Abstract
- The study was to study the effect of prolactin receptor (PRLR) gene SNPs polymorphism on reproductive performance in Shenxian pigs. The SNPs locus of PRLR gene was detected by PCR-RFLP technique combined with mixed pool generation sequencing technology, and association with the reproductive performance of 71 multiparous sows of shenxian pigs was analyzed. The results showed that there were two SNPs loci of PRLR gene in Shenxian pig population: A133T and T225C, both located in exon 8. There were three genotypes at A133T: AA genotype, AT genotype and TT genotype, in which AA genotype was the dominant genotype and A is the dominant allele. The total litter size, litter weight and live litter size of AA genotypes were significantly higher than those of TT genotypes (P<0.05). There were three genotypes at T225C: TT, TC and CC, in which C was the dominant allele and CC was the dominant genotype. The total litter size and litter weight of TC and CC genotypes were significantly higher than those of TT genotypes (P<0.05). Among the combined genotypes, ATTC and ATCC genotypes had higher reproductive performance as a whole. The two loci were in Hardy-Weinberg equilibrium. The polymorphism information content was 0.329 and 0.373, respectively. The heterozygosity was 0.416 and 0.495, and the homozygosity was 0.584 and 0.505, respectively. The study indicates that the reproductive performance of Shenxian pigs can be improved by selecting individuals with AA genotypes at A133T site and TC genotypes at T225C loci, and combining individuals with ATTC and ATCC genotypes.
