The purpose of this study was to investigate early stage dynamic changes in relevant indicators in neurocritical patients to identify biomarkers that can predict a poor prognosis at an early stage (1–4 days after admission). This study retrospectively collected clinical data, inflammatory indicators, and nutritional indicators from 77 patients at the neurology intensive care unit. The 3-month modified Rankin scale score was used as the outcome indicator. A linear mixed model was used to analyze changes in inflammatory indicators and nutritional indicators in neurocritical patients over time from 1–4 days after admission. Logistic regression was used to determine the independent risk factors for a poor prognosis in neurocritical patients and to construct a predictive model. The predictive efficacy of the model was verified using leave-one-out cross-validation and decision curve analysis methods. The analysis results showed that 1–4 days after admission, the inflammatory indicators of white blood cell and absolute monocyte counts and the nutritional indicators of body cell mass(BCM), fat-free mass, body cell mass/phase angle (BCM/PA), intracellular water, extracellular water, and skeletal muscle index increased overall, while the nutritional indicators of albumin and visceral fat area decreased overall. The logistic multivariate regression model showed that the Charlson comorbidity index (CCI) (odds ratio (OR) = 2.526, 95% CI [1.202, 5.308]), hemoglobin (Hb)(on admission)-Hb(min) (OR = 1.049, 95% CI [1.015, 1.083), BCM(on admission) (OR = 0.794, 95% CI [0.662, 0.952]), and the change in BCM/PA 1–4 days after admission (OR = 1.157, 95% CI [1.070, 1.252]) were independent risk factors for a poor prognosis in neurocritical patients. The predictive analysis showed that the predictive power of Model 1 with BCM/PA (area under the curve (AUC) = 0.95, 95% CI (0.90, 0.99)) was 93%, 65%, 141%, and 133% higher than that of Model 2 without BCM/PA, the CCI, the APACHE II score, and the NRS2002 score (all P < 0.05), respectively. The CCI, Hb(on admission)-Hb(min), BCM(on admission), and an increase in BCM/PA 1–4 days after admission were independently associated with a poor prognosis in neurocritical patients. Mapping soil organic carbon (SOC) accurately is essential for sustainable soil resource management. Hyperspectral data, a vital tool for SOC mapping, is obtained through both laboratory and satellite-based sources. While laboratory data is limited to sample point monitoring, satellite hyperspectral imagery covers entire regions, albeit susceptible to external environmental interference. This study, conducted in the Yuncheng Basin of the Yellow River Basin, compared the predictive accuracy of laboratory hyperspectral data (ASD FieldSpec4) and GF-5 satellite hyperspectral imagery for SOC mapping. Leveraging fractional order derivatives (FODs), various denoising methods, feature band selection, and the Random Forest model, the research revealed that laboratory hyperspectral data outperform satellite data in predicting SOC. FOD processing enhanced spectral information, and discrete wavelet transform (DWT) proved effective for GF-5 satellite imagery denoising. Stability competitive adaptive re-weighted sampling (sCARS) emerged as the optimal feature band selection algorithm. The 0.6FOD-sCARS RF model was identified as the optimal laboratory hyperspectral prediction model for SOC, while the 0.8FOD-DWT-sCARS RF model was deemed optimal for satellite hyperspectral prediction. The persistent utilization of synthetic nitrogen fertilizers poses a significant risk to the global climate, sustainable food production, and soil health. Exploiting the diverse assemblies of diazotrophic phyllosphere bacteria can reduce the severe risks of excessive use of artificial nitrogen fertilizers. Free-living diazotrophic bacteria in the leaf endosphere and episphere contribute tremendously to N input in terrestrial ecosystems and global N cycling. Understanding the ecological processes and biological mechanisms that shape N 2 -fixers and N 2 fixation in the phyllosphere will provide novel knowledge for future plant microbiome manipulation toward sustainable agricultural production. Recent advances in PANOMICS, gene editing, single-cell omics, and Raman spectroscopy have enhanced our understanding of the interactions between N 2 -fixers and plant hosts. The importance of biological nitrogen fixation (BNF) in securing food production for the growing world population with minimal environmental cost has been increasingly acknowledged. Leaf surfaces are one of the biggest microbial habitats on Earth, harboring diverse free-living N 2 -fixers. These microbes inhabit the epiphytic and endophytic phyllosphere and contribute significantly to plant N supply and growth. Here, we summarize the contribution of phyllosphere-BNF to global N cycling, evaluate the diversity of leaf-associated N 2 -fixers across plant hosts and ecosystems, illustrate the ecological adaptation of N 2 -fixers to the phyllosphere, and identify the environmental factors driving BNF. The corrosion behaviors of arc spraying 5083Al and Zn15Al coatings in a simulated marine environment were comparatively analyzed. The 5083Al coating exhibited lower corrosion rates and higher polarization resistance than the Zn15Al coating. Due to the lower solubility and higher supersaturation of Al(OH) 3 , the corrosion products of 5083Al coating presents a faster deposition rate and causes a denser and blocky corrosion products layer, which inhibits the penetration of Cl−, thereby enhancing corrosion resistances. Furthermore, in the case of coating cracking, the 5083Al coating, with lower potentials than EH36 steel matrix and lower self-discharge rate than the Zn15Al coating, acting as the sacrificial anode, is more suitable to provide durable protection for steels in marine environments. • 5083Al arc spraying coating has higher corrosion resistance than Zn15Al coating. • The faster deposition rate of Al(OH) 3 leads to a compact corrosion products layer. • 5083Al coating, acting as sacrificial anode, can protect steel matrix more durably. A coaxially integrated macro-micro composite actuator (MMCA) with large stroke and high accuracy is proposed by combining a voice coil motor (VCM) with the giant magnetostrictive actuator (GMA). The magnetic circuit model of the macro-motion part is established based on the driving principle of VCM, and the multi-field coupling model of the micro-motion part is established based on the Jiles-Atherton model. The finite element method was used to analyze the relationship between displacement, output force, velocity, acceleration, and time of the macro-motion part under different currents, the magnetic flux density, output force, and displacement curves of the micro-motion part, and the mutual influence between the macro and micro motion parts were analyzed. The prototype of the MMCA was developed, and an experimental test platform was built. The results show that the MMCA macro-motion displacement curve can fit the simulation curve well during open-loop positioning, and when the current size of the input macro-motion coil is 4 A, the experimental curve of the MMCA is the most consistent with the simulation curve. When closed-loop control, the motion curve of the drive can well follow the set displacement curve, in which the maximum stroke of the prototype developed is 50 mm, the positioning error of the macro-motion part is less than 20 μm, the maximum stroke of the micro-motion part is 40 μm, and the overall positioning accuracy of the MMCA is 0.14 μm. High-manganese steels, due to their unique combination of strength and elongation, have been widely used in aerospace, petrochemical and rail transportation. However, a prevalent challenge in advancing the utilization of high-manganese steel is the need for corresponding welding consumables. The deformation mechanism of high-manganese steel encompasses three primary mechanisms: martensitic phase transformation, twinning and dislocation