Experiment of the Effect of Arsenic Trioxide Plus Amlodipine on the Proliferation and Apoptosis of Human Liver Cancer HepG-2 Cell in Vitro.

Abstract
- Objective: To explore the effect of arsenic trioxide plus amlodipine on the proliferation and apoptosis of human liver cancer HepG2 cell line in vitro. Methods: Different concentrations of arsenic trioxide (4.0, 2.0, 1.0 μmol/L) and amlodipine (27*#215; 10³, 18× 10³, 12× 10³ mg/mL) were treated on human hepatoma HepG-2 cells in vitro, the changes of cell morphology were observed, CCK-8 method and flow cytometry were used to test the proliferation and apoptosis of HepG-2 cell respectively. Results: Arsenic trioxide and amlodipine significantly increased the growth inhibition rate and apoptotic rate ofHepG2 cells in a concentration dependent manner, and the combination of two drugs was more significant(P<0.05). Compared with the arsenic trioxide and amlodipine alone, combinati on of two drugs significantly blocked the HepG2 cells in phase G2 and S. Conclusion: Arsenic trioxide and amlodipine could significantly inhibit the proliferation and apoptosis of human liver cancer HepG2 cell line in vitro, which had a synergistic effect by combination.
