Abstract

De dissertatie van Petrus van Galen van zijn promotieonderzoek aan de Universiteit Utrecht uit 1830 is een vergeten vondst in de geschiedenis van het zwaartekracht onderzoek naar de aarde. In de vertaling van het Latijn naar het Nederlands geeft deze dissertatie ons een inzicht in de wetenschappelijke ontdekkingen rond de tijd van zijn publicatie. Petrus van Galen geeft in zijn dissertatie een volledig beeld van alle slingermetingen die gedaan waren met betrekking tot het bepalen van de vorm van de aarde. Deze unieke metingen werden gedaan door verschillende ontdekkingsreizigers en wetenschappers, van Noord-Rusland tot in de Vuurland-archipel in het verre zuiden van Zuid-Amerika, allemaal om zo een beter beeld te krijgen van de afplatting van onze planeet. In zijn tweede hoofdstuk presenteert Van Galen in een wiskundige uiteenzetting hoe een slingermeting gecorrigeerd moet worden om zo nauwkeurig mogelijk de zwaartekracht te kunnen bepalen, hierbij verder bouwend aan de theorie van Christiaan Huygens. Hierbij selecteert hij ook de apparatuur en experimenten die precies genoeg zijn voor de bepaling van de vorm. In zijn laatste hoofdstuk werkt hij de wiskundige theorie uit hoe de vorm van de aarde bepaald kan worden door de verschillende zwaartekrachtmetingen. Hij gebruikt daarbij de net bedachte kleinste kwadratenmethode, om zo tot een waarde voor de afplatting te komen van 1/286.23. Deze waarde ligt dicht bij de huidige berekende waarde, een bijzondere prestatie zo'n tweehonderd jaar geleden. Dit boek presenteert zo een nieuw puzzelstukje in de fascinerende geschiedenis van de Nederlandse wetenschap. Abstract

The DFG-Network ‘Dispositiv der Menge’ (= crowd, mass, multitude) is based on the recognition that ‘the crowd’ has been constituted, classified, regulated, or dispersed throughout history in various, heterogenous, conflictual ways, which make it impossible to hold onto any understanding of the crowd as a clearly delimited and substantiated entity or to the forms in which it is represented as such. This workshop extends this approach by taking ‘the crowd’ itself as a multitude or multiplicity. It will address phenomena related to more fluctuating and in/determinate intra/sections of collectivities in order to transform the image of ‘the crowd’ from an in-divi-dual-ized One into a mani-fold multiplicity. Such a multiplicity in the singular plural is characterized by movement and motion, fragmentation and friction, ongoingness and inherent contestation. By focusing on this multiplicity, the workshop seeks to un-work a persistent conceptual anthropocentrism of the ways in which agentiality is conceptualized and imagined, instead searching for more dynamic relations with/in a variety of agencies (human, animal, plant, things, propositions) that can be seen as ‘intra-active’. Yet, while ‘the crowd’ will be approached from a critique of anthropomorphism, the turn towards ecological or relational co-existence cannot be a turn away from the violent asymmetrical relations of power and the continued flexibilization and hierarchical re- ordering of global social structures. The workshop sounds out conceptual and phenomenal resonances between what in Western academic discourses has of late become known as ‘New Materialism’ (in its different strands) and the long tradition of (but also always newly emerging) indigenous and decolonial epistemologies. Abstract

Age has a major effect on brain volume. However, the normative studies available are constrained by small sample sizes, restricted age coverage and significant methodological variability. These limitations introduce inconsistencies and may obscure or distort the lifespan trajectories of brain morphometry. In response, we capitalized on the resources of the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Consortium to examine age-related trajectories inferred from cross-sectional measures of the ventricles, the basal ganglia (caudate, putamen, pallidum, and nucleus accumbens), the thalamus, hippocampus and amygdala using magnetic resonance imaging data obtained from 18,605 individuals aged 3–90 years. All subcortical structure volumes were at their maximum value early in life. The volume of the basal ganglia showed a monotonic negative association with age thereafter; there was no significant association between age and the volumes of the thalamus, amygdala and the hippocampus (with some degree of decline in thalamus) until the sixth decade of life after which they also showed a steep negative association with age. The lateral ventricles showed continuous enlargement throughout the lifespan. Age was positively associated with inter-individual variability in the hippocampus and amygdala and the lateral ventricles. These results were robust to potential confounders and could be used to examine the functional significance of deviations from typical age-related morphometric patterns.

