Introduction: NAC (NAM, ATAF, and CUC) transcription factor family, one of the important switches of transcription networks in plants, functions in plant growth, development, and stress resistance. Night lily (Hemerocallis citrina) is an important horticultural perennial monocot plant that has edible, medicinal, and ornamental values. However, the NAC gene family of night lily has not yet been analyzed systematically to date. Methods: Therefore, we conducted a genome-wide study of the HcNAC gene family and identified a total of 113 HcNAC members from the Hemerocallis citrina genome. Results: We found that 113 HcNAC genes were unevenly distributed on 11 chromosomes. Phylogenetic analysis showed that they could be categorized into 16 instinct subgroups. Proteins clustering together exhibited similar conserved motifs and intron-exon structures. Collinearity analysis indicated that segmental and tandem duplication might contribute to the great expansion of the NAC gene family in night lily, whose relationship was closer with rice than Arabidopsis. Additionally, tissue-specific pattern analysis indicated that most HcNAC genes had relatively higher expression abundances in roots. RNA-Seq along with RT-qPCR results jointly showedHcNACgenesexpresseddifferentlyunder drought andsalinity stresses. Interestingly, HcNAC35 was overexpressed in watermelon, and the stress resilience of transgenic lines was much higher than that of wild-type watermelon, which revealed its wide participation in abiotic stress response. This study presents a comprehensive model to simulate daylily growth and optimize planting patterns to maximize bud yield while minimizing land resource utilization. Methods: The model incorporates source-sink relationship specific to daylilies into physiological process modeling, considering environmental factors such as micro-light and temperature climate, and CO2 concentration. Spatial factors, including planting pattern, row spacing, plant spacing, and plant density were examined for their impact on light interception, photosynthesis, and resource efficiency. Employing partial least square path modeling (PLS-PM), we analyzed the interrelations and causal relationships between planting configurations and physiological traits of daylily canopy leaves and buds. Through in situ simulations of 36 planting scenarios, we identified an optimal configuration (Scenario ID5) with a density of 83,000 plants·ha−1, row spacing of 0.8 m, and equidistant planting with a plant spacing of 0.15 m. Results and discussion: Our research findings indicate that increased Wide+Narrow row spacing can enhance yield to a certain extent. Although planting patterns influence daylily yield, their overall impact is relatively minor, and there is no clear pattern regarding the impact of plant spacing on individual plant yield. This modeling approach provides valuable insights into daylily plant growth dynamics and planting patterns optimization, offering practical guidance for both farmers and policymakers to enhance daylily productivity while minimizing land use. Hemerocallis citrina Borani is a traditional folk food used to promote the lactation of postpartum mothers in China; however, the active ingredients and corresponding mechanisms are still unknown. In this study, the lactogenic effect of alcoholic and aqueous extracts of H. citrina was primarily evaluated, and the aqueous extract (1,000 and 2,000 mg/kg) displayed significant lactationpromoting effects. Three eluates of the aqueous extract (0%, 30%, and 50%HCW) were further evaluated for their lactogenic effect, and 30% and 50%HCWshowed significant lactation-promoting activity. Nineteen ingredients, including those with a high content of rutin and isoquercetin, were then identified from 30% and 50%HCW using the ultra-performance liquid chromatography-quadrupole-time-of-flight-mass spectrometry (UPLC-Q-TOF-MS) method. Finally, the lactogenic effect of rutin and isoquercetin was evaluated, and both compounds displayed significant lactation-promoting activity. The mechanisms relative to the lactation-promoting active ingredients for H. citrina extracts and compounds are to stimulate the release of prolactin (PRL) and progesterone (P), as well as to induce the expression of prolactin receptor (PRLR) and improve the morphology of mammary tissue. This study first clarified the lactation-promoting active ingredients of H. citrina and the corresponding mechanisms, which provide a new insight into the new lactation-promoting drug and promote the high-value utilization of H. citrina resources. Members of the genus Hemerocallis have significant value as ornamental, edible, and medicinal plants, particularly in China, where they have been utilized for thousands of years as both a vegetable and Traditional Chinese Medicine. Hemerocallis species exhibit strict control over flowering time, with individuals flowering either diurnally or nocturnally. However, our understanding of the evolutionary history of this genus, especially concerning important horticultural traits, remains limited. In this study, sequencing and assembly efforts were conducted on 73 samples within the Hemerocallis genus. All accessions were classified into two distinct groups based on their diurnal (daylilies) or nocturnal (nightlilies) flowering habits. Comparative analysis of the chloroplast genomes from these two groups identified fifteen variant hotspot regions, including fourteen SNPs and one deletion, which hold promise for the development of molecular markers for interspecific identification. Phylogenetic trees, generated through both maximum-likelihood and Bayesian inference methods using 76 shared protein-coding sequences, revealed that diurnal flowering evolved prior to nocturnal flowering. The divergence between the two groups is estimated to have occurred approximately 0.82 MYA (95% CI: 0.35-1.45 MYA). The ancestral state of Hemerocallis is hypothesized to have featured diurnal flowering with orange yellow petals. This study marks the first reconstruction of the evolutionary history and ancestral state of the genus Hemerocallis. [Objective] The effects of water and fertilizer coupling on the growth and yield of day lily were studied. [Method] This experiment took day lily as the research object, two factors experiments were set up to study the effects of water and fertilizer coupling on the growth and yield of day lily. [Result] Under the same drip amount, the yield of day lily was significantly different under different drip times, and the agronomic characters of day lily were the best under IV and V treatments, and the maximum yield was 5 983.96-6 000 kg/hm². Under the same fertilization