Airborne pathogens are the most important factor causing environmental issues on pig farms. Research on airborne swine-derived microbes has mainly concentrated on several specific microbes or pathogens. The present work was conducted to detect and identify the entire microbial community in piggery air and their dispersion by 16SrRNA sequencing. Fifteen faeces and eighty-four air samples were collected in swine barns and from different distances away from the barns, respectively. The results showed that the faeces and air share the most dominant bacteria. The top 10 genera belonged to Fimicutes, Proteobacteria and Bacteroidetes, and accounted for 54% and 76-84% of the total sequences. Moreover, great higher (P < 0.01) microbial diversity was detected in the faeces. This study indicated that a hygienic interval of 50 m should be set on swine farms to prevent the spreading infectious disease caused by airborne pathogens.

Two new phenolic glycosides (1 and 2), one known analogue (3), along with a new diterpene glucoside (4) were obtained from ethanolic extract of the stems of Eurya chinensis R. Br. The structures of these isolated compounds were identified by extensive analysis of HRESIMS and NMR spectroscopic data. The cytotoxicities of these compounds were evaluated on MCF-7, A549, HepG2, CaCo2 and 5-8 F cell lines by MTT method, but no obvious activities were observed.

Two new withaphysalin-type withanolides (18-O-ethylwithaphysalin R and 5-O-ethylphysaminimin C, 1 and 2), along with twelve known withanolides (3–14), were purified and identified from Physalis peruviana L. The chemical structures of these new isolates were elucidated through analyzing spectroscopic and HRESIMS data. All the obtained metabolites were appraised for their potential antiproliferative activity against the human breast cancer cell line MCF-7. Compound 7 was discovered to exhibit potent activity with an IC50 value of 3.51 µM and compounds 2, 6 and 14 showed weak cytotoxic effect.

Cajaninstilbene acid (CSA), Longistylin A (LLA) and Longistylin C (LLC) are three characteristic stilbenes isolated from pigeon pea, exhibiting antibacterial activities against Staphylococcus aureus and even methicillin-resistant Staphylococcus aureus (MRSA). The results showed that LLA had the highest antibacterial activity against the tested strains following by LLC and CSA. Besides, the results implied that these stilbenes effectively inhibited the levels of adhesion and virulence genes and downregulate the production of ɑ-hemolysin. This study showed that stilbenes from pigeon pea have anti-Staphylococcus potential, and they also inhibited bacterial proliferation, biofilm formation, and key gene expressions related to adhesion and virulence of MRSA. It is the first time that the anti-S. aureus and MRSA activities of the three stilbenes has been systematically reported. These current findings provide insight into the anti-MRSA mechanism of stilbenes from pigeon pea, indicating these compounds may be used as antimicrobial agents or additives for food with health functions, and contribute to the development as well as application of pigeon pea in food science.

BackgroundThe relationship between cuproptosis and HCC is still in the exploratory stage. Long noncoding RNAs (lncRNAs) have recently been linked to the progression of hepatocellular carcinoma (HCC). However, the clinical significance of lncRNAs associated with cuproptosis remains unclear.MethodsBased on The Cancer Genome Atlas (TCGA) liver hepatocellular carcinoma (LIHC) dataset, we identified characteristic prognostic lncRNAs by univariate, LASSO, and multifactorial regression analysis, and constructed a prognostic signature of cuproptosis-related lncRNAs in HCC. The role of lncRNAs were identified through CCK-8, clone formation in Huh-7 cells with high expression of FDX1. Prognostic potential of the characteristic lncRNAs was evaluated in each of the two cohorts created by randomly dividing the TCGA cohort into a training cohort and a test cohort in a 1:1 ratio. Immune profiles in defined subgroups of cuproptosis-related lncRNA features as well as drug sensitivity were analyzed.ResultsWe constructed a multigene signature based on four characteristic prognostic lncRNAs (AL590705.3, LINC02870, KDM4A-AS1, MKLN1-AS). These four lncRNAs participated in the development of cuproptosis. HCC patients were classified into high-risk and low-risk groups based on the median value of the risk score. The receiver operating characteristic curve area under the curve values for 1-, 3-, and 5-year survival were 0.773, 0.728, and 0.647, respectively, for the training cohort, and 0.764, 0.671, and 0.662, respectively, for the test cohort. Univariate and multifactorial regression analyses indicated that this prognostic feature was an independent prognostic factor for HCC. Principal component analysis plots clearly distinguished between low- and high-risk patients in terms of their probability of survival. Furthermore, gene set enrichment analysis showed that a variety of processes associated with tumor proliferation and progression were enriched in the high-risk group compared with the low-risk group. Moreover, there were significant differences in the expression of immune cell subpopulations, immune checkpoint genes, and potential drug screening, which provided distinct therapeutic recommendations for individuals with various risks.ConclusionsWe constructed a novel cuproptosis-associated lncRNA signature with a significant predictive value for the prognosis of patients with HCC. Cuproptosis-associated lncRNAs are associated with the tumor immune microenvironment of HCC and even the efficacy of tumor immunotherapy.

