Abstract

Atherosclerosis (AS), a multifactorial and chronic inflammatory disease, is considered a lifethreatening condition worldwide. As the predominant immunocytes in AS lesions, macrophages are of exceptional interest in exploring the pathogenesis of this disease. The proliferation, aggregation, aging and death of macrophages significantly contribute to the occurrence and development of AS. Iron is an essential microelement that plays a pivotal role in regulating diverse functions of macrophages under physiological conditions. However, abnormal iron metabolism can disrupt iron homeostasis in macrophages, leading to iron overload in macrophages, significantly contributing to the promotion of AS. From the perspective of macrophage iron overload and and its role in AS, this paper provides an extensive exposition on the phenomenon of macrophage iron overload and its underlying pathophysiological mechanism in promoting AS, providing novel insights for the prevention and treatment of AS.