movement. The stacking fault energy (SFE) is a critical factor in determining the dominant deformation mechanism in high-manganese austenitic steels. Furthermore, the magnitude of the SFE is principally influenced by the alloying elements present and the temperature at which deformation occurs. Alloying elements can significantly influence the microstructure and mechanical properties of wire arc additive manufacturing (WAAM) of high-manganese steels. The metal powder-cored wire of high-manganese steel with full austenitic microstructure was designed in this paper. The effects of C, Mn and Cu on the microstructure, solute segregation and properties of WAAM of high-manganese steels were systematically investigated by optical microscopy, electron microscopy and mechanical testing. The influence of SFE on the microstructure characteristics and work hardening behaviour were also studied. The results showed that as an increase of the C content, the tensile strength and elongation of deposited metals were improved. The corresponding low-temperature impact toughness increased at first and then decreased. The highest value of impact toughness was 68.5 J with 0.79%C. As the Mn increased, the strength decreased, the elongation increased and the low-temperature impact toughness value displayed an initial increase followed by a subsequent decrease. With the increase of Cu, the yield strength and elongation improved significantly. The tensile strength exhibited a marginal initial increase followed by a decrease, whereas the change was not substantial. In contrast, the low-temperature impact toughness value showed a substantial increase followed by a decrease. The developed M3 wire containing 1.10%C-21%Mn-0.3%Cu possessed the optimum performance (yield strength of 551 MPa, tensile strength of 909 MPa, elongation at break of 30.2%, impact toughness value of 57.5J), with the good mechanical stability and low solidification cracking sensitivity. ZnO:Al films were prepared by magnetron sputtering at room temperature. During the growth process, H radicals produced by radio frequency power sources were in situ introduced into ZnO:Al films. By injecting H radicals, the content of Al in ZnO:Al films increased from 3.4 at. % to 6.1 at. %, which could be ascribed to the reaction between H radicals and ZnO as well as the resputtering of the Zn element. Surface morphologies of ZnO:Al films were tuned from smooth shape to sunflower seed-like. Furthermore, ZnO:Al films without injecting H radicals showed highly c-axis preferential orientation and then nearly became amorphous with the addition of H radicals, which corresponds to the results of Raman spectra. The grain boundary potentials decreased due to the removal of oxygen at grain boundaries and an amount of Vo was produced, which was proved by Raman and x-ray photoelectron spectroscopy results. The optimal resistivity of as-grown ZnO:Al films co-doped with H radicals was 1.5 × 10−3 Ω cm at an H2 ratio of 2.3%. The variation of the carrier concentration for annealed ZnO:Al films at different H2 ratios supported the existence of H interstitials and Vo. The land surface albedo product of visible infrared imaging radiometer suite (VIIRS) in National Oceanic and Atmospheric Administration (NOAA)'s operational system provides real-time, global daily mean surface albedo, which is a required parameter in the estimation of the daily shortwave net radiation budget. The global gridded VIIRS albedo product is derived from the level-2 granule surface albedo product, which is generated using a Direct Estimation Method. Special gridding and compositing algorithms were developed for aggregating the granular albedo data into the gridded albedo product. This paper describes the design and evaluation of the NOAA VIIRS gridded daily surface albedo product. The cloudy condition, retrieval path, retrieval method, and observing geometry are the criteria in deciding the priority order in the composition processing. The proposed albedo product possesses a complete spatial coverage over global land and ice surface and provides a timely response to surface dynamics. The validation of satellite retrieved daily mean albedo against ground counterparts over a series of well-maintained networks demonstrates the reliability of the composed albedo considering the interference of the seasonal surface heterogeneity conditions around each site. The inter-comparison between the S-NPP and NOAA-20 VIIRS albedo shows good agreement, except for a minor bias related to solar/view angle differences. In this paper, Newton's iterative method to solve the nonlinear matrix equation is studied. For the given initial matrix Q, the main results that the matrix sequence generated by the iterative method is contained in a fixed open ball, and that the matrix sequence generated by the iterative method converges to the only solution of the nonlinear matrix equation in a fixed closed ball are proved. In the present study, diluted acetic acid was used for the pretreatment of rice straw in order to improve its bioconversion into biomethane. After pretreatment with 2% acetic acid for 24 h, cellulose and hemicellulose significantly reduced by 8.9% and 18.1%, respectively. The degradation of cellulose and hemicellulose resulted in significant increase of relative lignin ratio by 14.9%. Cumulative biomethane production after 28 days of anaerobic digestion of straw pretreated with 2%, 4% and 6% for 24 h was 24.3%, 15.2%, and 18.1%, respectively, higher than the untreated sample. Longer pretreatment for 36 and 48 h showed the same trend with relatively low biomethane yield. Thus, 24 h pretreatment with 2% acetic acid improved biomethanation over other pretreatments. Scanning electron microscopy and Fourier transform infrared spectroscopy confirmed the partial degradation of rice straw and modification of lignin structure by un-bonding the ether chain within lignin molecules as a result of pretreatment. Pretreatment with 2% acetic acid for 24 h resulted in the maximum biomethane energy yield of 6.29 MJ kg−1 dry straw, 47.3% energy conversion efficiency and the maximum net profit of US$ 106.4 ton−1 dry straw. In conclusion, application of 2% acetic acid for 24 h is a promising approach for high lignocellulose degradation efficiency at mild conditions due to the relatively short pretreatment time and enhanced biomethanation. Abstract The removal routes of pharmaceuticals especially biodegradation routes in the activated sludge process are still unclear. Some studies indicated pharmaceuticals were mainly removed via nitrification process (autotrophic biodegradation), while others suggested pharmaceuticals were mainly removed via COD degradation process (heterotrophic biodegradation). These unclear problems limited the improvements of pharmaceuticals removal. In this study, in order to elucidate three biodegradation routes (nitrification, COD degradation, or both nitrification and COD degradation), autotrophic and heterotrophic reactors were individually developed to separate nitrification and COD degradation form the activated sludge process (mix-trophic process including nitrification and COD degradation). Furthermore, the pharmaceuticals removal routes of adsorption, hydrolysis, and oxidation were also studied. Among six degradable pharmaceuticals, heterotrophic biodegradation and adsorption were the major removal routes. Two sulfonamides of five antibiotics were predominantly removed by COD degradation process, while nitrification and adsorption had no contributions. Adsorption, hydrolysis, nitrification, and COD degradation were the main elimination routes of cefalexin. COD degradation and adsorption were the dominant removal routes of norfloxacin. Tetracycline was mainly removed by the adsorption route, and hydrolysis and oxidation also played a role. For two drugs, ibuprofen was removed mainly via nitrification and COD degradation, and no adsorption occurred. Diclofenac could not be removed at all and was persistent in the aerobic conditions. Kinetic studies showed that