An afternoon heavy rainfall event occurred over coastal Nantong, an area of 70–100 km downstream from the Shanghai–Suzhou–Wuxi–Changzhou city belt over the Yangtze River Delta, under the influences of weak southwesterly monsoonal flows and lake/sea breezes on 26 July 2018. An observational analysis shows the emergence of pronounced urban heat island (UHI) effects along the city belt during the late morning hours. A series of nested-grid cloud-permitting model simulations with the finest grid spacing of 1 km are performed to examine the impacts of urbanization on convection initiation (CI) and the subsequent heavy rainfall event. Results reveal the generation of lake breezes and warm anomalies in the planetary boundary layer along the city belt and low-level convergence, thereby inducing upward motion for CI. The southwesterly flows of the monsoonal warm–moist air, enhanced by the UHI effects along the city belt, allow the development of convective cells along the city belt, some of which merge with convective clusters during their downstream propagation, and the ultimate generation of several distinct heavy rainfall centers by local convective clusters over coastal Nantong where atmospheric columns are more moist and potentially unstable under the influences of sea breezes. Sensitivity simulations show small contribution of Nantong's UHI effects to the heavy rainfall event. The above findings help elucidate how the UHI effects could assist the CI in a weak-gradient environment, and explain why urbanization can contribute to increased downwind mean and extreme precipitation under the influences of favorable regional forcing conditions. Significance Statement: The urban heat island (UHI) effects tend to produce more rainfall on its downwind side than that on the other sides, but alone could hardly account for the generation of heavy rainfall. This study examines the influences of the UHI effects associated with a city belt over the Yangtze River Delta on generating an afternoon heavy rainfall event over coastal Nantong that is 70–100 km downwind from the city belt. Results show (i) the downwind advection of the UHI effects; (ii) the initiation of convective storms along the city belt, and their subsequent downstream propagation, leading to the generation of heavy rainfall over Nantong; and (iii) an important role of sea breezes in generating the heavy rainfall event. The color of flour and its end-use products is an important quality trait of wheat. Understanding the genetic basis of this trait is essential for improving wheat quality. In this study, quantitative trait locus (QTL) mapping for 27 color-related traits of flour, starch, gluten, dough sheet, steamed bread, and baked bread was performed using a population of recombinant inbred lines (RILs) with 173 lines of F9:10, tested in four environments. For this purpose, a high-density genetic map constructed using a 90 K SNP array, Diversity Arrays Technology (DArT) and simple sequence repeat (SSR) markers was used. A total of 43 additive QTLs were detected, including 25 large-effect QTLs, located on chromosomes 1A, 1B, 2A, 4B, 5B, and 7B. Four of these major QTLs, namely QFa7B.7-1, QFb7B.7-2, QFb7B.7-1 and QGb4B.4, made the highest contribution and accounted for 29.60, 29.02, 27.45, and 22.24% of the phenotypic variation, respectively. Six stable QTLs (QSa4B.4, QSl4B.4-1, QFa7B.7-2, QFb7B.7-1, QFb7B.7-2, and QBca7B.7) were detected in more than one environment with high PVE values. In addition, two robust QTL clusters contribution to 13 evaluated traits were identified on chromosomes 4B (RAC875_c27536_611-wsnp_Ku_c28756_38667953) and 7B (wPt-1196-Kukri_c2348_2340). A locus within the interval Ex_c101685_705-RAC875_c27536_611 of 4B also harbored many genes previously reported to affect wheat quality and yield. Dopa and tetrahydrobiopterin (BH4) supplementation are recommended therapies for the dopa-responsive dystonia caused by GTP cyclohydrolase 1 (GCH1 , also known as GTPCH) deficits. However, the efficacy and mechanisms of these therapies have not been intensively studied yet. In this study, we tested the efficacy of dopa and BH4 therapies by using a novel GTPCH deficiency mouse model, Gch1 KI/KI , which manifested infancy-onset motor deficits and growth retardation similar to the patients. First, dopa supplementation supported Gch1 KI/KI mouse survival to adulthood, but residual motor deficits and dwarfism remained. Interestingly, RNAseq analysis indicated that while the genes participating in BH4 biosynthesis and regeneration were significantly increased in the liver, no significant changes were observed in the brain. Second, BH4 supplementation alone restored the growth of Gch1 KI/KI pups only in early postnatal developmental stage. High doses of BH4 supplementation indeed restored the total brain BH4 levels, but brain dopamine deficiency remained. While total brain TH levels were relatively increased in the BH4 treated Gch1 KI/KI mice, the TH in the striatum were still almost undetectable, suggesting differential BH4 requirements among brain regions. Last, the growth of Gch1 KI/KI mice under combined therapy outperformed dopa or BH4 therapy alone. Notably, dopamine was abnormally high in more than half, but not all, of the treated Gch1 KI/KI mice, suggesting the existence of variable synergetic effects of dopa and BH4 supplementation. An extensive urban agglomeration has occurred over the Yangtze River delta (YRD) region of East China as a result of rapid urbanization since the middle 1990s. In this study, a 44-yr (i.e., 1975–2018) climatology of the summertime extreme hourly precipitation (EXHP; greater than the 90th percentile) over the YRD is analyzed, using historical land-use data, surface temperature, and hourly rain gauge observations, and then the relationship between rapid urbanization and EXHP changes is examined. Results show significant EXHP contrasts in diurnal variation and storm type roughly before and after middle July. That is, tropical cyclones (TCs) account for 16.4% of the total EXHP hours, 80.5% of which occur during the late summer, whereas non-TC EXHP accounts for 94.7% and 66.2% during the early and late summer, respectively. Increasing trends in occurrence frequency and amount of the non-TC and TC-induced EXHP are detected over the urban agglomeration. Statistically significant larger increasing trends in both the EXHP and surface temperature are observed at urban stations than those at the nearby rural stations. An analysis of 113 locally developed non-TC extreme rainfall events during 2011–18 summers also suggests the contribution of the urban heat island effects to the more occurrences of EXHP, especially over a band-shaped urban region where several major cities are distributed. This study reveals a significant correlation between rapid urbanization and increased EXHP during the past two decades over the YRD region. This paper proposes a dynamic trajectory planning technique for six-degree-of-freedom (6-DOF) cable-suspended parallel robots (CSPRs). First, a passive mechanical system that is equivalent to the CSPR is introduced to provide insight and facilitate the design of trajectories that can extend beyond the robot's static workspace. The tilt-and-torsion angle convention is used to develop the mathematical model and impose restrictions for the rotational component of the trajectories. The dynamic differential equations that govern the translational component of the trajectories are shown to become linear under some conditions. Natural frequencies of an equivalent passive linear system of constant-stiffness springs are, thus, obtained and the set of linear differential equations associated with this system is integrated to produce a general solution for natural, periodic trajectories. This approach is