Effect of Bax Gene Expression on Cell Invasion in Hepatobiliary Carcinoma Cells

Abstract

There is a growing tendency of the morbidity of hilar cholangiocarcinoma in recent years. Some studies indicate that Bax gene expression was slightly increased in adjacent tissues of cholangiocarcinoma. Therefore, it is speculated that this condition may have certain correlation with the invasive ability of cholangiocarcinoma cell. This research aimed to detect the effect of Bax on cell invasive ability. The cholangiocarcinoma cell line RBE was cultured. Bax high-expression vector and Bax small interfere RNA (Bax-siRNA) were named pLBax and pLBax-siRNA, respectively. The constructed vector was transfected into the RBE cell line by using liposome transfection followed by analysis of the relationship between Bax expression and invasive ability of cholangiocarcinoma cell. The Bax gene expression in cells transfected with both vectors showed increased and decreased expression, respectively. The RBE cell apoptosis was significantly increased after high expression of Bax, while cell invasive ability was decreased with significant differences among pLBax, pLBax-siRNA and control group (p < 0.05). Bax was downregulated and RBE cell showed reversed changes with significant differences among different groups (p < 0.05). The change of the Bax expression in cholangiocarcinoma cell can affect cell invasive ability and apoptosis. Bax expression was negatively correlated with cell invasive ability.