Primary neuroendocrine breast carcinomas (NEBCs) are an extremely rare and underrecognized subtype of mammalian carcinoma. The prognostic factors for NEBCs remain controversial.In this multicenter retrospective study, the prognostic factors for patients with primary NEBCs who underwent surgery and had a pathologically confirmed diagnosis of neuroendocrine carcinoma in China and the United States were examined. The endpoints were disease-free survival (DFS) and overall survival (OS).A total of 51 Chinese patients and 98 US patients were included. In the Chinese cohort, tumor grade and Ki-67 levels were prognostic factors for DFS in univariate analysis (hazard ratio [HR] = 5.11 [1.67-15.60], p = 0.004; HR = 57.70 [6.36-523.40], p 0.001, respectively) and multivariate analysis (HR = 100.52 [1.33-7570.21], p = 0.037; HR = 31.47 [1.05-945.82], p = 0.047, respectively). In the US cohort, age was an important prognostic factor for OS in univariate analysis (HR = 1.09 [1.04-1.15], p = 0.001). The random effects model for the combined cohorts revealed age and positive expression of estrogen receptor (ER) as potential prognostic factors for OS (HR = 1.08 [1.01-1.14], p = 0.015; HR = 0.10 [0.02-0.44], p = 0.003, respectively).Tumor grade and Ki-67 levels are important prognostic factors for DFS of patients with primary NEBCs. Age and ER status are important prognostic factors for OS of patients with primary NEBCs.

The sixth generation (6G) wireless systems are envisioned to enable the paradigm shift from "connected things" to "connected intelligence", featured by ultra high density, large-scale, dynamic heterogeneity, diversified functional requirements and machine learning capabilities, which leads to a growing need for highly efficient intelligent algorithms. The classic optimization-based algorithms usually require highly precise mathematical model of data links and suffer from poor performance with high computational cost in realistic 6G applications. Based on domain knowledge (e.g., optimization models and theoretical tools), machine learning (ML) stands out as a promising and viable methodology for many complex large-scale optimization problems in 6G, due to its superior performance, generalizability, computational efficiency and robustness. In this paper, we systematically review the most representative "learning to optimize" techniques in diverse domains of 6G wireless networks by identifying the inherent feature of the underlying optimization problem and investigating the specifically designed ML frameworks from the perspective of optimization. In particular, we will cover algorithm unrolling, learning to branch-and-bound, graph neural network for structured optimization, deep reinforcement learning for stochastic optimization, end-to-end learning for semantic optimization, as well as federated learning for distributed optimization, for solving challenging large-scale optimization problems arising from various important wireless applications. Through the in-depth discussion, we shed light on the excellent performance of ML-based optimization algorithms with respect to the classical methods, and provide insightful guidance to develop advanced ML techniques in 6G networks.

