase over time. Concerning the shampoo's effectiveness, both formulations significantly reduced M. globosa burden and tissue damage. Conclusion. G. mellonella larvae were allowed to evaluate M. globosa superficial infection and antifungal effectiveness. The broad spectrum antimicrobial/antifungal zinc pyrithione (ZnPT) is used in products ranging from antifouling paint to antidandruff shampoo. The hazard profile of ZnPT was established based upon comprehensive toxicological testing, and products containing this biocide have been safely used for years. The purpose of this study was to create a dermal physiologically based pharmacokinetic (PBPK) model for ZnPT in the rat for improving dose–response analysis of ZnPT-induced toxicity where reversible hindlimb weakness was the endpoint used as the basis for ZnPT risk assessments. Previously, we developed a PBPK model which simulated the kinetics of pyrithione (PT) and its major metabolites 2-(methylsulfonyl)pyridine and S-glucuronide conjugates in blood and tissues of rats following oral ZnPT administration. The dermal model was optimized utilizing in vitro dermal penetration investigations conducted with rat skin and with historical data from a dermal repeat dose study using rats. The model replicated the observed temporal patterns and elimination kinetics of [14C]PT equivalents in blood and urine during and following repeated dermal dosing and replicated the observed dose-dependencies of absorption, blood [14C]PT equivalents and plasma PT concentrations. The model provided internal dosimetry predictions for a benchmark dose analysis of hindlimb weakness in rats that combined dermal, gavage and dietary studies into a single internal dose–response model with area-under-the-curve (AUC) for plasma PT, the toxic moiety in the rat, as the internal dose metric. Two polypropylene HVAC electret filters: a regular filter and an antimicrobial filter containing zinc pyrithione (ZPT), were compared for filtration performance. The study was conducted over 7 months in realistic conditions with semi-urban outdoor air. Several parameters were monitored over the study period: the average temperature was about 20 °C and relative humidity about 60%, the average inlet concentration of cultivable microorganisms was 50 CFU m−3, the average inlet concentration of particles was 10 μg m−3, the filter pressure drop increased moderately by about 30 Pa, and the particle collection efficiency of soda fluorescein (median diameter 0.35 μm) decreased in the first half of the study period by about 30% and then stabilized. The microbial concentration on the filters was quantified every 2 months using an innovative methodology based on media coupons in conjunction with microorganism quantification by CFU counting, with 5 culture media favorable to bacteria and/or fungi growth. The microbial concentrations on the filters were between 100 and 2000 CFU cm−2. The antimicrobial effect of zinc pyrithione was confirmed by the fungi cultivated with DRBC agar: no effects in the level of filter clogging were revealed in the range studied. Chromophoric dissolved organic matter (CDOM) is the main photosensitizer in seawater. CDOM can absorb solar radiation extensively and generate reactive intermediates (RIs), including singlet oxygen (1O 2), hydroxyl radicals (HO·), and CDOM triplet states (3CDOM*), that are favorable for indirect photodegradation of organic pollutants. In the current study, the effect and mechanism of CDOM components on zinc pyrithione (ZnPT) indirect photodegradation were studied. Among the three CDOM isolates (humic acid (JKHA), Suwannee River humic acid (SRHA), and Suwannee River natural organic matter (SRNOM)), JKHA had the highest fluorescence intensity per gram of carbon and the fastest indirect photodegradation rate for ZnPT. Furthermore, the fluorescent components were divided into four terrestrial humic-like components: C1, C2, C3, and C4, by using excitation–emission matrix spectroscopy combined with parallel factor analysis. A notable relationship was found between the four fluorescent components of CDOM and ZnPT indirect photodegradation. The indirect photodegradation rate of ZnPT varied in different types of CDOM solutions, which may be attributed to the different compositions of CDOM. The relative contents of C1 and C4 were significantly higher in JKHA than in the other two CDOMs, and they were the major contributors to the production of 3CDOM* and 1O 2 , which promoted ZnPT indirect photodegradation. The impact of seawater factors on ZnPT indirect photodegradation was also investigated. The ZnPT indirect photodegradation rate initially decreased and then increased as the pH increased from 5.00 to 11.00. The lowest rate of indirect photodegradation of ZnPT was observed at pH = 9.00. As the salinity was <25‰, in salinity the ZnPT indirect photodegradation rate increased concomitantly with an increase, and pronounced inhibition of ZnPT indirect photodegradation was observed when the salinity was >25‰. Nitrate had a promoting influence on ZnPT indirect photodegradation, whereas HCO 3 −, halogen ions, and metal ions all inhibited it. Bacterial skin diseases of livestock could be a serious global threat, especially in association with overcoming bacterial resistance. Combinatory action of antimicrobial agents proves to be an effective strategy to overcome the problem of increasing antibiotic resistance of microorganisms. In this study, the in vitro combined effect of zinc pyrithione with gentamicin against bacterial skin pathogens of livestock (Staphylococcus aureus, Streptococcus agalactiae, and Streptococcus dysgalactiae) was evaluated according to the sum of fractional inhibitory concentration indices (FICI) obtained by checkerboard method. The results showed that a combination of zinc pyrithione with gentamicin produced a strong synergistic effect (p < 0.001) against all tested streptococcal strains (with FICI values ranging from 0.20 to 0.42). Compared to that, only three out of eight S. aureus strains were highly susceptible to the combination of antimicrobial agents at single concentration (0.25 µg/mL) of zinc pyrithione with range of FICI 0.35–0.43. These findings suggest that interference between agents tested in this study can be used for the development of future veterinary pharmaceutical preparations for the treatment of bacterial skin infections of livestock.