time and different fertilization rates, S-IV treatment and S-V treatment showed the best agronomic characters such as height, number of flower buds and flower buds. Domestic export cut lily flowers are expensive in Japan when they are in bud state that has not yet bloomed and when no leaf yellowing has occurred. Predicting the blooming day of domestic cut lily flowers is essential to increase their commodity value. Thermal imaging, spectroscopic technologies, and hyperspectral cameras have recently been used for quality prediction. This study uses a hyperspectral camera, reflectance of wavelength, and a support vector machine (SVM) to evaluate the predictability of blooming days of cut lily flowers. While examining spectra at wavelengths of 750–900 nm associated with pollination, the resultant reflectance was over 75% during six to four days before blooming and 30% on a blooming day, indicating a decline in their reflectance toward blooming. Furthermore, SVM classification models based on kernel function revealed that the quadratic SVM had the highest accuracy at 84.4%, while the coarse Gaussian SVM had the lowest accuracy at 34.4%. The most crucial wavelength for the quadratic SVM was 842.3 nm, which was associated with water. The quadratic SVM's accuracy, verified using the area under the curve (ACU), was above 0.8, showing suitability for spectral classification based on blooming day prediction. Thus, this study shows that hyperspectral imaging can classify spectra based on the blooming day, indicating its potential to predict the blooming day, vase life, and quality of cut lily flowers. Background: Sucrose invertase is an important catalytic enzyme that is widely distributed in plants and can irreversibly hydrolyze sucrose into fructose and glucose. Daylily is an important perennial flower worldwide and a traditional vegetable in East Asia. Previous studies have suggested that sucrose invertase is involved in the aging of daylily flowers. However, knowledge about the number, physicochemical properties, and expression patterns of daylily sucrose invertases is still lacking. Identifying the daylily sucrose invertase family genes in the genome is highly important for understanding phylogenetic evolution and determining the genetic function of sucrose invertase. Methods: To obtain basic knowledge about the number, classification, sequence composition, and physicochemical properties of sucrose invertases in daylily, bioinformatics software was used to analyze the genome of Hemerocallis citrina (H. citrina), and the basic properties of sucrose invertase genes and proteins were obtained. Then, combined with transcriptome data from flower organs at different developmental stages, the expression patterns of each gene were clarified. Finally, the reliability of the transcriptome data was verified by quantitative real-time polymerase chain reaction (PCR). Results: Through software analysis, 35 sucrose invertases were identified from the H. citrina genome and named HcINV1-HcINV35; these enzymes belong to three subfamilies: cell wall invertases, vacuolar invertases, and chloroplast invertases. The amino acid composition, motif types, promoter composition, gene structure, protein physicochemical properties, gene chromosomal localization, and evolutionary adaptability of daylily invertases were determined; these results provided a comprehensive understanding of daylily invertases. The transcriptome expression profile combined with fluorescence quantitative reverse transcription-polymerase chain reaction (RT‒PCR) analysis suggested that almost all daylily invertase genes were expressed in flower organs, but even genes belonging to the same subfamily did not exhibit the same expression pattern at different developmental stages, suggesting that there may be redundancy or dissimilation in the function of daylily sucrose invertases. Daylily (Hemerocallis citrina) is a perennial herb of the genus Hemerocallis of Liliaceae. It is also an economically important crop and is widely cultivated. Daylily has nutritional, medicinal and ornamental values. The research literature shows that daylily is a high-quality food raw material rich in soluble sugars, ascorbic acid, flavonoids, dietary fiber, carotenoids, mineral elements, polyphenols and other nutrients, which are effective in clearing heat and diuresis, resolving bruises and stopping bleeding, strengthening the stomach and brain, and reducing serum cholesterol levels. This article reviews the main nutrients of daylily and summarizes the drying process of daylily. In addition, due to the existence of active ingredients, daylily also has a variety of biological activities that are beneficial to human health. This article also highlights the nutritional quality of daylily, the research progress of dried vegetable rehydration technology and dried daylily. In the end, the undeveloped molecular mechanism and functional research status of daylily worldwide are introduced in order to provide reference for the nutritional quality research and dried processing industry of daylily. Sugars will be eventually effluxed transporters (SWEETs) have been confirmed to play diverse physiological roles in plant growth, development and stress response. However, the characteristics and functions of the SWEET genes in Hemerocallis citrina remain unclear and poorly elucidated. In this study, the whole genome of Hemerocallis citrina was utilized to conduct bioinformatics analysis and a total of 19 HcSWEET genes were successfully identified. Analysis of the physicochemical properties indicated dominant differences among these HcSWEETs. A phylogenetic analysis revealed that HcSWEET proteins can be divided into 4 clades ranging from Clade I to IV, where proteins within the same clade exhibited shared conserved motifs and gene structures. Five to six exons were contained in the majority of HcSWEET genes, which were unevenly distributed across 11 chromosomes. The gene duplication analysis showed the presence of 4 gene pairs. Comparative syntenic maps revealed that the HcSWEET gene family might present more closed homology in monocotyledons than dicotyledons. Cis-acting element analysis of HcSWEET genes indicated key responsiveness to various hormones, light, and stresses. Additionally, transcriptome sequencing analysis suggested that most HcSWEET genes had a relatively higher expression in roots, and HcSWEET4a was significantly up-regulated under salt stress. Overexpression further verified the possibility that HcSWEET4a was involved in response to salt stress, which provides novel insights and facilitates in-depth studies of the functional analysis of HcSWEETs in resistance to