1. Influence of Viscosity and Thickener on the Effects of Bleaching Gels

Abstract

SUMMARY Objective This study investigated the influence of the viscosity and kind of thickener of 35% hydrogen peroxide bleaching gels on the tooth (color change, demineralization of enamel, and permeation) and on the gel [reactive oxygen species (ROS), pH, and peroxide concentration]. Methods and Materials Two hundred forty specimens were divided into groups of bleaching gels with different thickeners (CAR, carbomer; ASE, alkali swellable emulsion; MSA, modified sulfonic acid polymer; SSP, semisynthetic polysaccharide; PAC, particulate colloids) in three viscosities (low: 50,000 cP; medium: 250,000 cP; high: 1,000,000 cP). Color change (ΔEab), demineralization of enamel by Knoop microhardness (KHN) reduction analysis, and peroxide permeation (PP) were analyzed in the specimens, while pH, peroxide concentration (PC), and ROS were evaluated in the gels. Data were analyzed by two-way ANOVA (α=0.05). Results The higher viscosity gels reduced ΔEab, PP, enamel softening, and ROS in relation to the lower viscosity gels. However, the drop in pH and PC were higher in the more viscous gels. Gels with MSA produced higher ΔEab compared with SSP and ASE. The PP was higher for PAC, and smaller for SSP and CAR. The KHN reduction was higher for CAR and smaller for PAC. The higher pH reduction was seen for ASE and CAR, and the smaller for SSP. The PC reduction was higher for SSP and smaller for CAR. More ROS were observed for MSA and fewer for ASE. Conclusions Increased gel viscosity was associated with reduced color change, permeation, demineralization of enamel, and ROS, and led to increased peroxide decomposition and pH alteration during the treatment. The kind of thickener significantly interfered with the treatment effects.

Abstract

Rare genetic neurological disorders (RND; ORPHA:71859) are a heterogeneous group of disorders comprising >1700 distinct genetic disease entities. However, genetic discoveries have not yet translated into dramatic increases of diagnostic yield and indeed rates of molecular genetic diagnoses have been stuck at about 30–50% across NGS modalities and RND phenotypes [1, 2]. Existence of yet unknown disease genes as well as shortcomings of commonly employed NGS technologies and analysis pipelines in detecting certain variant types are typically cited to explain the low diagnosis rates. To increase the diagnostic yield in RNDs - one of the four focus disease groups in Solve-RD - we follow two major approaches, that we will here present and exemplify: (i) systematic state-of the art re-analysis of large cohorts of unsolved whole-exome/genome sequencing (WES/WGS) RND datasets; and (ii) novel-omics approaches. Based on the way Solve-RD systematically organizes researchers’ expertise to channel this approach [3], the European Reference Network for Rare Neurological Diseases (ERN-RND) has established its own Data Interpretation Task Force (DITF) within SOLVE-RD, which is currently composed of clinical and genetic experts from 29 sites in 15 European countries. The Solve-RD project has received funding from the European Union’s Horizon 2020 research and innovation programme under Grant Agreement No. 779257. 3. Proteomic features of skeletal muscle adaptation to resistance exercise training as a function of age

Abstract

Resistance exercise training (RET) can counteract negative features of muscle ageing but older age associates with reduced adaptive capacity to RET. Altered muscle protein networks likely contribute to ageing RET adaptation; therefore, associated proteome-wide responses warrant exploration. We employed quantitative sarcoplasmic proteomics to compare age-related proteome and phosphoproteome responses to RET. Thigh muscle biopsies were collected from eight young (25 ± 1.1 years) and eight older (67.5 ± 2.6 years) adults before and after 20 weeks supervised RET. Muscle sarcoplasmic fractions were pooled for each condition and analysed using Isobaric Tags for Relative and Absolute Quantification (iTRAQ) labelling, tandem mass spectrometry and network-based hub protein identification. Older adults displayed impaired RET-induced adaptations in whole-body lean mass, body fat percentage and thigh lean mass (P > 0.05). iTRAQ identified 73 differentially expressed proteins with age and/or RET. Despite possible proteomic stochasticity, RET improved ageing profiles for mitochondrial function and glucose metabolism (top hub; PYK (pyruvate kinase)) but failed to correct altered ageing expression of cytoskeletal proteins (top hub; YWHAZ (14–3-3 protein zeta/delta)). These ageing RET proteomic profiles were generally unchanged or oppositely regulated post-RET in younger muscle. Similarly, RET corrected expression of 10 phosphoproteins altered in ageing, but these responses were again different vs. younger adults. Older muscle is characterised by RET-induced metabolic protein profiles that, whilst not present in younger muscle, improve untrained age-related proteomic deficits. Combined with impaired cytoskeletal adhesion responses, these results provide a proteomic framework for understanding and optimising ageing muscle RET adaptation.

