In this study, one strain of Beauveria caledonica was isolated from wild fruiting bodies collected from Guizhou Province, China, and its species identification, biological characteristics, domestication, and cultivation methods were studied along with polysaccharide and adenosine content analysis. The mycelia were identified by ITS sequencing, and the fruiting bodies of B. caledonica were domestically cultivated for the first time using wheat and rice as basic cultivation media. The carbon sources, nitrogen sources, cultivation temperatures, and pH for mycelial growth were optimized through single-factor experiments and response surface methodology (RSM) experiments. The polysaccharide content was detected by the phenol–sulfuric acid method, and the adenosine content was measured by high-performance liquid chromatography (HPLC). The results confirmed that the identified mycelia were B. caledonica. The optimum medium for solid culture was 25.8 g/L glycerol, 10.9 g/L yeast extract, 1 g/L MgSO4·7H2O, 1 g/L KH2PO4, 10 mg/L vitamin B1, and 20 g/L agar; the optimum pH was 6.5, and the optimum culture temperature was 25 °C. The optimal liquid culture medium was 26.2 g/L glycerol, 11.1 g/L yeast extract, 1 g/L MgSO4·7H2O, 1 g/L KH2PO4, and 10 mg/L vitamin B1; the mycelia grew well at pH 6.6 and 25 °C. The average biological efficiencies of fruiting bodies on wheat and rice as culture media were 1.880% and 2.115%, respectively; the polysaccharide contents of fruiting bodies on the two media were 6.635% and 9.264%, respectively, while the adenosine contents were 0.145% and 0.150%, respectively. This study provides a valuable reference for further artificial cultivation and utilization of B. caledonica by investigating its biological characteristics, cultivation conditions for artificial domestication, and polysaccharide and adenosine contents in cultivated fruiting bodies. Subway stations are important components of subway system planning and urban rail transit where passengers can board, alight, transfer, or wait. With an increase in urban passenger flow, subway stations must meet greater transfer‐space demands commensurate with growing station structure sizes. In this study, the field‐monitoring and analysis method was adopted to investigate the Huaxing Street Station of the Chengdu Metro Line 19 project and explore the temperature‐variation rule in the process of mass‐concrete pouring in the station floor. Based on an ABAQUS large‐finite‐element calculation programme and secondary development of damage and failure research methods, a mechanical model of mass‐concrete pouring in subway‐station floor construction was established. The pouring temperature effect, damage, and failure mechanism of mass‐concrete pouring in subway stations were comprehensively studied. The main results were as follows: (1) the maximum temperature difference between the upper and middle layers of the floor was dominant and reached a peak within 1–3 d after the completion of pouring, and the temperature difference gradually decreased with a decrease in the hydration‐heat reaction. (2) At 0–10 d, the damage ratio of the floor increased by 1.8%, predominantly in the form of longitudinal damage. Within 10–30 d, the damage and failure ratio of the floor increased by 1.86%, primarily in the form of transverse damage and failure. Within 30–40 d, the temperature of the floor stabilized and gradually approached the ambient temperature. The damage and failure ratio of the floor increased by 0.32% at this time. (3) The lower the ambient temperature, the greater was the damage sustained by the floor at different ages, and the ambient temperature had a significant influence on floor damage. Background: Congenital absence of the pericardium (CAP) is a rare cardiac abnormality. As pericardial defects are usually asymptomatic, most cases are diagnosed during surgery or on autopsy. The patient in this case was found to have CAP during thoracoscope. Case: We present the unusual case of a 69-year-old patient with CAP who experienced sudden ventricular arrhythmia and developed ventricular fibrillation during left upper lobectomy. Surgical operations, the lateral decubitus position, and other external stimuli may be important risk factors for ventricular fibrillation. The patient regained sinus rhythm soon after intrathoracic cardiac compression and pharmacological treatment, including lidocaine spray (2%, 10 ml) administered to the heart surface. The surgery was then completed without any additional instances of ventricular arrhythmia. Conclusions: Patients with CAP are more susceptible to cardiac-related adverse events during thoracotomy or thoracoscopy. Tissue-resident innate lymphoid cells (ILCs) play a vital role in the frontline defense of various tissues, including the lung. The development of type 2 ILCs (ILC2s) depends on transcription factors such as GATA3, RORα, GFI1, and Bcl11b; however, the factors regulating lung-resident ILC2s remain unclear. Through fate mapping analysis of the paralog transcription factors GFI1 and GFI1B, we show that GFI1 is consistently expressed during the transition from progenitor to mature ILC2s. In contrast, GFI1B expression is limited to specific subsets of bone marrow progenitors and lung-resident ILC progenitors. We found that GFI1B+ lung ILC progenitors represent a multi-lineage subset with tissue-resident characteristics and the potential to form lung-derived ILC subsets and liver-resident ILC1s. Loss of GFI1B in bone marrow progenitors led to the selective loss of lung-resident IL-18R+ ILCs and mature ILC2, subsequently preventing the emergence of effector ILCs that could protect the lung against inflammatory or tumor challenge. Fibroblastic reticular cells (FRCs) construct microanatomical niches that support lymph node (LN) homeostasis and coordination of immune responses. Transcription factors regulating the functionality of FRCs remain poorly understood. Here, we investigated the role of the transcription factor SpiB that is expressed in LN FRCs. Conditional ablation of SpiB in FRCs impaired the FRC network in the T‐cell zone of LNs, leading to reduced numbers of FRCs and altered homeostatic functions including reduced CCL21 and interleukin‐7 expression. The size and cellularity