Abstract

In order to solve the problem of early field inspection of porcine epidemic diarrhea, the experiment established a colloidal gold immunochromatographic method for the detection of porcine epidemic diarrhea virus (PEDV) in pig feces using a new kind of colloidal golds prepared via 3-hydroxymethyl aminomethane (Tris) assisted reduction. The preparation conditions including labeled pH and protein concentration was optimized. The sensitivity, specificity and accuracy of the established method were finally evaluated. The results showed that the established PEDV immunochromatography method could detect PEDV in pig feces within 15 min, and the minimum detection limit was 1×102 TCID50/mL. No cross reaction with porcine infectious gastroenteritis virus, rotavirus, Salmonella and other common pathogens was observed. In addition, there was no significant difference between intra-batch and inter-batch detection results. The coincidence rate of 107 clinical samples and RT-PCR was 100%. The study indicates that the established PEDV immunochromatographic strip based on the new colloidal gold marker had high sensitivity, strong specificity, good repeatability, with simple and quick operation, and is suitable for the rapid diagnosis of PED in primary farms.