Malassezia restricta and Malassezia globosa are lipid dependent commensal yeasts associated with dandruff. Antifungal actives such as zinc pyrithione are commonly used in antidandruff shampoos, although their efficacy is not clearly demonstrated. In this study, we assessed the efficacy of antifungal treatments on scalp Malassezia via a combination of culturomic and genomic detection methods. Zinc pyrithione inhibited Malassezia growth at low minimum inhibitory concentrations (MICs). In a longitudinal pilot study, quantitative polymerase chain reaction (qPCR) analysis showed a decrease in M. restricta on the scalp after zinc pyrithione treatment. These findings validate the antifungal efficacy of zinc pyrithione as a dandruff treatment. Lay Abstract Malassezia yeasts are associated with dandruff and seborrheic dermatitis. Zinc pyrithione is an organometallic compound with antimicrobial activity used in many industrial products. In this study, cytotoxicity (cell viability, lactate dehydrogenase release, proliferation rate, and mitotic index) and genotoxicity (sister chromatid exchange and micronucleus test) of zinc pyrithione on human lymphocytes were determined. Intracellular zinc concentrations were determined by inductively coupled plasma-optical emission spectrometry. Intracellular antioxidant/oxidant status was evaluated by total antioxidant capacity and total oxidative status assays. Its irritation potential was investigated using the hen's egg test chorioallantoic membrane model. Up to the highest concentration of 1000 nmol/L, ZnPT did not cause genotoxicity and did not change the proliferation index. Above 500 nmol/L, zinc pyrithione caused an increase in lactate dehydrogenase, oxidative stress, and intracellular zinc levels, while decreasing cell viability and mitotic index level. At 1000 nmol/L, zinc pyrithione caused slight irritation. These results suggest that zinc pyrithione may exert toxic effects via increased oxidative stress on human cellular systems. The review is devoted to the pathogenetic substantiation of the use of the activated zinc pyrithione agent in atopic dermatitis. The main mechanisms of formation of the pathological process in atopic dermatitis are explained in more details: hereditary predisposition, violations of the barrier function of the skin and immunological dysfunction leading to the development and persistence of chronic inflammation. The role of zinc ions in the pathogenesis of atopic dermatitis is discussed. The article presents the detailed analysis of the main clinical studies of the activated zinc pyrithione agent in atopic dermatitis, its positive effect on such symptoms as itching, skin manifestations, skin microbioma and severity of the disease course.