of LNs remained intact in the absence of SpiB but the space between the reticular network increased, indicating that although FRCs were reduced in number they stretched to maintain network integrity. Following virus infection, antiviral CD8+ T‐cell responses were impaired, suggesting a role for SpiB expression in FRCs in orchestrating immune responses. The projected increase in sea surface temperature due to climate change is expected to substantially intensify future hurricanes. Wooden light-frame residential buildings are particularly vulnerable to hurricane damage, and their risk is expected to increase due to heightened exposure and intensifying hurricanes. Therefore, adaptation strategies need to be planned to reduce damage to such buildings while considering the impact of climate change on hurricanes. This study investigates the effectiveness of various climate change adaptation strategies for coastal wood-frame single-story residential buildings and demonstrates how these strategies can be planned. The study considers the four Representative Concentration Pathways (RCPs) proposed by the IPCC to investigate the impact of climate change on wind hazard and losses. Additionally, three locations in the coastal United States of varying sizes, exposure, and hurricane hazard levels are considered: Harris County, Texas; Mobile County, Alabama; and Miami-Dade County, Florida. The results show that the increase in wind speeds and losses will be non-linear with time. All considered adaptation strategies decreased losses, with some able to completely counter the increasing losses even under high emission scenarios. In this study, we present a numerical investigation into the phenomenon of rising droplets in immiscible fluids, focusing on the Marangoni effect induced by both solute and a combination of solute and soluble surfactants. We meticulously examine the interfacial behaviors of pure solute droplets and mixed droplets, with a particular interest on the intricate interplay among interfacial concentration, interfacial tension, Marangoni stress, and Marangoni convection. Our investigation provides insight into the influence of key physicochemical parameters, such as viscosity, diffusion coefficient, partition coefficient, and interfacial tension gradient, on the Marangoni instability. Furthermore, we conduct a comprehensive parametric exploration of the impact of dimensionless numbers such as the Langmuir number (La), the Damkohler number (Da), the Peclet number (Pe), and the elasticity number β on the stabilizing efficacy of surfactants. The research findings underscore the effectiveness of our numerical method in capturing the distinctive two-step acceleration characteristics of pure solute droplets and the stabilizing effect of surfactants on mixed droplets. Notably, our study reveals that the Marangoni instability may manifest even when the viscosity and diffusivity ratios of the two-phase fluids are closely matched. Partition coefficients below unity exhibit only a marginal influence on the re-acceleration time of the droplets. Systems characterized by extremely low interfacial tension gradients tend to exhibit no Marangoni instability. Moreover, an increase in La enhances the stability of mixed droplets, while a significant threshold is identified for Da to affect the stability of mixed droplets. The ascent speed of mixed droplets displays pronounced variation across varying Pe magnitudes. Finally, in scenarios involving a wide-ranging variation in β, mixed droplets transition between the states of pure solute droplets and rigid spheres, revealing a distinct-state transition point. Spaceborne interferometric synthetic aperture radar (InSAR) techniques are important for landslide detection and monitoring; however, several limitations and uncertainties, such as the unique north–south flying direction and side-look radar observing geometry, currently limit the ability of InSAR to credibly detect landslides, especially those related to high and steep slopes. Here, we conducted experimental and statistical analysis on the feasibility of time-series InSAR monitoring for steep slopes using ascending and descending SAR images. First, the theoretical (TGNSS), practical (PGNSS), and terrain (Hterrain) (T-P-H) indices for sensitivity evaluations of the slope displacement monitoring results from time-series InSAR were proposed for slope monitoring. Subsequently, two experimental and statistical studies were conducted for the cases with and without Global Navigation Satellite System (GNSS) monitoring data. Our experimental results of two high and steep open-pit mines showed that the defined theoretical and practical sensitivity indices can quantitatively evaluate the feasibility of ascending and descending InSAR observations in steep-slope deformation monitoring with GNSS data, and the terrain sensitivity index can qualitatively evaluate the feasibility of landslide monitoring results from ascending and descending Sentinel-1 satellite data without GNSS data. We further demonstrate the generalizability of these proposed indices using four landslide cases with both public GNSS and InSAR monitoring data and 119 landslide cases with only InSAR monitoring data. The statistical results indicated that greater indices correlated with higher reliability of the monitoring results, suggesting that these novel indices have wide suitability and applicability. The safety retaining wall is a critical infrastructure in ensuring the safety of both rock removal vehicles and personnel. However, factors such as precipitation infiltration, tire impact from rock removal vehicles, and rolling rocks can cause local damage to the safety retaining wall of the dump, rendering it ineffective in preventing rock removal vehicles from rolling down and posing a huge safety hazard. To address these issues, this study proposed a safety retaining wall health assessment method based on modeling and analysis of UAV point-cloud data of the safety retaining wall of a dump, which enables hazard warning for the safety retaining wall. The point-cloud data used in this study were obtained from the Qidashan Iron Mine Dump in Anshan City, Liaoning Province, China. Firstly, the point-cloud data of the dump platform and slope were