The synthesis of carbon dots (CDs) is gaining wide-ranging interest due to their broad applicability, owing to their small size and luminescence. CDs were prepared from charcoal via a one-step process using laser ablation in liquid without the use of reagents. The adopted method was based on the use of a commercially available continuous wave (CW) laser diode emitting a 450 nm wavelength and, for the liquid, a phosphate-buffered saline (PBS) solution, routinely used in the biological field. Photoluminescence analysis revealed fluorescence, at 480 nm, increasing with laser irradiation time. The atomic force microscopy (AFM) of the CDs revealed an average sphere shape with a size of about 10 nm. Biodegradable polycaprolactone (PCL), typically adopted in biomedicine applications, was used as a matrix to show the preserved luminescence, ideal for the non-invasive monitoring of implanted scaffolds in tissue engineering.

Vitamin D3 is a crucial fat-soluble pro-hormone essential for bolstering bone health and fortifying immune responses within the human body. Orodispersible films (ODFs) serve as a noteworthy formulation strategically designed to enhance the rapid dissolution of vitamin D, thereby facilitating efficient absorption in patients. This innovative approach not only streamlines the assimilation process but also plays a pivotal role in optimizing patient compliance and therapeutic outcomes. The judicious utilization of such advancements underscores a paradigm shift in clinical strategies aimed at harnessing the full potential of vitamin D for improved patient well-being. This study aims to examine the vitamin D3 ODF structure using spectroscopic techniques to analyze interactions with excipients like mannitol. Fourier-transform infrared spectroscopy (FTIR) and ultraviolet-visible (UV-Vis) spectroscopy were utilized to assess molecular composition, intermolecular bonding, and vitamin D3 stability. Understanding these interactions is essential for optimizing ODF formulation, ensuring stability, enhancing bioavailability, and facilitating efficient production. Furthermore, this study involves a translational approach to interpreting chemical properties to develop an administration protocol for ODFs, aiming to maximize absorption and minimize waste. In conclusion, understanding the characterized chemical properties is pivotal for translating them into effective self-administration modalities for Vitamin D films.

Carbon dots (CDs), owing to their excellent photoluminescent features, have been extensively studied for physics preparation methods and for biomedical and optoelectronic device applications. The assessment of the applicability of CDs in the production of luminescent polymeric composites used in LEDs, displays, sensors, and wearable devices is being pursued. The present study reports on an original, environmentally friendly, and low-cost route for the production of carbon dots with an average size of 4 nm by laser ablation in liquid. Jointly, to prove the significance of the study for a wide range of applications, a free-standing flexible polyvinyl alcohol (PVA) composite containing photoluminescent carbon dots was manufactured. CDs were prepared using targets of porose charcoal with a density of 0.271 g/cm 3 placed in phosphate-buffered saline (PBS) liquid solution and irradiated for 30 min by pulsed IR diode laser. The optical properties of the obtained suspension containing carbon dots were studied with UV-ViS and FTIR spectroscopies. The photoluminescence of the produced carbon dots was confirmed by the emission peak at 480 nm in the luminescence spectrum. A narrow luminescence band with a full width at half-maximum (FWHM) of less than 40 nm could be an asset in spectral emission analysis in different applications. Atomic force microscopy confirms the feasibility of manufacturing CDs in clean and biocompatible environments, paving the way for an easier and faster production route, crucial for their wider applicability.

Cyclic olefin copolymer (COC) is a novel type of thermoplastic polymer gaining the attention of the scientific community in electronic, optoelectronic, biomedicine and packaging applications. Despite the benefits in the use of COC such as undoubted optical transparency, chemical stability, a good water-vapor barrier and biocompatibility, its original hydrophobicity restricts its wider applicability and optimization of its performances. Presently, we report on the optical and morphological properties of the films of COC covered with Ti in selected areas. The layer of Ti on COC was deposited by pulsed lased deposition processing. The Ti/COC film was characterized by UV-Vis spectroscopy indicating that its transmittance in the visible region decreased by about 20% with respect to the pristine polymer. The quality of the deposited Ti was assessed with the morphology by scanning electron (SEM) and atomic force microscopies (AFM). The modification of the wettability was observed by the sessile drop method indicating a reduction of the native hydrophilicity.

SiC detectors based on a Schottky junction represent useful devices to characterize fast laser-generated plasmas. High-intensity fs lasers have been used to irradiate thin foils and to characterize the produced accelerated electrons and ions in the target normal sheath acceleration (TNSA) regime, detecting their emission in the forward direction and at different angles with respect to the normal to the target surface. The electrons' energies have been measured using relativistic relationships applied to their velocity measured by SiC detectors in the time-of-flight (TOF) approach. In view of their high energy resolution, high energy gap, low leakage current, and high response velocity, SiC detectors reveal UV and X-rays, electrons, and ions emitted from the generated laser plasma. The electron and ion emissions can be characterized by energy through the measure of the particle velocities with a limitation at electron relativistic energies since they proceed at a velocity near that of the speed of light and overlap the plasma photon detection. The crucial discrimination between electrons and protons, which are the fastest ions emitted from the plasma, can be well resolved using SiC diodes. Such detectors enable the monitoring of the high ion acceleration obtained using high laser contrast and the absence of ion acceleration using low laser contrast, as presented and discussed.

Thin sheets of ultra-high molecular weight polyethylene (UHMWPE), both in pristine form and containing carbon nanotubes (CNTs) or Fe 2 O 3 nanoparticles (NPs) at different concentrations, were prepared. The CNT and Fe 2 O 3 NP weight percentages used ranged from 0.01% to 1%. The presence of CNTs and Fe 2 O 3 NPs in UHMWPE was confirmed by transmission and scanning electron microscopy and by energy dispersive X-ray spectroscopy analysis (EDS). The effects of the embedded nanostructures on the UHMWPE samples were studied using attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy and UV-Vis absorption spectroscopy. The ATR-FTIR spectra show the characteristic features of the UHMWPE, CNTs, and Fe 2 O 3 . Concerning the optical properties, regardless of the type of embedded nanostructures, an increase in the optical absorption was observed. The allowed direct optical energy gap value was determined from the optical absorption spectra: in both cases, it decreases with increasing CNT or Fe 2 O 3 NP concentrations. The obtained results will be presented and discussed.