biodegradation of the two sulfonamides, cefalexin, norfloxacin, and ibuprofen followed first-order kinetics rather than zero-order or second-order kinetics. Abstract A novel series of 2-(phenoxyaryl)-3-urea derivatives were designed, synthesized, and biologically evaluated for their anti-thrombotic activity. Most of compounds exhibited good inhibition against P2Y 1 receptor. Among them, three compounds 11 , 12 , and 13 demonstrated good P2Y 1 receptor antagonistic potency in vitro (IC 50 = 0.62 μM, 0.82 μM, and 0.21 μM, respectively). In antiplatelet aggregation study, four compounds 2 , 3 , 9 , and 13 showed good antiplatelet activity. The possible binding modes of compounds with P2Y 1 receptor were also explored by molecular docking simulation. The docking studies demonstrated that compound 13 interacted well with Phe119 through hydrophobic interaction and modestly improved the P2Y 1 receptor antagonistic activity, making it justifiable for further investigation. To explore the structural and electronic properties of SnSe under pressure, we applied hydrostatic pressure from 0 to 8 GPa to a fully relaxed SnSe cell sample based on plane-wave pseudopotential density functional theory. The calculated results indicate that the structure of SnSe changes gradually from an irregular zigzag structure with low symmetry to a B1-like structure with regular arrangement and high symmetry under pressure. The lattice parameters and cell volume of SnSe decrease monotonically as the applied pressure increases. The energy band gap of SnSe becomes narrow under pressure and is finally closed at 6.1 GPa. Moreover, we found that SnSe exhibits non-magnetic and semi-metallic features based on analyzing its electronic state density and spin state density. This can be attributed to the decrease in the lattices constants and the enhancement of the Sn-Se bond interaction under pressure, which causes the density of electronic states to increase near the Fermi surface. Finally, the charge distribution between Se-Sn-Se along the c-axis changes gradually from asymmetric to symmetric as the pressure is increased to 6.1 GPa and beyond. This implies that enhancement of the structure symmetry of SnSe can lead to a symmetrical distribution of charges, which further affects the bonding characteristics of the Sn-Se bond. Traditional Chinese medicine consists of complex phytochemical constituents. Selecting appropriate analytical markers of traditional Chinese medicine is a critical step in quality control. Currently, the combination of fingerprinting and efficacy evaluation is considered as a useful method for screening active ingredients in complex mixtures. This study was designed to develop an orthogonal partial least squares model for screening bioactive quality control markers of QishenYiqi dripping pills based on the fingerprint-efficacy relationship. First, the chemical fingerprints of 49 batches of QishenYiqi dripping pill samples were established by ultra-high performance liquid chromatography coupled with a photodiode array detector. Second, ultra-high performance liquid chromatography coupled with quadrupole-time-of-flight mass spectrometry was exploited to systematically investigate the 36 copossessing fingerprint components in QishenYiqi dripping pills. The vascular protective activity of QishenYiqi dripping pills was determined by using a cell counting kit-8 assay. Finally, fingerprint-efficacy relationship was established by orthogonal partial least squares model. The results indicated that ten components exhibited strong correlation with vascular protective activity, and these were preliminarily screened as quality control markers. In this paper we propose an iteration method to solve the multiple constrained least squares matrix problem. We first transform the multiple constrained least squares matrix problems into the multiple constrained matrix optimal approximation problem, and then we use the idea of Dykstra’s algorithm to derive the basic iterative pattern. We observe that we only need to solve multiple single constrained least squares matrix problems at each iteration step of the proposed algorithm. We give a numerical example to illustrate the effectiveness of the proposed method to solve the original problems. To increase the optical path of gas absorption in a multi-pass cell (MPC), a dense spot pattern has been obtained on MPC to improve the accuracy of trace gas detection. In this study, an algorithm was proposed to calculate the spot patterns of a new type of dense optical MPC. In this novel algorithm, the reflected ray equation and spherical equation were combined to obtain the position coordinates of light spots on a mirror surface. The calculation was repeated by changing the reflected optical path parameters until a new dense light spot pattern was obtained. To verify the accuracy of the proposed algorithm, a series of spot numerical simulations and methane (CH 4) concentration detection experiments were conducted. Results showed that in an MPC with a double spherical mirror, a series of dense spot patterns located on the mirror surface was obtained by calculating off-axis incident rays using the new algorithm, effectively increasing optical path length, and obtaining an MPC with a long optical path of 12.92 m. CH 4 concentration was detected by combining the long optical path MPC with a tunable diode laser absorption spectroscopy. A good linear relationship was established between the calibrated CH 4 concentration and the peak of the second harmonic signal detected by the system. The minimum detection limit of the system for CH 4 was 2.33 ppb at an integration time of 20 s obtained via Allan deviation analysis. This study demonstrated that a combined heterotrophic and autotrophic denitrification (HAD) process is highly effective for the simultaneous removal of acetate, nitrate, and sulfide at an efficiency of 100, 80, and 100 %, respectively. In the HAD system, simultaneous sulfide, acetate, and nitrate removals were observed, which indicated that heterotrophic and autotrophic denitrification occurred simultaneously. When the sulfide was existed in HAD reactor, the main product of sulfide biooxidation was S. Once the sulfide was exhausted, the sulfate concentration in the HAD reactor increased and became the main end product. These results provided an alternative method to control the end sulfide biooxidation product by online monitoring sulfide concentration. Nearly half (43 %) of the total clones in our mix-trophic reactor were amphitrophy denitrifiers. The autotrophic denitrifiers, heterotrophic denitrifiers, and amphitrophy denitrifiers coexisted in the HAD reactor to complete the denitrification process. Coarse-resolution satellite albedo products are important for climate change and energy balance research because of their capability to characterize the spatiotemporal patterns of land surface parameters at both the regional and global scales. The accuracy of coarse-resolution products is usually assessed via comparison with in situ measurements. The key issue in the comparison of remote sensing observations with in situ measurements is scaling and uncertainty. This paper presents a strategy for validating 1-km-resolution remote sensing albedo products using field measurements and high-resolution remote sensing observations. Field measurements were collected to calibrate the high-resolution (30 m) albedo products derived from HJ-1a/b images. Then, the calibrated high-resolution albedo maps were resampled (i.e., upscaled) to assess the accuracy of the coarse-resolution albedo products. The samples of field measurements and high-resolution pixels are based on an uncertainty analysis. Two types of coarse-resolution albedo datasets, from global land surface satellite (GLASS) and moderate-resolution imaging spectroradiometer (MODIS), are validated over the middle reaches of the Heihe River in China. The results indicate that the upscaled HJ (Huan Jing means environment in Chinese and this refers to a satellite constellation designed for