used to produce pure translation trajectories and more complex motion that includes changes in position and orientation. This paper proposes a point-to-point dynamic trajectory planning technique for reaching a series of poses with a six-degree-of-freedom (six-DOF) cable-suspended parallel robot. Each trajectory segment is designed to have zero translational and rotational velocity at its endpoints; transitions between segments have translational and rotational acceleration continuity. This formulation facilitates the synthesis of trajectories that extend beyond the static workspace of the robot. A basis motion is introduced, which is a mathematical function that can be adapted for each coordinate direction along each trajectory segment. Kinematic constraints are satisfied through the selection of the coefficients for this function. Dynamic constraints are imposed by defining feasible regions within the workspace for each segment endpoint, based on the previous endpoint. Spherical linear interpolation (SLERP) is used to produce singularity-free, optimally interpolated rotational trajectory segments. Agropyron elongatum (Host.) Neviski (synonym, Thinopyrum ponticum Podp., 2n = 70) has been used extensively as a valuable source for wheat breeding. Numerous chromosome fragments containing valuable genes have been successfully translocated into wheat from A. elongatum. However, reports on the transfer of powdery mildew resistance from A. elongatum to wheat are rare. In this study, a novel wheat-A. elongatum translocation line, 11-20-1, developed and selected from the progenies of a sequential cross between wheat varieties (Lankaoaizaoba, Keyu 818 and BainongAK 58) and A. elongatum, was evaluated for disease resistance and characterized using molecular cytogenetic methods. Cytological observations indicated that 11-20-1 had 42 chromosomes and formed 21 bivalents at meiotic metaphase I. Genomic in situ hybridization analysis using whole genomic DNA from A. elongatum as a probe showed that the short arms of a pair of wheat chromosomes were replaced by a pair of A. elongatum chromosome arms. Fluorescence in situ hybridization, using wheat D chromosome specific sequence pAs1 as a probe, suggested that the replaced chromosome arms of 11-20-1 were 5DS. This was further confirmed by wheat SSR markers specific for 5DS. EST-SSR and EST-STS multiple loci markers confirmed that the introduced A. elongatum chromosome arms belonged to homoeologous group 5. Therefore, it was deduced that 11-20-1 was a wheat-A. elongatum T5DL∙5AgS translocation line. Both resistance observation and molecular marker analyses using two specific markers (BE443538 and CD452608) of A. elongatum in a F2 population from a cross between line 11-20-1 and a susceptible cultivar Yannong 19 verified that the A. elongatum chromosomes were responsible for the powdery mildew resistance. Background: In recent years, a relationship between depression and basal dehydroepiandrosterone (DHEA) and dehydroepiandrosterone sulfate (DHEA-S) levels has frequently been suggested, but responses of these adrenal steroids to psychosocial stress have not been examined in individuals with depressive disorders. In this study, we examined salivary DHEA, DHEA-S, and cortisol/DHEA response to the Trier Social Stress Test (TSST) in individuals with depressive disorders and in healthy controls to discover whether the responses of DHEA and DHEA-S to acute psychosocial stress could be a more sensitive marker of HPA dysfunction in depressive disorders.Methods: We compared salivary cortisol, DHEA, DHEA-S, and cortisol/DHEA levels to the TSST tests between 38 individuals with depression and 43 healthy controls aged 18.4-25.9 years. Depression severity was assessed by the self-reported Beck Depression Inventory-II (BDI-II). Salivary samples were evaluated at four time points: the baseline (-10 time point), before the TSST started (0 time point), the end of the TSST (+20 time point), and the recovery (+50 time points).Results: No significant differences existed in the basal adrenal hormonal levels between subjects with depressive disorders and controls; however, at the end of TSST, attenuated DHEA and DHEA-S response was identified in subjects with depressive disorders compared to that found in healthy subjects. The differences in the DHEA and DHEA-S levels at the +20 time point, as well as the differences in the cortisol/DHEA at the +50 time point, exhibited negative correlations with depression severity.Conclusion: Attenuated DHEA and DHEA-S response to acute psychosocial stress was identified in subjects with depressive disorders. These findings help us to discover the bi-directional relationship between depression and the hypothalamic-pituitary-adrenal (HPA) axis function, hence furthering our understanding of whether altered DHEA and DHEA-S response to psychosocial stress may be a more sensitive method than basal adrenal steroid analysis for detecting HPA axis dysfunction in depressive disorders.Limitations: As this is a case control study, we could only draw the conclusion of the bi-directional relationship between the depression and the altered DHEA (S) response to stress, and could not identify whether depression was due to the HPA dysfunction, or vice versa. Prospective studies such as such as cohort studies or epidemiology experiments are needed to further test the cause of depression or HPA dysfunction; and the mechanisms responsible for altered DHEA and DHEA-S in response to acute psychosocial stress in individuals with depressive disorders are also needed to be clarified. This paper proposes a dynamic trajectory planning method for point-to-point motion of three-degree-of-freedom (three-DOF) cable-suspended parallel robots. Natural frequencies as well as associated periodic trajectories that can be obtained from the integration of the dynamic model of an equivalent passive mechanical system are used to design point-to-point trajectories. The trajectories can be used to connect consecutive points in sequence that may lie beyond the static workspace of the robot. The technique ensures zero velocity at each of the target points and continuity of the accelerations. Based on the cable tension constraints, attainable regions can be determined to search for the next target point, while feasible regions of intermediate points are generated in cases for which a given point cannot be directly attained. An example trajectory is performed to illustrate the approach. This paper proposes an interval-analysis-based algorithm to determine the trajectory-reachable workspace (TRW) of planar two-degree-of-freedom (DOF) cable-suspended parallel robots and planar three-DOF cable-suspended parallel robots. The TRW is a set of target points that the end-effector can reach along a given trajectory under the cable tension constraint. The method includes two sufficient conditions for a continuous set of target points to lie fully inside or outside the TRW. The two conditions can be verified by the implementation of interval-analysis-based methods. On the basis of these two verifiable sufficient conditions, a TRW determination method using branch-and-prune algorithms is presented. This method avoids the discretization of a continuous trajectory set, which guarantees the accuracy of TRW determination. Background: Recent studies have indicated that low serum testosterone levels are associated with increased risk of developing hepatic steatosis; however, the mechanisms mediating this phenomenon have not been fully elucidated. To gain insight into the role of testosterone in modulating hepatic steatosis, we investigated the effects of testosterone on the development of hepatic steatosis in pigs fed a high-fat and high-cholesterol (HFC) diet and profiled hepatic gene expression by RNA-Seq in