In the present study, graphene oxide foils 10 μm thick have been irradiated in vacuum using same charge state (one charge state) ions, such as protons, helium and oxygen ions, at the same energies (3 MeV) and fluences (from 5 × 10 11 ion/cm 2 to 5 × 10 14 ion/cm 2 ). The structural changes generated by the ion energy deposition and investigated by X-ray diffraction have suggested the generation of new phases, as reduced GO, GO quantum dots and graphitic nanofibers, carbon nanotubes, amorphous carbon and stacked-cup carbon nanofibers. Further analyses, based on Rutherford Backscattering Spectrometry and Elastic Recoil Detection Analysis, have indicated a reduction of GO connected to the atomic number of implanted ions. The morphological changes in the ion irradiated GO foils have been monitored by Transmission Electron, Atomic Force and Scanning Electron microscopies. The present study aims to better structurally, compositionally and morphologically characterize the GO foils irradiated by different ions at the same conditions and at very low ion fluencies to validate the use of GO for radiation detection and propose it as a promising dosimeter. It has been observed that GO quantum dots are produced on the GO foil when it is irradiated by proton, helium and oxygen ions and their number increases with the atomic number of beam gaseous ion.

Typically, polymeric composites containing nanoparticles are realized by incorporating pre-made nanoparticles into a polymer matrix by using blending solvent or by the reduction of metal salt dispersed in the polymeric matrix. Generally, the production of pre-made Au NPs occurs in liquids with two-step processes: producing the gold nanoparticles first and then adding them to the liquid polymer. A reproducible method to synthetize Au nanoparticles (NPs) into polydimethylsiloxane (PDMS) without any external reducing or stabilizing agent is a challenge. In this paper, a single-step method is proposed to synthetize nanoparticles (NPs) and at the same time to realize reproducible porous and bulk composites using laser ablation in liquid. With this single-step process, the gold nanoparticles are therefore produced directly in the liquid polymer. The optical properties of the suspensions of AuNPs in distilled water and in the curing agent have been analyzed by the UV-VIS spectroscopy, employed in the transmission mode, and compared with those of the pure curing agent. The electrical dc conductivity of the porous PDMS/Au NPs nanocomposites has been evaluated by the I-V characteristics. Scanning electron microscopy (SEM) and energy dispersive X-ray (EDX) analysis have monitored the composition and morphology of the so-obtained composites and the size of the fabricated Au nanoparticles. Atomic force microscopy (AFM) has been used to determine the roughness of the bulk PDMS and its Au NP composites.

The conformational variation of the viral capsid structure plays an essential role both for the environmental resistance and acid nuclear release during cellular infection. The aim of this study was to evaluate how capsid rearrangement in engineered phages of M13 protects viral DNA and peptide bonds from damage induced by UV-C radiation. From in silico 3D modelling analysis, two M13 engineered phage clones, namely P9b and 12III1, were chosen for (i) chemical features of amino acids sequences, (ii) rearrangements in the secondary structure of their pVIII proteins and (iii) in turn the interactions involved in phage capsid. Then, their resistance to UV-C radiation and hydrogen peroxide (H 2 O 2 ) was compared to M13 wild-type vector (pC89) without peptide insert. Results showed that both the phage clones acquired an advantage against direct radiation damage, due to a reorganization of interactions in the capsid for an increase of H-bond and steric interactions. However, only P9b had an increase in resistance against H 2 O 2 . These results could help to understand the molecular mechanisms involved in the stability of new virus variants, also providing quick and necessary information to develop effective protocols in the virus inactivation for human activities, such as safety foods and animal-derived materials.

This work describes the development of a new method for ion implantation induced crystal damage recovery using multiple XeCl (308 nm) laser pulses with a duration of 30 ns. Experimental activity was carried on single phosphorus (P) as well as double phosphorus and aluminum (Al) implanted 4H-SiC epitaxial layers. Samples were then characterized through micro-Raman spectroscopy, Photoluminescence (PL) and Transmission Electron Microscopy (TEM) and results were compared with those coming from P implanted thermally annealed samples at 1650-1700-1750 °C for 1 h as well as P and Al implanted samples annealed at 1650 °C for 30 min. The activity outcome shows that laser annealing allows to achieve full crystal recovery in the energy density range between 0.50 and 0.60 J/cm 2 . Moreover, laser treated crystal shows an almost stress-free lattice with respect to thermally annealed samples that are characterized by high point and extended defects concentration. Laser annealing process, instead, allows to strongly reduce carbon vacancy (V C ) concentration in the implanted area and to avoid intra-bandgap carrier recombination centres. Implanted area was almost preserved, except for some surface oxidation processes due to oxygen leakage inside the testing chamber. However, the results of this experimental activity gives way to laser annealing process viability for damage recovery and dopant activation inside the implanted area.

