The recent growing interest in the biological properties of lichen metabolites has evidenced different needs and challenges for further exploration, including the development of green processing with safer solvents and more efficient use of energy. Microwave assisted hydrothermal processing, applied after supercritical CO2 extraction, was proposed for the sequential extraction of bioactives and biopolymer fractions. Alternatively, it was combined with natural deep eutectics (NaDES) as cosolvents. Lichenic acids, antioxidants and oligosaccharides were simultaneously extracted using NaDES, and the recovered polysaccharides showed adequate mechanical properties for the formulation of films with antimicrobial action against Gram positive bacteria. An environmental assessment of the three different processes using the Eco-Scale suggested that the NaDES microwave extraction was, due to its low toxicity and good extraction yield of polysaccharides, the most sustainable of the three processes. This study aimed to obtain a betalain-rich extract (BRE) from Opuntia ficus-indica peels and protect them through a holistic approach involving microencapsulation via spray-drying. To enhance the stability of the microencapsulated betalains, an antioxidant-rich extract (ARE) obtained through hydrothermal extraction from the residual material remaining after the conventional extraction of betalains was used as a stabilizer during the spray-drying process. The antioxidant activity of the ARE was evaluated by measuring its IC 50 value for 2,2-Diphenyl-1-picrylhydrazyl (DPPH) of 65.8 μg/mL, its Trolox equivalent antioxidant capacity (TEAC) of 60.4 g Trolox eq. /100 g, and its reducing power of 16.6 g butylated hydroxytoluene (BHT) eq./100 g. Spray-drying parameters were optimized using a Box-Behnken design to enhance the total betalain content and minimize by-product formation. The results demonstrated that the addition of the ARE significantly improved the stability of betalains during spray-drying, leading to the formation of microparticles with high betalain content and enhanced stability. The optimal conditions (0.72% starch, 1.50% BRE and 0.25% ARE) resulted in microparticles with a maximum concentration of 6.74 ± 0.09 mg Betalains /g. The predictive model exhibited high accuracy in estimating experimental values. This research provides a natural and sustainable solution for food coloring by effectively utilizing discarded Opuntia ficus-indica peel. Orbital shaking in a glass vial is a commonly used forced degradation test to evaluate protein propensity for agitation-induced aggregation. Vial shaking in horizontal orientation has been widely recommended to maximize the air-liquid interface area while ensuring solution contact with the stopper. We evaluated the impact of shaking orbit diameter and frequency, and glass vial orientation (horizontal versus vertical) on the aggregation of three proteins prepared in surfactant-free formulation buffers. As soon as an orbit-specific frequency threshold was reached, an increase in turbidity was observed for the three proteins in vertical orientation only when using a 3 mm agitation orbit, and in horizontal orientation only when using a 30 mm agitation orbit. Orthogonal analyses confirmed turbidity was linked to protein aggregation. The most turbid samples had a visually more homogeneous appearance in vertical than in horizontal orientation, in line with the predicted dispersion of air and liquid phases obtained from computational fluid dynamics agitation simulations. Both shaking orbits were used to assess the performance of nonionic surfactants. We show that the propensity of a protein to aggregate in a vial agitated in horizontal or vertical orientation depends on the shaking orbit, and confirm that Brij® 58 and FM1000 prevent proteins from agitation-induced aggregation at lower concentrations than polysorbate 80.