extracted separately using elevation gradient filtering. Then, the point-cloud data of the unloading rock boundary was obtained via the ordered crisscrossed scanning algorithm. Subsequently, the point-cloud data of the safety retaining wall were extracted using the range constraint algorithm, and surface reconstruction was conducted to construct the Mesh model. The safety retaining wall mesh model was isometrically profiled to extract cross-sectional feature information and to compare the standard parameters of the safety retaining wall. Finally, the health assessment of the safety retaining wall was carried out. In this study, we present numerical simulations examining the impact of soluble surfactant on the interface dynamics of a rising droplet. To achieve this, the droplet interface is tracked using an arbitrary Lagrangian–Eulerian approach, and the bulk and interfacial surfactant concentration evolution equations fully coupled with the incompressible Navier–Stokes equations are solved. We systematically evaluate the boundary of interfacial dynamics evolution by varying certain dimensionless parameters. Specifically, we study the effects of changes in parameters such as the Langmuir number, the Biot number, the Damkohler number, the bulk Peclet number, and the elastic number on interfacial tangential velocity, interfacial concentration and its gradient, interfacial viscous shear stress, and droplet rising velocity. Our findings confirm the validity of the stagnant-cap model for describing the interfacial fluidity of a surfactant-laden rising droplet. Increasing the Langmuir number and decreasing the Damkohler number can inhibit interface fluidity, but there is a threshold for the Damkohler number. Additionally, the overall increase in interface tension may mask the hindering effect of the locally increased concentration gradient on the interfacial fluidity. The Biot number has no impact on the steady state of the interface, but a slow adsorption rate may result in a bimodal retardation before the interface reaches a steady state. A clear threshold exists for the Peclet number to hinder the interface velocity, and a too high Peclet number leads to strong nonlinearity in the interface physical quantities. Variations in the elastic number significantly affect the evolution of the interface, causing the interface velocity to pass through several states, ranging from almost no retardation, uniform retardation, stagnant-cap retardation to complete retardation. Background: Rhabdomyosarcoma (RMS) is the most common soft tissue sarcoma with poor prognosis in children. The 5-year survival rate for early RMS has improved, whereas it remains unsatisfactory for advanced patients. Urine can rapidly reflect changes in the body and identify low-abundance proteins. Early screening of tumor markers through urine in RMS allows for earlier treatment, which is associated with better outcomes. Methods: RMS patients under 18 years old, including those newly diagnosed and after surgery, were enrolled. Urine samples were collected at the time points of admission and after four cycles of chemotherapy during follow-up. Then, a two-stage workflow was established. (1) In the discovery stage, differential proteins (DPs) were initially identified in 43 RMS patients and 12 healthy controls (HCs) using a data-independent acquisition method. (2) In the verification stage, DPs were further verified as biomarkers in 54 RMS patients and 25 HCs using parallel reaction monitoring analysis. Furthermore, a receiver operating characteristic (ROC) curve was used to construct the protein panels for the diagnosis of RMS. Gene Ontology (GO) and Ingenuity Pathway Analysis (IPA) software were used to perform bioinformatics analysis. Results: A total of 251 proteins were significantly altered in the discovery stage, most of which were enriched in the head, neck and urogenital tract, consistent with the most common sites of RMS. The most overrepresented biological processes from GO analysis included immunity, inflammation, tumor invasion and neuronal damage. Pathways engaging the identified proteins revealed 33 common pathways, including WNT/β-catenin signaling and PI3K/AKT signaling. Finally, 39 proteins were confirmed as urinary biomarkers for RMS, and a diagnostic panel composed of 5 candidate proteins (EPS8L2, SPARC, HLA-DRB1, ACAN, and CILP) was constructed for the early screening of RMS (AUC: 0.79, 95%CI = 0.66 ~ 0.92). Conclusions: These findings provide novel biomarkers in urine that are easy to translate into clinical diagnosis of RMS and illustrate the value of global and targeted urine proteomics to identify and qualify candidate biomarkers for noninvasive molecular diagnosis. The dense networks of conventional hydrogel hinder the efficient cell penetration and thus tissue regeneration. In this study, a macroporous cryogel with excellent mechanical properties was designed and fabricated through cryogelation and crosslinking of methacrylated gelatin (Gel-MA) and gelatin grafted with 2-ureido-4[1H]-6-methyl-pyrimidinone (Gel-UPY). The successful modification of the two functional units on the side chain of gelatin was confirmed by 1H-NMR and infrared spectroscopy. The macroporous structure of cryogel was verified by scanning electron microscopy and confocal microscopy. The compressive modulus of cryogel (110 kPa) was 2 magnitudes higher than that of the conventional hydrogel of the same chemical compositions (8 kPa). Both the hydrogel and cryogel exhibited excellent stress-recovery ability. The cryogel could well maintain the viability of chondrocytes and facilitated their migration into the scaffold interior. After being implanted into osteochondral defects in rabbits for 12 w, the Gel-MA/Gel-UPY cryogel better facilitated the regeneration of upper cartilage and subchondral bone compared to the conventional hydrogel. Background: Dasatinib and imatinib are the recommended tyrosine kinase inhibitors (TKIs) for treating pediatric Philadelphia-positive acute lymphoblastic leukemia (Ph + ALL), and the one which has been approved indication in China is imatinib. Recently, clinical demand for Ph + ALL treatment is becoming unmet gradually with the increasing resistance of imatinib. There are some studies reporting the