Aim: A study on the possibility to use gold nanoparticles in mammography, both for a better image diagnostics and radiotherapy, is presented and discussed. We evaluate quantitatively the increment of dose released to the tumor enriched with Au-NPs with respect to the near healthy tissues, finding that for X-rays the increase can reach two orders of greater intensity., Background: Gold nanoparticles continue to be investigated for their potential to improve existing therapies and to develop novel therapies. They are simple to obtain, can be functionalized with different chemical approaches, are stable, non-toxic, non-immunogenic and have high permeability and retention effects in the tumor cells. The possibility to use these for breast calcified tumors to be better treated by radiotherapy is presented as a possible method to destroy the tumor., Materials and Methods: The nanoparticles can be generated in water using the top-down method, should have a size of the order of 10-20 nm and be treated to avoid their coalescence. Under diagnostic X-ray monitoring, the solution containing nanoparticles can be injected locally inside the tumor site avoiding injection in healthy tissues. The concentrations that can be used should be of the order of 10 mg/ml or higher., Results: An enhancement of the computerized tomography diagnostics using 80-150 keV energy is expected, due to the higher mass X-ray coefficient attenuation with respect to other contrast media. Due to the increment of the effective atomic number of the biological tissue containing the gold nanoparticles, also an improvement of the radiotherapy effect using about 30 keV X-ray energy is expected, due to the higher photoelectric cross sections involved., Conclusions: The study carried out represents a feasibility proposal for the use of Au-nanoparticles for mammographic molecular imaging aimed at radiotherapy of tumor nodules but no clinical results are presented.

Background: Gold nanoparticles, 5-20 nm in diameter, were generated with a pulsed Nd: YAG laser at 1010 W/cm2 at solution concentrations ranging between 1-100 mg/ml. The incremental X-ray contrast imaging using gold nanoparticles was investigated and measured. The study was performed with the aim to enhance the massive absorption coefficient of X-ray radiation in the tumor for medical image quality and to improve traditional X-ray radiotherapy or proton therapy. A simulation of proton therapy improvement was conducted using a human ocular melanoma model, placed 3 cm behind the eye lens, and testing 60 MeV protons. Calculations suggest that the local injection of a solution containing Au-NPs may increase the proton energy released in the tumor above 50%, with the dose in the surrounding tissues leading to an increased probability of tissue healing. A discussion on recent patents in the ambit of the preparation and use of Au nanoparticles in medical imaging and therapy is presented., Methods: Au nanoparticles were characterized using optical absorbance, X-ray fluorescence, SEM, and TEM microscopies. Biocompatible nanoparticle solutions were injected intravenously into tail veins of mice followed by X-ray imaging using 20-45 keV photons to evaluate the uptake and the clearance by different organs of the nanoparticles., Results: Diagnostic X-ray images of mice in which the Au-NPs were injected showed high spatial resolution contrast of different organs having high up-take. A calculation of the dose released by X-rays, electrons and protons to the tumor site demonstrates that an increment of the order of 50% can be obtained using adapt solution concentration., Conclusion: The use of Au-NPs in biocompatible solutions injected in living organism permits their blood transport up to different organs. The NPs can be employed as contrast medium to enhance the medical image resolution and to prepare the cancer tissues to be exposed to ionization radiations in order to enhance the dose released to the tumor cells. The wetting ability of water and of some biological liquids was measured on different biocompatible surfaces with and without different colloidal metals. Insoluble nanoparticles disperse in biological tissues enhance some properties, such as the interface adhesion between two surfaces, the X-ray contrast of medical images and the absorbed dose during radiotherapy treatments. The introduction of nanoparticles in the liquids generally improves the wetting ability and changes other properties of the solution, due to the different distribution of the adhesion forces, to the nature, morphology and concentration of the added nanoparticles. An investigation on the contact angle of the liquid drops, physiological liquids, including the human blood, placed on different substrates (polymers, ceramics and metals) with and without the use of metallic nanoparticles is presented, evaluated and discussed.

The physical properties of biocompatible titanium surfaces were modified using different techniques of surface treatment. Particularly the measurements of roughness and wetting ability were controlled using six different techniques: polishing, sandblasting, acid attack, laser ablation, ion implantation and nanoparticle deposition. The titanium surface wetting can be modified drastically depending on the used treatment to enhance the hydrophilic or the hydrophobic behaviour of the metallic biocompatible surface. The study demonstrates that a linear relation between roughness and contact angle occurs. Possible applications to permanent or removable prosthesis titanium based are presented and discussed.

The insertion of gold nanoparticles in biological liquids, tissues and organs permits to increase the equivalent atomic number of the medium that, if used as target to be irradiated by ionizing radiation, permits an increment of the absorbed dose. No toxic nanoparticles, such as the Au, can be injected in the cancer tissues at different concentrations before using a localized treatment that uses energetic proton beams for radiotherapy. Due to the high density and atomic number of the used gold nanoparticles, the absorbed radiation dose can be increased to about a factor six per cent using relatively low concentration of nanoparticles injectable as solution in the tumor tissue. This means to reduce the exposition to ionizing radiation or to increase the dose to the tumor site. Simulation programs of proton energy loss in tissues, using SRIM Code, are employed to evaluate the Bragg peak enhancing in presence of Au nanoparticles, so it will be presented and discussed. Some research findings and patents in the gold nanoparticle preparation and application to Medicine are reviewed in the present paper.

Au nanoparticles can be prepared inside biological solutions and incorporated in special molecules for their transport through blood, drugs and proteins up to the tumour sites or directly injected in their volume when it is possible. The Au nanoparticles are biocompatible and can be accepted locally in the organism also at relatively high concentrations. The use of Au nanoparticles injected in the tumour site enhances significantly the effective atomic number of the medium, depending on the used concentration, and consequently the proton and electron energy loss and the X-ray absorption coefficient determining an increment of the local absorbed dose during radiotherapy. Traditional radiotherapy using electrons, X-rays and gamma rays, and innovative protontherapy can benefit the increment of the effective atomic number of the tissue in the presence of Au-nanoparticles embedded in the tumour volume with an adaptive up-take procedure. This method decreases the dose released to the healthy tissues permitting a better cantering of the irradiated targets and shielding the healthy tissue placed behind the tumour. The presented theoretical study approach permits to evaluate an enhancement of the radiotherapy dose of the order of 1 % using 60 MeV protons, of the order of 10% using 6 MeV electrons and of the order of 100 % using 100 keV X-ray photons. Here, we also disccused for patents relaed to the topic.

