Pimaric acid reduces vasoconstriction via BK Ca channel activation and VDCC inhibition in rat pulmonary arterial smooth muscles.

Abstract

Pulmonary vessels play a pivotal role in oxygen circulation. We previously demonstrated that pimaric acid (PiMA) activated large-conductance Ca <superscript>2+</superscript> -activated K <superscript>+</superscript> (BK <subscript>Ca</subscript> ) channels and inhibited voltage-dependent Ca <superscript>2+</superscript> channels (VDCCs). In the present study, PiMA attenuated vasoconstriction induced by high K <superscript>+</superscript> or endothelin-1 in rat pulmonary arterial smooth muscles (PASMs). PiMA also reduced high K <superscript>+</superscript> -induced cytosolic [Ca <superscript>2+</superscript> ] increase in PASM cells. PiMA increased BK <subscript>Ca</subscript> currents and decreased VDCC currents. BK <subscript>Ca</subscript> channels and VDCCs were formed by the α/β <subscript>1</subscript> and α <subscript>1C</subscript> /α <subscript>1D</subscript> /β <subscript>2</subscript> /β <subscript>3</subscript> subunits, respectively. These results indicate that PiMA induces vasorelaxation through the dual effects of BK <subscript>Ca</subscript> channel activation and VDCC inhibition in PASMs.<br />Competing Interests: Declaration of competing interest The authors indicated no potential conflicts of interest.<br /> (Copyright © 2023 The Authors.
