Evaluation of long-read sequencing for Ostreid herpesvirus type 1 genome characterization from Magallana gigas infected tissues.

Abstract
- This article discusses the use of long-read sequencing technology for the characterization of the Ostreid herpesvirus type 1 (OsHV-1) genome from infected tissues of the Pacific oyster Magallana gigas. The complex genomic architecture of OsHV-1 makes it difficult to accurately assemble and characterize the genome using traditional short-read sequencing methods. The study evaluates different methods for extracting high molecular weight (HMW) DNA and compares the effectiveness of long-read sequencing from Oxford Nanopore Technologies (ONT) with Illumina technology. The results show that the HMW DNA extraction kit coupled with ONT sequencing and dedicated bioinformatics tools allows for the production of accurate OsHV-1 genomes, including the characterization of genomic isomers. This sequencing technology shows promise as a diagnostic tool for studying aquatic viruses directly from host tissues. [Extracted from the article]