environment and disaster monitoring by China) albedo, which was calibrated using field measurements, can provide accurate reference values for validating coarse-resolution satellite albedo products. Punicalagin, a bioactive ellagitannin isolated from pomegranate, has been reported to have anti-inflammatory property. In the present study, we analyzed the role of punicalagin against acute respiratory distress syndrome (ARDS) induced by lipopolysaccharide (LPS) in mice. Male BALB/c mice with ARDS, induced by intranasal instillation of LPS, were treated with punicalagin 1 h prior to LPS exposure. The effects of punicalagin on pro-inflammatory cytokines, myeloperoxidase activity, nuclear factor kappa B (NF-κB) activation, and the histopathological changes were evaluated. The results showed that punicalagin treatment attenuated LPS-induced lung edema, elevating TNF-α, IL-6, and IL-1β levels in the bronchoalveolar lavage fluid (BALF). Meanwhile, punicalagin significantly inhibited LPS-induced increases in the macrophage and neutrophil infiltration of lung tissues and myeloperoxidase activity. Furthermore, punicalagin inhibits Toll-like receptor 4 (TLR4) expression and NF-κB activation induced by LPS. In conclusion, this is the first study to demonstrate that punicalagin protects against LPS-induced ARDS in mice. Nitrite reduction is regulated by nitrite reductase encoded by nirK and nirS genes. This study aimed to investigate the abundance and composition of nirK- and nirS-containing denitrifiers in response to Spartina alterniflora invasion at the Jiulong River estuary, China. The sediment samples (depth: 0–5.0 and 5.1–20 cm) were collected from 3 vegetation zones, 1 dominated by the exotic plant S.alterniflora, 1 dominated by the native plant Kandelia candel, and 1 dominated by the native plant Cyperus malaccensis, and from an unvegetated flat zone. nirK- and nirS-containing denitrifier population sizes were lower in the invaded and nonvegetated zones than in those dominated by native K.candel and C.malaccensis, which were impacted by depth – vegetation species interaction. The ratios of nirS to nirK abundance ranged from 42.10 to 677.27, with the lowest ratio found for the upper layer in the invaded zone. The nirK-containing denitrifier compositions showed a 35% similarity between invaded zone and others. Most of the sequences of nirK genes recovered from the S.alterniflora zone were specific and distinct from those of nirK genes recovered from other vegetation types; nirS genes in the invaded zone were highly divergent. These results reveal that S.alterniflora invasion has a significant effect on the abundance and composition of both nirK- and nirS-containing denitrifiers, and nirS-containing denitrifiers were less responsive to invasion than nirK-containing denitrifiers. In this study, the cellulase gene celD from Clostridium thermocellum was cloned into expression vectors pET-20b(+) and pHsh. While high expression can be achieved by means of both these expression systems, only the pHsh expression system gives soluble proteins. By weakening the mRNA secondary structure and replacing the rare codons for the N-terminal amino acids of the target protein, the expression level of CelD was increased from 4.1 ± 0.3 to 6.4 ± 0.4 U ml in LB medium. Recombinant CelD was purified by heat treatment followed by Ni-NTA affinity. The purified CelD exhibited the highest activity at pH 5.4 and 60°C, and retained more than 50% activity after incubation at 70°C for 1 h. The cellulase activity of CelD was significantly enhanced by Ca but inhibited by EDTA. The favorable properties of CelD offer the potential for genetic modification of strains for biomass degradation. Presently, one of the major bottlenecks for industrial cellulase users is the high cost of enzyme production. The high level expression of soluble enzymes from the pHsh expression system offers a novel approach for the production of cellulases to be used in various agro-industrial processes such as chemical, food and textile. Abstract: The thermostable DNA ligase of Thermotoga maritima (Tma DNA ligase) was expressed in Escherichia coli, and purified by heat treatment followed by metal affinity chromatography. Purified Tma DNA ligase exhibited activity on DNA fragments with cohesive termini, and no activity was detected on blunt-end DNA. The ligase reaction required NAD+, and a divalent cation including Mg2+, Mn2+or Ca2+. The highest activity of Tma DNA ligase occurred at 60°C and pH 8.0 when Hind III digested plasmid was used as substrate. The purified enzyme had a half-life of over 30min at 95°C, and retained over 80% of its activity after holding a pH ranging from 7.2 to 8.8 for 1h at 80°C. When the enzyme was employed in PCR cycles, Tma DNA ligase promoted the amplification of long DNA fragments from the genomic DNA of T. maritima. [Copyright &y& Elsevier]

Abstract: The trifunctional enzyme (XAR–XYN) associating the Thermoanaerobacter ethanolicus xylosidase-arabinosidase (XAR) with the Thermomyces lanuginosus xylanase (XYN) was produced in E. coli to study the effect of the physical association of the fusion partners on the enzymatic efficiency. Recombinant XAR, XYN and XAR–XYN were purified to homogeneity and characterized. The optimal pH and temperature of the XAR–XYN were found to be similar to those of the XAR and XYN, except for less temperature optimum of α-arabinosidase activity. Its pH and xylanase activity exhibited more stable than those of the XAR and XYN. Finally, the XAR–XYN was tested for degradation of oat spelt xylan and wheat bran, the XAR–XYN was found to be more facile than the corresponding free enzyme degradation of wheat bran but provided little or no advantage on purified xylan. Furthermore cooperation within a trifunctional enzyme containing linker SAGSSAAGSGSG between each partner was achieved, leading to a trifunctional enzyme with enhanced enzymatic efficiency on arabinoxylan. The Ignavibacteria are ubiquitously abundant in paddy soils, but their functional role is poorly known. Here, we applied a targeted meta-genomic/transcriptomic approach to elucidate genetic potential and functional activity of the Ignavibacteria in straw-amended paddy soils from Italy. Eight high-quality metagenome-assembled genomes (MAGs) were recovered, being the largest ones (∅ 4.8 Mbp) among all known Ignavibacteria genomes (∅ 3.7 Mbp). Functional annotation revealed that the Ignavibacteria in Italian paddy soil are facultative anaerobes, represent a novel lineage in the Ignavibacteriae phylum, and have the ability for aerobic-hybrid phenylacetate degradation. Their putative functional roles under anoxic and micro-oxic conditions were assessed by mapping environmental mRNA onto the MAGs. Transcript analysis showed that primary fermentation of sugars to acetate and hydrogen is a major energy-yielding pathway under anoxic conditions. Although the phenylacetate degradation pathway is widely distributed among taxonomically diverse paddy soil bacteria, Ignavibacteria almost exclusively expressed this pathway under micro-oxic conditions. In particular, the transcript level of the paaABCDE gene cluster increased significantly. It encodes a monooxygenase, which is the key enzyme of phenylacetate degradation. This was linked to a significant decrease in transcripts encoding fermentation pathways and oxygen-sensitive hydrogenases, but increase in transcripts encoding TCA cycle, respiratory chain, and proteins involved in oxygen stress response (SOD1, AhpC, Bfr). More than half of the carbohydrate-active enzymes (CAZymes) expressed under anoxic and micro-oxic conditions possessed signal peptides, thereby indicating that Ignavibacteria contribute to the decomposition of complex polymers, such as cellulose, hemicellulose, and chitin. The Agro-pastoral ecotone of northern China is a typical ecologically vulnerable area. There is limited research on managing multiple ecosystem services to maximize supply based on understanding the complex relationships among ecosystem services. This study used the