Dual action of palmitoyl trifluoromethyl ketone (PACOCF3) on Ca2+ signaling: activation of extracellular Ca2+ influx and alteration of ATP- and bradykinin-induced Ca2+ responses in Madin Darby canine kidney cells

Abstract

The effect of the phospholipase A<subscript>2</subscript> inhibitor palmitoyl trifluoromethyl ketone (PACOCF<subscript>3</subscript>) on Ca<superscript>2+</superscript> signaling in Madin Darby canine kidney (MDCK) cells was examined using fura-2 as the fluorescent Ca<superscript>2+</superscript> indicator. At a concentration of 20 µM, PACOCF<subscript>3</subscript> did not change basal cytosolic free calcium concentrations ([Ca<superscript>2+</superscript>]<subscript>i</subscript>), but at concentrations of 50–250 µM PACOCF<subscript>3</subscript> induced an increase in [Ca<superscript>2+</superscript>]<subscript>i</subscript> by activating extracellular Ca<superscript>2+</superscript> entry which was partly suppressed by 50 µM La<superscript>3+</superscript>. The effect of PACOCF<subscript>3</subscript> was abolished by removal of extracellular Ca<superscript>2+</superscript>. PACOCF<subscript>3</subscript> (10 µM) enhanced both the peak value and the area under the curve of the [Ca<superscript>2+</superscript>]<subscript>i</subscript> increase induced by 10 µM ATP and 1 µM bradykinin by potentiating extracellular Ca<superscript>2+</superscript> influx without affecting internal Ca<superscript>2+</superscript> release. Several other phospholipase A<subscript>2</subscript> inhibitors had no effect on basal [Ca<superscript>2+</superscript>]<subscript>i</subscript> or agonist-induced [Ca<superscript>2+</superscript>]<subscript>i</subscript> increases. Collectively, the results suggest that PACOCF<subscript>3</subscript> alters Ca<superscript>2+</superscript> signaling in renal tubular cells in a manner independent of phospholipase A<subscript>2</subscript> inhibition.