HFC-fed intact male pigs (IM), castrated male pigs (CM), and castrated male pigs with testosterone replacement (CMT). Results: Serum testosterone levels were significantly decreased in CM pigs, and testosterone replacement attenuated castration-induced testosterone deficiency. CM pigs showed increased liver injury accompanied by increased hepatocellular steatosis, inflammation, and elevated serum alanine aminotransferase levels compared with IM pigs. Moreover, serum levels of total cholesterol, low-density lipoprotein cholesterol, and triglycerides were markedly increased in CM pigs. Testosterone replacement decreased serum and hepatic lipid levels and improved liver injury in CM pigs. Compared to IM and CMT pigs, CM pigs had lower serum levels of superoxide dismutase but higher levels of malondialdehyde. Gene expression analysis revealed that upregulated genes in the livers of CM pigs were mainly enriched for genes mediating immune and inflammatory responses, oxidative stress, and apoptosis. Surprisingly, the downregulated genes mainly included those that regulate metabolism-related processes, including fatty acid oxidation, steroid biosynthesis, cholesterol and bile acid metabolism, and glucose metabolism. KEGG analysis showed that metabolic pathways, fatty acid degradation, pyruvate metabolism, the tricarboxylic acid cycle, and the nuclear factor-kappaB signaling pathway were the major pathways altered in CM pigs. Conclusions: This study demonstrated that testosterone deficiency aggravated hypercholesterolemia and hepatic steatosis in pigs fed an HFC diet and that these effects could be reversed by testosterone replacement therapy. Impaired metabolic processes, enhanced immune and inflammatory responses, oxidative stress, and apoptosis may contribute to the increased hepatic steatosis induced by testosterone deficiency and an HFC diet. Complete diallel crosses with five parents of common wheat ( Triticum aestivum L.) were conducted to analyse inheritance of 17 amino acid contents by using the genetic model including seed, cytoplasmic, maternal and environment interaction effects on quantitative traits of seeds in cereal crops. The results showed that inheritance of 17 amino acid contents, except tyrosine, was controlled by several genetic systems including seed, cytoplasmic, and maternal effects, and by significant gene × environment interaction effects. Seed-direct additive and maternal effects constituted a major part of genetic effects for lysine, tyrosine, arginine, methionine, and glutamic acid content. Seed-direct additive effect formed main part in inheritance of isoleucine and serine contents. Threonine content was mainly governed by maternal additive effect. The other nine amino acid contents were almost entirely controlled by dominance effects. High general heritability of tyrosine (36.3%), arginine (45.8%), lysine (24.7%) and threonine (21.4%) contents, revealed that it could be effective to improve them by direct selection in progenies from appropriate crosses. Interaction heritability for phenylalanine, proline, and histidine content, which was 36.1%, 39.5% and 25.7%, respectively, was higher than for the other amino acids. [ABSTRACT FROM AUTHOR]

Abstract: Quickly and sensitively detection of antibiotic residues in animal products often requires time-consuming techniques and expensive instrumentation. To address these limitations, a high sensitive aptasensor for sulfadimethoxine (SDM) using unmodified gold nanoparticles (AuNPs) was developed in the present study. The assay sensitivity was improved by optimizing several key parameters such as the amount of aptamer adsorbed to AuNPs, the concentration of salt and the AuNPs pH value. To our knowledge, this is the first time to investigate the effect of AuNPs pH value on the sensitivity of aptamer–AuNPs based label-free assay. The best procedure for SDM analysis in our system was: the pH value at 8.0, and the concentration of aptamer at 2μM, and the concentration of salt at 2M. The linear range for the current analytical system was from 50ng/mL to 1μg/mL, and the detection limit was 50ng/mL, which is lower than the Maximum Residue Limit (MRL) (100ng/mL) in edible animal tissues regulated by China and European commission. Our study provides a simple, fast, and easy to read method for SDM analysis with high sensitivity, which can be applied in future on-site detection in animal products. [Copyright &y& Elsevier]

Thyrotropin-releasing hormone receptor (TRHR) is a G-protein-coupled receptor that plays a crucial role in regulating the hypothalamic-pituitary-thyroid axis by conveying the action of the hypothalamic tripeptide TRH, which is the primary central activator of this hormonal cascade. In the present study, the porcine TRHR (pTRHR) gene was localized to chromosome 4 by Radiation hybrid mapping. Quantitative trait loci affecting average backfat thickness, daily gain, and carcass and meat quality traits have been mapped to the region containing this gene. Further, the full-length cDNA of pTRHR was cloned and sequenced. pTRHR contains an open reading frame encoding 398 amino acids and shares 96.2% amino acid identity to human TRHR. Real-time quantitative RT-PCR showed that the mRNA of pTRHR is expressed in a variety of tissues, with high expression in the brain, hypothalamus, pituitary, testis, and fat tissue. The considerable expression level of TRHR mRNA found in fat tissue indicates potential direct action of TRH on lipocyte might exist. Additionally, two alternative spliced transcript variants of pTRHR were also isolated in this study. Abstract: Oxidative stress plays a crucial role in both initiation and progression of liver injury in almost all experimental and clinical liver diseases. Antioxidative therapy is therefore an effective means of preventing and attenuating oxidative stress related liver diseases. Human paraoxonase 3 (hPON3) is a lipid-associated enzyme with antioxidant activity. In the present study, hPON3 cDNA gene was cloned into pcDNA3.1 plasmid and electro-transferred into mouse skeletal muscle to maintain a higher serum PON3 activity. After gene delivery, serum PON3 activity was about 1.4 times higher than those of control and PON3 mRNA expression was also detected in mouse skeletal muscle. To investigate the role of hPON3 in protecting mice against liver injury, subacute liver injury model was induced by repeated CCl4 administration and hPON3 gene was delivered into mouse skeletal muscle before progression or recovery phase, respectively, of liver injury. Afterwards, the mice were euthanized to evaluate liver marker enzymes, degrees of oxidative stress and liver histological architecture in order to reveal the effects of PON3 on subacute liver injury. In both damage phases, delivery of hPON3 gene significantly reduced serum aminotransferase level and improved liver histological architecture. Moreover, transgene expression of hPON3 attenuated oxidative stress by increasing hepatic glutathione content, superoxide dismutase (SOD) activity, total antioxidant capability (T-AOC), and reducing malondialdehyde (MDA) level. Abstract: Understanding the mechanism of energy flux may be critical for explaining how obesity has emerged as a public health epidemic. It is known that changes in caloric intake predictably alter physical activity levels (PA) in mammals. Here, our goal was to test the hypothesis that fasting induces a biphasic pattern of change in PA by measuring PA before and after long-term food deprivation in zebrafish. Compared to control-fed fish, food-deprived fish showed a significant increase in PA levels during the first 2 days of food deprivation. Subsequently, however, fasted fish showed a significant chronic decrease in PA compared to fish fed at weight-maintenance levels. These data are comparable