better efficacy and comparative safety of dasatinib compared with imatinib, but no economic comparison has been published. This study aims to supplement economic evidence by comparing the cost-effectiveness between imatinib and dasatinib in treating pediatric patients with Ph+ ALL in China, and to help clinical rational drug use via multi-dimensional value assessment. Methods: A decision tree model combined with a 10-year Markov model were established based on the disease progression. The parameters were collected from published literatures and our hospital's electronic medical records. From the health system perspective, the incremental cost-effectiveness ratio (ICER) between the two treatment groups was calculated through cost-effectiveness analysis and then compared with the willingness-to-pay (WTP) threshold. The set WTP threshold in this study was 1 times per capita gross domestic product (GDP) of China, as recommended by the World Health Organization. Direct medical costs and quality-adjusted life years (QALYs) were calculated and discounted at 5%. The sensitivity analyses were conducted to assess the uncertainty and robustness of the results. Results: The total costs were CNY 1,020,995.35 and CNY 1,035,788.50 in imatinib group and dasatinib group during the 10-year simulation, and the total QALYs were 2.59 and 4.84. Compared with the imatinib treatment group, the ICER was around CNY 6,575.78/ QALY, which was less than the set threshold CNY 70,892/ QALY. The sensitive analyses indicated the robustness of the results. Conclusions: The cost-effectiveness analysis shows the potential cost-effective advantages of adding dasatinib comparing with adding imatinib for pediatric Ph + ALL patients in China under the set WTP threshold, which indicates that those patients could achieve more QALYs by paying acceptable fee. The level set method was used to numerically simulate the droplet formation process in T‐junction microchannels with different junction angle. The effects of flow rate ratio, capillary number, and contact angle on droplet formation under different junction angles were discussed. The numerical simulation results show that the dimensionless length and generation frequency of microdroplets increase with the flow rate ratio. It was observed that the droplet transforms from the squeezing regime to the dripping regime when the capillary number Ca is ≥ 0.02. For Ca ≤ 0.04, the smallest droplet was generated in the microchannel with 120° junction angle. Finally, the results also indicate that the effect of the contact angle is discrepant at different junction angles. [ABSTRACT FROM AUTHOR]

Step-feature lines are one of the important geometrical elements for drawing the status quo maps of open-pit mines, and the efficient and accurate automatic extraction and updating of step-feature lines is of great significance for open-pit-mine stripping planning and analysis. In this study, an automatic extraction method of step-feature lines in an open-pit mine based on unmanned-aerial-vehicle (UAV) point-cloud data is proposed. The method is mainly used to solve the key problems, such as low accuracy, local-feature-line loss, and the discontinuity of the step-feature-line extraction method. The method first performs the regular raster resampling of the open-pit-mine cloud based on the MLS algorithm, then extracts the step-feature point set by detecting the elevation-gradient change in the resampled point cloud, further traces the step-feature control nodes by the seed-growth tracking algorithm, and finally generates smooth step-feature lines by fitting the space curve to the step-feature control nodes. In this study, the probability integral method, Synthetic Aperture Radar Interferometry (InSAR), and the Okada dislocation model were collaboratively used to analyze deformation in the Alhada Pb-Zn mine. The predicted deformation values of the subsidence centers in three subsidence areas were 107 mm, 120 mm, and 83 mm, respectively, as predicted using the probability integral method. The coherent scatterer InSAR technique was used to analyze the time-series deformation of the mining area, and the same subsidence center locations and similar deformation values were observed. The Okada dislocation model was used to invert the optimal parameters of the underground-mining ore body causing the surface subsidence, indicating that the surface subsidence is mainly caused by the mining of ore bodies in the 888 and 848 middle sections. We further simulated ground deformation using the multi-source Okada model. The results showed that the predicted and modeled deformations are highly correlated with the observed deformation. Through the analysis and comparison of the InSAR results, it was concluded that the three subsidence areas do not threaten the stability of the main buildings in the mining area. Using theoretical, observational, and modeling methods, the development and evolution of the subsidence area in mines can be established, which could provide basic data for subsidence control work and guarantee mine production safety. Existing structures may suffer from resistance deterioration due to repeated attacks. The modeling of resistance deterioration is a critical ingredient in the reliability assessment and service life prediction of these degraded structures. In this paper, an explicit compound Poisson process-based model is developed to describe the shock deterioration of structural resistance, where the magnitude of each shock deterioration increment is modeled by a Gamma-distributed random variable. The moments (mean value and variance) and the distribution function of the cumulative shock deterioration are derived in a closed form, based on a proposed W-function. A method for the efficient calculation of the W-function is presented, which reduces to the Bessel type I function if the shock deterioration increment is exponentially distributed (a special case of Gamma distribution). The proposed shock deterioration model is applicable to either a stationary or a nonstationary Poisson process of random jumps. Subsequently, the overall resistance deterioration is modeled as the linear combination of gradual and shock deteriorations, based on which the proposed model can be used in the