Abstract

Objective To investigate the humoral and cellular immunity effects of liposome lyophilized powder prepared from different dilutions of rabies vaccine solution, to screen for the optimal dilution. Methods The rabies vaccine solution was diluted to 40 IU/mL, 30 IU/mL, 25 IU/mL, 20 IU/mL, 15 IU/mL, 10 IU/mL and 5IU/ml respectively then made with freeze- thaw- drying method into the vaccine liposome freeze- dried powder. Mice were immunized by intraperitoneal injection after reconstitution. MTT assay was used to detect the proliferation of splenic lymphocytes and ELISA assay was used to detect mice body fluid concentration of RV- IgG, to screen for the optimal dilution. Re s ults Compared with the solution group (P<0.05), when diluted to 15 IU/mL or less, there is a higher stimulus index SI (1.3313 ± 0.1082), which can stimulate the body to produce a higher level of cellular immunity and is capable of stimulating the body to produce more RV- IgG antibody (31.84 ± 2.09 IU/mL) in the early immunization. Conclusions Rabies vaccine liposome prepared by dilution of vaccine stocks to 15 IU/ml provides the best cellular and humoral immunity. [ABSTRACT FROM AUTHOR]