to those seen with mammals, which also show a biphasic response of PA to caloric restriction. In a separate group of fish, long-term food deprivation, associated with decreases in PA, induced a significant increase in brain preproorexin mRNA levels compared to fed controls. No change in orexin mRNA was seen after 2 days of food deprivation. The finding that orexin mRNA expression is altered only after long-term starvation suggests that orexin may be coupled with the changes in PA seen at this time. Thus, the association between negative energy balance and reductions in PA occurs across genera in biology and is associated with predictable neurological changes in brain gene expression. We previously reported that an elevated level of matrix metalloproteinase-1 (MMP-1) gene expression in patients with chondrosarcoma has a strong statistical correlation with recurrence and in vitro invasion. In the present study, we used an antisense RNA strategy for MMP-1 inhibition to determine if this would affect the invasive characteristics of the cells. We transfected a human chondrosarcoma cell line with a retroviral plasmid expressing a 770 bp genomic fragment of the human MMP-1 gene in the sense or antisense orientation. The results show that cells transfected with the MMP-1 antisense fragment had a significant decrease in both MMP-1 protein and enzyme activity (p<0.05) as compared to cells transfected with an empty plasmid or the parental cells. Cells transfected with the MMP-1 antisense fragment demonstrated a significant decrease in their ability to invade the collagen I barrier (p<0.05). The gene expression for MMP-8 and MMP-13 were unaffected in cells transfected with the MMP-1 antisense fragment, MMP-1 sense fragment, or empty plasmid. These results support the hypothesis that MMP-1 facilitates tumor cell egress from chondrosarcoma tissue and demonstrate the potential of MMP-1 as a promising target for a novel biologic therapy in chondrosarcoma. This study analyzes the spatiotemporal distributions and climate trends of tropical cyclone (TC)-induced extreme hourly precipitation (EXHP) in the warm season (May–September) during 1975–2018 over China and the involved mechanisms. Each TC is classified as a high-, mid-, and low-EXHP TC according to the total amount of EXHP it produces over China during its lifetime. Results show that low-EXHP TCs have a greater contribution to the total TC-induced EXHP over southern and southwestern China, whereas high-EXHP TCs make greater contributions over eastern and northeastern China as they tend to move northwestward after making landfall. It is shown that, although the total frequency of EXHP-producing TCs displays a decreasing trend, the total frequency of TC-induced EXHP over China shows a significant increasing trend, which is largely contributed by the high-EXHP TCs. To explore the possible mechanisms responsible for the different characteristics in EXHP-producing TC groups, we further analyze the large-scale environmental conditions with respect to three groups by composite analysis. The cooperation of large-scale environmental fields between high and low levels provides favorable conditions for the intensification of TCs and the enhancement of the TC-induced precipitation for the high-EXHP group, such as stronger divergence at high levels, weaker vertical wind shear, more sufficient water vapor, and more conducive steering airflow over the southeastern and eastern area of mainland China. The westward and northward movement of western North Pacific subtropical high is conducive to the northward shift of TC tracks, thus contributing to the high frequency of TC-induced EXHP over the eastern area of mainland China. Fatigue damage affects both durability and safety, and it has been the most important distress in asphalt concrete. Fatigue damage occurs as a result of repeated traffic loading. An asphalt mixture is a typical viscoelastic material, and its fatigue damage is related to its viscoelastic properties. Under repeated traffic loading, the combined effects of creep damage and fatigue damage will shorten its fatigue life. Currently, the evaluation of the fatigue damage of asphalt mixtures rarely considers the combined effects of creep damage and fatigue damage. To solve this problem, a viscoelastic fatigue damage prediction model of an asphalt mixture considering the combined effects of creep damage and fatigue damage is put forward by introducing parameter β and a displacement factor based on theoretical derivations and testing. The results show that the model can embody the viscoelastic fatigue damage essence of asphalt mixtures, and it can also consider the effects of aging degree, temperature, load frequency and stress on fatigue damage of asphalt mixtures. Down syndrome (DS), also known as trisomy 21, is one of the most common chromosomal disorders associated with intellectual disability. Mouse models are valuable for mechanistic and therapeutic intervention studies. The purpose of this study was to investigate astroglial anomalies in Dp16, a widely used DS mouse model. Brain sections were prepared from one-month-old Dp16 mice and their littermates, immunostained with an anti-GFAP or anti-S100B antibody, and imaged to reconstruct astroglial morphology in three dimensions. No significant difference in the number of astrocytes was found in either the hippocampal CA1 region or cortex between Dp16 and WT mice. However, the average astroglial volume in Dp16 was significantly (P < 0.05) greater than that in WT, suggesting the astroglial activation. Reanalysis of the single-nucleus RNA sequencing data indicated that the genes differentially expressed between WT and Dp16 astrocytes were associated with synapse organization and neuronal projection. In contrast, in vitro cultured neonatal astrocytes did not exhibit significant morphological changes. The expression of Gfap in in vitro cultured Dp16 astrocytes was not increased as it was in in vivo hippocampal tissue. However, after treatment with lipopolysaccharides, the inflammatory response gene IFNβ increased significantly more in Dp16 astrocytes than in WT astrocytes. Overall, our results showed that the increase in astrogliogenesis in DS was not apparent in the early life of Dp16 mice, while astrocyte activation, which may be partly caused by increased responses to inflammatory stimulation, was significant. In this study, the total days, mean duration, and intensity of extreme hot events over southern China during the 1971–2020 warm seasons are analyzed on the basis of daily maximum and minimum temperatures, by comparing the newly proposed independent hot day (IHD), independent warm night (IWN), and compound extreme (CMPD; i.e., the continuous occurrences of hot days and hot nights) with the traditionally defined hot day and warm night. Relationships between the hot extremes and urbanization are explored with 1-km-resolution population density data. Results show obvious differences in the spatial distributions among IHD, IWN, and CMPD over southern China. Positive correlations of 0.43, 0.41, and 0.37 are found between the population density and the total days, mean duration, and mean intensity of CMPD, respectively, which are qualitatively similar to those using the traditional hot days and warm nights. In contrast, negative correlations between the IHD and IWN indices and the population density are found, because those indices are more apparent over rural areas. Moreover, total days, mean duration, and mean intensity of CMPD increase significantly, with trends of approximately 103%, 21%, and 38% decade−1, respectively, during the rapid urbanization period from the mid-1990s to 2020, which are about 4.9, 2.1, and 2.4 times their counterparts from 1970 to the mid-1990s, and less significant and smaller differences