abiotic stress. Sleep disorders are widespread. Inhaled aromatherapy has shown advantages over medication in treating insomnia. Daylily (Hemerocallis fulva) has been found to positively affect sleep either by ingestion or inhalation, but there is a lack of research on the sleep aid effect of its inhalation route. At present, the floral scent characteristics of daylily are not obvious, and the extraction rate of essential oil is low. Based on the above problems, this study developed daylily essence which replicates the natural aroma of plants and investigated the effect of inhaling daylily essence aroma on sleep activity by using Pittsburgh Sleep Quality Index (PSQI) and electroencephalogram (EEG). According to PSQI results, inhaling the aroma of daylily essence improved participants' subjective sleep quality. The difference between men and women was statistically significant (p <.05). The slow‐wave sleep activity of EEG in the experimental group was significantly stronger than that in the control group (p <.01). In addition, the frontal and central regions were more responsive to aroma stimuli (p <.05). The results of this study show that the aroma of daylily can promote the subjective and objective sleep quality of healthy college students, and the use of fragrance intervention is a potential scheme to improve sleep quality and insomnia. Abstract. Salicylic acid (SA) is an essential phytoregulator that is widely used to promote the synthesis of high-value nutraceuticals in plants. However, its application in daylily, an ornamental plant highly valued in traditional Chinese medicine, has not been reported. Herein, we investigated the exogenous SA-induced physiological, transcriptional and biochemical changes in long yellow daylily (LYD). We found that 2 mg/L foliar SA treatment significantly improved LYD plant growth and yield. Transcriptome sequencing and differentially expressed genes (DEGs) analysis revealed that the phenylpropanoid biosynthesis, isoquinoline alkaloid biosynthesis, sulfur metabolism, plant hormone signal transduction and tyrosine metabolism were significantly induced in SA-treated leaves. Many transcription factors and antioxidant system-related DEGs were induced under the SA treatment. Biochemical analyses showed that the leaf contents of soluble sugar, soluble protein (Cpr), ascorbic acid (AsA) and colchicine were significantly increased by 15.15% (from 30.16 ± 1.301 to 34.73 ± 0.861 mg/g), 19.54% (from 60.3 ± 2.227 to 72.08 ± 1.617 mg/g), 30.45% (from 190.1 ± 4.56 to 247.98 ± 11.652 μg/g) and 73.05% (from 3.08 ± 0.157 to 5.33 ± 0.462 μg/g), respectively, under the SA treatment. Furthermore, we identified 15 potential candidate genes for enhancing the growth, production and phytochemical content of LYD. Our results provide support for the bioaccumulation of colchicine in yellow daylily and valuable resources for biotechnological-assisted production of this important nutraceutical in Hemerocallis spp. [ABSTRACT FROM AUTHOR]

Hemerocallis citrina is an edible and medicinal homologous plant, which is rich in many nutrients and bioactive components. Terpenoids are a kind of important bioactive component in H. citrina. To understand the dominant terpenoid metabolites in H. citrina roots, stems, leaves, and flowers, respectively, this study analyzed the terpenoid metabolites in these 4 organs based on the ultra performance liquid chromatography-tandem mass spectrometry technology of widely targeted metabolomics. A total of 43 terpenoids were identified, including 7 kinds of monoterpenoids (16.2%), 18 kinds of ditepenoids (41.9%), 16 kinds of triterpene (37.2% ), and 2 kinds of triterpene saponin (4.7%). Principal component analysis and clustering analysis indicated that the mass spectrometry data were reliable, and the accumulation of terpenoids in different organs was significantly different. The dominant terpenoid metabolites in roots included isopimaradieneone, pimaradienone, abietic acid, scutebarbolide C, and isopimaric acid. The dominant terpenoid metabolites in stems included 3, 9-dihydroxy-13(14)-labden-16,15-olide and 9,19-cyclolanost-24-en-3-ol (cycloartenol). The dominant terpenoid metabolites in leaves included viteagnusin A and ent-3β-acetoxyisopimar-15-8β-ol. The dominant terpenoid metabolites in flowers included vitexilactone and 7-Ocoumaroyl- loganic acid. This study listed the relative contents of all terpenoids in roots, stems, leaves, and flowers of H. citrina in detail, which provided the foundation for further investigation of the different usages of various plant organs of H. citrina, and the information is important for the food industry and medical treatment. Because the water content of fresh daylily is high, the quality of fresh daylily decreases rapidly after harvest. It is very important to explore the suitable processing methods for extending the shelf life of daylily. The effects of different vacuum pulsating preventive technology (PVD) conditions on nutritional composition, color, flavor, rehydration rate and microstructure of daylily were investigated, so as to improve the quality of dried daylily. The effects of different drying temperatures (65, 70, 75, 80 " and 85 ") on the quality of daylilies were studied under the conditions of vacuum holding time (10 min) and atmospheric holding time (5 min) using PVD equipment. Furthermore, principal component analysis and partial least squares method were used to discriminate the drying process, flavor composition and characteristic taste of daylily. The results showed that the content of free amino acids in naturally dried (CK) daylily was the highest, at 0.858 mg/g. The !E value was the highest, at 16.47, further indicating that the overall quality of the sample was poor. After PVD drying, daylily has a bright and vibrant color, good rehydration performance, and significantly increased protein and flavonoid content. When PVD was 80 ", the primary color retention rate was the highest (!E= 7.28), and the free amino acid content was the highest (0.818 mg/g). When PVD was 75 ", the protein (7.331 g/L) and Ga (1072.445 "g/kg) content in the sample was the highest. When PVD was 70 the proportion of /3-fold and "helix content in the daylily sample was the highest, at 35%. When PVD was 65 the flavonoid content (1.653 mg/g) and rehydration rate are the highest (3.04%), and inorganic sulfides, nitrogen oxides, and terpenoids can be well retained. In addition, partial least squares discriminant analysis revealed that aspartic acid, histidine, serine, glutamic acid, and alanine are characteristic taste substances of daylily. Conclusion: Different PVD drying temperatures lead to differences in the