Deuterated polyethylene targets have been irradiated by means of a 1016 W/cm2 laser using 600 J pulse energy, 1315 nm wavelength, 300 ps pulse duration and 70 micron spot diameter. The plasma parameters were measured using on-line diagnostics based on ion collectors, SiC detectors and plastic scintillators, all employed in time-of-flight configuration. In addition, a Thomson parabola spectrometer, an X-ray streak camera, and calibrated neutron dosimeter bubble detectors were employed. Characteristic protons and neutrons at maximum energies of 3.0 MeV and 2.45 MeV, respectively, were detected, confirming that energy spectra of reaction products coming from deuterium-deuterium nuclear fusion occur. In thick advanced targets a fusion rate of the order of 2 × 108 fusions per laser shot was calculated.

The investigation on the differences occurring in the manufacture of silver coins allows to get information on their elemental composition and represents a powerful support to the methodology to identify the producing technologies, workshops being also instrumental to distinguish between original and counterfeit ones. Aim of the present work is to study recent and old silver coins through non-destructive X-Ray Fluorescence (XRF) analysis. The XRF was applied to extend the analysis to the deepest layers of the coins; for surface layers an X-ray tube or an electron beam were employed to induce the atom fluorescence to obtain information on the surface elemental composition. Moreover, a detailed study has been performed to evaluate the influence of the surface curvature on the measurement, by deducing a proper corrective factor to keep into account in the data analysis. The elemental atomic composition was measured for each coin, mainly by means of the X-ray tube excitation for the bulk and the electron Scanning Electron Microscope (SEM) microbeam probe for the surface patina analysis. Ionization was induced by an X-ray tube using an Ag anode for the bulk and by an electron microprobe for the surface composition. X-ray detection was performed by using a semiconductor Si device cooled by a Peltier system. The Ag L-lines X-ray yield is affected by coin surface morphology and geometry. The comparison between coin spectra and standard samples, shows that the Ag quantitative analysis is influenced by error of the atomic concentration lower that 10%.

Gold nanoparticles (GNPs) are a promising radiosensitizer agent in radiotherapy. Through a simulation performed with the Geant4 Monte Carlo code, we evaluated the dose enhancement effect of GNPs during therapies with an x-ray tube operating at 150 kV (E = 55 keV and E(max) = 150 keV) and we studied the impact of GNP diffusion out of the tumour vessels, in terms of antiangiogenic and cytotoxic effects. Firstly, a single x-ray beam was assumed to irradiate a parallelepiped volume of soft tissue, in which a GNP-doped "target" volume was placed at different depths. Average dose enhancement factors (DEF) in presence of GNPs were obtained as a function of the target depth and GNP concentration, uniformly distributed; values ranging between 1.6 for 10 mg Au/g at 0 cm and 7.2 for 200 mg Au/g at 5 cm were determined. Furtherly, a second geometry was adopted, in which a blood capillary vessel (10 μm thick and 10 μm of inner radius) was placed at the centre of a cubic volume of soft tissue; doses and DEFs to the capillary endothelium as well as to the surrounding viable tumour were evaluated, for different models of GNP diffusion. Our results indicate that the radial DEF profiles around the vessel are in close relationship with the radial profiles of GNP concentration assumed, except for at sharp gradients of concentration. DEFs at the endothelium ranged from 1.6 to 6.5, for GNP concentrations in the blood of 10 and 200 mg/ml, respectively. These data can be helpful for the development of new and more specific GNP-based radiosensitizers of potential interest in radiotherapy, exploiting the combined benefit of anti-angiogenic and cytotoxic dose enhancement effects.

In this paper a very sensitive ion collection device, for diagnostic of laser ablated-target plasma, is described. It allows for reducing down to few microvolts the signal threshold at digital scope input. A standard ion collector is coupled to a transimpedance amplifier, specially designed, which increases data acquisition sensitivity by a gain ≈1100 and does not introduce any significant distortion of input signal. By time integration of current intensity, an amount of charge as small as 2.7 × 10(-2) pC can be detected for photopeak events.

Ge crystals were prepared by means of laser-induced ion implantation technique. A Nd:YAG pulsed laser (repetition rate: 10 Hz; pulse duration: 3.5 ns; pulse energy: ∼0.5 J) was used both as an ion source and to carry out the ablation processes. The optimization of the laser-generated ion beam parameters in a broad energy and current density range has been obtained controlling the electrostatic field parameters. Numerical simulations of the focusing system, performed adopting an OPERA 3D code, and an investigation of the ion characteristics, using the ion time-of-flight method, have allowed to optimize the preparation parameters. The structural properties of the samples were investigated by means of x-ray photoelectron, micro-Raman spectroscopies, and scanning electron microscopy techniques. Experimental results show that, by appropriately varying the ion implantation parameters and by a post-preparation annealing treatment, it is possible to achieve the development of a micrometer-sized crystalline Ge phase and∕or an amorphous one.

An overview of the last experimental campaigns on laser-driven ion acceleration performed at the PALS facility in Prague is given. Both the 2 TW, sub-nanosecond iodine laser system and the 20 TW, femtosecond Ti:sapphire laser, recently installed at PALS, are used along our experiments performed in the intensity range 10(16)-10(19) W∕cm(2). The main goal of our studies was to generate high energy, high current ion streams at relatively low laser intensities. The discussed experimental investigations show promising results in terms of maximum ion energy and current density, which make the laser-accelerated ion beams a candidate for new-generation ion sources to be employed in medicine, nuclear physics, matter physics, and industry.