between the two eras are found in IHD and IWN. These results will provide a new scientific basis for evaluating climate models of hot extremes in southern China and have important implications for the other urbanized regions as well. Abstract Hepatocellular carcinoma (HCC) is one type of the most common malignancies. However, the underlying molecular mechanisms involved in the development of HCC remain unknown. To identify the candidate genes in the progression of HCC, gene expression profiles GSE14520, GSE54236, GSE57957 and GSE64041 were downloaded from the Gene Expression Omnibus database (GEO). A total of 405 tumor and 399 para-carcinoma samples from patients with HCC were examined to identify the differentially expressed genes (DEGs), followed by function enrichment analyses including Gene Ontology (GO) functions and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. A total of 78 DEGs were screened, including 62 downregulated genes and 16 upregulated genes. Subsequently, the protein-protein interaction network (PPI) was constructed using the Search Tool for Retrieval of Interacting Genes (STRING) database. The module analysis and Hub genes validation were performed using Cytoscape software. Hierarchical clustering of hub genes was evaluated using UCSC Cancer Genomics Browser. Survival analyses of Hub genes were performed using Kaplan Meier Plotter database. Genes specifically expressed in the liver were analyzed using GENEVESTIGATOR database. CYP2C8 was identified as one of the most promising molecules among all the candidate genes. The expression profile of CYP2C8 in HCC was analyzed using ONCOMINE and UALCAN database. The expression levels of CYP2C8 in HCC samples and hepatoma cells were verified using reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and immunohistochemistry analysis. In summary, DEGs and hub genes were identified in the present study, which provides novel insight on the development of HCC. CYP2C8 was downregulated in HCC and could be a potential prognostic biomarker. MicroRNAs (miRNAs) play a prominent role in post-transcriptional gene expression regulation and have been involved in various biological and metabolic processes to regulate gene expression. For Brassica napus, improving seed-weight and oil-content is the main breeding goal. In order to better understand the regulation mechanism of miRNAs during seed-weight formation and oil-content accumulation in B. napus, in this study, a high-throughput sequencing technology was used to profile miRNAs expression of Brassica napus immature seeds from one to six weeks after flowering. A total of 1,276 miRNAs, including 1,248 novel and 28 known miRNAs, were obtained from both the high-seed-weight with low-oil-content RNA pool (S03) and the low-seed-weight with high-oil-content RNA pool (S04). Analysis of their expression profiles disclosed that 300 novel and two known miRNAs were differentially expressed between S03 and S04. For degradome analysis, 57 genes with 64 degradation sites were predicted to be targeted for degradation by these miRNAs. Further bioinformatics analysis indicated that these differentially expressed miRNAs might participate in regulation of myriad cellular and molecular processes, during seed development and oil synthesis. Finally, 6 target genes with potential roles in regulation of seed development and 9 other targets in seed oil synthesis, were further confirmed as candidate genes from small RNA and degradome sequencing. The fall armyworm (FAW), Spodoptera frugiperda (JE Smith) (Lepidoptera: Noctuidae), is a major migratory invasive pest that poses a severe threat to agricultural production and food security in China. In this study, we estimated the effects of six cereal crops on the development, survival, and fecundity of FAW using the age‐stage, two‐sex life table: maize (Zea mays L.), wheat (Triticum aestivum L.), sorghum [Sorghum bicolor (L.) Moench], oats (Avena sativa L.), millet (species, authority) (all Poaceae), and quinoa (Chenopodium quinoa Willd.) (Amaranthaceae). In a six‐choice test using detached leaf material, FAW larvae preferentially occurred on maize, followed by sorghum, within 6–24 h, and occurred the least on millet and quinoa. After 24 h, they had consumed the most leaf material from sorghum and maize, and the least from quinoa. In no‐choice tests using detached leaf material, they performed best on maize in terms of development (e.g., short development time), survival (e.g., high larval survival and pupation rate), and fecundity (e.g., high number of oviposition days and number of eggs laid per female), resulting in the highest predicted population growth (e.g., high intrinsic rate of increase and net reproductive rate). Although maize was found to be the most appropriate host plant, FAW was able to complete its life cycle on all six tested cereals. Our findings can help in predicting population dynamics and deepening our understanding of the potential damage incurred by FAW infestation, especially in regions under maize cultivation. Sparassis latifolia is a valuable edible mushroom, growing on fresh pine wood sawdust substrate. However, the mechanistic bases are poorly understood. The gene expression profiles of S. latifolia were analyzed from submerged cultures with fresh pine wood sawdust substrate for different time (0 h, 1 h, 6 h, 1 day, 5 days, and 10 days, respectively). The total number of differentially expressed genes (DEGs) identified under pine sawdust inducing was 2,659 compared to 0 h (CK). And 1,073, 520, 385, 424, and 257 DEGs were identified at the five time points, respectively. There were 34 genes in common at all inoculated time points, including FAD/NAD(P)-binding domain-containing protein, glucose methanol choline (GMC) oxidoreductase, flavin-containing monooxygenase, and taurine catabolism dioxygenase. Weighted gene co-expression analysis (WGCNA) was then used to compare the molecular characteristics among the groups and identified that the blue module had the highest correlation with the time induced by pine wood sawdust. There were 102 DEGs out of 125 genes in the blue model, which were most enriched in nitronate monooxygenase activity, dioxygenase activity, and oxidation–reduction process GO terms (p < 0.05), and peroxisome in KEGG pathway. What Is Known and Objective: A previous randomized clinical trial concluded that an optimal concentration of 0.3% ropivacaine could provide satisfactory analgesia for breast cancer patients undergoing modified radical mastectomy. We wondered if a smaller volume (30 ml vs. 40 ml) of 0.3% ropivacaine could still provide adequate analgesia in an ultrasound‐guided PECS II block in modified radical mastectomy. Methods: We performed a prospective parallel randomized double‐blind controlled clinical trial. Eligible patients were assigned to either the P30 or P40 group (30 or 40 ml of 0.3% ropivacaine, respectively). The skin area of hypoesthesia, anaesthetic plane determined with ultrasound, pain visual analogue scale (VAS), anaesthetic dosages, and complications were recorded. Serum levels of interleukin‐1β and interleukin‐6 were measured postoperatively. Results and Discussion: A total of 40 patients completed the trials, with 20 patients in each group. Although the skin area of hypoesthesia and the anaesthetic planes were significantly larger in the P40 group compared with the P30 group (p < 0.05), the VAS, analgesic and opioid doses, serum cytokine levels, anaesthetic toxicity, and complications had no significant differences between the two groups. This paper analyzes a set of dynamic trajectories for planar two-dof redundantly actuated cable-suspended parallel mechanisms. In recent literature, the global dynamic trajectory planning problem of cable-suspended mechanisms was addressed and some of the