Abstract

The article presents a study on the overproduction, purification and biochemical characterization of 7-dimethylallyltryptophan synthase from Aspergillus fumigatus. It focuses on the identification of Afu3g12930, a putative prenyltransferase gene from the genome sequence of Aspergillus fumigatus. The coding sequence was cloned and the overexpression in Escherichia coli was apparent with the soluble protein purified in homogeneity. The enzyme catalyzed the prenylation of Trp at the indole moiety in the presence of dimethylallyl diphosphate (DMAPP). The roles of the putative genes in the cluster are explained.
