The current study investigated the effects of stocking density (SD) on the performance, tibia mineralization, and the hypothalamic appetite genes expression in broilers. A total of 2,800 1-d-old male broilers (Cobb 500) were distributed in a completely randomized design to 1 of 5 SD treatments with 8 replicate cages for each treatment. The SD treatments were 12.5, 15.0, 17.5, 20.0, and 22.5 birds/m2, corresponding to 50, 60, 70, 80, and 90 birds per cage (4 m2/cage), respectively. The concentration of tibia phosphorus was determined by the ammonium metavanadate colorimetric method and the mRNA abundance in different tissues was measured by the real-time quantitative PCR method. The data were analyzed by the one-way and/or two-way analysis of variance and polynomial contrasts were used to determine the effect of increasing SD. Feed intake linearly decreased (P < 0.05) with increasing SD during d 1-42 production period. On d 42, body weight and tibia breaking strength were significantly lower in the groups of 17.5, 20.0 and 22.5 birds/m2 than in the groups of 12.5 and 15 birds/m2 (P < 0.01). Concentrations of ash and phosphorus in the tibia of broilers linearly decreased (P < 0.03) with increasing SD on d 42. The SD of 22.5 birds/m2 decreased the mRNA abundance of neuropeptide Y (NPY), NPY-receptor (NPYR) 1, and NPYR2 (P < 0.05), while it increased melanocortin receptor 4 mRNA abundance (P = 0.012) in the hypothalamus of broilers as compared with the SD of 12.5 birds/m2 on d 21 and 42. The mRNA abundance of hypothalamic cocaine and amphetamine-regulated transcript (CART), corticotrophin-releasing factor (CRF), and CRF-receptor 1 (CRFR1) were higher (P < 0.05) in the group of 22.5 birds/m2 than in the group of 12.5 birds/m2 on d 21. We concluded that increasing stocking density beyond 15 birds/m2 (corresponding to the 45 kg/m2 at 42 days of age) suppressed final BW and bone mineralization of broilers raised in multitier cage system. Objective: The objective of this study was to determine whether a dietary vitamin E (VE) supplement could alleviate any detrimental effects of aged corn on lipid metabolism and antioxidant status in laying hens. Methods: The experiment consisted of a 2×3 factorial design with two corn types (normal corn and aged corn (stored for 4 yr) and three concentrations of VE (0, 20, and 100 IU/kg). A total of 216 Lohmann laying hens (50 wk of age) were randomly allocated into six treatment diets for 12 wk. Each treatment had 6 replicates of 6 hens per replicate. Results: The results show that aged corn significantly decreased the content of low-density lipoprotein cholesterol (p<0.05), and reduced chemokine-like receptor 1 (CMKLR1) mRNA expression (p<0.05) in the liver compared to controls. Diet with VE did not alter the content of crude fat and cholesterol (p>0.05), or acetyl-CoA carboxylase, lipoprotein lipase, fatty acid synthase or CMKLR1 mRNA expression (p>0.05) in the liver among treatment groups. Aged corn significantly increased the content of malondialdehyde (MDA) (p<0.05) and decreased superoxide dismutase (SOD) activity (p<0.05) in the liver. The VE increased the content of MDA (p<0.05) but decreased glutathione peroxidase (GSH-Px) activity in serum (p<0.01) and in the ovaries (p<0.05). Adding VE at 20 and 100 IU/kg significantly increased GSH-Px activity (p<0.05) in liver and in serum (p<0.01), 100 IU/kg VE significantly increased SOD activity (p<0.05) in serum. Aged corn had no significant effects on GSH-Px mRNA or SOD mRNA expression (p<0.01) in the liver and ovaries. Addition of 100 IU/kg VE could significantly increase SOD mRNA expression (p<0.01) in the liver and ovary. Conclusion: Aged corn affected lipid metabolism and decreased the antioxidant function of laying hens. Dietary VE supplementation was unable to counteract the negative effects of aged corn on lipid metabolism. As the understanding of the pathways involved in such effect are quite limited, we investigated the gene pathways that modulate lipid metabolism in layers and the fatty acid profiles of the yolk of layers that were challenged with dietary vanadium (V) and supplemented with epigallo-catechin-3-gallate (EGCG). For