time-dependent reliability assessment of aging structures efficiently. A numerical example is presented to demonstrate the applicability of the proposed deterioration model by estimating the time-dependent reliability of an aging bridge. It is found that a smaller threshold for the degraded resistance leads to greater mean value and standard deviation of the time to failure, and this effect is enhanced by a smaller occurrence rate of the shock deterioration. We show that diverse cell types express the cell surface marker PDPN in the spleen, including three TRC subsets, glial cells, mesothelial cells, and ARCs, despite that PDPN has been widely used as a marker of TRC in the spleen. The spleen is a complex organ that performs multiple functions during homeostasis and during immune responses. We defined strategies to isolate and analyze stromal cell subsets from mice and human spleens and performed RNA-seq, scRNA-seq, flow cytometry, and confocal microscopy to interrogate splenic stromal cells and to compare these with stromal cells from LNs. Background: Drug-induced thrombocytopenia (DITP) is a severe adverse reaction and a significantly under-recognized clinical problem in children. However, for post-marketing pharmacovigilance purposes, detection of DITP signals is crucial. This study aimed to develop a signal detection model for DITP using the pediatric electronic medical records (EMR) data. Methods: This study used the electronic medical records collected at Beijing Children's Hospital between 2009 and 2020. A two-stage modeling method was developed to detect the signal of DITP. In the first stage, we calculated the crude incidence by mining cases of thrombocytopenia to select the potential suspected drugs. In the second stage, we constructed propensity score–matched retrospective cohorts of specific screened drugs from the first stage and estimated the odds ratio (OR) and 95% confidence interval (CI) using conditional logistic regression models. The novelty of the signal was assessed by current evidence. Results: In the study, from a total of 839 drugs, 21 drugs were initially screened as potentially inducing thrombocytopenia. In total, we identified 18 positive DITP associations. Of these, potential DITP risk of nystatin (OR: 1.75, 95% CI: 1.37–2.22) and latamoxef sodium (OR: 1.61, 95% CI: 1.38–1.88) were two new DITP signals in both children and adults. Six associations between thrombocytopenia and drugs including imipenem (OR: 1.69, 95% CI: 1.16–2.45), teicoplanin (OR: 4.75, 95% CI: 3.33–6.78), fusidic acid (OR: 2.81, 95% CI: 2.06–3.86), ceftizoxime sodium (OR: 1.83, 95% CI: 1.36–2.45), ceftazidime (OR: 2.16, 95% CI: 1.58–2.95), and cefepime (OR: 5.06, 95% CI: 3.77–6.78) were considered as new signals in children. Conclusion: This study developed a two-stage algorithm to detect safety signals of DITP and found eighteen positive signals of DITP, including six new signals in a pediatric population. The durability design of reinforced concrete (RC) structures that are exposed to aggressive environmental attacks (e.g., corrosion due to chloride ingress in marine environment) plays a vital role in ensuring the structural serviceability within a reference period of interest. Existing approaches for the durability design and assessment of RC structures have, for the most part, not considered the spatial distribution of corrosion-related structural properties. In this paper, a closed-form approach is developed for durability assessment of RC structures, where the structural dimension, spatial variability, and correlation of structural properties such as the concrete cover thickness and the chloride diffusion coefficient are taken into account. The corrosion and crack initiations of an emerged tube tunnel segment that was used in the Hong Kong-Zhuhai-Macau bridge project were assessed to demonstrate the applicability of the proposed approach. The accuracy of the method was verified through a comparison with Monte Carlo simulation results based on two-dimensional random field modeling. The proposed method can be used to efficiently assess the durability performance of RC structures in the marine environment and has the potential to become an efficient tool to guide the durability design of RC structures subjected to corrosion. Dendritic cells (DCs) and macrophages are at the forefront of immune responses, modifying their transcriptional programs in response to their tissue environment or immunological challenge. Posttranslational modifications of histones, such as histone H3 lysine-27 trimethylation (H3K27me3) by the Polycomb repressive complex 2 (PRC2), are tightly associated with epigenetic regulation of gene expression. To explore whether H3K27me3 is involved in either the establishment or function of the mononuclear phagocyte system, we selectively deleted core components of PRC2, either EZH2 or SUZ12, in CD11c-expressing myeloid cells. Unexpectedly, EZH2 deficiency neither prevented the deposition and maintenance of H3K27me3 in DCs nor hindered DC/macrophage homeostasis. In contrast, SUZ12 deficiency markedly impaired the capacity of DCs and macrophages to maintain H3K27me3. SUZ12 ablation induced a rapid loss of the alveolar macrophage and Langerhans cell networks under both steady state and inflammatory conditions because these cells could no longer proliferate to facilitate their self-renewal. Despite the reduced H3K27me3, DC development and function were unaffected by SUZ12 ablation, suggesting that PRC2-mediated gene repression was dispensable for DC homeostasis. DC-SCRIPT takes center stage: Dendritic cells (DCs) initiate diverse and context-specific adaptive immune responses. Zhang et al. define a role for the transcription factor DC-SCRIPT in the development and function of type 1 conventional DCs (cDC1s) by generating DC-SCRIPT reporter and knockout mouse strains. DC-SCRIPT was required for establishment of the cDC1 gene expression program, as well as cross-presentation of cell-associated antigen and production of IL-12, which are important for stimulating effective type 1 immune responses. The cell type–specific role of DC-SCRIPT was likely due at least in part to its ability to directly control expression of the transcription factor IRF8 in cDC1s, highlighting