Abstract

This study aimed to determine the distribution, morphology and staining characteristics of mast cells in the lower respiratory tract (upper and lower trachea, primary bronchus, lung) of sheep (ovis Aries). Toluidine Blue (TB) (pH 0.5), Thionin (pH 0.3), Alcian Blue (pH 0.3) / Safranin O (pH 1.0) (AB / SO) and Berberine sulfate staining methods were applied to count and demonstrate the mast cells. The numerical density of mast cells in all the slides was determined by means of an ocular micrometer. The obtained data were evaluated in a semiquantitative manner. It has been found that mucosal mast cells (MMCs) generally outnumbered the connective tissue mast cells (CTMCs) in all the studied regions. TB, Thionin, and AB / SO and Berberine sulfate staining revealed that the mast cell density was the largest in lung and primary bronchus. Mast cells were found to be in the spindle, oval, round or amorphous-shaped and in varying sized. Mast cells stained metachromatically with Thionin and Toluidine Blue. In the AB/SO staining, mast cells stained red with SO (+), blue with AB (+), and purple with AB/SO (+) in all the examined regions. Mast cells showed yellow fluorescence Berberine sulfate staining. In conclusion, it is thought that mast cells may contribute to distribution and heterogeneity, allergic diseases of respiratory tract, cancer diagnosis and treatment, pathological studies.
