Abstract

The methods described below were adapted from the small molecule protocol described in Lian et al.1, and have been optimized for the differentiation of cardiomyocytes in a glass 96-well plate format using the Allen Cell Collection, derived from the hiPSC WTC parental line released by the Conklin Laboratory2 at the J. David Gladstone Institute. Differentiation on glass allows high resolution imaging without replating steps. It also allows observationsat the earliest transitions and cell states during differentiation.