this purpose, a total of 120 hens were divided into four groups which were fed the following experimental diets for a period of 8 weeks: control (basal diet), V10 (control + 10 mg/kg V), EGCG130 (V10 + 130 mg/kg EGCG), and EGCG217 (V10 + 217 mg/kg EGCG). Blood total cholesterol, triglyceride, glucose, and very low-density lipoprotein-cholesterol concentration were lower in V10, EGCG130, and EGCG217 groups compared to the control group, while total cholesterol and triglyceride content in blood were lower in the EGCG217 group than in V10 group (P < 0.05). Hens consumed V10 diet had the highest triglyceride content in liver among treatments, whereas EGCG130 and EGCG217 groups had lower values when compared to those observed in the control group (P < 0.01). Dietary inclusion of V increased yolk polyunsaturated fatty acid (PUFA) and total unsaturated fatty acid (UFA) content compared to the control group (P < 0.05), whereas the addition of either 130 or 217 mg/kg EGCG in V containing diet resulted in similar yolk PUFA and UFA contents with those observed in the control group. Treatment with V alone upregulated the expression of hepatic fatty acid synthase (FAS) and sterol-regulator element-binding protein 1 (SREBP1), while EGCG downregulated FAS and SREBP1 expressions in contrast to V10 treatments (P < 0.01). Liver gene expression peroxisome proliferator-activated receptor gamma (PPARγ) was lower in the V10 than in the control group while EGCG inclusion groups upregulated their expression (P < 0.05). In conclusion, the data gathered in this study indicate that dietary V and EGCG alter the layers' lipid metabolism and fat deposition pattern in egg yolk, which might be associated with their modulatory effect on lipogenesis-related gene (FAS, SREBP1, and PPARγ) expression. This study investigated the effects of maternal canthaxanthin ( CX, 6 mg/kg) and 25-hydroxycholecalciferol (25- OH-D3, 0.069 mg/kg) supplementation on the performance of Cherry Valley ducklings under two different vitamin regimens. A total of 780 duck breeder females and 156 males were randomly allotted to two diets with or without the addition of the mixture of CX and 25- OH-D3 (CX+25-OH-D3) for 32 weeks. Ducklings (males and females separately) hatched from eggs laid at 24 weeks of the duck breeder trial were fed with a NRC vitamin regimen, and ducklings (males and females separately) hatched from eggs laid at 32 weeks of the duck breeder trial were fed with a HIGH vitamin regimen (had higher levels of all vitamins except biotin than NRC vitamin regimen), for 14 days. The results showed that, maternal CX+25-OH-D3 supplementation increased the shank pigmentation for 7-days post hatch in ducklings under a NRC vitamin regimen, and for 14-days post hatch in ducklings under a HIGH vitamin regimen. Growth performance, antioxidant status and serum phosphorus of ducklings under a NRC vitamin regimen were increased by maternal CX+25-OH-D3 supplementation; however, these positive effects were not observed in ducklings under a HIGH vitamin regimen. Males revealed increased growth performance in ducklings under both NRC and HIGH vitamin regimens. Sexual differences in shank pigmentation, antioxidant status, tibia strength and serum phosphorus were not consistent as they were dependent on maternal CX+25- OH-D3 status or dietary vitamin regimens. This study assessed the effect of dietary vanadium (V) and vitamin C ( VC) on production performance, egg quality and antioxidant status in laying hens. A total of 360 laying hens (31-week-old) were randomly allotted into a 3 × 3 factorial arrangement treatments (four replicates and 10 chicks per replicate) with three levels of dietary V (0, 5 and 10 mg/kg) and three levels of vitamin C (0, 50 and 100 mg/kg) for 12 weeks. The effect of V and VC did not alter egg production, egg weight, average daily feed intake and feed conversion ratio during 1-12 week. Albumen height and Haugh unit value were linearly decreased (p < 0.001) by addition of V, whereas the effect of 100 mg/kg VC was observed to counteract (p < 0.05) this effect in V-containing treatments during 1-12 week. Hens fed V-containing diet laid lighter (linear