Abstract
- Objective To observe the drug resistance of human epithelial ovarian cancer cell line SKOV3/DDP added with reversal dose of mifepristone (MIF). Methods The proliferation inh伽tion rate ofM IF on SKOV3 and SKOV3/ DDP cell in logarithmic growth phase were detected byM TT method, and we selected the noncytotoxic dose ofM IF as reversal dose. The resistance index (RI) of SKOV3/DDP cells to cisplatin (DDP) was further measured by MTT. SKOV3/ DDP cells were distributed into the group A and group B. C ells in the group A were added with DDP, group B with DDP and 5 μmoVLM IF. The IC 50 of SKOV3/DDP cells was measured byM TT at 24, 48, and 72 h, respectively, and then we calculated the reversal fold (RF). The cell cycle of SKOV3/DDP was detected by flow cytometry (FCM). Results MTT assay results showed that mifepristone inhibited the proliferation of SKOV3 and SKOV3/DDP cells in a dose-dependent manner. The reversal dose ofM IF on SKOV3/DDP was 5 μmoVL. The IC50 of SKOV3 and SKOV3/DDP to DDP were 3. 22 and 14. 83 μg/ml, respectively, and the RI of SKOV3/DDP to DDP was 4. 61. The IC50 of group A was (23. 13 土 0. 83), (14. 83 土0.52), (10. 07 土0.56) μg/mL at 24, 48 and 72 h after culture, and (17. 06 土0.53), (7.77 土 0.47) and (4.06 土 0.69) μg/mL in the group B, respectively, and the RF ofM IF to SKOV3/DDP were 1. 36, 1. 91, and 2. 48, respectively. After 24, 48, and 72 hours of culture, the proportion of SKOV3/DDP cells added with 5 μmol/L MIF in the G0/G1 phase increased gradually (48. 7%, 53. 9%, 55. 1 %, and 59. 0%), while the proportion of cells in the S phase decreased gradually (13. 5%, 9. 6%, 7. 7%, and 5. 8%). C onclusion MIF (5 μmol/L) could inhibit the proliferation of SKOV3/DDP cells and reduce the resistance of SKOV3/DDP cells to DDP, and the possible mechanism might be that MIF can block the cell cycle of SKOV3/DDP in the G0/G¹ phase [ABSTRACT FROM AUTHOR]