characteristic properties of such robots were revealed. In this paper, actuation redundancy is introduced and the dynamic trajectory planning is addressed using a series of periodic trajectories (i.e. straight line and circular periodic trajectories) and the application of the antipodal theorem. The experimental results obtained show that introducing actuation redundancy increases the dynamic capabilities of the robots. Also, cable tensions acquired via tension sensors confirm that cables always remain taut during all experimental verifications at feasible frequencies and that they are consistent with the tension variations predicted by theory. Furthermore, special frequencies are specified that are similar to those encountered with non-redundant mechanisms. Additionally, an alternative architecture is proposed to deal with cable interferences and it is shown that the novel architecture leads to improved dynamic capabilities when compared to the original architecture. Trisomy 21 (Down syndrome, DS) is the most common human genetic anomaly associated with heart defects. Based on evolutionary conservation, DS-associated heart defects have been modeled in mice. By generating and analyzing mouse mutants carrying different genomic rearrangements in human chromosome 21 (Hsa21) syntenic regions, we found the triplication of the Tiam1- Kcnj6 region on mouse chromosome 16 (Mmu16) resulted in DS-related cardiovascular abnormalities. In this study, we developed two tandem duplications spanning the Tiam1- Kcnj6 genomic region on Mmu16 using recombinase-mediated genome engineering, Dp( 16) 3Yey and Dp( 16) 4Yey, spanning the 2.1 Mb Tiam1- Il10rb and 3.7 Mb Ifnar1- Kcnj6 regions, respectively. We found that Dp( 16) 4Yey/+, but not Dp( 16) 3Yey/+, led to heart defects, suggesting the triplication of the Ifnar1- Kcnj6 region is sufficient to cause DS-associated heart defects. Our transcriptional analysis of Dp( 16) 4Yey/+ embryos showed that the Hsa21 gene orthologs located within the duplicated interval were expressed at the elevated levels, reflecting the consequences of the gene dosage alterations. Therefore, we have identified a 3.7 Mb genomic region, the smallest critical genomic region, for DS-associated heart defects, and our results should set the stage for the final step to establish the identities of the causal gene(s), whose elevated expression(s) directly underlie this major DS phenotype. Background: Kernel number per spike (KNS) and spike length (SL) are important spike-related traits in wheat variety improvement. Discovering genetic loci controlling these traits is necessary to elucidate the genetic basis of wheat yield traits and is very important for marker-assisted selection breeding. Results: In the present study, we used a recombinant inbred line population with 248 lines derived from the two founder genotypes of wheat, Bima4 and BainongAK58, to construct a high-density genetic map using wheat 55 K genotyping assay. The final genetic linkage map consists of 2356 bin markers (14,812 SNPs) representing all 21 wheat chromosomes, and the entire map spanned 4141.24 cM. A total of 7 and 18 QTLs were identified for KNS and SL, respectively, and they were distributed on 11 chromosomes. The allele effects of the flanking markers for 12 stable QTLs, including four QTLs for KNS and eight QTLs for SL, were estimated based on phenotyping data collected from 15 environments in a diverse wheat panel including 384 elite cultivars and breeding lines. The positive alleles at seven loci, namely, QKns.his-7D2–1, QKns.his-7D2–2, QSl.his-4A-1, QSl.his-5D1, QSl.his-4D2–2, QSl.his-5B and QSl.his-5A-2, significantly increased KNS or SL in the diverse panel, suggesting they are more universal in their effects and are valuable for gene pyramiding in breeding programs. The transmission of Bima4 allele indicated that the favorite alleles at five loci (QKns.his-7D2–1, QSl.his-5A-2, QSl.his-2D1–1, QSl.his-3A-2 and QSl.his-3B) showed a relatively high frequency or an upward trend following the continuity of generations, suggesting that they underwent rigorous selection during breeding. At two loci (QKns.his-7D2–1 and QSl.his-5A-2) that the positive effects of the Bima4 alleles have been validated in the diverse panel, two and one kompetitive allele-specific PCR (KASP) markers were further developed, respectively, and they are valuable for marker-assisted selection breeding. This paper presents a dynamic trajectory planning scheme for passing through singular orientations of a three degree-of-freedom (DOF) rotational cable-driven parallel robot (CDPR) that uses a rigid link to restrict its translational DOFs. The kinematics, dynamics and actuation singularity loci of this CDPR are analyzed. Singularity loci divide the workspace into four independent parts, which significantly limit the motion performance of the mechanism. To this end, consistency conditions that can allow the robot to steadily pass through singular orientations are studied. The vector inner product form of consistency conditions is derived, which can simplify the determination of the conditions of a more complex parallel robot. A sequence of target orientations are reached by planning dynamic trajectories using the unit quaternion. A modified spherical linear interpolation with a fifth degree polynomial as the time function is used for the generation of a trajectory which does not contain a singular orientation. Moreover, a transition segment using a seventh degree polynomial is designed to merge into the fifth degree polynomial in order to satisfy the consistency conditions and pass through singular orientations. Background: Hajdu-Cheney syndrome (HCS) is a rare inherited skeletal disorder caused by pathogenic mutations in exon 34 of NOTCH2. Its highly variable phenotypes make early diagnosis challenging. In this paper, we report a case of early-onset HCS with severe phenotypic manifestations but delayed diagnosis.Case Presentation: The patient was born to non-consanguineous, healthy parents of Chinese origin. She presented facial anomalies, micrognathia and skull malformations at birth, and was found hearing impairment, congenital heart disease and developmental delay during her first year of life. Her first visit to our center was at 1 year of age due to cardiovascular repair surgery for patent ductus arteriosus (PDA) and ventricular septal defect (VSD). Skull X-ray showed wormian bones. She returned at 7 years old after she developed progressive skeletal anomalies with fractures. She presented with multiple wormian bones, acro-osteolysis, severe osteoporosis, bowed fibulae and a renal cyst. Positive genetic test of a de novo heterozygous frameshift mutation in exon 34 of NOTCH2 (c.6426dupT) supported the clinical diagnosis of HCS.Conclusion: This is the second reported HCS case caused by the mutation c.6426dupT in NOTCH2, but presenting much earlier and severer clinical expression. Grain-size is one of the yield components, and the first 14 days after pollination (DAP) is a crucial stage for wheat grain-size formation. To understand the mechanism of grain-size formation at the whole gene expression level and to identify the candidate genes related to grain pattern formation, cDNA libraries from immature grains of 5 DAP and 14 DAP were constructed. According to transcriptome analysis, a total of 12,555 new genes and 9,358 differentially expressed genes (DEGs) were obtained. In DEGs, 2,876, 3,357 and 3,125 genes were located on A, B and D subgenome respectively. 