Development of Nano Ache Enzyme Based Sensor for Determination of Nerve Agents Used in Chemical Weapons

Abstract

In this study; Nano enzyme system based sensor system has been developed for detection of nerve agents used in chemical weapons production. The chemical substances forming the nerve agents are included in the group of organophosphorus compounds. Paraoxon was used as the organophosphorus compound. Paraoxon molecule irreversibly inhibits the acetylcholinesterase enzyme (AChE). In this respect, it is preferred because of its similarity to nerve agents. In the sensor system, nano enzyme system synthesized according to photosensitive cross-linking method with ruthenium based amino acid monomers was used as sensor layer. The fluorescence spectrum of the obtained nano enzyme was taken and observed to show fluorescence. As a result of the analyzes, working conditions were determined as wavelength of 300 nm and pH of 7,4. The fluorescence intensities of the paraoxon samples, which ranged from 0.01 ppm to 200 ppm, were measured after interaction with the nano-enzyme at 300 nm wavelength and 7.4 pH. Calibration graph is drawn by using the obtained data. According to the experimental results, the LOD value of the sensor system based on the Nano AChE enzyme was calculated as 6,0x10-8 M, and the LOQ was calculated as 6,98x10-8M.