Zinc pyrithione (ZPT) is the antimicrobial substance and has been applied for various fields, including non-fouling coating paints and cosmetics. It also used for the tub part of the electronic machine with polypropylene (PP). Since the released ZPT can cause skeletal deformity or abnormality to coastal organisms, it is essential to quality check of ZPT in PP for environmental safety. However, as far as we knew, no previous reports about the analysis of ZPT in PP samples have been published; hence, we have developed a simple preparation method via extraction and precipitation. As assume that ZPT is just entrapped in the hydrocarbon chain of PP, ZPT was drawn out from the composites and homogeneously existed in the solution after untying the chain only precipitate PP with an organic solvent. Zinc pyrithione (ZnPT) and copper pyrithione (CuPT) are widely used antifouling booster biocides in the shipping industry as a replacement for the banned tributyltin (TBT). Sub-lethal effects of zinc pyrithione (ZnPT; 10 μg L−1), copper pyrithione (CuPT; 10 μg L−1) and combined copper–zinc pyrithione (CZPT; 5 + 5 and 10 + 10 μg. L−1) to freshwater indicator mussel Unio crassus (5.49 ± 0.85 cm in size) were determined by investigation of the total hemocyte counts, biochemical analysis mussel tissues and histopathological observations after 48-h and 7-d exposures. Mean total hemocyte counts differed significantly among all groups, for both exposure durations. In the 7 d, lipid peroxidation levels (malondialdehyde, MDA nmol g−1 wet tissue) and reduced glutathione levels (GSH μmol g−1 wet tissue) were lower than 48 h. However, there were no significant differences among groups. Total protein levels (mg mL−1) were approximately in the same range and differed only among 7-d exposure groups. The gill tissues showed hemocytic infiltration and lipofuscin like structures, while digestive gland showed loss of digestive tubules in the treatments of pyrithione compounds. Histopathological damages were most severe in the combined exposure groups. In conclusion, ZnPT, CuPT and combined CZPT5/10 are highly toxic on a freshwater indicator mussel species U. crassus. Since the ban of tributyltin in antifouling paints, many alternative biocides have been introduced to prevent settlement and growth of marine organisms on ship hulls. Zinc pyrithione (ZnPT) is one of the most frequently used alternative biocides in antifouling paints. This paper reviewed the overall chemical properties, toxicological characteristics, and environmental fates of ZnPT, as well as the analytical challenges of studying pertinent processes. ZnPT is generally toxic to a wide range of marine organisms, including algae, bivalves, sea urchins, polychaetes, crustaceans, and fish, typically at μg/L levels. ZnPT can be transchelated into other compounds in the presence of metal ions, and photodegrades when exposed to UV light. ZnPT is also reported to be biodegraded or hydrolyzed forming several metabolites of their own toxicity and stability. However, ZnPT accumulates in the water column or sediment, if it does not degrade at certain environmental conditions. To determine potential risks caused by ZnPT in the marine environment, studies have evaluated the environmental distribution of ZnPT with various chromatographic or voltammetry methods. Unfortunately, rapid transchelation and degradation of ZnPT in both the marine environment and laboratory interfered with most of the methods employed, making it difficult to evaluate its environmental distribution. More robust and sensitive analytical methods need to be developed to reliably describe the environmental release and distribution of ZnPT. To comprehensively understand the risk posed by the input of ZnPT into the marine environment, total degradation processes and its potential products also need to be adequately addressed. The purpose of the present study was to evaluate the antimicrobial potential of styrene-ethylene/butylene-styrene based thermoplastic elastomers (TPE) incorporated with zinc pyrithione (ZnPT) and silver nanoparticles (AgNano). Japan Industrial Standard was applied to evaluate the antimicrobial potential of incorporated TPE compounds against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). Antifungal action was evaluated against Aspergillus niger, Candida albicans and Cladosporium cladosporioides. Samples prepared with ZnPT eliminated 99.9% of the E. coli and 99.7 % of the S. aureus population, and presented an inhibition zone in the fungal assay. Samples prepared with AgNano eliminated 99.7% of the E. coli and 95.5 % of the S. aureus population. There was no inhibition zone in samples containing AgNano; however, these samples did not present fungal growth on their surfaces. TPE samples containing ZnPT showed biocidal activity against the microorganisms tested and can be used to develop antimicrobial products.