9,937 (79.15%) new genes and 9,059 (96.80%) DEGs were successfully annotated. For DEGs, 4,453 were up-regulated and 4,905 were down-regulated at 14 DAP. The Gene Ontology (GO) database indicated that most of the grain-size-related genes were in the same cluster. The Kyoto Encyclopedia of Genes and Genomes (KEGG) database analysis showed that 130, 129 and 20 DEGs were respectively involved in starch and sucrose metabolism, plant hormone signal transduction and ubiquitin-mediated proteolysis. Expression levels of 8 randomly selected genes were confirmed by qRT-PCR, which was consistent with the transcriptome data. The present database will help us understand the molecular mechanisms underlying early grain development and provide the foundation for increasing grain-size and yield in wheat breeding programs. A fibre-based Mach-Zehnder (MZ) temperature sensor based on an ultra-long polymer waveguide integrated in an open-cavity multimode optical fibre is presented and analysed. A two-photon polymerization method was used to fabricate the polymer waveguide in the centre of the open-cavity. The air cavity and polymer waveguide constitute the reference arm and the sensing arm of the interferometer. Measurements show that the temperature sensitivity of the sensor can reach up to -205 pm/°C owing to the high thermo-optical coefficient of the polymer waveguide. We believe that the proposed sensor could have useful applications in biomedicine, environmental science, and other industries owing to its high sensitivity, biocompatibility, and good stability. Previous studies revealed that leptin induces the growth and proliferation and inhibits the apoptosis of lung cancer cells. However, the effect of leptin on epithelial-to-mesenchymal transition (EMT) is not yet clear. In the present study, the effect of leptin on EMT was investigated as well as its underlying mechanisms in A549 cells. The ability of leptin to induce EMT was investigated by microscopic examination and western blotting. The impacts of leptin on cell migration, invasion and tumorigenesis were evaluated by wound healing, Transwell and colony formation assays, respectively. It was demonstrated that leptin induced EMT-associated morphological changes, namely a decrease in cell-cell contact and a more elongated morphological shape. Leptin decreased the expression levels of epithelial phenotype markers E-cadherin and keratin, increased the expression of mesenchymal phenotype marker Vimentin, and raised the expression of EMT-induced transcription factor ZEB-1. In addition, leptin activated the extracellular signal regulated kinase (ERK) signaling pathway and did not affect the activation of the protein kinase B signaling pathway in A549 cells. Leptin also promoted EMT-induced migration, invasion and tumorigenesis in vitro in A549 cells. The present study provides evidence that leptin induced EMT via the activation of the ERK signaling pathway and increased EMT-induced tumor phenotypes in lung cancer cells. Meat traceability based on molecular markers is exerting a great influence on food safety and will enhance its key role in the future. This study aimed to investigate and verify the polymorphism of 23 microsatellite markers and select the most suitable markers for individual identification and meat traceability of six swine breeds in the Chinese market. The mean polymorphism information content value of these 23 loci was 0.7851, and each locus exhibited high polymorphism in the pooled population. There were 10 loci showing good polymorphism in each breed, namely, Sw632, S0155, Sw2406, Sw830, Sw2525, Sw72, Sw2448, Sw911, Sw122 and CGA. When six highly polymorphic loci were combined, the match probability value for two random individual genotypes among the pig breeds (Beijing Black, Sanyuan and Taihu) was lower than 1.151 E-06. An increasing number of loci indicated a gradually decreasing match probability value and therefore enhanced traceability accuracy. The validation results of tracing 18 blood and corresponding meat samples based on five highly polymorphic loci (Sw2525, S0005, Sw0107, Sw911 and Sw857) were successful, with 100% conformation probability, which provided a foundation for establishing a traceability system for pork in the Chinese market. Microsatellite markers have been shown to be a useful tool in individual identification and meat traceability. Aiming at developing a genetic tracing system for beef cattle breeds in the Chinese market, this study identified a set of 16 specific microsatellite markers within six breeds, including Japanese Black, Anduo yak, Limousin, Jiaxian Red, Nanyang Yellow and Luxi Yellow. A total of 180 alleles have been detected with an average number of 11.2 per locus, and the average polymorphism information content (PIC) is 0.7696 for all loci. The 16-loci set could successfully distinguish all the individuals of the six breeds. When the six most polymorphic markers were chosen for each breed, the matching probability (MP) value was found to be about seven in one million, excluding the extremely high value in Limousin. As the number of markers increased, the MP value was gradually lowered, and the accuracy was also enhanced. Meanwhile, the traceability validation test was conducted with the seven most polymorphic markers (ETH10, ETH225, ILSTS006, INRA032, INRA035, INRA037 and TGLA122), the conforming probabilities of genotypes for 28 blood and corresponding tissue samples were 100%. The results of this study could partly prevent the food fraud incidence in the Chinese market, and they also showed further evidence in the applications of genetic markers to meat traceability based on animal identification to ensure food safety. Light is a necessary environmental factor for fruit body formation and development of the cauliflower mushroom Sparassis latifolia, a well-known edible and medicinal fungus. In this study, we firstly characterized the SP-C strain, which belonged to S. latifolia. And then we cloned and sequenced a photoreceptor gene ( Slwc-1) from S. latifolia. The product of Slwc-1, SlWC-1 (872 aa residues) contained a coiled-coil region, a LOV domain, and two PAS domains. Phylogenetic tree result showed that SLWC-1 was most close to GfWC-1 from Grifola frondosa in edible and medicinal fungus. The Slwc-1 gene was found to be enhanced by light. The coiled-coil domain-containing protein 43 (CCDC43) is essential to promote gastric cancer (GC) proliferation and invasion, while four and a half LIM domains 1 (FHL1) involves GC cells apoptosis. We attempted to address inter-relationship between CCDC43 and FHL1 in modulating GC cells growth and apoptosis. Levels of protein expression were assessed by western blot, immunofluorescence. Using EdU, plate colony formation, Matrigel invasion and animal models, we evaluated the function in vitro and in vivo. Apoptosis was evaluated by flow cytometry and Hoechst 33258 staining. Reciprocal co-immunoprecipitation (co-IP) analyses indicated that CCDC43 physically interacted with FHL1. The expression of CCDC43 was negatively correlated with FHL1. Moreover, up-regulation of CCDC43 resulted in FHL1 level decline, and the reverse is also true. CCDC43 expressed jointly with FHL1 group significantly decreases the ability of the growth, metastasis and invasion of GC cells compared with that of the CCDC43 group. Furthermore, siRNA-mediated repression of CCDC43 results in dissociation from FHL1 and causes suppression of GC cell proliferation and metastasis. CCDC43 repression mediates the stability of FHL1 protein. In addition, CCDC43 interacts with FHL1. Knockdown of CCDC43 plus FHL1 overexpression inhibits proliferation and migration and induces apoptosis of GC cells in vitro and vivo. • CCDC43 interacts and colocates with FHL1 in GC. • The decrease of FHL1 by CCDC43 promotes the growth and migration of GC cells. • CCDC43 inhibits the stability of FHL1 protein and promotes apoptosis in GC cells. The ability to detect sequence-specific single-strand DNA (ssDNA) in complex, contaminant-ridden samples, using a fluorescent method directly without a DNA extraction and PCR