DC-SCRIPT as a key component of the gene regulatory networks coordinating cDC1 development and function. The functional diversification of dendritic cells (DCs) is a key step in establishing protective immune responses. Despite the importance of DC lineage diversity, its genetic basis is not fully understood. The transcription factor DC-SCRIPT is expressed in conventional DCs (cDCs) and their committed bone marrow progenitors but not in plasmacytoid DCs (pDCs). We show that mice lacking DC-SCRIPT displayed substantially impaired development of IRF8 (interferon regulatory factor 8)–dependent cDC1, whereas cDC2 numbers increased marginally. The residual DC-SCRIPT–deficient cDC1s had impaired capacity to capture and present cell-associated antigens and to secrete IL-12p40, two functional hallmarks of this population. Genome-wide mapping of DC-SCRIPT binding and gene expression analyses revealed a key role for DC-SCRIPT in maintaining cDC1 identity via the direct regulation of cDC1 signature genes, including Irf8. A new type of Ag3PO4/UiO-66-NH2 Z-scheme heterojunction nanocomposite with visible light catalytic activity was successfully prepared. The synthesis of the composite was confirmed by XRD, FT-IR, SEM, TEM, EDS, XPS. The Ag3PO4/UiO-66-NH2 Z-scheme heterojunction can effectively improve the utilization of visible light. The optimal Ag/U/1/4 composite has the highest degradation efficiency for methylene blue (MB, 30 mg/L) in 90 min (80%) and Norfloxacin (NOR, 10 mg/L) in 150 min (85%). The decrease of Nyquist radius and the enhancement of photocurrent response indicated that the combination of photogenerated electron–hole pair in Ag3PO4/UiO-66-NH2 composite material is significantly inhibited. N2 adsorption–desorption and zeta potential results indicated that biggish specific surface area and negative zeta potential value are more favorable for the adsorption of the cationic dye MB. The cyclic test verified the good stability of Ag3PO4/UiO-66-NH2 heterojunction material. Furthermore, the photocatalytic degradation mechanism of MB was discussed by introducing different traps to inhibit the possible active substances in the degradation process. The immune system plays a fundamental role at mucosal barriers in maintaining tissue homeostasis. This is particularly true for the gut where cells are flooded with microbial-derived signals and antigens, which constantly challenge the integrity of the intestinal barrier. Multiple immune cell populations equipped with both pro- and anti-inflammatory functions reside in the gut tissue and these cells tightly regulate intestinal health and functions. Dysregulation of this finely tuned system can progressively lead to autoimmune disease and inflammation-driven carcinogenesis. Over the last decade, the contribution of the adaptive immune system in controlling colorectal cancer has been studied in detail, but the role of the innate system, particularly innate lymphoid cells (ILCs), have been largely overlooked. By sensing their microenvironment, ILCs are essential in supporting gut epithelium repair and controling bacterial- and helminth-mediated intestinal infections, highlighting their important role in maintaining tissue integrity. Accumulating evidence also suggests that they may play an important role in carcinogenesis including intestinal cancers. The first 10-kV regional compensation dynamic voltage restorer (DVR) has been installed in a 110-kV substation in Shenzhen power grid, to protect plenty of sensitive loads in a wide area simultaneously from the disturbances caused by voltage sags. The location selection and performance indicators of the DVR are determined based on the voltage sag statistic results in recent years of Shenzhen power grid. The implementation schemes of main circuit elements, such as the energy storage unit, inverter and LC filter are presented. To further improve the DVR's cost-effectiveness, a novel virtual impedance control strategy is proposed to provide the DVR with a function of a Fault Current Limiter (FCL) or a Series Compensator (SC) besides the function of voltage sag compensation. Leg inductors which connect a shunt Active Power Filter (APF) to power network play a crucial role in guaranteeing desired performances of the APF. Optimal tuning of the inductances is still a tricky problem that has not yet been tackled systematically and theoretically. This paper proposes a novel inductance optimisation method that is universally applicable to four-wire shunt APFs with arbitrary topologies and PWM strategies. Based on the unified mathematic model, indexes used to quantitatively evaluate the abilities of a four-wire APF in compensating phase harmonic currents and depressing neutral line current are formally formulated. The upper and lower value constraints of the inductances are derived by comprehensively taking into account the characteristics of load harmonic profile and PWM strategy. Inductances are further optimised in the feasible value ranges with concern of several engineering application issues. The currently used hysteresis method is of low control accuracy, and the PWM-based strategy fails to provide a high enough response speed; thus, there is always a trade-off between the performances of control accuracy and response speed for a four-leg shunt active power filter (APF) to cope with the large variation in the load harmonic profile. First, an improved SVPWM strategy is proposed in this paper; with the aim to increase the response speed of a four-leg APF by simplifying the implementation of current 3D-SVPWM methods, the principle and realisation of this strategy are explained in detail. Based on these works, a compound PWM algorithm that effectively integrates the improved SVPWM and a PI-based current hysteresis method is proposed to simultaneously guarantee a higher response speed and a higher control accuracy for a four-leg APF. The realisation and parameter tuning of the compound PWM are elaborated. The correctness and effectiveness of the improved SVPWM and the compound PWM strategy are verified by simulations using Matlab/Simulink software. A dynamic voltage restorer (DVR) can be installed in a middle-voltage (MV) power grid, to concurrently protect a cluster of sensitive loads from voltage