InVEST model to quantify habitat quality (HQ), soil conservation (SC), carbon sequestration (CS), and water yield (WY) in the Agro-pastoral ecotone of northern China in 2000, 2010, and 2020 at the 5 km grid and county scales. Their trade-off and synergistic effects were explored using Spearman correlation analysis and the Geographically Weighted Regression (GWR) model. In addition, self-organizing mapping was employed to identify ecosystem service bundles at two scales, whereas the geodetector was employed to determine the primary social-ecological drivers of ES. Finally, management strategies are proposed based on the research results to promote regional ecological sustainability. The results indicate that: (1) The four ESs exhibited an increasing trend during the study period. Except for WY, the other three ES demonstrated a similar spatial distribution while remaining relatively stable over time; (2) there is a synergistic effect between CS, HQ, and SC, whereas WY-related ES show trade-off effects; (3) six ES bundles were identified at the 5 km grid scale, and four ES bundles were identified at the county scale; (4) precipitation is a major driving factor for WY and an important driving factor for CS, HQ, and SC. In summary, natural factors have a greater impact on ES than socio-economic factors. Based on the differences in ecosystem service bundles and drivers at different scales, and in conjunction with China's existing environmental policies, corresponding ecological management strategies were proposed, potentially providing a reference for sustainable development of the Agro-pastoral ecotone in northern China. Background: Definitive concurrent chemoradiotherapy (CCRT) is the standard treatment for locally advanced, inoperable non-small cell lung cancer (NSCLC). Previous studies have mainly focused on examining local failure and recurrence patterns after surgery and the principles of lymph node metastasis (LNM) in surgical candidates with NSCLC. However, these studies were just only able to guide postoperative radiotherapy (PORT) and the patterns of LNM in patients with resected NSCLC was inadequate to represent that in locally advanced inoperable NSCLC patients for guiding target volume delineation of CCRT. In this study, we aimed to analyze the metastasis regularities and establish the correlations between different lymph node levels in NSCLC patients without any intervention using positron emission tomography/computed tomography (PET/CT) images. Methods: Overall, 358 patients with N1–N3 NSCLC admitted in our hospital between 2018 and 2022 were retrospectively analyzed. The diagnosis of metastatic lymph nodes was reviewed and determined using the European Organization for Research and Treatment of Cancer standard and the standardized value of the PET/CT examination. Univariate and multivariate analysis were performed to investigate the correlations between the different levels were evaluated by using of the chi-square test and logistic regression model. Results: The lymph nodes with the highest metastasis rates in patients with left lung cancer were in order as follows: 10L, 4L, 5, 4R, and 7; while in those with right lung cancer they were 10R, 4R, 7, 2R, and 1R. Notably, we found left lung patients were more likely to have contralateral hilar, mediastinal and supraclavicular lymph nodes involved, and the right lung group exhibited a higher propensity for ipsilateral mediastinum and supraclavicular lymph node invasion. Furthermore, correlation analysis revealed there were significant correlative patterns in the LNM across different levels. Conclusions: This study elucidated the patterns of primary LNM in patients with NSCLC who had not undergone surgery (without any treatment interventions) and the correlations between lymph node levels. Highly transparent indium-free multilayers of TiO2/Cu/TiO2 were obtained by means of annealing. The effects of Cu thickness and annealing temperature on the electrical and optical properties were investigated. The critical thickness of Cu mid-layer with optimal electrical and optical properties was 10 nm, with the figure of merit reaching as high as 5 × 10−3 Ω−1. Partial crystallization of the TiO2 layer enhanced the electrical and optical properties upon annealing. Electrothermal experiments showed that temperatures of more than 100 °C can be reached at a heating rate of 2 °C/s without any damage to the multilayers. The ecologically fragile agro-pastoral ecotone in northern China is characterized by relatively poor arable land quality. Yunzhou District in Datong City, which is situated within this transitional zone, boasts over 600 years of Hemerocallis citrina Baroni cultivation. Exploring the effects of soil physicochemical properties on daylily yield and related agronomic traits is essential for enhancing the ecological and economic value of dominant crops in ecologically fragile areas. Therefore, in this study, we focused on the daylily, a characteristic cash crop that is grown in the agro-pastoral ecotone in Yunzhou District. Physicochemical property measurement and yield estimation were performed using soil samples collected from 37 sites, with Spearman's correlation analysis, one-way analysis of variance with multiple comparisons, path analysis, and stepwise regression analysis used to analyze the generated data. The results showed the following: (1) The pathway analysis of daylily yield with each agronomic trait showed that the BN and PH directly affected the yield of daylily with direct pathway coefficients of 0.844 and 0.7, respectively, whereas the SN indirectly affected the yield of daylily through the BD and PH, with indirect pathway coefficients of 0.827 and 0.566, respectively. (2) A total of four principal components were extracted for the soil factors, of which SMC, ST and BD had large loadings on PC1; OM, TN and pH had large loadings on PC2; AK had large loadings on PC3; and AP had large loadings on PC4. (3) From the principal component regression and stepwise regression, it can be seen that SMC is the most critical factor affecting the yield of daylily, as well as the related agronomic traits, and the results also show that yield prediction was affected by OM, ST, and AK, while BN was influenced by OM and ST, and SN and PH were influenced by AP. Ice albedo feedback amplifies climate change signals and thus affects the global climate. Global long-term records on sea-ice albedo are important to characterize the regional or global energy budget. As the successor of MODIS (Moderate Resolution Imaging Spectroradiometer), VIIRS (Visible Infrared Imaging Radiometer Suite) started its observation from October 2011 on S-NPP (Suomi National Polar-orbiting Partnership). It has improved upon the capabilities of the operational Advanced Very High Resolution Radiometer (AVHRR) and provides observation continuity with MODIS. We used a direct estimation algorithm to produce a VIIRS sea-ice albedo (VSIA) product, which will be operational in the National Oceanic and Atmospheric Administration's (NOAA) S-NPP Data Exploration (NDE) version of the VIIRS albedo product. The algorithm is developed from the angular bin regression method to simulate the sea-ice surface bidirectional reflectance distribution function (BRDF) from physical models, which can represent different sea-ice types and vary mixing fractions among snow, ice, and seawater. We compared the VSIA with six years of ground measurements at 30 automatic weather stations from the Programme for Monitoring of the Greenland Ice Sheet (PROMICE) and the Greenland Climate Network (GC-NET) as a proxy for sea-ice albedo. The results show that the VSIA product highly agreed with the station measurements with low bias (about 0.03) and low root mean square error (RMSE) (about 0.07) considering the Joint Polar Satellite System (JPSS) requirement is 0.05 and 0.08 at 4 km scale, respectively. We also evaluated the VSIA using two datasets of field measured sea-ice albedo from previous field campaigns. The comparisons suggest that VSIA generally