El contingut del currículum de química no s’ha modificat en els últims setanta anys. Com a resultat, els estudiants perceben que la química escolar no està ben relacionada amb les seves vides. Hem utilitzat pràctiques autèntiques com a contextos per seleccionar, especificar, reemplaçar i/o modificar continguts químics específics. En aquest article es descriu el procediment de recerca basat en el disseny que hem seguit per desenvolupar nous esbossos per a un currículum i per dissenyar un marc d’ensenyament juntament amb unitats curriculars exemplificadores. El marc i les unitats es van adaptar com a resultat de diversos cicles d’experimentació.paraules clau: Pràctiques autèntiques, context, procés de recerca basat en el disseny, unitats didàctiques exemplars., The content of the chemistry curriculum has not been changed in the past seventy years. As a result, students perceive that school chemistry is not well connected to their lived worlds. We used authentic practices as contexts to select, specify, replace and/or modify specific chemistry content. This paper describes the design-based research procedure we followed to develop new sketches for a curriculum and to design an instructional framework together with exemplary curriculum units. Abstract

The midbrain dopamine (mDA) system is composed of molecularly and functionally distinct neuron subtypes that mediate specific behaviors and show select disease vulnerability, including in Parkinson’s disease. Despite progress in identifying mDA neuron subtypes, how these neuronal subsets develop and organize into functional brain structures remains poorly understood. Here we generate and use an intersectional genetic platform, Pitx3-ITC, to dissect the mechanisms of substantia nigra (SN) development and implicate the guidance molecule Netrin-1 in the migration and positioning of mDA neuron subtypes in the SN. Unexpectedly, we show that Netrin-1, produced in the forebrain and provided to the midbrain through axon projections, instructs the migration of GABAergic neurons into the ventral SN. This migration is required to confine mDA neurons to the dorsal SN. These data demonstrate that neuron migration can be controlled by remotely produced and axon-derived secreted guidance cues, a principle that is likely to apply more generally.

Abstract

Cells can enter quiescence in adverse conditions and resume proliferation when the environment becomes favorable. Prolonged quiescence comes with a cost, reducing the subsequent speed and potential to return to proliferation. Here, we show that a similar process happens during Caenorhabditis elegans development, providing an in vivo model to study proliferative capacity after quiescence. Hatching under starvation provokes the arrest of blast cell divisions that normally take place during the first larval stage (L1). We have used a novel method to precisely quantify each stage of postembryonic development to analyze the consequences of prolonged L1 quiescence. We report that prolonged L1 quiescence delays the reactivation of blast cell divisions in C. elegans, leading to a delay in the initiation of postembryonic development. The transcription factor DAF‐16/FOXO is necessary for rapid recovery after extended arrest, and this effect is independent from its role as a suppressor of cell proliferation. Instead, the activation of DAF‐16 by decreased insulin signaling reduces the rate of L1 aging, increasing proliferative potential. We also show that yolk provisioning affects the proliferative potential after L1 arrest modulating the rate of L1 aging, providing a possible mechanistic link between insulin signaling and the maintenance of proliferative potential. Furthermore, variable yolk provisioning in embryos is one of the sources of interindividual variability in recovery after quiescence of genetically identical animals. Our results support the relevance of L1 arrest as an in vivo model to study stem cell‐like aging and the mechanisms for maintenance of proliferation potential after quiescence.

Abstract

We organized 10Kin1day, a pop-up scientific event with the goal to bring together neuroimaging groups from around the world to jointly analyze 10,000+ existing MRI connectivity datasets during a 3-day workshop. In this report, we describe the motivation and principles of 10Kin1day, together with a public release of 8,000+ MRI connectome maps of the human brain.