Here, we aimed to analyse the correlations between inflammatory cells and organ injury parameters in AMI patients and compared between male and female groups.Methods: A total of 603 AMI patients who underwent percutaneous coronary intervention (PCI) within 24 hours of the onset were analysed retrospectively. Basic information and hematological parameters detected 6 hours before the PCI were collected, neutrophil-to-lymphocyte ratio (NLR) and monocyte-to-lymphocyte ratio (MLR) were calculated. Renal, liver function indicators, and myocardial enzymes were measured. Left ventricular ejection fraction (EF) and fractional shortening (FS) on days 5– 7 after PCI were obtained. Western blot was performed to detect iNOS, eNOS and nNOS expression in H9C2 rat cardiomyocytes treated with IL-6 with and without estrogen and testosterone.Results: WBC, NEU, MON, MLR, CK, ALT and CREA of male patients were significantly higher than females, but FS was lower in females. NEU, MON and MLR were positively correlated with CK, CK-MB, AST, and ALT in males, whereas LYM were correlated with these parameters in female. NEU and NLR were inversely correlated with EF or FS only in female. Estrogen and testosterone reduced IL-6-induced iNOS protein expression in H9C2 cardiomyocytes, estrogen enhanced IL-6-induced nNOS protein expression.Conclusion: NEU, MON, MLR in male AMI patients, and LYM in female patients were associated with organ injury parameters. Background In China, health technology assessment (HTA) has recently been adopted in pricing negotiation for medicine listing in the National Reimbursement Drug List. At present, how HTA is applied to inform the decision-making process remains underreported. In order to explore how the adoption of HTA was translated into listing and price negotiation results in light of the confidential nature of the negotiating process, this study aimed to compare the negotiated price and the clinical benefit of selected targeted anticancer medicines (TAMs) involved in the 2019 negotiation. Main text Among 16 TAMs successfully negotiated, only four TAMs representing four indication groups had appropriate reference medicines for comparison and were, therefore, included in the analysis. The price and clinical benefit of the four TAMs were compared against one or two reference medicines with the same initial indications. The sales prices for nine TAMs before and after the negotiation were extracted from the centralized medication procurement system. Clinical benefits were evaluated based on evidence from published articles and clinical guidelines. The results suggested that, despite the application of HTA, both rational and irrational decisions had been made about the reimbursement of TAMs in the 2019 negotiation, warranting further investigation. Conclusion While the development and adoption of HTA has seen significant progress in China, actions are needed to ensure that the adoption of HTA is effectively applied in decisions on the reimbursement of medicines.

F-doped ZnO (FZO) films were prepared by reactive sputtering using Zn/ZnO/ZnF2 mixture as the target at substrate temperature (Ts) of 150℃ and 300℃ and sputtering atmosphere of Ar+O2 and Ar+H2. The effect of gas flux, Ts, and sputtering atmosphere on the structure and transparent conductive properties of the thin film was investigated. The results show that for FZO films prepared under Ar+O2, Ts=300℃ is conductive to the preparation of films with (002) preferred orientation, high crystallinity, low compressive stress and good transparent conductive properties. For the films prepared under Ar+H2, with the increase of Ts to 300℃, the crystallinity and transmittance of the films improve and the compressive stress reduces, but the film thickness significantly reduces and leads to the deterioration of the conductive properties of the films. The FZO films prepared in the two atmospheres are compared, and it is found that the films prepared in Ar+H2 can obtain better transparent conductive properties (the resistivity of 3.5×10-3 Ω·cm and the average transmittance in visible range of 87%) at 150℃ and in H2 flux range of 0.8-3.2 mL·min-1. The etching effect of H plasma and H doping in the Ar+H2 atmosphere, the bombardment of O ions and the change of oxygen defects of the films in the Ar+O2 atmosphere, the enhancement in reaction activity and mobility of deposited atoms with increasing Ts, and the relationship between Eg and carrier concentration are discussed in detail.

Microcystins (MCs) are hepatotoxic heptapeptides identified in cyanobacterial bloom-impacted waters and soils. However, their environmental fate in soils is poorly understood, preventing reliable site assessment. This study aims to clarify the variant-specific adsorption, desorption, and dissipation of MC-LR and MC-RR in agricultural soils. Results revealed that their adsorption isotherms followed the Freundlich model (R2 ≥ 0.96), exhibiting a higher nonlinear trend and lower adsorption capacity for MC-LR than for MC-RR. The soils had low desorption rates of 8.14-21.06% and 3.06-34.04%, respectively, following a 24 h desorption cycle. Pairwise comparison indicated that soil pH and clay played key roles in MC-LR adsorption and desorption, while organic matter and cation exchange capacity played key roles in those of MC-RR. MC-LR dissipation half-lives in soils were 27.18-42.52 days, compared with 35.19-43.87 days for MC-RR. Specifically, an appreciable decrease in MC concentration in sterile soils suggested the significant role of abiotic degradation. This study demonstrates that the minor structural changes in MCs might have major effects on their environmental fates in agricultural soil and indicates that the toxic effects of MCs should be of high concern due to high adsorption, low desorption, and slow dissipation.