At intensities of the order of 10(10) W∕cm(2), ns pulsed lasers can be employed to ablate solid bulk targets in order to produce high emission of ions at different charge state and kinetic energy. A special interest is devoted to the production of protons with controllable energy and current from a roto-translating target irradiated in repetition rate at 1-10 Hz by a Nd:Yag pulsed laser beam. Different hydrogenated targets based on polymers and hydrates were irradiated in high vacuum. Special nanostrucutres can be embedded in the polymers in order to modify the laser absorption properties and the amount of protons to be accelerated in the plasma. For example, carbon nanotubes may increase the laser absorption and the hydrogen absorption to generate high proton yields from the plasma. Metallic nanostrucutres may increase the electron density of the plasma and the kinetic energy of the accelerated protons. Ion collectors, ion energy analyzer, and mass spectrometers, used in time-of-flight configuration, were employed to characterize the ion beam properties. A comparison with traditional proton ion source is presented and discussed.

A 10(16) W∕cm(2) Asterix laser pulse intensity, 1315 nm at the fundamental frequency, 300 ps pulse duration, was employed at PALS laboratory of Prague, to irradiate thick and thin primary CD(2) targets placed inside a high vacuum chamber. The laser irradiation produces non-equilibrium plasma with deutons and carbon ions emission with energy of up to about 4 MeV per charge state, as measured by time-of-flight (TOF) techniques by using ion collectors and silicon carbide detectors. Accelerated deutons may induce high D-D cross section for fusion processes generating 3 MeV protons and 2.5 MeV neutrons, as measured by TOF analyses. In order to increase the mono-energetic proton yield, secondary CD(2) targets can be employed to be irradiated by the plasma-accelerated deutons. Experiments demonstrated that high intensity laser pulses can be employed to promote nuclear reactions from which characteristic ion streams may be developed. Results open new scenario for applications of laser-generated plasma to the fields of ion sources and ion accelerators.

The iodine laser at PALS Laboratory in Prague, operating at 1315 nm fundamental harmonics and at 300 ps FWHM pulse length, is employed to irradiate thin hydrogenated targets placed in vacuum at intensities on the order of 10(16) W∕cm(2). The laser-generated plasma is investigated in terms of proton and ion emission in the forward and backward directions. The time-of-flight technique, using ion collectors and semiconductor detectors, is used to measure the ion currents and the corresponding velocities and energies. Thomson parabola spectrometer is employed to separate the contribution of the ion emission from single laser shots. A particular attention is given to the proton production in terms of the maximum energy, emission yield, and angular distribution as a function of the laser energy, focal position, target thickness, and composition. Metallic and polymeric targets allow to generate protons with large energy range and different yield, depending on the laser, target composition, and target geometry properties.

A pulsed neodymium-doped yttrium aluminum garnet laser ion source has been used as proton beams generator. The laser wavelength is 1064 nm, the pulse duration is 9 ns and the intensity reaches 10(10) W/cm(2). Laser irradiates hydrogenated polymers targets located in a chamber at 10(-7) mbar. The ions are post-accelerated in a suitable chamber by 30 kV of voltage between the target, positively biased, and the following ground electrode. The extracted beams is characterized through a time-of-flight technique. Possible applications to the field of nuclear physics, such as nuclear excitation and de-excitations, nuclear reactions and nuclear fusion, will be presented and discussed.

Ultrafast plasma light ion streams have been produced using the 300 ps, kJ-class iodine laser, operating at PALS Centre in Prague. Ion detection was performed through standard ion collectors (IC) in time-of-flight configuration (TOF), shielded by thin metallic absorbers. This new diagnostics technique has been theoretically studied and experimentally tested in order to cut the long photopeak contribution and to analyze the ultrafast particle signal. Processing the obtained experimental IC-TOF data, including deconvolution processes of the TOF signals, UV/soft-x-ray photopeak absorption, and ion transmission calculations for different metallic filters, is shown. Mainly amorphous carbon (graphite) targets have been irradiated in order to limit the maximum number of ion charge states and to focus our study on demonstrating the validity of the proposed investigation technique. Maximum ion energy and acceleration gradient estimations as a function of the laser energy and focal spot diameter are reported.

Ultrahigh molecular weight polyethylene (UHMWPE) was modified by a 5-MeV energy electron beam at different temperatures before, during, and after irradiation, both in air and in high vacuum. Wear resistance, hardness, and tensile strength of irradiated polyethylene were compared with those of untreated one. Physical analyses (like infrared spectroscopy and calorimetric analysis) were carried out to investigate about the changes in the material induced by irradiation. Experimental results suggested that structural changes (double bonds, crosslinks, and oxidized species formation) occur in the polymer depending on the environmental conditions of the irradiation. Mechanical behavior is related to the structural modifications. A temperature of 110 degrees C before, during, and after the in vacuum irradiation of UHMWPE produces a high amount of crosslinks and improves polymeric tensile and wear resistance, compared to that of the untreated material.

Results of recent studies on highly charged Au ion generation, using the intense long pulses of the PALS high power iodine laser (lambda=1.315 microm, E(L)=800 J400 ps), operating under variable experimental conditions (1omega, 3omega, varying target thickness and changing focus positions), are presented. Both the ion collectors and the ion electrostatic analyzers were applied for the identification of ions in a large distance from the target. The time-of-flight collector signals were treated by a means of peak deconvolution assuming a shifted Maxwell-Boltzmann form of the constituent ion current peaks. Attention was paid to the influence of pulse precursor, which becomes evident, especially, if using thinner targets and 1omega. The results for 3omega point to the presence of several groups of ions with the highest recorded charge state Au(53+).

Ultra high molecular weight polyethylene, both pure and electron beam irradiated, was exposed to air and hyaluronic acid. Physical and mechanical analyses were performed in order to check the polymeric change due to the treatment. Pure UHMWPE, in fact, was modified by the hyaluronic acid that plasticizes the polymer. The electron irradiation cross links the chains and prevents their sliding in acid presence. A low irradiation dose is preferable rather than an high one. Low doses induce low amount of free radicals, witch react with oxygen and hyaluronic acid inducing low polymer degrade maintaining the initial mechanical performance. High doses degrade significantly the polymer properties.100 kGy irradiation in air can be applicable on the polyethylene as well as a compromise between the low mechanical performance improving and the material degradation.