Abstract

[EN]: With the explosion of genome sequencing technologies, mechanisms such as aneuploidy, polyploidy, and hybridization are emerging as being more frequent and relevant in genomes evolution than what was considered earlier. An interesting model to study such mechanisms are Saccharomyces yeasts as much data is available on its genome structure and evolution and they are easily manipulated in laboratory. Yeast from this genus have small and compact genomes that make them an interesting model for genomics studies on eukaryotic organisms. Moreover, hybrids between the different species of the genus are known and relevant for industrial processes. In this doctoral thesis, we aimed to investigate different aspects of the adaptive value of aneuploidy and interspecific hybridization in Saccharomyces. In the first chapter, we were interested in studying what genomic differences were underlying the different ethanol tolerance observed in S. cerevisiae strains. The most interesting genomic change we observed was a shared polysomy of chromosome III in the highest ethanol tolerant strains. We could determine that this correlation between ethanol tolerance and chromosome III copy number also appeared in different background and we confirmed it was an adaptive mechanism on ethanol stress. In the second chapter of this work, we asked how S. cerevisiae x S. kudriavzevii hybrids mate and how this mechanism would influence the genomic and adaptability outcome of these hybrids. We found that rare-mating was the most frequent but not the only mechanism use. As most of the hybrid were triploid and had a diploid heterozygous contribution of S. cerevisiae, this mating mechanism was the most probable. However, two hybrids were tetraploid. One had an extreme reduction of the S. kudriavzevii subgenome and heterozygous S. cerevisiae. This structure seemed more compatible with the outcome of artificial crossing in strain improvement programs. The last strain had a diploid homozygous contributi, [ES]: Las levaduras del género Saccharomyces son de reconocida importancia tanto por ser un modelo genético básico como por su uso biotecnológico. Tienen un genoma muy compacto. En sus aproximadas 12Mb tienen codificados 6000 genes. Además, muy pocos de estos genes contienen intrones. Esto convierte a estos organismos en unos modelos simples para el estudio genético y genómico. El género tiene 9 especies. La divergencia nucleotídica entre especies es interesante ya que puede cubrir la divergencia entre humanos y aves (S. cerevisiae vs S. kudriavzevii) como entre humanos y primates (S. cerevisiae vs S. paradoxus). De estas especies, S. cerevisiae es la más estudiada y usada biotecnológicamente. Sin embargo las otras especies tienen distintas propiedades fenotípicas que están levantando interés para la industria. Una de ellas es la capacidad de crecimiento a bajas temperaturas que se observa en Saccharomyces no cerevisiae. Estas diferencias fenotípicas y la divergencia entre especie convierten al género en un buen modelo para estudios genéticos y evolutivos. Los mecanismos que generan diversidad genómica son muy diversos. Si un genoma es heterocigoto tiene distintos alelos en el genoma y puede usar mecanismos como la pérdida de heterocigosidad para fijar uno u otro alelo en respuesta a estreses. Con el avance de las tecnologías de secuenciación del ADN y la explosión de estudios genómicos, la importancia de mecanismos de diversificación genómica como las aneuploidías, las polyploidías y la hibridación está quedando más clara. En Saccharomyces múltiples estudios están encontrando que variar el número de copias de ciertos cromosomas es un mecanismo muy frecuente en ambientes variables como son los ambientes industriales. Un hecho interesante es que en estos ambientes también se encontraron distintos híbridos interespecíficos del género. Abstract

Being confronted with concerns from both inside Belgium and abroad, the operation of Belgian nuclear power stations constitutes a controversial issue of cross-border relevance. Drawing on stakeholder theory and borrowing core concepts from the Advocacy Coalition Framework (ACF), this paper offers a systematic stakeholder-centered analysis of the present policy dispute as well as an evaluation of prospects for a Belgian nuclear phase-out. It identifies key stakeholders in the decision-making process and analyzes their diverging positions and beliefs. Eight semi-structured interviews with crucial stakeholders and political insiders shed light upon the policy debate and reveal major lines of argumentation and conflict. Theoretically deduced preconditions for policy change are consequently compared to the current situation in Belgium. Results indicate that despite the Belgian Federal Government’s commitment to stop domestic nuclear energy generation by 2025, this endeavor cannot be taken for granted. Further lifetime extensions of nuclear power plants appear to be a realistic future scenario under given circumstances. Although there seems to be increasing agreement on a major sectoral transformation toward renewable energies, members of the pro- and anti-nuclear coalition differ drastically in their assessment of an adequate and manageable temporal horizon for phasing out nuclear power. Based on theoretical suggestions and analytical findings, tangible recommendations for future stakeholder action to support policy change in Belgium are given.