BACKGROUND Currently, there is a lack of sepsis screening tools that can be widely used worldwide. Pulmonary sepsis can be of sufficient concern to physicians due to their noticeable symptoms, which usually rely less on screening tools. AIM To investigate the efficiency of the international normalized ratio (INR) for the early rapid recognition of adult nonpulmonary infectious sepsis. METHODS This is a prospective observational study. A total of 108 sepsis patients and 106 nonsepsis patients were enrolled according to relevant inclusion and exclusion criteria. Commonly used clinical indicators, such as white blood cell, neutrophil count, lymphocyte count, neutrophil-lymphocyte count ratio (NLCR), platelets (PLT), prothrombin time, INR, activated partial thromboplastin time, and quick Sequential “Sepsis-related” Organ Failure Assessment (qSOFA) scores were recorded within 24 h after admission. The diagnostic performances of these clinical indicators were analyzed and compared through multivariate logistic regression analysis, Spearman correlation, and receiver operating characteristic curve analysis. RESULTS The INR value of the sepsis group was significantly higher than that of the nonsepsis group. INR has superior diagnostic efficacy for sepsis, with an area under the curve value of 0.918, when those preexisting diseases which significantly affect coagulation function were excluded. The diagnostic efficacy of the INR was more significant than that of NLCR, PLT, and qSOFA (P < 0.05). Moreover, INR levels of 1.17, 1.20, and 1.22 could be used to categorize the relative risk of nonpulmonary infections sepsis into three categories: low, medium and high risk, respectively. CONCLUSION The INR is a promising and easily available biomarker for diagnosis, and it can be used as one of the indicators for early screening of adult nonpulmonary infectious sepsis. When its value is higher than the optimal cutoff value (1.22), high vigilance is required for adult nonpulmonary infectious sepsis.

A small-scale plasma ablation facility was employed to test the C/C-SiC composite material for investigating the thermal performance and ablation characteristics under two heat flux conditions, 3593.54 kW·m−2 and 5644.86 kW·m−2. The morphology of post-test specimens was analyzed with the ablation rates calculated. The average mass ablation rates of two group specimens were 0.01735 and 0.10620 g·s−1 respectively with average linear ablation rate of 0.00680 and 0.09407 mm·s−1. Specimen surface could be divided into three regions with typical layered structure characteristics. For the stagnation point ablation test, the structural deformation in the ablation surface area featured in vertical layering and lateral regionality, forming an ablation pit near the stagnation point. In the center region, sublimation occured primarily, accompanied by a serious jet scouring of the molten liquid phase, as well as a small amount of oxidation reaction; Jet erosion with thermal sublimation was the main factor for the mass loss in the transitional region; Thermochemical reactions were mainly carried out in the marginal region. The SiO2 generated from the thermochemical reaction of the material filled the interspace well and prevented the thermochemical reaction from penetrating deeper through the crack. The protective layer in the molten state with high viscosity reduced the damage of the high-speed jet impact material.

Social hierarchy greatly influences behavior and health. Both human and animal studies have signaled the medial prefrontal cortex (mPFC) as specifically related to social hierarchy. Dopamine D1 receptors (D1Rs) and D2 receptors (D2Rs) are abundantly expressed in the mPFC, modulating its functions. However, it is unclear how DR-expressing neurons in the mPFC regulate social hierarchy. Here, using a confrontation tube test, we found that most adult C57BL/6J male mice could establish a linear social rank after 1 week of cohabitation. Lower rank individuals showed social anxiety together with decreased serum testosterone levels. D2R expression was significantly downregulated in the dorsal part of mPFC (dmPFC) in lower rank individuals, whereas D1R expression showed no significant difference among the rank groups in the whole mPFC. Virus knockdown of D2Rs in the dmPFC led to mice being particularly prone to lose the contests in the confrontation tube test. Finally, simultaneous D2R activation in the subordinates and D2R inhibition in the dominants in a pair switched their dominant–subordinate relationship. The above results indicate that D2Rs in the dmPFC play an important role in social dominance. Our findings provide novel insights into the divergent functions of prefrontal D1Rs and D2Rs in social dominance, which may contribute to ameliorating social dysfunctions along with abnormal social hierarchy.