sags. To further improve its efficiency and reduce the difficulty in its implementation, a novel control strategy for operating such a DVR as a virtual impedance in series with sensitive loads is proposed in this paper. In addition to its usual function of compensating for voltage sags, such a DVR can also operate as a virtual inductance, to function as a fault current limiter (FCL) during a downstream fault, or a virtual capacitance, to function as a series compensator (SC) to compensate the voltage loss along the feeder line during heavy load. Based on a dual-loop control design, strategies for operating a DVR as a series virtual inductance and a virtual capacitance are proposed, and methods for tuning the parameter values and a stability analysis of the whole system are presented. The feasibility and effectiveness of the proposed method are verified by simulations using the PSCAD software, and experimental results obtained using a prototype DVR are presented. Online detuning state detection and the subsequent detuned element identification of a passive filter bank is of great concern in regard to achieving condition-based maintenance of an HVDC transmission system. In this paper, a novel method that can rapidly detect the online detuning state of filter banks with arbitrary structures of an HVDC system and accurately identify the detuning element is proposed. The method is based on the calculated amplitude change rates (ACRs) of several deliberately selected harmonic components of the measured real-time current in each branch of the filter. The principle and the implementation of the proposed method are presented, and the relationship between the ACR of the harmonic current and the detuning degree of an individual element is deduced theoretically. Based on the above works, the criteria for identifying the detuning state of the filter bank, the detuned element in the series and parallel part of the filter, and the detuning degree of the element are given. Diverse simulation results, such as applications on a single- and a double-tuned DC filter bank, sensitivity analysis and performance comparison, are provided to show the correctness and the effectiveness of the proposed method. This paper describes the design of a dynamic voltage restorer (DVR) that can simultaneously protect several sensitive loads from voltage sags in a region of an MV distribution network. A novel reference voltage calculation method based on zero-sequence voltage optimisation is proposed for this DVR to optimise cost-effectiveness in compensation of voltage sags with different characteristics in an ungrounded neutral system. Based on a detailed analysis of the characteristics of voltage sags caused by different types of faults and the effect of the wiring mode of the transformer on these characteristics, the optimisation target of the reference voltage calculation is presented with several constraints. The reference voltages under all types of voltage sags are calculated by optimising the zero-sequence component, which can reduce the degree of swell in the phase-to-ground voltage after compensation to the maximum extent and can improve the symmetry degree of the output voltages of the DVR, thereby effectively increasing the compensation ability. In-service civil structures are often subjected to repeated external loads (shock events), which may lead to an accumulation of structural damage and further an increase in the probability of structural failure. The shock actions are by nature stochastic with randomness arising from their intensities and occurrence times. A varying trend in future intensity and/or occurrence rate of many types of shock action has been projected in the literature, such as cyclone actions due to the potential impact of climate change. This article proposes a method to assess the structural reliability in the presence of uncertainty and non-stationarity in external shock events, with a limit state that the cumulative damage exceeds the permissible level. The mean value and variance of the cumulative damage are estimated with proposed closed-form solutions, and the gamma distribution is suggested to describe the probabilistic characteristics of the cumulative damage, whose accuracy is verified through a comparison with Monte Carlo simulation. Furthermore, the cumulative density function of structural service life is derived explicitly. The Conducted Electromagnetic Interference (CEI) characteristics in the primary circuit and at the ports of the secondary devices of the converter station of a UHVDC transmission system are researched comprehensively and systematically in this paper, by taking the Zhalute-Qingzhou ±800 kV/10000MW UHVDC project in East Inner Mongolia of China as an example. The primary circuit equipment parameters of the target system are designed systematically at first, and the overall broadband equivalent model of the main circuit of the UHVDC system, which is composed of converter valve, converter transformer, filter banks and smoothing reactor, is developed. The CEI characteristics in the primary circuit under various conditions of the UHVDC system are analyzed based on the simulations carried out on the built BEC, and the influences of several primary circuit elements on the propagation of the CEI characteristics are researched. To improve the accuracy of the analysis of the CEI characteristics in the secondary device circuit, accurate BECs of the Capacitor Voltage Transformer (CVT), Current Transformer (CT) and secondary signal cable are established. The CEI characteristics at the ports of secondary devices under different operation modes are studied, and the influences of the cable length and burden rate on the CEI characteristics are analyzed. Severe hurricanes in coastal areas have caused enormous human and economic losses. Furthermore, the intensity and frequency of future hurricanes may increase due to the potential impact of climate change. Due to common underlying climatological causes, interrelations may exist between successive hurricane events. This paper proposes a novel approach for modeling the temporal correlation in hurricane frequency and develops an analytical method that reflects this correlation to estimate the statistical properties of cumulative