Aislamiento e identificación de bacterias en el reactor RP-10 del Centro Nuclear 'RACSO'

El objetivo de esta investigación fue aislar e identificar las bacterias que viven en el fondo de la pileta auxiliar del reactor nuclear RP-10 del Centro Nuclear RACSO. Se colectaron muestras de bacterias a partir del agua y de la superficie de un tapón de aluminio y se inocularon sobre diferentes medios de cultivo para su aislamiento. Para la caracterización fenotípica se usaron técnicas microbiológicas como la coloración de Gram, coloración de esporas y pruebas bioquímicas. Esto ha permitido identificar presencia de diferentes géneros bacterianos tales como Bacillus, Staphylococcus y bacilos gram negativos. A su vez se muestran avances en la identificación molecular así como en la estandarización de la extracción de ADN y amplificación parcial del gen 16S rRNA por Reacción en Cadena de la Polimerasa (PCR). The aim of this study was isolation and identification of bacteria living in the bottom of the auxiliary pool of the RP-10 in the Nuclear Center RACSO. Samples of bacteria from water and from the surface of an aluminum stopple were collected and inoculated into different media for culturing and isolation. For phenotypical characterization, microbiological techniques as Gram staining, spore staining and biochemical assays were used. This permitted to us to identify presence of different bacterial genus like Bacillus, Staphylococcus and Gram-negative bacteria. At the same time, advances in molecular identification, DNA extraction standardization and partial amplification of gene 16S rRNA using PCR technology are shown.