Background Tumor metabolic reprogramming has attracted extensive attention, and mitochondria play a vital role in this process as a metabolic hub. Despite increasing evidence that mitochondria are involved in breast tumorigenesis, the impact of mitochondria-related genes on breast cancer remains unclear. In this study, we aimed to construct a novel mitochondria-related gene signature through bioinformatic strategies to predict and stratify the prognosis, immune infiltration, and treatment response of breast cancer patients. Methods The transcriptomic data and clinical features of breast cancer samples were extracted from The Cancer Genome Atlas and Molecular Taxonomy of Breast Cancer International Consortium databases. We constructed a mitochondria-related gene signature to predict survival, and Cox regression and receiver operating characteristic were used to evaluate the overall predictive performance. Subsequently, we combined the risk scores with corresponding clinical features to construct a nomogram model and evaluated the model’s accuracy by clinical calibration curve and decision curve analysis. After dividing patients based on risk scores, several immune-related analyses were performed to compare the immune infiltration status between different groups. Finally, we compared the mutation status and drug sensitivity to explore the treatment response and underlying mechanism of the difference in prognosis. Results We constructed an eight mitochondria-related gene risk signature by bioinformatic strategies and verified it by Cox regression, receiver operating characteristic, calibration curve and decision curve analyses. Patients with low-risk score have a better prognosis, enhanced immune infiltration, significantly different mutation landscapes, and a more sensitive response to antitumor drugs, which may account for the favorable survival. Conclusion The mitochondria-related gene signature is a novel prognostic risk signature that can be used as a predictor for patient stratification in breast cancer. In addition, this signature can effectively distinguish the immune infiltration and drug sensitivity status of patients. This may provide a new perspective for the treatment of breast cancer focusing on mitochondria.

We revisit $M$-ary classification of Gutman (TIT 1989), where one is tasked to determine whether a testing sequence is generated with the same distribution as one of the $M$ training sequences or not. Our main result is a two-phase test, its theoretical analysis and its optimality guarantee. Specifically, our two-phase test is a special case of a sequential test with only two decision time points: the first phase of our test is a fixed-length test with a reject option, the second-phase of our test proceeds only if a reject option is decided in the first phase and the second phase of our test does \emph{not} allow a reject option. To provide theoretical guarantee for our test, we derive achievable error exponents using the method of types and derive a converse result for the optimal sequential test using the techniques recently proposed by Hsu, Li and Wang (ITW, 2022) for binary classification. Analytically and numerically, we show that our two phase test achieves the performance of an optimal sequential test with proper choice of test parameters. In particular, similarly as the optimal sequential test, our test does not need a final reject option to achieve the optimal error exponent region while an optimal fixed-length test needs a reject option to achieve the same region. Finally, we specialize our results to binary classification when $M=2$ and to $M$-ary hypothesis testing when the ratio of the lengths of training sequences and testing sequences tends to infinity so that generating distributions can be estimated perfectly., submitted to IEEE Trans. Perfluorooctanesulfonate (PFOS) as an accumulative emerging persistent organic pollutant in crops poses severe threats to human health. Lettuce varieties that accumulate a lower amount of PFOS (low-accumulating crop variety, LACV) have been identified, but the regarding mechanisms remain unsolved. Here, rhizospheric activation, uptake, translocation, and compartmentalization of PFOS in LACV were investigated in comparison with those of high-accumulating crop variety (HACV) in terms of rhizospheric forms, transporters, and subcellular distributions of PFOS. The enhanced PFOS desorption from the rhizosphere soils by dissolved organic matter from root exudates was observed with weaker effect in LACV than in HACV. PFOS root uptake was controlled by a transporter-mediated passive process in which low activities of aquaporins and rapid-type anion channels were corrected with low expression levels of PIPs (PIP1-1 and PIP2-2) and ALMTs (ALMT10 and ALMT13) genes in LACV roots. Higher PFOS proportions in root cell walls and trophoplasts caused lower root-to-shoot transport in LACV. The ability to cope with PFOS toxicity to shoot cells was poorer in LACV relative to HACV since PFOS proportions were higher in chloroplasts but lower in vacuoles. Our findings provide novel insights into PFOS accumulation in lettuce and further understanding of multiprocess mechanisms of LACV.

