Abstract

Introduction: Peritoneal metastases from colorectal cancer (PM-CRC) have a poor prognosis. For eligible patients, the only possible cure is cytoreductive surgery (CRS) and hyperthermic intraperitoneal chemotherapy (HIPEC). Unlike primary colorectal tumours and liver metastases, few studies with good clinical data have explored the mutational landscape in PM-CRC. In this thesis, clinical and tumoral characteristics in PM-CRC were studied in patients treated at the National Centre for CRS-HIPEC at the Norwegian Radium Hospital. Methods: Two-hundred-and-thirty fresh frozen tumour samples with sufficient quality for subsequent analysis using next generation sequencing were collected during CRS-HIPEC for PM-CRC. Patients’ data were entered into the departments’ MedInsight database. Clinicopathological data and mutations were described using medians and percentages. Hazard ratios (HR) and Kaplan-Meier plots described associations between clinicopathological data/mutations and overall survival (OS). Results: The cohort median age was 61 years, 68,7% were women. Primary tumours were in the right colon in 47,4% and 56,5% were T4. Univariables associated with outcome were: Peritoneal cancer index (PCI): PCI > 12; HR 3,12 (2,23-4,35), vs PCI ≤ 12, p