matches the magnitude of the ground measurements, with a bias of 0.09 between the instantaneous albedos in the central Arctic and a bias of 0.077 between the daily mean albedos near Alaska. The discrepancy is mainly due to the scale difference at both spatial and temporal dimensions and the limited sample size. The VSIA data will serve for weather prediction applications and climate model calibrations. Combined with the historical observations from MODIS, current S-NPP VIIRS, and NOAA-20 VIIRS observations, VSIA will dramatically contribute to providing high-accuracy routine sea-ice albedo products and irreplaceable records for monitoring the long-term sea-ice albedo for climate research. Bathyarchaeia (formerly Bathyarchaeota) is a group of highly abundant archaeal communities that play important roles in global biogeochemical cycling. Bathyarchaeia is predominantly found in sediments and hot springs. However, their presence in arable soils is relatively limited. In this study, we aimed to investigate the spatial distributions and diversity of Bathyarchaeia in paddy soils across eastern China, which is a major rice production region. The relative abundance of Bathyarchaeia among total archaea ranged from 3 to 68% in paddy soils, and Bathy-6 was the dominant subgroup among the Bathyarchaeia (70–80% of all sequences). Bathyarchaeia showed higher migration ability and wider niche width based on the neutral and null model simulations. Bathy-6 was primarily assembled by deterministic processes. Soil pH and C/N ratio were identified as key factors influencing the Bathyarchaeia composition, whereas C/N ratio and mean annual temperature influenced the relative abundance of Bathyarchaeia. Network analysis showed that specific Bathyarchaeia taxa occupied keystone positions in the archaeal community and co-occurred with some methanogenic archaea, including Methanosarcina and Methanobacteria, and ammonia-oxidizing archaea belonging to Nitrososphaeria. Methane generation has a significant impact on climate change. Aquatic plants have been widely studied for their role in methane emissions. Most research has focused on the contribution of the plant itself whereas the potential involvement of the phyllosphere microbiota associated with aquatic plants in methanogenesis remains largely unexplored. This study investigated the phyllosphere microbiota on both the top and bottom sides of lotus (Nelumbo nucifera), a commonly found aquatic plant in Asia, sampled from two urban ponds, using sequencing for both the 16S rRNA gene and the methanogenesis functional marker gene. Methanogenesis potential was assessed through laboratory-based 13C labeling experiments. Results show that methanogenic archaea were successfully detected on the floating lotus leaves but not on the standing leaves. Among the methanogens detected, Methanobacterium was identified as the predominant group, with significantly higher relative abundance on the bottom sides of floating leaves compared to the surrounding pond water. This study demonstrates the metabolic activity of the phyllosphere methanogens of floating lotus. The microbiota associated with the bottom sides of floating leaves exhibited the ability to produce methane and incorporate 13C carbon into methane during incubation. Overall, this study provides the first compelling genetic and functional evidence for the existence of epiphytic methanogens on the phyllosphere of aquatic plants. Hence, the phyllosphere of floating-leaved macrophytes represents a previously unrecognized habitat for methanogens and a potential microbial source of methane at the water–atmosphere interface. To investigate the risk factors and predictive model of diarrhea among patients with severe stroke. The study analyzed the retrospective clinical data of patients with new-onset stroke who had been admitted to the intensive care unit at the Department of Neurology of X Hospital, between September 2017 and April 2018. All data were analyzed with a binary logistic regression, and a logistic regression equation was used to build a predictive model of diarrhea among patients with severe stroke. A total of 153 patients with severe stroke were included in this study, including 45 patients (29.41%) with diarrhea. The binary logistic multivariate analysis showed that the National Institutes of Health Stroke Scale score at admission (odds ratio [OR], 1.123; 95% confidence interval [CI], 1.016–1.242), the Glasgow Coma Scale score at admission (OR, 1.563; 95% CI, 1.048–2.330), antibiotic use (OR, 2.168; 95% CI, 1.041–4.514), gavage feeding time (OR, 1.260; 95% CI, 1.098–1.445), and hospital stay before the occurrence of diarrhea (OR, 0.652; 95% CI, 0.552–0.770). The receiver operating characteristic curve was 0.862 (95% CI, 0.799–0.925), the specificity was 0.778, and the sensitivity was 0.843. The National Institutes of Health Stroke Scale score at admission, the Glasgow Coma Scale score at admission, antibiotic use, gavage feeding time, and hospital stay before the occurrence of diarrhea independently predict diarrhea among patients with severe stroke. A recent study published in the journal Nature used a data-driven machine-learning approach to examine the relationship between microbial carbon use efficiency (CUE) and soil organic carbon (SOC) storage. The study concluded that CUE promotes global SOC storage based on a positive correlation between CUE and SOC content. However, the study has been criticized for excluding important data sets and lacking mechanistic consideration of the complexities of SOC formation. The critics argue that the stabilization process, rather than CUE, matters more for SOC formation. They also caution against inferring causality from large data sets and emphasize the need for more sophisticated empirical measurements before establishing a globally causal link between CUE and SOC. When grown according to the conventional one-step annealing process at 550 °C, WO3 exhibits low photocurrent density and inferior photocurrent retention. In contrast, after a two-step annealing process at first 550 °C and second 700 °C, WO3 contains a higher concentration of oxygen deficiencies acting as shallow donors, thus leading to improved charge separation. Importantly, this results in a substantial photocurrent increase and higher material stability, two criteria that are of utmost importance for efficient photoelectrochemical water splitting. Objectives: Intestinal mucositis is the major side effect during abdominal or pelvic radiotherapy, but the underlying immunogen remains to be further characterised and few radioprotective agents are available. This study investigated the role of dsDNA‐triggered inflammasomes in intestinal mucositis during radiotherapy. Methods: Pro‐inflammatory cytokines were detected by ELISA. Radiation‐induced intestinal injury in mice was analyzed by means of survival curves, body weight, HE staining of intestines, and intestinal barrier integrity. Western blot, immunofluorescence staining, co‐immunoprecipitation assay and flow cytometry were used to investigate the regulatory role of dsDNA on inflammasomes. Results: Here, we show that a high level of IL‐1β and IL‐18 is associated with diarrhoea in colorectal cancer (CRC) patients during radiotherapy, which accounts for intestinal radiotoxicity. Subsequently, we found that the dose‐dependently released dsDNA from the intestinal epithelial cells (IECs) serves as the potential immunogenic molecule for radiation‐induced intestinal mucositis. Our results further indicate that the released dsDNA transfers into the macrophages in an HMGB1/RAGE‐dependent manner and then triggers absent in melanoma 2 (AIM2) inflammasome activation and the IL‐1β and IL‐18 secretion. Finally, we show that the FDA‐approved disulfiram (DSF), a newly identified inflammasome inhibitor, could mitigate intestinal radiotoxicity by controlling inflammasome. Conclusion: These findings indicate that the extracellular self‐dsDNA released from the irradiated IECs is a potential immunogen to stimulate immune