Abstract

Analysis of four probable mismatch sets possessing 8 single-base-pair mismatches obtained from isolated mutations helped assess the potential of T4 endonuclease VII to identify heteroduplex mismatches. Cleavage occurs in at least one strand of each set that contains a mismatch while an A.A T.T set remains uncleaved in any mismatched strand. This technique facilitates the efficient detection and location of all mutational changes upon DNA screening for mutations.