step could simplify the detection of pathogens in the field and in the clinic. Here, we have demonstrated a simple label-free sensing strategy to detect ssDNA by employing its complementary ssDNA, S1 nuclease and nucleic acid fluorescent dyes. Upon clearing away redundant complementary ssDNA and possibly mismatched double strand DNA by using S1 nuclease, the fluorescent signal-to-noise ratio could be increased dramatically. It enabled the method to be adaptable to three different types of DNA fluorescent dyes and the ability to detect target ssDNA in complex, multicomponent samples, like tissue homogenate. The method can distinguish a two-base mismatch from avian influenza A (H1N1) virus. Also, it can detect the appearance of 50 pM target ssDNA in 0.5 µg·mL−1 Lambda DNA, and 50 nM target ssDNA in 5 µg·mL−1 Lambda DNA or in tissue homogenate. Abstract: Gold nanoparticles (AuNPs) based fluorescence quenching or colorimetric aptasensor have been developed for many analytes recently largely because of the ease of detection, high sensitivity, and potential for high-throughput analysis. However, the effects of remnant non-AuNPs components in the colloid gold solution on these assays performance remain unclear. For the first time, we demonstrated that the remnant sodium citrate and the reaction products of three acids play counteractive roles in AuNPs based fluorescence quenching and colorimetric aptasensor in three ways in this study. First, the remnant sodium citrate in the colloid gold solution could increase the fluorescence intensity of FAM labeled on the aptamer that reduce the efficiency of AuNPs fluorescent quenching. Second, the reaction products of citric acid, HCl and ketoglutaric acid reduce the fluorescence recovery by quenching the fluorescence of FAM labeled on the aptamer dissociated from the surface of AuNPs upon addition of target. Lastly, the reaction products of three acids reduce the pH value of the colloid gold solution that reduce the sensitivity of AuNPs based colorimetric aptasensor by increasing the adsorption of aptamer to surface of AuNPs. With sulfadimethoxine and thrombin as model analytes, we found that water resuspended AuNPs can significantly increase the sensitivity by more than 10-fold for AuNPs based fluorescence quenching aptasensor. In the AuNPs based colorimetric aptasensor for sulfadimethoxine using the water resuspended AuNPs, the sensitivity also was increased by 10-fold compared with that of original AuNPs. The findings in this study provide theoretical guidance for further improving AuNPs based fluorescent quenching and colorimetric aptasensor by adjusting the composition of AuNPs solution. A simple method to extract DNA from hair shafts was developed by using enzymatic laundry powder at the first step of the process. The whole extraction can be finished in less than 2 hours. The simple extraction reagent proposed here contains only two cheap components: ordinary enzymatic laundry powder and PCR buffer. After extraction, an ultra sensitive fluorescent nucleic acid stain, PicoGreen, was used for quantifying trace amount of double-stranded DNA in the solution extracted. For further validation of DNA extraction, four primers were employed to amplify DNA microsatellite loci. Both fluorescence spectroscopy and PCR results suggested that this method can extract DNA from hair shafts with good efficiency and repeatability. Platinum-based drugs that induce DNA damage are commonly used first-line chemotherapy agents for testicular, bladder, head and neck, lung, esophageal, stomach, and ovarian cancers. The inherent resistance of tumors to DNA damage often limits the therapeutic efficacy of these agents, such as cisplatin. An enhanced DNA repair and telomere maintenance response by the Mre11/Rad50/Nbs1 (MRN) complex is critical in driving this chemoresistance. We hypothesized therefore that the targeted impairment of native cellular MRN function could sensitize tumor cells to cisplatin. To test this, we designed what we believe to be a novel dominant-negative adenoviral vector containing a mutant RAD50 gene that significantly downregulated MRN expression and markedly disrupted MRN function in human squamous cell carcinoma cells. A combination of cisplatin and mutant RAD50 therapy produced significant tumor cytotoxicity in vitro, with a corresponding increase in DNA damage and telomere shortening. In cisplatin-resistant human squamous cell cancer xenografts in nude mice, this combination therapy caused dramatic tumor regression with increased apoptosis. Our findings suggest the use of targeted RAD50 disruption as what we believe to be a novel chemosensitizing approach for cancer therapy in the context of chemoresistance. Abstract: Background: The association of hepatitis B virus (HBV) genotypes with clinical course of infection is increasingly recognized. Objectives: In order to investigate the genetic diversity of HBV and its clinical implications, 241 HBV-infected patients including 34 with hepatocellular carcinoma (HCC) were enrolled in this study. Methods: HBV genotyping was performed with an ELISA assay. HBV subgenotypes were determined by PCR-RFLP. HBV core promoter/precore/core mutations were analyzed by direct sequencing. Results: The overall prevalence of HBV/B and C was 65% and 33%, respectively. Among HBV/C, 42% were Cs/C1 and 58% were Ce/C2. The HBV/C1 was only found in the patients originating from Southern China (p =0.0001). Among HCC patients, HBV/C2 was only found in the elder age group (≥51 years; p <0.05) and HBV/Ba was associated with young HCC patients (<35 years). Mutations associated with HCC were V1753 and T1762/A1764 (p <0.01). The prevalence of the V1753 was higher in HBV/C1 strains (p <0.04), A1898 was only found among HBV/C1 (p =0.056). T1762/A1764 was frequently demonstrated in both subgenotypes. The T1858 (90%) and A1896 (40%) mutations were most frequent in HBV/C2 (p <0.008). Conclusions: HBV/C1 and HBV/C2 have distinct geographic distributions in China. V1753 in addition to T1762/A1764 double mutation in the basal core promoter region seems to be associated with HCC development, especially in the patients with HBV/C1. [Copyright &y& Elsevier]

Light is an essential environmental factor for Sparassis latifolia primordia formation, but the molecular mechanism is still unclear. In this study, differential expression profiling of light-induced primordia formation (LIPF) was established by integrating the assay for transposase accessible chromatin by sequencing (ATAC-seq) and RNA-seq technology. The integrated results from the ATAC-seq and RNA-seq showed 13 down-regulated genes and 17 up-regulated genes in both the L vs. D and P vs. D groups, for both methods. According to the gene ontology (GO) annotation of these differentially expressed genes (DEGs), the top three biological process categories were cysteine biosynthetic process via cystathionine, vitamin B6 catabolic, and glycine metabolic; the top three molecular function categories were 5-methyltetrahydropteroyltriglutamate-homocysteine S-methyltransferase activity, glycine binding, and pyridoxal phosphate binding; cellular component categories were significantly enriched in the glycine cleavage complex. The KEGG (Kyoto Encyclopedia of Genes and Genomes) enrichment analysis revealed that these genes were associated with vitamin B6 metabolism; selenocompound metabolism; cysteine and methionine metabolism; glycine, serine, and threonine metabolism; and glyoxylate and dicarboxylate metabolism pathways. The expression of most of the DEGs was validated by qRT-PCR. To the best of our knowledge, this study is the first integrative analysis of ATAC-seq and RNA-seq for macro-fungi. These results provided a new perspective on the understanding of key pathways and hub genes in LIPF in S. latifolia.
