Abstract

【Objective】The present paper aimed to study the localization and expression pattern of histone deacetylases(HDACS) family members HDAC3 and HDAC8 in growing follicles of Tan sheep, in order to provide reference for further research on the epigenetics mechanism of follicular development and oocyte maturation regulation in Tan sheep. 【Method】The ovarian tissues of healthy tan sheep were collected and the following growth follicles were mechanically separated: Small antral follicles(diameter 0.5-3.0 mm), large antral follicles(diameter 3.0-5.0 mm) and preovulatory follicles(diameter>5.0 mm). The localization and expression of HDAC3 and HDAC8 in different follicles were detected by immunohistochemistry and immunofluorescence respectively. At the same time, the expression of HDAC3 and HDAC8 in the follicles of Tan sheep was detected by Western blotting.【Result】HDAC3 was mainly located in the granular cells of primitive follicles, primary follicles and secondary follicles, granular cells of antral follicles and cumulus cells of Tan sheep, and its expression was up-regulated with follicular development. The localization of HDAC8 in growing follicles was different from that of HDAC3, which was not only distributed in the oocyte and granulosa cytoplasm of primordial follicles, primary follicles and secondary follicles, it was also localized in the cytoplasm of oocytes, granulosa cells and cumulus cells with antral follicles.【Conclusion】These results indicated that the localization and expression of HDAC3 and HDAC8 in growing follicles of Tan sheep were different, suggesting that HDAC3 and HDAC8 may be involved in the regulation of follicular development and oocyte maturation in different ways.
