高脂饮食诱导肥胖大鼠睾丸组织 circRNA-miRNA-mRNA三元转录网络分析.

Abstract

Objective To investigate the differential expressions of circRNAs (circRNAs) and microRNAs (miRNAs) in testicular tissues of high-fat diet-induced rat models with obesity・ Methods Rat models with obesity were induced and established by high-fat diet. The histopathological changes of their testicular tissue were observed by HE staining and microscope・ Their sperm concentration, motility, DFI and CMA3 + were respectively measured. Differential expressions of circRNAs and miRNAs in testicular tissues of rat models with obesity were analyzed by high-throughput sequence・ The main biological processes of the identified differential circRNAs and miRNAs were obtained by GO and KEGG analysis. Finally, circRNA-miRNA-mRNA network was constructed. Results Significant changes in testicular tissue structure of rat models with obesity were observed compared with the normal controls. Sperm concentration and motility were obviously decreased (P <0.05), whereas DFI and CMA3 + were significantly increased (P < 0. 05)・ Total of 32 differential circRNAs and 4 differential miRNAs were identified, and these differential cicRNAs and miRNAs may be associated with obesity-cuased male reproductive disorders through Wnt signaling pathway, TNF signaling pathway and insulin secretion* CircRNAs and miRNAs with significant differences and targeting relationships were as follows: novel_circ_0003226/mo-niiR-3544, novel_ circ_0015575/mo-miR-9a-5p, and circRNA-miRNA-mRNA networks were finally constnicted. Conclusion There are differentially expressed circRNAs and miRNAs in testicular tissues of obese rats, and these difierential genes may regulate obesity-caused reproductive disorders through circRNA-miRNA-mRNA network.
