Structural and functional characterization of the ClpXP1/2 protease complex from Listeria monocytogenes

The ClpXP protease complex is important for targeted protein degradation in bacteria and in organelles of eukaryotic cells. The complex consists of the AAA+ chaperone ClpX and the peptidase ClpP. While the majority of bacteria utilize only a single ClpP, Listeria monocytogenes expresses two isoforms. LmClpPs assemble into either a LmClpP2 homocomplex or a LmClpP1/2 heterocomplex. In this thesis, the function of the ClpP1/2 heterocomplex and the structure of the ClpXP1/2 complex is examined.
Der ClpXP-Proteasekomplex ist wichtig für gezielte Proteolyse in Bakterien und in Organellen von eukaryotischen Zellen. Der Komplex besteht aus dem AAA+ Chaperon ClpX und aus der Peptidase ClpP. Während die meisten Bakterien nur ein ClpP haben, besitzt Listeria monocytogenes zwei Isoformen. Sie bilden entweder einen ClpP2-Homokomplex, oder einen ClpP1/2-Heterokomplex. In dieser Dissertation werden die Funktion des ClpP1/2-Heterokomplexes und die Struktur des ClpXP1/2-Komplexes untersucht.