An asymmetric [3 + 2] cycloaddition of vinyl ethylenecarbonates (VECs) and (E)-3-arylvinyl substituted benzo[d] isothiazole 1,1-dioxides has been developed using the Pd complex of a bidentate phosphoramidite (Me-BIPAM) as the catalyst, providing a wide variety of chiral multistereogenic vinyltetrahydrofurans in good yields with excellent diastereo- and enantioselectivities (up to >20:1 dr, 99% ee).

Carbon steel welding is a major manufacturing method for nuclear power construction. For the moment, there are two sets of standards of welding material for carbon steel which used in pressurized water reactor nuclear power plants of China. The coexistence of standards of two different technical routes brings unnecessary cost and waste to pressurized water reactor nuclear power plant manufacturing. With the rapid development of pressurized water reactor nuclear power plants in China, the research of developing a set of unified standards for carbon steel electrodes and rods for gas shielded arc welding and carbon steel covered electrodes is necessary. At present, two main standards will be revised and published. Through the interpretation of “Welding material for pressurized water reactor nuclear power plants — Part 1 Carbon steel covered electrodes” and “Welding material for pressurized water reactor nuclear power plants — Part 6 Carbon steel electrodes and rods for gas shielded arc welding” and the introduction to their compilation thoughts and technical points, the article guides engineers to understand the compilation thoughts of these two standards quickly. The standards specify classification, technical requirements, test methods and testing of carbon steel electrodes and rods for carbon steel in pressurized water reactor nuclear power plants.

Inflammatory arthritis is an inflammatory disease that involves the joints and surrounding tissues. Synovial hyperplasia often presents when joints become inflamed due to immune cell infiltration. Synovial membrane is an important as well as a highly specific component of the joint, and its lesions can lead to degeneration of the joint surface, causing pain and joint disability or affecting the patients’ quality of life in severe cases. Synovial macrophages (SMs) are one of the cellular components of the synovial membrane, which not only retain the function of macrophages to engulf foreign bodies in the joint cavity, but also interact with synovial fibroblasts (SFs), T cells, B cells, and other inflammatory cells to promote the production of a variety of pro-inflammatory cytokines and chemokines, such as TNF-α, IL-1β, IL-8, and IL-6, which are involved in the pathogenic process of inflammatory arthritis. SMs from different tissue sources have differently differentiated potentials and functional expressions. This article provides a summary on studies pertaining to SMs in inflammatory arthritis, and explores their role in its treatment, in order to highlight novel treatment modalities for the disease.

This research aims to investigate the association between hyperthyroidism and age, sex, and infraction of the myocardium in the Hong Kong population in order to detect hyperthyroidism-induced infraction earlier (e.g., thyrocardiac disease) through chemiluminescent microparticle immunoassay (CMIA). A total of 7616 random blood specimens from males and females were collected and examined by Architect i immunoassay platform to obtain quantitative laboratory values of Free FT4, TSH, and hs-cTnI in samples serum differentiating into overt and subclinical hyperthyroidism cases. The age was between 19 and 81 years old. In this research, for population characteristics, 57 cases confirmed as hyperthyroidism (e.g. FT4 > 1.48 ng/dL, according to Abbott’s suggested reference interval) were used for analysis and 22 cases were examined in the laboratory. The highest number of hyperthyroidisms including overt and subclinical cases in female was 43 (75.44%), rather than males. For prevalence data of hyperthyroidism between September 2019 to January 2020 in Hong Kong, the range was from 0.62% to 0.93% (Median percentage = 0.75%). The research will explore the in-depth impact of gender difference on thyrocardiac disease, risk of thyrocardiac disease by age group, and the possible relationship between FT4 and hs-cTnI.

