Rapid Monitoring of Aquatic Organism Biodiversity Based on Environmental DNA in a Medium-Sized Reservoir.

Abstract

Environmental DNA (eDNA) has emerged as a highly sensitive and efficient tool for the biomonitoring of aquatic ecosystems. In this study, we investigated fish and benthic species communities using eDNA techniques in a medium-sized reservoir (about 3 square kilometers) in Anhui, China. A total of 12 water samples and 11 sediment samples were analyzed by 12S and 18S primers, respectively. We analyzed the composition of species diversity and the effect of seven environmental factors using the Mantel test. A total of 42 fish taxa were present in the water samples, and 188 benthic taxa in the sediment samples. Species composition was different in disparate stations. We found that water temperature and salinity are pivotal factors influencing the composition of fish communities, while chlorophyll-a is a primary environmental determinant for benthic species assembly structures across different zones. Biodiversity information generated by eDNA techniques can be used to reflect the resource status of this reservoir.