physicochemical properties and nutritional components of daylily. Hemerocallis L. possesses abundant germplasm resources and holds significant value in terms of ornamental, edible, and medicinal aspects. However, the quality characteristics vary significantly depending on different varieties. Selection of a high-quality variety with a characteristic aroma can increase the economic value of Hemerocallis flowers. The analytic hierarchy process (AHP) is an effective decision-making method for comparing and evaluating multiple characteristic dimensions. By applying AHP, the aromatic character of 60 varieties of Hemerocallis flowers were analyzed and evaluated in the present study. Headspace solid-phase microextraction gas chromatography–mass spectrometry (HS-SPME-GC-MS) was employed to identify volatile components in Hemerocallis flowers. Thirteen volatile components were found to contribute to the aroma of Hemerocallis flowers, which helps in assessing their potential applications in essential oil, aromatherapy, and medical treatment. These components include 2-phenylethanol, geraniol, linalool, nonanal, decanal, (E)-β-ocimene, α-farnesene, indole, nerolidol, 3-furanmethanol, 3-carene, benzaldehyde and benzenemethanol. The varieties with better aromatic potential can be selected from a large amount of data using an AHP model. This study provides a comprehensive understanding of the characteristics of the aroma components in Hemerocallis flowers, offers guidance for breeding, and enhances the economic value of Hemerocallis flowers. The ecologically fragile agro-pastoral ecotone in northern China is characterized by relatively poor arable land quality. Yunzhou District in Datong City, which is situated within this transitional zone, boasts over 600 years of Hemerocallis citrina Baroni cultivation. Exploring the effects of soil physicochemical properties on daylily yield and related agronomic traits is essential for enhancing the ecological and economic value of dominant crops in ecologically fragile areas. Therefore, in this study, we focused on the daylily, a characteristic cash crop that is grown in the agro-pastoral ecotone in Yunzhou District. Physicochemical property measurement and yield estimation were performed using soil samples collected from 37 sites, with Spearman's correlation analysis, one-way analysis of variance with multiple comparisons, path analysis, and stepwise regression analysis used to analyze the generated data. The results showed the following: (1) The pathway analysis of daylily yield with each agronomic trait showed that the BN and PH directly affected the yield of daylily with direct pathway coefficients of 0.844 and 0.7, respectively, whereas the SN indirectly affected the yield of daylily through the BD and PH, with indirect pathway coefficients of 0.827 and 0.566, respectively. (2) A total of four principal components were extracted for the soil factors, of which SMC, ST and BD had large loadings on PC1; OM, TN and pH had large loadings on PC2; AK had large loadings on PC3; and AP had large loadings on PC4. (3) From the principal component regression and stepwise regression, it can be seen that SMC is the most critical factor affecting the yield of daylily, as well as the related agronomic traits, and the results also show that yield prediction was affected by OM, ST, and AK, while BN was influenced by OM and ST, and SN and PH were influenced by AP. Comparing the goodness of fit and significance of the two models, it can be concluded that the stepwise regression model is the optimal model for this study. [ABSTRACT FROM AUTHOR]

Simple Summary: This research article investigates the ploidy's impact on daylilies' (Hemerocallis spp.) response to water deficit conditions. By analyzing the morphological and physiological changes in diploid and tetraploid plants under drought stress, this study aims to elucidate the adaptive mechanisms that contribute to their differential tolerance levels. The results reveal distinct responses in the chlorophyll content, flavonoid accumulation, and nitrogen balance between the two ploidy groups, highlighting the superior stress resistance of tetraploid daylilies. This study aimed to understand the differences in the performance of diploid and tetraploid daylily cultivars under water deficit conditions, which are essential indicators of drought tolerance. This research revealed that tetraploid daylilies performed better than diploid varieties in arid conditions due to their enhanced adaptability and resilience to water deficit conditions. The analysis of the results highlighted the need to clarify the specific physiological and molecular mechanisms underlying the enhanced drought tolerance observed in tetraploid plants compared to diploids. This research offers valuable knowledge for improving crop resilience and sustainable floricultural practices in changing environmental conditions. The morphological and physiological parameters were analyzed in 19 diploid and 21 tetraploid daylily cultivars under controlled water deficit conditions, and three drought resistance groups were formed based on the clustering of these parameters. In a high drought resistance cluster, 93.3% tetraploid cultivars were exhibited. This study demonstrates the significance of ploidy in shaping plant responses to drought stress. The farming–pastoral ecotone in northern China is an ecologically vulnerable area with low-quality arable land, and cash crops are an important economic source for local farmers. Although local governments have introduced supportive policies, there are still several factors that hinder the implementation of the policies: there is a lack of sufficient research on the distribution of specialty crops, and the driving factors for agricultural planting structure adjustment are not yet clear. In this study, the specialty cash crop of the daylily planting industry in Yunzhou District, in the Farming–Pastoral Ecotone in northern China, was selected as the research object. Field surveys were conducted to collect sample points and village-level survey data, which were further combined with Sentinel-1 and Sentinel-2 data, and vegetation indices. Support vector machine (SVM) and random forest (RF) classifiers were utilized to identify daylilies and compare the accuracy using different combinations of input data. Furthermore, the classification results were counted by village, and spatial autocorrelation was used to analyze the spatial distribution pattern of daylilies. Finally, in conjunction with the village-level survey data, Spearman correlation analysis, multiple regression trees (MRT), and random forests were employed to explore the driving