effect, p < 0.05) coloured eggs (higher lightness value, lower redness and yellowness value), and the VC exerted no influence on it during 1-12 week. The serum superoxide dismutase ( SOD), catalase ( CAT) and glutathione peroxidase ( GSH-Px) activities, ability to inhibit hydroxyl radical, were significantly decreased, and the malondialdehyde ( MDA) and V contents were increased (p < 0.05) by effect of V during 4, 8 and 12 week. The effect of VC alone and the interactive effect between VC and V were shown to increase serum (p < 0.05) SOD activity in 4 week and decrease MAD levels in 12 week. The result indicate that V decreased the egg quality and caused the oxidative stress at level of 5 mg/kg and 10 mg/kg, and the addition of 100 mg/kg vitamin C can alleviate its egg quality reduction effect and can mitigate the oxidative stress to some extent. Two Schiff base copper( II) complexes, bis(N-furfurylsalicylaldiminato)copper( II) ( I) and bis(N-tetrahydrofurfurylsalicylaldiminato)copper( II) ( II), were synthesized and their solid state structures were determined by X-ray crystallography. Complex I has a square planar geometry. In contrast, complex II displays a distorted square planar geometry. Thus, the geometry around copper in the solid state structures of I and II is determined by a combination of steric and electronic effects, as well as by crystal packing forces. The most stable structures of gaseous Si m C n (3 ⩽ n+ m ⩽ 6) clusters in their ground electronic states are determined with the high level electronic correlation method QCISD(T)/g3large. Thermodynamic properties on heat capacity ( C ), entropy ( S ), Gibbs energy function (−[ G Θ − H Θ( T r )]/ T) and enthalpy function ( H Θ− H Θ( T r )) are predicted with standard statistical thermodynamics using the structure parameters and vibrational frequencies obtained with B3PW91/6-31G( d) method combined with the electronic excitation energies determined with time dependent density functional (TD DFT) method at B3PW91/6-31G( d) level. The electronic energies are calculated with the accurate model chemistry method at G3(QCI) level of theory and the Δ f H (0 K), Δ f H (298.15 K) and Δ f G (298.15 K) values are predicted. The heat capacities C ( T) as a function of temperature within 298.15-2000 K are fitted into analytical equations. The thermodynamic functions at higher temperatures are determined classically by using these equations. Most of the results obtained in this work are consistent with the available experiments. The effective elastic properties of carbon fibre reinforced silicon carbide composite (C/SiC) with multilayered matrix manufactured by chemical vapour infiltration method are studied in the present paper. Based on the material tomography of scanning electron microscopy, distributions of voids and microcracks are identified and taken into account in the finite element model of such C/SiC composite. It is shown the proposed two scale finite element models of multilayered C/SiC composites can characterise physically the real situation of involved voids and microcracks related to the chemical vapour infiltration process. The transcription factor early growth response gene (Egr-1) is a stress response gene activated by various forms of stress. The effect of silica on transcription and expression of Egr-1 was investigated in rat lung and in RAW264.7 cells. Silica induced the expression of Egr-1 in vivo and was mainly located in alveolar macrophage cells and lung epithelial cells. Furthermore, silica induced Egr-1 mRNA and protein expression in cultured RAW264.7 cells. Immunofluorescence microscopy revealed translocation of Egr-1 to the nucleus in response to silica. The contribution of the extracellular signal-regulated kinase (ERK) pathway to the activation of Egr-1 in response to silica was examined. Exposure to silica resulted in a rapid phosphorylation of ERK 1/2 kinases in RAW264.7 cells. MAP Kinase Kinase (MEK) inhibitor U0126 prevented Egr-1 induction by silica. The results suggest that silica could induce Egr-1 activation in macrophages in vivo and in vitro and that phosphorylated ERK 1/2 may be involved in this action. [ABSTRACT FROM AUTHOR]