In this work composite materials, based on quartz fibers and epoxy resins, were employed with the aim to restore damaged teeth. The composite materials were chosen because they show biomechanical features very similar to that of the dentine, the main constituent of the tooth. Extracted teeth were rebuilt with two different restorative procedures: in the first, the composite material was pre-formed in a conical trunk shape abutment (PA) and then bonded to a fiber quartz post with a dental bonder. In the second rebuilt system the abutment was prepared by cross linking the resin on the fiber quartz post with a halogen lamp (CRA). The restored teeth were then mechanically tested and observed with a Scanning Electron Microscope (SEM) with the aim to study the interaction between the reconstructive materials. Wetting and roughness measurements were also carried out in order to study the interface adhesion between the post and the abutments. Characterization analysis evidenced that the CRA restorative procedure improves the adhesion between the substitutive materials and shows higher fracture strength than the PA ones. Anyway both the rebuilt systems are able to support the masticator load. An explanation of the interfacial post-abutment interaction phenomenon is discussed.

The significant wear of the UHMWPE bearings of explanted knee prostheses is produced mainly by micrometric debris ("third-body" wear) that diffuse toward the mobile metal-polymer interface. Here debris is crushed during the movement producing scratches in the metal and in the polymeric surfaces. Mechanical stress and the biological effects change the physical polymeric properties. In order to evaluate the area of UHMWE bearings submitted to high load stresses, in this work physical investigations are performed on the explanted knee prosthesis. Particularly, the roughness profile analysis (RPA) and the micro-hardness measurements (MHM) resulted suitable for the localisation of the mechanical and biological wear area. In the stressed zone, surface treatments could be applied in order to improve the mechanical resistance of the polymeric material. Particularly, the ion implantation with heavy ions is proposed to enhance the polymeric wear resistance. The Scanning Electron Microscopy (SEM), Energy Dispersive X-ray Spectrometry (EDX) and Infrared absorption spectroscopy (FTIR) techniques were also applied and results discussed.

Over the last 20 years the JAK/STAT signal transduction pathway has been extensively studied. An enormous amount of data on different cell signal transduction pathways is now available. The JAK/STAT signal transduction pathway is one of the intracellular signaling pathways activated by cytokines and growth factors that was first studied in the hematopoietic system, but recent data demonstrate that this signal transduction is also greatly utilized by other systems. The JAK/STAT pathway is a signaling cascade that links the activation of specific cell membrane receptors to nuclear gene expression. This review is focused on the role of JAK/STAT signal transduction pathway activation in the central nervous system (CNS).

The present work analyzes possible increases in resistance to wear of nickel-titanium endodontic instruments that have undergone a process of ionic implantation. ProFile .04 taper #25 instruments were subjected to ionic implantation with bands of nitrogen ions of 250 KeV, currents in the order of 10 microA/cm2, and doses of 2 x 10(17) ions/ cm2. The instruments were used to make preparations in acrylic endodontic training blocks. Scanning electron microscopic investigations showed that after 60 s of work inside the endodontic training blocks nonionic implanted control instruments showed small modifications in their blades, and their tips showed the first signs of wear. After 240 s the control instruments showed consistent signs of wear and frequent changes to their surfaces. After 240 s of use the ionic-implanted instruments did not present any significant changes in the micromorphology of their surfaces. The implanted instruments did not manifest the typical signs of wear and did not show the surface changes that quickly affect the working life of untreated endodontic instruments manufactured from nickel-titanium.

The production of nitric oxide by the calcium-independent inducible nitric oxide synthase (iNOS) in glial cells has been implicated in the neuropathogenesis of various diseases. It is well known that in response to lipopolysaccharide (LPS) and cytokines, such as IFN-gamma, glial cells are induced to synthesize large amount of nitric oxide (NO) (Bolaños et al., 1996; Nicoletti et al., 1998). The signaling transduction pathways for iNOS transcription in astroglial cells have however not yet been established. Because IFN-gamma receptor chains are associated with Janus tyrosine kinases (JAK1 and JAK2) (Darnell et al., 1994), we analyzed the involvement of the JAK/STAT signal transduction pathway in iNOS expression. Our study shows increased JAK2 and STAT1 alpha/beta tyrosine phosphorylation in primary astroglial cell culture after treatment with IFN-gamma and LPS. A temporal correlation was observed between JAK2 and STAT1 alpha/beta tyrosine phosphorylation, the appearance of interferon-regulatory factor-1 (IRF-1) mRNA and the iNOS expression. Inhibition experiments showed that JAK2 and STAT1 alpha/beta tyrosine phosphorylation were necessary for IFN gamma-mediated iNOS induction in astroglial cells. We conclude that JAK2 and STAT1 alpha/beta tyrosine phosphorylation is an early event involved in the expression of iNOS in astroglial cells., (Copyright 2001 Wiley-Liss, Inc.)