factors of daylily cultivation. The results indicate that using an RF classification tree of 300 resulted in the optimal method, as it achieved the highest accuracy for crop classification. The overall accuracy and daylily classification accuracy were 94.6% and 94.75%, respectively. Daylily distributions were mainly concentrated near the Sanggan River, urban areas, and the tourism industry. The distribution area of daylilies in each village was concentrated in 13.4–38.8 hm2. Spatial clustering showed more aggregation of low–low and high–high types. Labor force and daylily yield were identified as the most significant influencing factors. Further analysis of the different regions revealed the importance of industry support policies and technical training. The daylily (Hemerocallis citrina Baroni) flower is a traditional raw food material that is rich in a variety of nutrients. In particular, the content of polysaccharides in daylily is abundant and has been widely used as a functional component in food, cosmetics, medicine, and other industries. However, studies on the structure-effective relationship of daylily flower polysaccharides are still lacking. In view of this, daylily flower polysaccharides were isolated and purified, and their physical and chemical properties, structure, antioxidant activity, and adhesion-promoting effect on probiotics were evaluated. The results showed that a novel water-soluble polysaccharide (DPW) with an average molecular weight (Mw) of 2.224 kDa could be successfully isolated using column chromatography. Monosaccharide composition analysis showed that DPW only comprised glucose and fructose, with a molar ratio of 0.242:0.758. Through methylation and nuclear magnetic resonance (NMR) analysis, it was inferred that DPW belonged to the fructans group with a structure of α-D-Glcp-1→2-β-D-Fruf-1→(2-β-D-Fruf-1)n→. Antioxidant analysis showed that DPW showed strong 2-Phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl-3-Oxide (PTIO-scavenging activity with IC50 of 1.54 mg/mL. DPW of 1.25 to 5 mg/mL could significantly increase the adhesion rate of Lactobacillus acidophilu, Lactobacillus casei, Bifidobacterium adolescentis, and Lactobacillus plantarum on Caco-2 cells. Considering the above results, the present study provides a theoretical basis and practical support for the development and application of daylily polysaccharides as a functional active ingredient. The perennial herbaceous night lily, Hemerocallis citrina Baroni, is an important vegetable crop with an increasing production and consumption in China. The long lifecycle and slow growth of the night lily are becoming bottlenecks for the large-scale production of elite lines and various genetic and breeding studies. There is a lack of a protocol for rapid and efficient micropropagation for this crop. Here, we reported the systematic investigation and optimization of in vitro plant regeneration through tissue-culture-based organogenesis in the night lily variety 'Datong Huanghua'. We evaluated various factors affecting the efficiency of callus induction and subculture, shoot regeneration, rooting and plantlet establishment, including explant type and age, inoculation methods, basal culture media and the type and concentration of plant growth regulator (phytohormones) in various growth media. We developed an optimized protocol, as follows. The highest efficiency of callus induction was observed on Murashige and Skoog (MS) medium supplied with 22.7 µM TDZ (thidiazuron) using the young scape (flower stem or stalk) as the explant, which was cut longitudinally in half to produce a segment approximately 0.5 cm in length. Callus subculture and proliferation were more efficient on MS medium containing 9.0 µM 2,4-D (2,4-dichlorophenoxyacetic acid) under light culture conditions. Shoot regeneration showed the highest efficiency on MS medium supplemented with 8.9 µM 6-BA (6-benzylaminopurine) + 5.4 µM NAA (α-naphthaleneacetic acid), while the best rooting medium was MS medium containing 2.7 µM NAA. After transplanting, the transplanted regenerated seedlings showed the highest survival rate (96%) on a substrate mixture with a 2:1:1 ratio of peat/perlite/vermiculite. A protocol and flowchart for the rapid in vitro micropropagation of night lily plants is proposed that will facilitate various genetic, genomic and breeding studies on this crop. Background: Hemerocallis citrina Baroni is a traditional vegetable crop widely cultivated in eastern Asia for its high edible, medicinal, and ornamental value. The phenomenon of codon usage bias (CUB) is prevalent in various genomes and provides excellent clues for gaining insight into organism evolution and phylogeny. Comprehensive analysis of the CUB of mitochondrial (mt) genes can provide rich genetic information for improving the expression efficiency of exogenous genes and optimizing molecular-assisted breeding programmes in H. citrina. Results: Here, the CUB patterns in the mt genome of H. citrina were systematically analyzed, and the possible factors shaping CUB were further evaluated. Composition analysis of codons revealed that the overall GC (GCall) and GC at the third codon position (GC3) contents of mt genes were lower than 50%, presenting a preference for A/T-rich nucleotides and A/T-ending codons in H. citrina. The high values of the effective number of codons (ENC) are indicative of fairly weak CUB. Significant correlations of ENC with the GC3 and codon counts were observed, suggesting that not only compositional constraints but also gene length contributed greatly to CUB. Combined ENC-plot, neutrality plot, and Parity rule 2 (PR2)-plot analyses augmented the inference that the CUB patterns of the H. citrina mitogenome can be attributed to multiple factors. Natural selection, mutation pressure, and other factors might play a major role in shaping the CUB of mt genes, although natural selection is the decisive factor. Moreover, we identified a total of 29 high-frequency codons and 22 optimal codons, which exhibited a consistent preference for ending in A/T. Subsequent relative synonymous codon usage (RSCU)-based cluster and mt protein coding gene (PCG)-based phylogenetic analyses suggested that H. citrina is close to Asparagus officinalis, Chlorophytum comosum, Allium cepa, and Allium fistulosum in evolutionary terms, reflecting a certain correlation between CUB and evolutionary relationships. Conclusions: There is weak CUB in the H. citrina mitogenome that is subject to the combined effects of multiple factors, especially natural selection. H. citrina was found to be closely related to Asparagus officinalis, Chlorophytum comosum, Allium cepa, and Allium