Abstract

• Classification and excellent properties of PONs for tumor theranostics are highlighted. • Promising strategies for enhanced tumor monotherapy are presented. • PONs-based combined therapies and imaging-guided tumor therapies are summarized. • Potential challenges and clinical perspectives for tumor theranostics are outlooked. Cancer nanomedicines offer the effective solutions to overcome the difficulties of traditional cancer therapies. Porous organic nanoplatforms (PONs) are anticipated to be an optimal choice for tumor therapy due to their distinctive properties, including self-versatility, controllable pore size, high loading capacity, ease of functionalized modification, morphological diversity, metal-free composition as well as superior biodegradability and biocompatibility. In this review, we initially provide an overview of the classification and excellent properties of PONs. Then, with a specific focus on their applications in tumor monotherapy, combined therapy and theranostics, as well as the advancements in therapeutic optimization strategies. Lastly, we succinctly summarize the current challenges and future directions of PONs within this field to provide valuable references for further investigations. [ABSTRACT FROM AUTHOR]