A number of observations point to chromosome 15 as a good candidate to harbor genes involved in epilepsy. This hypothesis is supported by at least two lines of evidence: one is the finding that haploinsufficiency of the 15q11-q13 region, of maternal origin, is responsible for Angelman syndrome, one of the cardinal manifestations of which is epilepsy; the second is the observation that extra copies of this same genomic region, in the form of inv-dup(15) or intrachromosomal duplications, again of maternal origin, are usually associated with a severe neurological phenotype characterized by developmental delay and untreatable seizures. Therefore, both reduced and increased dosage of genes from the 15q11-q13 region, possibly subjected to maternal imprinting, appear to be causally involved in severe forms of epilepsy. We tested the hypothesis that submicroscopic rearrangements of this genomic region might be responsible for nonsyndromic epilepsy in both familial and sporadic forms. To this purpose, we genotyped 118 epileptic patients and their parents with closely spaced microsatellite markers mapped within the 15q11-q13 region. Previous work on the anaerobic treatment of olive oil mill effluents (OME) have shown: (a) lipids, even if more easily degraded than phenols, were potentially capable of inhibiting methanogenesis more strongly; (b) a pretreatment based on addition of Ca(OH)2 and bentonite removed lipids almost quantitatively; (c) preliminary biotreatability tests performed on the pretreated OME showed high bioconversion into methane at very low dilutions ratios, especially when the mixture (OME, Ca(OH)2 and bentonite) was fed to the biological treatment without providing an intermediate phase separation. This paper was directed towards two main aims: (a) to optimize pretreatment: the best results in terms of methane production were obtained by addition of Ca(OH)2 up to pH 6.5 and of 10 g L-1 of bentonite; (b) to evaluate the enhancement of anaerobic treatability of OME pretreated under optimized conditions in a lab-scale continuous methanogenic reactor fed with the substrate without intermediate solid/liquid separation: very satisfactory performances were obtained (at an organic load of 8.2 kg COD m-3 d-1 and at a dilution ratio of 1:1.5 total COD removal was 91%, biogas production was 0.80 g CH4 (as COD)/g tot. COD, lipids removal was 98%, phenols removal was 63%). The results confirm the double role played by bentonite (adsorption of the inhibiting substances and release of the adsorbed biodegradable matter in the methanogenic reactor).

Duplications of chromosome 15 have been reported in individuals with atypical autism, varying degrees of mental retardation, and epilepsy. The authors report the molecular analysis, neurophysiologic, and clinical evaluation of a 12-year-old boy with atypical autism and epilepsy due to a maternally derived 15q11-q13 duplication. Their findings suggest that this chromosomal region harbors genes for autism and possibly for partial epilepsy that may act in a dose-dependent manner.

Mosaicism for trisomy 17 in amniocyte cultures is a rare finding, whilst postnatal cases are exceptional. In order to gain insight into the possible effects of the distribution of the trisomic line and of uniparental disomy (UPD) on embryofoetal development, we have performed follow-up clinical, cytogenetic and molecular investigations into three newly detected prenatal cases of trisomy 17 mosaicism identified in cultured amniotic fluid. In the first case, the pregnancy ended normally with the birth of a healthy girl, and analysis of newborn lymphocytes and of multiple extra-embryonic tissues was indicative of confined placental mosaicism. The second case was also associated with a normal pregnancy outcome and postnatal development, and only euploid cells were found in peripheral blood after birth. However, maternal isodisomy 17 consequent to a meiosis II error and loss of a chromosome 17 homologue was detected in peripheral lymphocytes postnatally. In the third case, pathological examination after termination of pregnancy showed growth retardation and minor dysmorphisms, and the trisomic line was detected in foetal skin fibroblasts. In addition, biparental derivation of chromosome 17 was demonstrated in the euploid lineage. These results, together with previously reported data, indicate that true amniotic trisomy 17 mosaicism is more commonly of extra-embryonic origin and associated with normal foetal development. Phenotypic consequences may arise when the trisomic line is present in foetal tissues. Case 2 also represents the first observation of maternal UPD involving chromosome 17; the absence of phenotypic anomalies in the child suggests that chromosome 17 is not likely to be subject to imprinting in maternal gametes.

Background: Reconstructions with aesthetical materials neither finished nor polished can be extremely irregular and wrinkled. For this reason they represent an ideal basis for the growing of pigmentation which come from food remainings and the nestle of bacteria on the plaque. The finishing of aesthetical materials is a fundamental step in conservative dentistry. The finishing session regarding the aesthetical restorations should be considered and planned at the moment of their insertion in the prepared hollows. The finishing should not be considered an option, but the conclusion of all the conservative treatment. Purpose of the search is to examine and assess, through a technical equipment measuring the superficial wrinkledness of the materials, the action of 4 systems of finishing and polishing on two aesthetical materials widely used in the daily practice by dental surgeons: a compomer (Compoglass, Vivadent) and a composite (Spectrum, Dentsply)., Methods: For each of these two materials some little slabs model have been prepared, choosing the universal colour in the colorimetric scale. The two types of filling materials have been compared with 4 systems of finishing and polishing: Sof-Lex Pop-on (3M), Enhance system (Dentsply), Hawe Micro Disc (Howe Neos), Heawe Gommini Polisher (Heawe Neos). The total working time did not overcome 1 minute. The little slabs have been obtained, pressing the resin between two slides used in microscopy. In the hope to guarantee in the different samples, a uniform thickness, we have used a technical device. On each covering slide we have put a weight of 0.5 kg for 5 minutes. The thickness of the little slabs obtained was 2 mm. Thus, pressed between the two slides, the aesthetical material has been photopolymerized according to the time suggested by Manufacturing Industries. The sizes concerning the wrinkledness of samples subjected to different treatments have been carried out using a pointed profilometer with high sensitivity. (Tencor-P10). This instrument used in the National Laboratory of Catania, called INFN, is able to graphitize the wrinkledness of a surface "survbeying" it with a diamond ultramicrometric point., Results: All tested systems gradually produce the upper layers of the materials less suitable to resist the assault of plaque bacteria as time passed. The 3M coarse and medium grain Disks are very abrasive and for this reason the surface of the materials is ill-shaped. Those disks with fine and extra fine grain, smooth the tracks left by previous disks. As they have been always used according to their decreasing granulometry, the disks are used only for removing small composite pieces in excess and to improve micromorphology of the restauration. The "Gommini" are less abrasive than Disks. In a few minutes and often with only one step they produce a much regular and polished fillings surface., Conclusions: The "Gommini" have a preference when the last photopolymerization has left a regular layer, with a very good micromorphology. Actually, "Gommini" do not remove much material, but they continue to smooth the outline of the reconstruction. Disks are not classified as being of first quality in the finishing of composites and compomers.