Accumulating evidence indicates that abnormalities in the composition of gastrointestinal (GI) microbiota play a vital role in stress-related disorders. Both human beings and animals perceive stressful events differently, i.e., resilience or susceptibility. However, the role of GI microbiota in stress resilience/susceptibility and the underlying mechanisms remain largely unknown.Sixty male C57BL/6J mice were exposed to 10-day chronic social defeat stress (CSDS), and 28 were found to be resilient to CSDS. We next analyzed microbiota compositions in the cecum using 16S rDNA gene sequencing, which revealed a significant increase in the relative abundance of Lactobacillus at the genus level in the resilient mice. In subsequent experiments, we found that oral administration of a strain of Lactobacillus (Lactobacillus murinus) for 2 weeks attenuated the increased levels of stress-induced corticosterone and anxiety-like behavior in stress-susceptible mice. The mRNA expression of tryptophan hydroxylase 2 (a rate-limiting enzyme in 5-HT synthesis) was also significantly increased in the dorsal raphe nucleus (DR) of stress-susceptible mice.Lactobacillus contributes to stress resilience, and the DR 5-HT system may play an important role during this process. The above results suggest that certain organisms in the GI tract may play an essential role in stress response and be useful in the prevention and treatment of some stress-related psychiatric disorders, such as depression.

Polycystic ovary syndrome (PCOS) is a heterogeneous disorder with evidence of polygenetic components, and obesity may be a risk factor for hyperandrogenism. Previous studies have shown that LHCGR is enriched in the ovary and LHCGR deficiency causes infertility without typical PCOS phenotypes. ALMS1 is implicated in obesity and hyperandrogenism, the common phenotypes among PCOS patients. Through whole-exome sequencing of 22 PCOS families and targeted candidate gene sequencing of additional 65 sporadic PCOS patients, we identified potential causative mutations in LHCGR and ALMS1 in a sibling-pair PCOS family and three sporadic PCOS patients. The expression of LHCGRL638P in granulosa-like tumor cell line (KGN) cells promoted cyclic adenosine monophosphate production and granulosa cell proliferation, indicating that LHCGRL638P is an activating mutation. LhcgrL642P/L642P mice showed an irregular estrous cycle, reduced follicles with dynamic folliculogenesis, and increased testosterone (T), estradiol (E2), and dehydroepiandrosterone. Lhcgr+/L642PAlms1+/PB mice displayed increased T and E2 but decreased late secondary and preovulatory follicles. We showed that activating mutation of LHCGR likely plays important roles in the pathophysiology of PCOS involving abnormal reproductive physiology, whereas ALMS1 deficiency may promote anovulatory infertility via elevated androgens, suggesting that the disturbed LHCGR and ALMS1 cooperatively induce PCOS phenotypes, characterized as anovulation and hyperandrogenemia frequently observed in PCOS patients with obesity.

This preclinical study aimed to evaluate whether using transferred mosaic embryos (primarily selected by embryonic morphology assessment (EMA) and compared by the noninvasive preimplantation genetic testing for aneuploidy (niPGT-A) on cell-free DNA in blastocoel fluid (BF)) increases the rates of clinical pregnancies (CPs) and healthy live births (HLBs) and to investigate whether niPGT-A could provide valuable genetic information for the EMA-selected transferred mosaic embryos.This study collected 215 blastocyst culture samples and 182 BF samples. Cell-free DNA from the BF was amplified and examined by next-generation sequencing-based niPGT-A. All 182 patients underwent EMA. However, only 147 underwent in vitro fertilization and embryo transfer, and only 113 clinical outcomes were followed up. Comprehensive chromosome screening for the chorionic villus sampling of spontaneous miscarriages and noninvasive prenatal testing for ongoing pregnancies were also performed.The implantation rate was 77.55% in 147 transferred high-quality embryos selected by EMA. Among 113 CPs, 16 led to spontaneous miscarriage (14.16%), and 97 resulted in HLBs (85.84%). According to the niPGT-A results for 113 patients with clinical outcomes, 80.4% had CP (euploid, 20.54%; single aneuploid, 1.79%; mosaic chromosome aneuploid and/or segmental aneuploid, 58.04%). Of all the mosaic aneuploids, 90.76% were false positive, transforming to euploid.Transferred EMA-selected embryos showed higher implantation rates. The niPGT-A of BF provided valuable genetic status ("-ploid") information, which helped reduce aneuploid-induced implantation failure and miscarriage, thereby increasing the CP and HLB rates. Additionally, majority of the transferred embryos with complex/chaotic mosaic aneuploid would likely develop HLBs.