fistulosum in terms of their evolutionary relationships as well as the CUB patterns of their mitogenomes. Background: Hemerocallis citrina Baroni (Huang hua cai in Chinese) is a perennial herbaceous plant grown for its flower buds that are eaten fresh or dried and is known as the vegetarian three treasures. The nuclear genome of H. citrina has been reported, but the intraspecific variation of the plastome (plastid genome) has not yet been studied. Therefore, the panplastome of this species collected from diverse locations is reported here for the first time. Results: In this study, 65 H. citrina samples were resequenced, de novo assembled, and aligned with the published plastome of H. citrina to resolve the H. citrina panplastome. The sizes of the 65 newly assembled complete plastomes of H. citrina ranged from 156,048 bp to 156,263 bp, and the total GC content ranged from 37.31 to 37.34%. The structure of the complete plastomes showed a typical tetrameric structure, including a large single copy (LSC), a small single copy (SSC), and a pair of inverted repeat regions (IRA and IRB). Many nucleotide variants were identified between plastomes, among which the variants in the intergenic spacer region were the most abundant, with the highest number of variants concentrated in the LSC region. Based on the phylogenetic tree constructed using the ML method, population structure analysis, and principal component analysis (PCA), the panplastome data were subdivided into five genetic clusters. The C5 genetic cluster was mostly represented by samples from Qidong, Hunan Province, while samples from Shanxi and Shaanxi Provinces were classified into the C4 genetic cluster. The greatest genetic diversity was found in the C1 genetic cluster, and the greatest genetic distance between any two clusters was found between the C4 and C5 clusters. Conclusion: The resolution of the panplastome and the analysis of the population structure of H. citrina plastomes provide important data for future breeding projects and germplasm preservation. Plant polysaccharides are important for anti-aging research. Polysaccharides from Hemerocallis citrina Baroni (H. citrina) have been reported to have antioxidant activity; however, their anti-aging roles and mechanisms are not clear. In this study, we extracted polysaccharides from H. citrina by an ultrasonic-assisted water extraction–alcohol precipitation method and chemically determined the physicochemical properties such as extraction yield, content, and in vitro antioxidant properties of H. citrina polysaccharide-rich extract (HCPRE). Using Caenorhabditis elegans (C. elegans) as a model animal, the anti-aging effect of HCPRE was investigated, and the mechanism of action of HCPRE was explored by the in vivo antioxidant level assay of C. elegans and the related gene expression assay. The extraction yield of HCPRE was 11.26%, the total polysaccharide content was 77.96%, and the main monosaccharide components were glucose and galactose. In addition, HCPRE exhibited good antioxidant activity both in vitro and in vivo. Under normal thermal stress and oxidative stress conditions, being fed 1200 µg/mL of HCPRE significantly prolonged the life span of C. elegans by 32.65%, 17.71%, and 32.59%, respectively. Our study showed that HCPRE exerted an anti-aging effect on C. elegans, and its mechanism involves increasing the activities of catalase (CAT) and superoxide dismutase (SOD), reducing the level of reactive oxygen species (ROS) and regulating the expression of related genes. Hemerocallis citrina Baroni (daylily) is an important cash crop in many regions of China. Daylily rust caused by Puccinia hemerocallidis (Ph) is a major disease in H. citrina. Rust infection on the leaves of two resistant and susceptible varieties of H. citrina was examined. Transcriptome sequencing and defense enzyme activity were used to analyze the differential expression of related genes after 0, 72, 144, 192, and 288 h, to reveal the response mechanism of H. citrina to rust infection and screen-related resistance genes. The results suggested that 72 h after Ph infection was the key time point for determining differences between resistant and susceptible varieties. DEGs were significantly enriched in phenylpropanoid biosynthesis, plant–pathogen interaction, plant hormone signal transduction, and the MAPK signaling pathway. TFs WRKY33 and CYP98A are key genes closely related to the disease-resistance response of H. citrina. This study is the first large-scale transcriptomic analysis of H. citrina. Simple Summary: To date, few studies in China and abroad have investigated the utilization of daylily. Currently, there is no literature on the evaluation of the feed utilization value of daylily. By using biological fermentation technology, we developed a formula for replacing corn silage with daylily as a feed source for Tan sheep by analyzing coated silage production, diet formulation optimization, growth performance indicators, slaughter performance indicators, carcass quality, and blood biochemical indicators. Based on the feeding test and safety evaluation, the effect of daylily silage on the growth performance and slaughter performance of Tan sheep was evaluated, and the significance of daylily for feed utilization was determined, which provided a scientific basis for further scientific and rational utilization of daylily stems and leaves as feed resources. The shortage of high-quality coarse feed resources is the main factor that restricts the development of animal husbandry in many developing countries. The present study aimed to investigate the effects of replacing corn silage with daylily silage on the growth performance, slaughter performance, blood biochemical indicators, meat quality, and muscle amino acid composition of Tan sheep. A total of 72 healthy Tan sheep were randomly assigned to four groups. In each group, 0%, 20%, 40%, and 60% of corn silage were replaced with daylily silage (denoted as CON, HC20, HC40, and HC60, respectively). Tan sheep fed with daylily silage showed no significant adverse effects on their growth performance, meat quality, and muscle amino acid composition (p > 0.05). Some increase was observed in the carcass fat content value (GR-value, p < 0.05), thickness of backfat (p < 0.05), and the blood urea level (p < 0.05). These findings indicate that the utilization of daylily silage instead of whole-plant corn silage has no adverse effects on the growth performance and meat quality of Tan sheep, thus indicating that it can partially replace whole-plant corn feed as a feed resource for Tan sheep. Hemerocallis is a characteristic vegetable with outstanding edible and medicinal