cells and trigger the subsequent intestinal mucositis, while blunting the dsDNA‐triggered inflammasome in macrophages may represent an exciting therapeutic strategy for side effects control during abdominal radiotherapy. The effect of different heat inputs of 1.45 kJ/mm, 1.78 kJ/mm and 2.31 kJ/mm on the microstructure and mechanical properties of deposited metals of the self-developed AWS A5.28 E120C-K4 high strength steel flux-cored wire was studied by optical microscope, scanning electron microscope and mechanical property test. With the increase in heat input, the results showed that the microstructure of deposited metals became coarse. Acicular ferrite increased at first and then decreased, granular bainite increased and degenerated upper bainite and martensite decreased slightly. Under the low heat input of 1.45 kJ/mm, the cooling rate was fast and the element diffusion was uneven, which caused composition segregation and easy to form large size inclusions SiO2-TiC-CeAlO3 with weak binding to the matrix. Under the middle heat input of 1.78 kJ/mm, the composite rare earth inclusions in dimples were mainly TiC-CeAlO3. The dimples were small and uniformly distributed, and the dimple fracture mainly depended on the wall-breaking connection between medium-sized dimples rather than an intermediate media. Under the high heat input of 2.31 kJ/mm, SiO2 was easy to adhere to high melting point Al2O3 oxides to form irregular composite inclusions. Such irregular inclusions do not need to absorb too much energy to form necking. Finally, the integrated effects of microstructure and inclusions resulted in the optimum mechanical properties of deposited metals with a heat input of 1.78 kJ/mm, which was a tensile strength of 793 MPa and an average impact toughness at −40 °C of 56 J. Land surface albedo (LSA) is an essential component of the surface radiation budget, and has been retrieved extensively as a basic remote sensing product; however, daily LSA products suffer from extensive data gaps primarily caused by cloud cover. Accordingly, several gap-filling methods were developed (e.g., spatiotemporal interpolation and data fusion with albedo climatology), although the traditional methods are limited by cloud scale and surface heterogeneity. Further, as the largest varying surface landscape feature, seasonal snow cover substantially influences LSA and represents a major uncertainty factor of gap recovery because previous studies failed to employ actual surface signals to capture such ephemeral but intense albedo changes under cloud cover. To address this issue, a three-step framework was proposed for estimating 1 km cloudy-sky LSA using passive microwave (PMW) data, albedo climatology, and Visible Infrared Imaging Radiometer Suite (VIIRS) clear-sky albedo: (1) All-sky snow albedo was estimated from PMW brightness temperatures using a statistical model, (2) Continuous albedo dynamics were generated by combining the all-sky snow albedo with snow-free climatological albedo, and (3) The 1 km cloudy-sky LSA was predicted after filtering 1 km VIIRS clear-sky LSA by the albedo dynamic series. PMW-derived snow albedo was assessed over the Contiguous US (CONUS), and the final 1 km cloudy-sky LSA was validated across 10 sites from SURFRAD and Core AmeriFlux in 2013. Based on the comparison with high-quality MODIS pixels, the estimated snow albedo yielded an overall RMSE of 0.064 over CONUS, with a bias of −0.010 (R2 = 0.845). The recovered 1 km cloudy-sky LSA produced RMSEs of 0.074 (0.137) for all (snow) samples, a significant improvement over the Global Land Surface Satellite (GLASS) gap-free albedo products especially on snow cases (p-value = 0.027). Corresponding RMSE in calculating surface net radiation was also decreased by 38.91 W·m−2; and anomalous snow samples were corrected as well. The temporal analysis and all-sky LSA mapping suggest that the recovered LSA has satisfactory spatiotemporal continuity, and successfully captured details of spatiotemporal variability, especially for ephemeral snow events. This study provides an innovative solution to recover gaps in LSA data, and considerably improves the LSA accuracy under cloud cover, which can inform snow melting modeling, hazard forecasting, and irrigation management. Iron oxide @ biochar (FeO/C) promotes bacterial growth and facilitates electron transfer, thereby effectively promoting malathion degradation by Shewanella oneidensis MR-1 (S. oneidensis MR-1). This study elucidated the underlying mechanism of FeO/C-enhanced malathion degradation by S. oneidensis MR-1 through a combination of metabolomics and proteomics analysis. The kinetic fitting results from the degradation experiment indicated that 0.1 g/L FeO/C exerted the most significant enhancement effect on malathion degradation by S. oneidensis MR-1. Observations from Scanning Electron Microscopy and Laser Scanning Confocal Microscopy, along with physiological and biochemical analysis, showed that FeO/C enhanced the growth and oxidative response of S. oneidensis MR-1 under malathion stress. In addition, metabolomics and proteomics analysis revealed an increase in certain electron transfer related metabolites, such as coenzymes, and the upregulation of proteins, including coenzyme A, sdhD, and petC. Overall, spectroscopic analysis suggested that Fe2+, which was reduced from Fe3+ by S. oneidensis MR-1 in FeO/C, promoted electron transfer in S. oneidensis MR-1 to enhance the degradation of malathion. The Visible Infrared Imaging Radiometer Suite (VIIRS) Land Surface Temperature (LST) has been operationally produced for a decade since the Suomi National Polar-orbiting Partnership (SNPP) launched in October 2011. A comprehensive evaluation of its accuracy and precision will be helpful for product users in climate studies and atmospheric models. In this study, the VIIRS LST is validated with ground observations from multiple high-quality radiation networks, including six stations from the Surface Radiation budget (SURFRAD) network, two stations from the Baseline Surface Radiation Network (BSRN), and 13 stations from the Atmospheric Radiation Measurement (ARM) network, to evaluate its performance over various land-cover types. The VNP21A1 LST was validated against the same ground observations as a reference. The results yield a close agreement between the SNPP VIIRS LST and ground LSTs with a bias of −0.4 K and a RMSE of 1.96 K over six SURFRAD sites; a bias of −0.2 K and a RMSE of 1.93 K over two BSRN sites; and a bias of −0.1 K and a RMSE of 1.7 K over the 13 ARM sites. The time series of the LST errors over individual sites indicate seasonal cycles. The data anomaly over the BSRN site in Cabauw and the SURFRAD site in Desert Rock is revealed and discussed in this study. In addition, a method using Landsat-8 data is applied to quantify the heterogeneity level of each ground station and the results provide promising insights. This work investigated the effects of retained austenite (RA) stability on electrochemical activity and stress corrosion cracking (SCC) of ultrafine bainitic (UFB) steel in marine environment. Under pre-strained condition, RA underwent transformation and strain-induced martensite formed, which increased the number of crystal defects in steels. Moreover, the micro-galvanic effect and the high dislocation density in the multiphase complex structure increased the electrochemical activity, which further promoted the SCC susceptibility and crack initiation of the pre-strained steels. However, high pre-strain promoted the transformation of highly stable film-like retained austenite and formed plastic lath martensite, which relatively delayed the crack propagation. Background and aims: Harnessing soil microbiomes is a major demand for development of sustainable and productive agriculture. Here we aimed to assess the impact of two different types of organic material amendments in combination with chemical fertilizer on the plant-soil microbiota in maize farming and its link to soil fertility and crop productivity. Methods: Soils