To observe the clinical effect of cluster acupuncture at scalp points in treating limb spasm after stroke on the basis of conventional exercise therapy.A total of 72 patients with limb spasm after stroke were randomly divided into an observation group (36 cases, 5 cases dropped off) and a control group (36 cases, 6 cases dropped off). The control group was treated with exercise therapy. In the observation group, on the basis of the control group, penetrating technique of acupuncture was exerted at Qianding (GV 21) to Baihui (GV 20), Xinhui (GV 22) to Qianding (GV 21), etc. once a day, 5 days a week for 4 weeks. Before and after treatment, the changes of the modified Ashworth scale (MAS), simplified Fugl-Meyer motor assessment (FMA), and modified Barthel index (MBI) scores of the two groups were compared.After treatment, the MAS scores of upper and lower limbs in the two groups were lower than before treatment (On the basis of conventional exercise therapy, cluster acupuncture at scalp points can reduce the spasm, improve motor function and activities of daily living in patients with limb spasm after stroke.

The Qingchengzi ore field is an important gold-polymetallic center of the North China Craton. It has been recognized that the gold deposits in Qingchengzi were controlled by structures like lithological interfaces and fractures along mechanically weak bedding and foliation planes, but it still remains poorly understood how the structures affected the localization of the gold deposits. Finite element based numerical modeling was used to reproduce the deformation process of the Baiyun gold deposit during the mineralization period. Paleoproterozoic schist and marble are widely exposed in Qingchengzi, and a large part of the Baiyun gold ores occurs along the interfaces between the schist and the marble. The modeling results suggest that the mechanical contrast between the schist and the marble may be a major reason why the stress was localized along their lithological interfaces under a compressional stress regime. Two parts of their lithological interfaces were identified to be easily stress-localized and first fractured: the interface between the schist and its underlying marble at shallower levels and the one between the schist and its overlying marble at deeper levels. Stress concentration in these two parts is independent on the dipping angle and direction of the interfaces. Therefore, mineralizing fluids may have been concentrated into these two parts. The first one is consistent with the present ore bodies of the Baiyun gold deposit, and the second one could be considered for deep prospecting. These findings also provide implications for the structural controls of lithological interfaces on the mineralization in other gold deposits of this region.

The reliability-based design optimization (RBDO) problem involving discrete design variables and multi-working constraints is tackled with the consideration of the uncertainty factors in the optimal design of crawler crane’s lattice boom. In the optimization model, the uncertainties of material property, geometric parameters and loads are represented by random variables. The geometrical dimensions, performance functions’ reliability indexes and the minimum weight of lattice boom are defined respectively as the random design variables, probabilistic constraints and objective function. Integrating the reliability analysis method based on response surface method (RSM) with multi-island genetic algorithm (MIGA), an efficient reliability-based design optimization process for the lattice boom is developed under the typical constrained working conditions. Several verification cases are presented to demonstrate the effectiveness of optimization results. It is shown that the reliability-based design optimization method can not only satisfy the reliability constraints of the lattice boom, but also ensure the safety and economy of crawler crane boom as well as minimize the structure weight. The developed method is capable to be extended to the reliability-based design optimization of other large and complex construction machinery structures.

A novel periodic phase modulation method is proposed and verified for the fast diagnosis of phased array. Taking the diagnosis of receiving paths as an example, its principle is as follows. Periodic phase modulations with different frequencies are added to each phase shifter of the phased array. Because of the periodic modulation, harmonic components are generated in all the elements. As modulation frequencies added to all the elements are different from each other, the generated harmonic components have different frequencies correspondingly. If some elements suffer failure, the corresponding harmonic components will vanish from the output of the power combiner. Therefore, the faulty elements can be located by analyzing the spectrum of the received signal after the power combiner. The proposed method needs no the auxiliary hardware and its signal processing is concise. An eight-element $S$ -band phased array is fabricated to examine its effectiveness.

Enantioselective copper-catalyzed cascade inter- and intramolecular amidation was achieved between ethynyl benzoxazinanones and α-halohydroxamates in the presence of an indapybox ligand. The one-pot cascade transformation was triggered by the attack of hydroxamates to dipolar copper–allenylidene intermediates, followed by a nucleophilic annulation reaction. Thus, a series of exo-methylene 3-aminoindoline derivatives were obtained in good yields with high enantioselectivities under mild reaction conditions.
