Abstract

The skin, the largest organ in the human body, is anatomically quite complex and serves a diverse array of biologic functions in all species. In this chapter, we provide an overview of the structure and function of the skin. Some basic concepts of dermatotoxicology are discussed. The various responses to injury of the skin including inflammation, irritation and corrosion, and hypersensitivity reactions are presented. Phototoxicity and metabolic and endocrine effects on the skin are addressed. Various tumors of the skin as they relate to drug development are provided. Some of the important animal models for studying cutaneous disease, including the pros and cons of each, are discussed. There are several case examples presented to highlight applications of dermatopathology in nonclinical species.