hurricane damage. The method provides a closed-form solution to the mean and variance of hurricane damage, and can be used to assess the impacts of temporal correlation and nonstationarity in hurricane events quantitatively. The applicability of the method is demonstrated through a case study of Miami-Dade County, Florida, in which it is found that the nonstationarity in hurricane events increases the mean value of hurricane damage over a future period of 100 years, while the temporal correlation increases its variance. The successful past performance of an existing bridge is evidence of its resistance, which reduces the uncertainty associated with the resistance and so increases the bridge reliability. However, the resistance deterioration and increases in traffic load may negate this effect. In this paper, statistical techniques are used to combine the prior estimate of time-dependent resistance with new information from the service load history, thereby obtaining an updated estimate of bridge resistance, as well as an updated reliability for subsequent years. The application of the method is illustrated using an existing bridge. The influences of the intensity and nonstationarity of service load history and the shape and variance of deterioration function on updated estimates of bridge resistance and reliability are examined using parametric examples. It is found that the updated resistance of an existing bridge with ever-increasing traffic load is close to that of the same bridge with a stationary traffic load process if the load intensities at the latest stage of the two-load process are identical; however, the updated reliability of the bridge is greatly reduced considering the increase of traffic load with time. The statistics of the deterioration function has a significant effect on both the updated resistance and reliability of the bridge, while the shape of the deterioration function has a significant effect on the updated reliability only. The estimate of initial bridge resistance can also be updated by the proposed method, but the updating effect is insignificant compared with the updating of current resistance. Sperm protein 17 (Sp17) is selectively overexpressed in several human malignancies including ovarian carcinoma, but is absent or expressed at low levels in most normal tissues. Previous work from our group characterized an anti-Sp17 monoclonal antibody (clone 3C12) and showed that it specifically targeted tumor cells. In this report, we investigated whether a novel immunoconjugate containing 3C12 linked to the chemotherapeutic agent doxorubicin [(DOX) Adriamycin] had antitumor activity against ovarian cancer cell lines and tumor models. DOX was conjugated to 3C12 using a linker, and the specificity of 3C12-DOX was examined in Sp17-positive SKOV3 and Sp17-negative COC2 ovarian cancer cells using cell-based ELISA and internalization assays. The cytotoxicity of 3C12-DOX was assessed with the MTT assay, and its therapeutic effectiveness was evaluated in immunodeficient mice bearing SKOV3 cells. In vitro, the 3C12-DOX immunoconjugate specifically bound to and was internalized by Sp17-positive SKOV3 cells but did not bind to Sp17-negative cells. Treatment with 3C12-DOX (0.001 to 10 μg/mL) decreased the viability of SKOV3 cells in a Sp17-specific manner. In vivo, 3C12-DOX (3 mg/kg) induced the regression of established SKOV3 xenograft tumors in BALB/c mice compared with control treatment. The antitumor effects of 3C12-DOX were significantly associated with the induction of apoptosis in tumor cells. In addition, 3C12-DOX showed no observable adverse effects or toxicity when compared with DOX alone in mice bearing ovarian tumor xenografts. We performed 31P-NMR measurements on LaFe(As1-xPx)O to investigate the relationship between antiferromagnetism and superconductivity. The antiferromagnetic (AFM) ordering temperature TN and the moment μord are continuously suppressed with increasing P content x and disappear at x = 0.3 where bulk superconductivity appears. At this superconducting x = 0.3, quantum critical AFM fluctuations are observed, indicative of the intimate relationship between superconductivity and low-energy AFM fluctuations associated with the quantum-critical point in LaFe(As1-xPx)O. The relationship is similar to those observed in other isovalent-substitution systems, e.g., BaFe2(As1-xPx)2 and SrFe2(As1-xPx)2, with the "122" structure. Moreover, the AFM order reappears with further P substitution (x > 0.4). The variation of the ground state with respect to the P substitution is considered to be linked to the change in the band character of Fe-3d orbitals around the Fermi level. [ABSTRACT FROM AUTHOR]

The recent discovery of high-temperature superconductivity in iron-based pnictides (chalcogenides) not only triggers tremendous enthusiasm in searching for new superconducting materials, but also opens a new avenue to the study of the Kondo physics. CeFeAsO is a parent compound of the 1111-type iron-based superconductors. It shows 3d-antiferromagnetic (AFM) ordering below ∼ 139 K and 4f-AFM ordering below ∼ 4 K. On the other hand, the phosphide CeFePO is a ferromagnetically correlated heavy-fermion (HF) metal with Kondo scale TK ∼ 10 K. These properties set up a new platform for research of the interplay among magnetism, Kondo effect, and superconductivity (SC). In this review, we present the recent progress in the study of chemical pressure effect in CeFeAsO1−yFy (y = 0 and 0.05). This P/As-doping in CeFeAsO serves as an effective controlling parameter which leads to two magnetic critical points, xc1 ≃ 0.4 and xc2 ≃ 0.92, associated with suppression of 3d and 4f magnetism, respectively. We also observe a turning point of AFM-FM ordering of Ce3+ moment at xc3 ≃ 0.37. The SC is absent in the phase diagram, which is attributed to the destruction to Cooper pair by Ce-FM fluctuations in the vicinity of xc1. We continue to investigate CeFeAs1−xPxO0.95F0.05. With the separation of xc1 and xc3, this chemical pressure results in a broad SC region 0≤ x ≤ 0.53, while the original HF behavior is driven away by 5% F− doping. Different roles of P and F dopings are addressed, and the interplay between SC and Ce-4f magnetism is also discussed. [ABSTRACT FROM AUTHOR]
