Abstract

A PCR detection method for mutations in receptor protein genes from Busseola fusca midgut material potentially involved in Bt-resistance Busseola fusca (Fuller) (Lepidoptera: Noctuidae) is a major insect pest to Bt-maize in South Africa. No study has yet been done to determine the molecular mechanism of B. fusca resistance to Cry1Ab. Aminopeptidase N (1-6), cadherin and alkaline phosphatase receptor-protein genes were amplified and sequences of PCR products were determined. BLAST searches showed similarity to the respective receptor-protein genes. Mutations can thus be identified by comparing sequences of receptor-protein genes from resistant and susceptible larvae.