The main properties of NiTi alloys are presented, discussed and compared with respect to those of stainless steels. Special regard is given to the use of NiTi alloy as orthodontic wires and endodontic instruments. The superelastic properties of orthodontic wires and endodontic files are measured as a function of the applied stress, of the strain and of the temperature. Both biomedical devices are submitted to surface analysis to control the interface with the biological environment in which they will be immersed.

The aim of this study was to investigate the Doppler effect of the 511 keV gamma peak from positron annihilation in biological matter. The broadening of the annihilation peak is due to positron annihilation with electrons that have high momentum. In aqueous solutions annihilation depends on the temperature and it is linked positronium formation. Measurements in vivo, on human brain, were taken during the diagnosis of positron emission tomography (PET) on healthy patients by injecting them with the beta emitter of short lifetime 18F. The Doppler-broadening in biological tissues rich in water content decreased significantly compared to biological solutions and water.

Swift ions with 50 MeV energy are used to irradiate bio-polymeric films. Lithium, carbon and oxygen beams, with fluencies ranging between 10(8) and 10(13) ions/cm2, are swept on large surfaces of hydrogenated and fluorated polymers. The electronic stopping power of the ion induces high rates of ionization, excitation and radical formation along the ion path with production of chain-scission and cross-linking processes which can be investigated by the molecular emission from the irradiated polymer. The radiation damage increases the chemical solubility of the material around the ion track up to distances of the order of 100 nm. A suitable chemical etching removes the damaged material leaving micrometric holes in the polymeric film. This process gives rise to micro-perforated films which can be used as microfilters, with sub-micrometric pore dimension, high selectivity and controllable porosity. Their use is very interesting for special applications in ultra- and microfiltration of biological liquids.

Nitrogen implantation and thermal nitridation processes are investigated to improve the wear resistance of titanium medical devices. Ion implantation uses 80 keV nitrogen ions and doses ranging between 1 x 10(17) and 5 x 10(17)/cm2 to modify the surface layers up to 140 nm depth. Stable surface layers are obtained at high ion doses by post-annealing at 550 degrees C for 30 min. Thermal processes are developed as a function of the time (20-90 min) and annealing temperature (950-1100 degrees C) in a dry nitrogen atmosphere. The amount of titanium nitride formed during the thermal process increases with the annealing temperature and time. Samples are analysed by Rutherford Backscattering Spectroscopy (RBS), Auger Electron Spectroscopy (AES) and Scanning Electron Microscopy (SEM). Ion implantation and thermal nitridation processes increase the surface hardness and wear resistance improving the mechanical properties of titanium used for movable devices for total joint replacements, such as the hip and knee protheses.

Metallic screws are becoming more and more useful to join bones or for prosthesis support in orthopaedic and dental surgery. High biocompatible materials such as titanium alloys and hydroxyapatite ceramics are making possible the realization of stable fixation devices utilizable in load-bearing applications. The mechanical and biological anchorage of metallic screws to the bone depends on many factors: mechanical screws-bone thread matching, use of cements between bone and screw, chemical-physical treatments of screw surface, use of screw coverage films based on osteointegrating ceramics or active bioglasses, use of porous coverage films to induce bone ingrowth into the pore, and so on. The first step of the research, the aim of the present paper, is the comparison among screws of different shapes and geometric characteristics in order to find the best macromechanical system versus the different load conditions. Static and dynamic tests are applied to the screws mounted on segments of pig spine, in order to measure the mechanical characteristics of the system under tension (pull-out), bending on the principal planes along the axis of the screw and fatigue loads. Physical analysis, obtained using energetic charged particles, such as SEM, RBS, and AES, are used to characterize the screw surface compositions and morphology.

Thin films of bio-polymers, such as polymethylmetacrylate, polytetrafluoroethylene, polyethylene-terephthalate and polyimide, are efficiently etched in air by UV laser pulses. A different sharp laser fluence threshold for significant etching is measured for each polymer. The thresholds are correlated to the chemical structure and to the wavelength-dependent absorption coefficient of the irradiated polymer. The polymers, having low absorption coefficients, show thresholds of the order of some J/cm2, about one magnitude higher than polymers having high absorption coefficients. Measurements on the polymer etching rates, in terms of removed molecules per eV at the used 347 nm wavelength, and a discussion on the laser-polymer interaction are presented. An interesting application of the laser ablation process to bio-polymers, concerning the ability to realize microfilters for the bio-medical field by irradiating the polymer surface through a perforated metal mask, is discussed.

Fluoride content in the enamel and dentine of ancient teeth has been measured using a nuclear analysis technique. Analysed teeth were found in a vast necropolis of Sicily (Italy) which is archeologically datable between the 13th and the 8th century B.C. Measurements gave high fluoride concentrations, of the order of 10 mg per gram of hydroxyapatite, comparable with the content of fluorotic teeth of patients living nowadays in regions rich in fluorinated drinking water. Results suggest that the dental caries process was probably unknown to the tribal population living in this geographical area.

Pulsed laser deposition of hydroxyapatite is applied to cover titanium medical prosthesis. Laser irradiation of ceramics by power-pulsed ruby laser produces high velocity micron-size particles emission with a narrow ejection plume directed normally to the target surface. Photoemitted particles are deposited on a metal surface at 400 degrees C to obtain a granular film, stoichiometric, polycrystalline, and very adherent to the substrate. Deposited films are highly biocompatible and may be used to induce osteointegration processes of the implanted prosthesis with the bone tissues. The comparison between laser and plasma spray depositions of bioceramics is discussed.