value. Flavonoids are important bioactive components of Hemerocallis. To improve the extraction efficiency and detection accuracy of flavonoids from Hemerocallis, we established a high-performance liquid chromatography (HPLC) detection system, which can simultaneously detect multiple flavonoids. In addition to the previously developed organic solvent extraction method, an ultrasonic-assisted extraction technique that uses fewer samples was established to extract flavonoids from Hemerocallis. The extraction conditions of the ultrasonic-assisted extraction were optimized via a single-factor experiment and a response surface experiment. The HPLC system detected and determined the contents of rutin, isoquercetin, myricetin, quercetin, apigenin, and diosmetin from 70 Hemerocallis germplasm resources. In addition, we evaluated the antioxidant activity of flavonoids in Hemerocallis using DPPH free radical scavenging capacity with ascorbic acid (Vc) as a positive control. The results showed that the optimum conditions for the ultrasonic extraction process were as follows: sample weight of 0.25 g, ethanol volume fraction of 72%, ethanol volume of 2.5 mL, and ultrasonic extraction time of 17 min. Under these conditions, flavonoid extraction had a strong scavenging effect on DPPH. With the increase in the sample solutions' concentrations, its antioxidant capacity was gradually enhanced, and the DPPH scavenging rate reached 70.2%. The optimized ultrasonic-assisted extraction technology can increase the total content of six flavonoids in day lily bud by 59.01%, especially the content of rutin (increased by 64.41%) in Hemerocallis flower buds. Among 70 Hemerocallis plant resources, we selected materials H0087 and H0059 with high and stable flavonoid content, with the total content of six substances being 4390.54 ug/g and 3777.13 ug/g. Thus, this study provides a reference for extracting and determining flavonoid contents in Hemerocallis materials. It also provides a theoretical basis for high-quality individual plant breeding. Chromosome doubling in ornamental plants, as shown by our study in daylilies (Hemerocallis spp.), has great potential to increase tolerance to abiotic stress. Drought is the most critical growth-limiting factor in a changing climate. Drought tolerance is one of the decisive factors for the survival, productivity, and appearance of perennial ornamental plants. Understanding and elucidating the molecular mechanisms that determine plant response to abiotic stress is essential. De novo transcriptome assembly of diploid and autotetraploid Hemerocallis spp. cv. Trahlyta was performed under artificially induced stress to elucidate the molecular mechanisms related to plant response to drought. In daylily mRNA, 237,886 transcripts were detected, and 42.4% of them were identified as annotated unigenes. In the experiment, diploid plants were more stressed, with 2871 upregulated or downregulated DEGs (differentially expressed genes) responding to drought, while tetraploid plants had 1599 DEGs. The proportion of upregulated DEGs differed by 1.3 times between diploid and autotetraploid genotypes, whereas the proportion of downregulated DEGs was 1.8 times greater in diploid plants. Signaling pathways related to the drought response were activated in daylilies, and key candidate genes were identified in both ploidy genotypes. In autotetraploid plants, more drought-related pathways were activated than in diploids—43 and 19, respectively. The most abundant DEGs in both cases were KEGG (Kyoto Encyclopedia of Genes and Genomes), metabolic (ko01100), and biosynthesis of secondary metabolites (ko01110) pathways. Summarizing the data, it was found that autotetraploid plants of the daylily have a wider potential for adaptation to drought stress. Therefore, they adapt faster and better to adverse drought conditions by activating alternative signaling pathways. The comparative transcriptome analysis of diploid and autotetraploid plants allows us to understand the molecular mechanisms of drought resistance and it is also essential for daylily breeding programs to develop drought-resistant genotypes in the future. Hemerocallis citrina Baroni with different maturity levels has different uses for food and medicine and has different economic benefits and sales value. However, the growth speed of Hemerocallis citrina Baroni is fast, the harvesting cycle is short, and the maturity identification is completely dependent on experience, so the harvesting efficiency is low, the dependence on manual labor is large, and the identification standard is not uniform. In this paper, we propose a GCB YOLOv7 Hemerocallis citrina Baroni maturity detection method based on a lightweight neural network and attention mechanism. First, lightweight Ghost convolution is introduced to reduce the difficulty of feature extraction and decrease the number of computations and parameters of the model. Second, between the feature extraction backbone network and the feature fusion network, the CBAM mechanism is added to perform the feature extraction independently in the channel and spatial dimensions, which improves the tendency of the feature extraction and enhances the expressive ability of the model. Last, in the feature fusion network, Bi FPN is used instead of the concatenate feature fusion method, which increases the information fusion channels while decreasing the number of edge nodes and realizing cross-channel information fusion. The experimental results show that the improved GCB YOLOv7 algorithm reduces the number of parameters and floating-point operations by about 2.03 million and 7.3 G, respectively. The training time is reduced by about 0.122 h, and the model volume is compressed from 74.8 M to 70.8 M. In addition, the average precision is improved from 91.3% to 92.2%, mAP@0.5 and mAP@0.5:0.95 are improved by about 1.38% and 0.20%, respectively, and the detection efficiency reaches 10 ms/frame, which meets the real-time performance requirements. Background: Hemerocallis citrina Baroni is a traditional medical and edible plant. It is rich in flavonoid compounds, which are a kind of important bioactive components with various health benefits and pharmaceutical value. However, the flavonoid metabolomics profile and the comparison of flavonoid compounds from different parts of H. citrina is scarce. Results: In this study, flavonoid metabolites were investigated from roots, stems, leaves and flowers of H. citrina. A total of 364 flavonoid metabolites were identified by UPLC-MS/MS based widely targeted metabolomics, and the four plant parts showed huge differences at flavonoid metabolic level. Compared to roots, 185, 