and roots were collected from a long-term wheat-maize rotation system involving three experimental treatments: chemical fertilizer (CF); chemical fertilizer plus seasonal application of manure (OM); and chemical fertilizer plus one-time application of peat and vermiculite (PV). Crop residues were returned in all three treatments each season. Bacterial 16S rRNA gene and fungal ITS sequencing were conducted to elucidate the treatment-specific response of the microbiota in bulk soil, rhizosphere soil, and root compartment. Results: Relative to CF and OM treatments, PV amendment led to significant increases in soil organic carbon (SOC) content, aboveground plant biomass, and grain yield over the five-year field study. The PV-induced changes in microbial composition involved the greatest treatment-specific "effect size" on indicator ASVs (amplicon sequence variants) in bulk and rhizosphere soils. The number of interactions was more than doubled in the PV co-occurrence network relative to those in the CF and OM co-occurrence networks. Potential beneficial microbes, such as Glomeromycota (arbuscular mycorrhiza), Basidiomycota, and various members of the Actinobacteria and Burkholderiales, were most enriched in the root compartment of the PV treatment. Conclusions: Peat-vermiculite enhanced microbiota-driven soil fertility and crop productivity, thereby providing new insights into plant-soil-microbiota interactions that can be harnessed for smart farming. Previous studies have reported the correlations between bacterial communities and coral bleaching, but the knowledge of fungal roles in coral bleaching is still limited. In this study, the taxonomic and functional diversities of fungi in unbleached, partly bleached and bleached stony coral Acropora intermedia were investigated through the ITS-rRNA gene next-generation sequencing. An unexpected diversity of successfully classified fungi (a total of 167 fungal genera) was revealed in this study, and the partly bleached coral samples gained the highest fungal diversity, followed by bleached and unbleached coral samples. Among these fungi, 122 genera (nearly 73.2%) were rarely found in corals in previous studies, such as Calostoma and Morchella , which gave us a more comprehensive understanding of coral-associated fungi. Positively correlated fungal genera (Calostoma , Corticium , Derxomyces , Fusicolla , Penicillium and Vishniacozyma) and negative correlated fungal genera (Blastobotrys , Exophiala and Dacryopinax) with the coral bleaching were both detected. It was found that a series of fungal genera, dominant by Apiotrichum , a source of opportunistic infections, was significantly enriched; while another fungal group majoring in Fusicolla , a probiotic fungus, was distinctly depressed in the bleached coral. It was also noteworthy that the abundance of pathogenic fungi, including Fusarium , Didymella and Trichosporon showed a rising trend; while the saprotrophic fungi, including Tricladium , Botryotrichum and Scleropezicula demostrated a declining trend as the bleaching deteriorating. The rising of pathogenic fungi and the declining of saprotrophic fungi revealed the basic rules of fungal community transitions in the coral bleaching, but the mechanism of coral-associated fungal interactions still lacks further investigation. Discovering more novel antimicrobial compounds has become a keen research problem. In this study, YA215 genome was sequenced by the Illumina HiSeq + PacBio sequencing platform. Genome assembly was performed by Unicycler software and the gene clusters responsible for secondary metabolite biosynthesis were predicted by antiSMASH. The genome comprised 3976514 bp and had a 46.56% G + C content. 3809 coding DNA sequences, 27 rRNAs, 86 tRNAs genes, and 79 sRNA were predicted. Strain YA215 was re-identified as Bacillus velezensis based on ANI and OrthoANI analysis. In the COG database, 23 functional groups from 3090 annotations were predicted. In the GO database, 2654 annotations were predicted. 2486 KEGG annotations linked 41 metabolic pathways. Glycosyl transferases, polysaccharide lyases, auxiliary activities, glycoside hydrolases, carbohydrate esterases, and carbohydrate-binding modules were predicted among the 127 annotations in the CAZy database. AntiSMASH analysis predicted that B. velezensis YA215 boasted 13 gene clusters involved in synthesis of antimicrobial secondary metabolites including surfactin, fengycin, macrolactin H, bacillaene, difficidin, bacillibactin, bacilysin, and plantazolicin. Three of the gene clusters (gene cluster 5, gene cluster 9, and gene cluster 10) have the potential to synthesize unknown compounds. The research underscore the considerable potential of secondary metabolites, identified in the genomic composition of B. velezensis YA215, as versatile antibacterial agents with a broad spectrum of activity against pathogenic bacteria. Summary: Autotrophic carbon dioxide (CO2) fixation by microbes is ubiquitous in the environment and potentially contributes to the soil organic carbon (SOC) pool. However, the multiple autotrophic pathways of microbial carbon assimilation and fixation in paddy soils remain poorly characterized. In this study, we combine metagenomic analysis with 14C‐labelling to investigate all known autotrophic pathways and CO2 assimilation mechanisms in five typical paddy soils from southern China. Marker genes of six autotrophic pathways are detected in all soil samples, which are dominated by the cbbL genes (67%–82%) coding the ribulose‐bisphosphate carboxylase large chain in the Calvin cycle. These marker genes are associated with a broad range of phototrophic and chemotrophic genera. Significant amounts of 14C‐CO2 are assimilated into SOC (74.3–175.8 mg 14C kg−1) and microbial biomass (5.2–24.1 mg 14C kg−1) after 45 days incubation, where more than 70% of 14C‐SOC was concentrated in the relatively stable humin fractions. These results show that paddy soil microbes contain the genetic potential for autotrophic carbon fixation spreading over broad taxonomic ranges, and can incorporate atmospheric carbon into organic components, which ultimately contribute to the stable SOC pool. Humic acids (HAs) can function as electron mediators for contaminants transformation in different environments. The humus respiration can facilitate pentachlorophenol (PCP) dechlorination during different biowastes composting. However, different characteristics of synthetic pathways of redox functional groups within HAs during different biowastes composting have never been characterized. Herein, we assessed the synthetic pathways of redox functional groups within HAs from protein-, lignocellulose-, and lignin-rich composts that facilitated the microbially reductive dechlorination of PCP, respectively. The results show that the aromatic systems are the major electron-accepting moieties of HAs and function as electron shuttles to facilitate the PCP dechlorination. Amino acid and reducing sugar are the major precursors for the synthesis of redox functional groups within HAs in protein-rich composts, and polyphenols and amino acids are discerned as the significant components to synthesize redox functional groups of HAs in lignocellulose- and lignin-rich composts. Seven groups of bacterial communities based on relationships among remarkable precursors, key bacterial communities, and redox functional groups within HAs are classified as participants in the precursors' catabolism and aromatic system' anabolism. Furthermore, the significant environmental factors on the synthetic pathways of redox functional groups within HAs in composting are confirmed by structural equation models. Conclusively, the regulating methods for promoting PCP dechlorination by HAs during different biowastes composting are proposed. Our results can help in understanding the distinct formative mechanisms of redox functional groups within HAs during different biowastes composting, providing insights into a classification-oriented approach for recycling utilization of different biowastes.
