Ketoconazole increases atovaquone exposure following concomitant administration with Malarone® in healthy subjects.

With an aim to assess the pharmacokinetics of atovaquone, 10 healthy subjects were administered Malarone® alone and in combination with ketoconazole in a crossover study. The results indicated 68% higher Cmax, 74% higher AUC and 34% lower CL/F of atovaquone following co-administration with ketoconazole compared to administration of atovaquone alone.
