Properties of the cellulase and xylanase enzyme complexes of Trichoderma harzianum UzCF-28.

Abstract

The properties of cellulase and xylanase enzyme complexes formed in the culture fluid of T. harzianum UzCF-28 local fungal strain selected by screening from cellulose-degrading micromycetes have been studied. These enzymes showed their maximum activities in the pH range of 4.0-9.0: at pH 5.0 endo-1,4-β-glucanase and xylanase had maximum activities of 3.8 and 11.5 units /ml, respectively, while exo-1,4-β-glucanase - 2,92 units/ml at pH 5.5. 50°C was found to be the optimum temperature for these enzymes. Incubation time for endo-1,4-β-glucanase and xylanase enzymes was 30 min, while it was 60 min for exo-1,4-β-glucanase enzyme. Optimum concentration of substrates for endo-1-4-β-glucanase and xylanase enzymes was 1%, while it was equal to 50 mg for exo-1,4-β-glucanase (filter paper). At 60°C the activities of exo-1,4-β-glucanase and endo-1,4-β-glucanase enzymes decreased sharply and began to inactivate.