In this study, potential immunological and hematological effects of different concentrations (0, 1, 10, and 50 μg L-l) of waterborne zinc pyrithione (ZnPT) were studied in the blood of the olive flounder Paralichthys olivaceus over 30 days. Reduced alternative complement activity (ACH50) and lysozyme activity were measured in fish exposed to 10 and/or 50 μg L-l of ZnPT for 20 days. Decreased levels of total Ig were also observed in response to 10 and/or 50 μg L-l ZnPT during the exposure period. Levels of cortisol, a marker of stress, were significantly increased by 10 and 50 μg L-l ZnPT from day 10, and by 1 μg L-l exposure on day 30. The levels of red blood cells (RBCs) and white blood cells (WBCs) decreased following exposure to 10 and/or 50 μg L-l ZnPT, while no significant change was observed in hemoglobin level. Concentrations of total protein and albumin were significantly reduced with 50 μg L-l ZnPT at day 20. Alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase activities were significantly increased following exposure to 10 and/or 50 μg L-l ZnPT. Lipid peroxidation was induced by ZnPT, and higher concentrations (10 and 50 μg L-l) significantly increased intracellular malondialdehyde levels during exposure. Regarding the subsequent antioxidant response, intracellular glutathione levels increased significantly in response to 10 and 50 μg L-l ZnPT on days 20 and 30. Similarly, catalase and superoxide dismutase activity was significantly increased in response to 10 and 50 μg L-l ZnPT after day 10. Nowadays there are various groups of biocidal chemical agents, which can be used in diverse areas, such as personal hygiene, disinfection, antiparasitic action, and also in antifouling mixtures or paints. The versatility and efficacy of some of these agents favors their use and ultimate release into the aquatic environment, where they may still exert toxic activity. Zinc pyrithione is classified as a metal biocide with bactericidal, algicidal, and fungicidal actions. It has been formulated in antifouling paints, which prevent the formation of biofilms in submerged structures, and has also been used for dermocosmetic purposes, in shampoos for the treatment of dandruff and seborrhea. Some of the uses of zinc pyrithione are responsible for its direct release as flakes that reach the bottom sediments, especially in estuarine areas. Considering this fate, the ecotoxicity assessment of its effects towards sediment organisms, namely Polychaetous species, is extremely important. The present study characterized the acute potential toxicity of zinc pyrithione in terms of parameters of oxidative stress (catalase, glutathione S-transferases (GSTs), and thiobarbituric acid reactive substances (TBARS)), and neurotoxicity (acetylcholinesterase) which were evaluated in individuals of the polychaete Hediste diversicolor. Regarding the results obtained, only the activity of GSTs and AChE was significantly altered in relation to non-exposed animals. This set of results indicates that oxidative stress did not occur. Objective: Sex‐related differences in skin properties may be expected to impact dandruff formation and treatment. A meta‐analysis approach was undertaken to investigate potential differences between males and females in response to zinc pyrithione (ZnPT) treatment vs. non‐anti‐dandruff (AD) shampoo. A separate pooled statistical analysis of ceramide and total protein loss endpoints was also undertaken to assess potential sex‐related differences in stratum corneum properties that might influence response to ZnPT vs. non‐AD shampoo in subjects with dandruff. Methods: The meta‐analysis approach included data from 17 half‐head, double‐blind, randomized studies (N = 2088) undertaken in Asia to assess the effectiveness of 1% ZnPT shampoo and/or non‐AD shampoo in reducing dandruff severity, as assessed by Total Weighted Head Score Adherent Flake (TWHS‐AF) methodology. Treatment duration was 4 weeks, with TWHS‐AF measured at weekly intervals. Data from an additional three studies (N = 143) conducted in Asia were included in the pooled analysis of ceramide levels and protein loss from scalp skin of subjects with dandruff. Results: Response to 1% ZnPT vs. non‐AD shampoo was greater in males than in females at all time points; after 4 weeks, the between‐treatment difference in TWHS‐AF was −17.5 (95% confidence interval [CI] −19.5, −15.5) in males and −11.1 (−13.2, −8.9) in females. Sex‐related differences were observed between males and females in response to both 1% ZnPT and non‐AD shampoos. Males had a stronger response than females to treatment with 1% ZnPT shampoo, while dandruff decreased to a greater extent in females than in males when using non‐AD shampoo. Statistically significant sex‐related differences in ceramide levels and total protein loss were observed (both P < 0.01). Ceramide levels were 0.76 times lower (95% CI 0.60, 0.97) in males than in females, while total protein loss was 1.4 times greater (95% CI 1.1, 1.9) in males than in females. Conclusion: Males show a greater response than females to 1% ZnPT shampoo, while females show a greater response than males to non‐AD shampoo. The SOS response is a conserved response to DNA damage that is found in Gram-negative and Gram-positive bacteria. When DNA damage is sustained and severe, activation of error-prone DNA polymerases can induce a higher mutation rate than is normally observed, which is called the