234, and 119 metabolites accounted for upregulated differential flavonoid metabolites (DFMs) in stems, leaves, and flowers, respectively. Compared to stems, 168 and 29 flavonoid metabolites accounted for upregulated DFMs in leaves and flowers, respectively. Compared to leaves, only 29 flavonoid metabolites accounted for upregulated DFMs in flowers. A number of 35 common flavonoid metabolites were observed among six comparison groups, and each comparison group had its unique differential metabolites. Timely detection and management of daylily diseases are crucial to prevent yield reduction. However, detection models often struggle with handling the interference of complex backgrounds, leading to low accuracy, especially in detecting small targets. To address this problem, we propose DaylilyNet, an object detection algorithm that uses multi-task learning to optimize the detection process. By incorporating a semantic segmentation loss function, the model focuses its attention on diseased leaf regions, while a spatial global feature extractor enhances interactions between leaf and background areas. Additionally, a feature alignment module improves localization accuracy by mitigating feature misalignment. To investigate the impact of information loss on model detection performance, we created two datasets. One dataset, referred to as the 'sliding window dataset', was obtained by splitting the original-resolution images using a sliding window. The other dataset, known as the 'non-sliding window dataset', was obtained by downsampling the images. Experimental results in the 'sliding window dataset' and the 'non-sliding window dataset' demonstrate that DaylilyNet outperforms YOLOv5-L in mAP@0.5 by 5.2% and 4.0%, while reducing parameters and time cost. The flowers of daylily (Hemerocallis fulva Linn.) have been used as vegetable and medicinal herb for thousands of years in Taiwan and eastern Asia. Daylily flowers have been demonstrated to exert several biomedical properties. In this study, we provided the evidences show that daylily flowers exert anti inflammatory activity in vitro and improved the sleep quality in vivo. We demonstrated that adult volunteers received water extract of daylily flowers improved sleep quality, sleep efficiency and daytime functioning, while sleep latency was reduced, compared to the adult volunteers received water. In addition, we demonstrated that aqueous and ethanol extracts of daylily flowers inhibited nitric oxide and interleukin 6 production in lipopolysaccharide activated macrophages. Furthermore, the quantitative high performance liquid chromatography based analysis showed the rutin content of the aqueous extract, ethanolic extract, ethyl acetate fractions of ethanolic extract, and water fractions of ethanolic extract were 7.27, 23.30, 14.71, and 57.43 ppm, respectively. Exploring microRNA (miRNA) expression patterns in Hemerocallis under low-temperature stress helps understanding the mechanisms of miRNA-mediated stress responses. Here, eight separate libraries were generated from the leaves and roots of two different varieties, cold-tolerant 'Jinyan 7' and cold-sensitive 'Lucretius 4', which were exposed to 20°C and −4°C for 8 h. In total, 333 cold-responsive miRNAs, including 85 putative novel and 248 conserved miRNAs, were identified during different temperature treatments. Under cold-stress conditions, two miRNAs were down-regulated and one was up-regulated in the roots, and no differentially expressed miRNAs were detected in the leaves in 'Lucretius 4'. In 'Jinyan 7', there were two novel and one conserved differentially expressed miRNAs in leaves, whereas three conserved and two novel miRNAs were differentially expressed in roots. The target genes were mainly involved in metabolic processes and cellular components. The differential expression levels of 10 miRNAs were validated using quantitative RT–PCR. This article discusses the occurrence of the daylily gall midge, Contarinia quinquenotata, in New Brunswick. The species is originally from Asia and has been a garden pest in the United Kingdom since 1989. It has been spreading rapidly in Europe and was first reported in North America in 2001. The midge lays eggs in daylily flower buds, causing them to become deformed and fail to open. While infestations do not kill the plants, they damage the ornamental blooms. Pesticide trials have had limited success in controlling the infestations. The article reports the first presence of C. quinquenotata in New Brunswick, with deformed flower buds observed in a residential garden. Daylily (Hemerocallis spp) rust caused by Puccinia hemerocallidis is a common disease that has become a global problem. This study identified and characterized the causal agent of daylily rust in east China. Based on the internal transcribed spacer (ITS) sequences, a phylogenetic tree of the P. hemerocallidis from China and other geographical regions was constructed, and the result showed that P. hemerocallidis from daylilies in east China grouped with other P. hemerocallidis isolates collected from North-, Central-American and Europe. In addition, the effects of incubation condition, temperature, and light on urediniospores germination were investigated in vitro. The results showed that the P. hemerocallidis urediniospores could germinate at 10–40 ℃ under light intensity of 10–100 Klux, but they hardly germinated at 5 ℃. W-WA medium (water agar medium covered with sterile water) was the most suitable in vitro germination substrate, combined with the temperature of 20–30 ℃ and light intensity of 40–70 Klux. The photolysis of colchicine under ultraviolet and visible light irradiation was studied by ultraviolet (UV) scanning and HPLC-MS. The photoproduct was proposed and the cytotoxicity change before and after irradiation was investigated. Results showed that both ultraviolet and visible light irradiation could effectively degrade colchicine into deacetamido-lumicolchicine. The process conformed to first-order kinetics, in which a high degradation rate (K = 0.5862 h− 1) was observed when colchicine was dissolved in ethanol and irradiated by UV light. Cell viability and cell cycle studies proved that a photolysis treatment of colchicine could weaken the cytotoxicity effectively. Colchicine inhibited the division of BRL 3 A cells in G2/M phase with an IC50 value of 0.48 µg/mL, while the toxic effect could be reduced significantly with IC50 2.1 µg/mL when colchicine was exposed to UV irradiation. Results are beneficial to the toxicity elimination of colchicine in the processing of daylily flower in food industry, and can also provide photochemistry reference for colchicine-related studies.
