Abstract

OBJECTIVE: To establish the method for mass concentration of lobaplatin in human plasma and hydrothorax by HPLC. The method was used for patients with lung cancer, and could provide basis for the clinical therapeutic drug monitoring (TDM). METHODS: Human plasma and hydrotliorax samples were centrifuged and extracted with acetonitrile. The Inertsil-ODS-SP column (250 mm x4. 6 mm, 5 μm) was used, the mobile phase were consisted of phosphate buffer (pH =6. 5) and acetonitrile (containing 0. 03% triethylamine) (V : V= 96 : 4) and flushed at a flow rate of 1 0 ml/min, the wavelength was 215 nm. RESULTS: The calibration curves were linear over a concentration range of 0. 1 μg/ml-80. 0 μg/ml in the hydrothorax and 0. 1 μg/ml-50. 0 μg/ml in the plasma respectively. The limit of detection (LOD) and limit of quantification (LOQ) was 0. 1 μgml and 0. 5 μgml in hydrotiiorax and plasma. CONCLUSIONS: A rapid HPLCmethod to detect the concentration of lobaplatin in human plasma and hydrothorax was established. Themethod is sensitive, stable with good reproducibility, selectivity and high accuracy, and could be successfully used for the determination of lobaplatin concentration in human plasma and hydrothorax so as to conduct the therapeutic drug monitoring and provide individualized therapy for lung cancer patients.