SOS mutator phenotype or hypermutation. We previously showed that zinc blocked the hypermutation response induced by quinolone antibiotics and mitomycin C in Escherichia coli and Klebsiella pneumoniae. In this study, we demonstrate that zinc blocks the SOS-induced development of chloramphenicol resistance in Enterobacter cloacae. Zinc also blocked the transfer of an extended spectrum beta-lactamase (ESBL) gene from Enterobacter to a susceptible E. coli strain. A zinc ionophore, zinc pyrithione, was ~100-fold more potent than zinc salts in inhibition of ciprofloxacin-induced hypermutation in E. cloacae. Other divalent metals, such as iron and manganese, failed to inhibit these responses. Electrophoretic mobility shift assays (EMSAs) revealed that zinc, but not iron or manganese, blocked the ability of the E. coli RecA protein to bind to single-stranded DNA, an important early step in the recognition of DNA damage in enteric bacteria. Zinc pyrithione (ZPT) is widely used as a substitute booster biocide for tributyltin and is also an additive to antidandruff shampoos and medical cosmetic products. ZPT and pyrithione have been detected in different environmental matrices and biota, suggesting that it may pose health threats to aquatic organisms and even humans. The present study used HepG2 cells, a human hepatoma cell line, to study the hepatotoxicity of ZPT (0.1–5.0 μM). ZPT treatment caused marked viability reduction and induced apoptosis depending on its dose used. ZPT-induced apoptosis involved an increased Bax/Bcl-2 ratio, loss of mitochondrial membrane potential, cytochrome c release, and enhanced caspase-9/-3 activity. In addition, a significant elevation in the amount of zinc ions and oxidative stress was evident. The involvement of these in ZPT-induced apoptosis was confirmed by toxicity comparison with analogs of ZPT and the observation that pretreatment with antioxidants afforded protection. Overall, these results suggest that ZPT induces zinc accumulation, oxidative stress, and subsequent apoptosis by causing mitochondrial dysfunction. A simple, rapid, and sensitive liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the determination of zinc pyrithione (ZnPT) and pyrithione (PT) in shampoos. The method consisted of a liquid-liquid extraction for sample preparation. The mass spectrometer was operated in multiple reaction monitoring (MRM) mode via the positive electrospray ionization interface. A linear regression (weighted 1/x) was used to fit calibration curves over the concentration range of 50-2000 ng/mL for both ZnPT and PT. Excellent linearity (r2 ≥ 0.9996) was achieved for all. The method was validated and found to be accurate (95.9-108.2% for ZnPT and 94.9-110.4% for PT), precise, and selective. Analytes in shampoos were found to be stable in the autosampler (6 °C for 6 h), in room temperature (for 6 h), and after three freeze-thaw cycles, and recovery of analytes was reproducible (90.8-94.6% for ZnPT and 90.2-96.3% for PT). Antimicrobial activity of Zinc oxide partially coated with silver nanoparticles produced by Flame Spray Pyrolysis (FSP) as an additive to include in waterborne paints with promising applications in areas as schools, clinics or hospital environments and in food industry equipment and facilities has been demonstrated. Due to the current increase of multi-resistant microorganisms, the results obtained in this research would enable an enhanced sanitary conditions to avoid health problems of professionals and improve the Hazard Analysis Critical Control Point (HACCP) in the Food Industry like a way to control the biofilm formation on surfaces and beyond. The nanomaterial was prepared by the use of Flame Spray Pyrolysis process obtaining nanoparticles of about 35 nm coated with ca. 5% (w/w) of silver (Ag). Antimicrobial trials showed that these ZnO-Ag nanoparticles has a potential effect against different bacteria ( Staphylococcus aureus , Pseudomona spp, Salmonella spp, Bacillus subtilis , Listeria monocytogenes ) and molds as Aspergillus niger . Furthermore, these nanoparticles were evaluated as an antimicrobial waterborne paints additive, and formulations with 0,15% (w/w) showed >5 mm inhibition zone against the main bacteria and molds tested. The mechanisms of sublethal toxicity of the antifouling biocide, zinc pyrithione (ZnPT), have not been well-studied. This investigation demonstrates that 14-d sublethal exposure to ZnPT (0.2 or 2 μM, alongside inorganic Zn and sea water controls) is genotoxic to mussel haemocytes but suggests that this is not caused by oxidative DNA damage as no significant induction of oxidised purines was detected by Fpg-modified comet assay. More ecologically relevant endpoints, including decreased clearance rate (CR), cessation of attachment and decreased tolerance of stress on stress (SoS), also showed significant response to ZnPT exposure. Our integrated approach was underpinned by molecular analyses (qRT-PCR of stress-related genes, 2D gel electrophoresis of proteins) that indicated ZnPT causes a decrease in phosphoenolpyruvate carboxykinase (PEPCK) expression in mussel digestive glands, and that metallothionein genes are upregulated; PEPCK downregulation suggests that altered energy metabolism may also be related to the effects of ZnPT. Significant relationships were found between % tail DNA (comet assay) and all higher level responses (CR, attachment, SoS) in addition to PEPCK expression.