Abstract

Recognition of the untapped potential of photosynthesis to improve crop yields has spurred research to identify targets for breeding. The CO2-fixing enzyme Rubisco is characterised by a number of inefficiencies and frequently limits carbon assimilation at the top of the canopy, representing a clear target for wheat improvement. Two bread wheat lines with similar genetic backgrounds and contrasting in vivo maximum carboxylation activity of Rubisco per unit leaf nitrogen (Vc,max,25/Narea) determined using high throughput phenotyping methods were selected for detailed study from a panel of 80 spring wheat lines. Detailed phenotyping of photosynthetic traits in the two lines using glasshouse-grown plants showed no difference in Vc,max,25/Narea determined directly via in vivo and in vitro methods. Detailed phenotyping of glasshouse-grown plants of the 80 wheat lines also showed no correlation between photosynthetic traits measured via high throughput phenotyping of field-grown plants. Our findings suggest that the complex interplay between traits determining crop productivity and the dynamic environments experienced by field-grown plants needs to be considered when designing strategies for effective wheat crop yield improvement when breeding for particular environments

5. Psychometric properties of the oral feeding assessment in premature infants scale

Abstract

Professionals that work in neonatal units need to identify the strengths and weaknesses of the premature infant who is in the transition process from feeding through a gastric tube to oral feeding. The main aim of this study was to validate the Oral FEeding Assessment in premaTure INfants (OFEATINg) instrument. A psychometric validity and reliability study was conducted in Neonatal Intensive Care Units of two public, metropolitan, university hospitals. The study population were premature infants at a postconceptional age of 31-35 weeks. The study included evaluation of the reliability, convergent, discriminant and construct validity, sensitivity and specificity of the OFEATINg instrument. A total of 621 feedings of 56 preterm infants were evaluated. Confirmatory factor analysis identified 3 factors and 13 indicators with a good fit to the model. Cronbach's alpha coefficient was 0.78. The instrument showed high indices of inter-rater reliability (Pearson 0.9 and intraclass correlation coefficient 0.95). The OFEATINg scale is a valid and reliable instrument for evaluating the readiness for oral feeding of preterm infants. It may enable clinicians to evaluate the physiological and behavioral abilities involved in the oral feeding process and help them make decisions related to the transition to full oral feeding.Clinical trial registration: This study was prospectively registered at the two Institutional review boards.

6. The use of Multi-Sensory Environments with autistic children: Exploring the effect of having control of sensory changes

Abstract

Multi-Sensory Environments (also called sensory or Snoezelen® rooms) are specialised spaces that contain equipment to modify the environment. They are commonly used in special-needs schools with autistic children, but empirical investigation into how to best use Multi-Sensory Environments with autistic children has been limited. Based on converging evidence that autistic children may benefit from having control, we tested 41 autistic children (8 female) aged 4–12 years (mean = 8 years, standard deviation = 2.05 years) who used the Multi-Sensory Environment both with and without control over the sensory changes. Behavioural coding of video data showed that having control was associated with increased attention and reduced repetitive motor behaviours, sensory behaviours, activity levels, stereotyped speech and vocalisations. Social behaviour, anxiety, positive affect and arousal were not significantly affected by condition. Our findings demonstrate that how a Multi-Sensory Environment is used can affect autistic children’s behaviours. They also suggest that providing control of sensory changes to autistic children may help create better conditions for learning.

Abstract

Novel Corona Virus Disease-19 (nCov-19, COVID-19) was recognised as a pandemic by the World Health Organization on March 11, 2020. As of June 14, 2020, this contagious viral disease has afflicted 188 out of 195 countries in the world with 7,893,700 confirmed cases and 432,922 global deaths. Canada has 98,787 people infected and 8,146 deaths. COVID-19 is thought to transmit through contact, droplets and aerosolization. A rapid review showed limited information on the benefits of conducting lung ultrasound (LUS) versus chest radiograph (CXR) or studies correlating lung ultrasound to chest computed Tomography (CT) in patients positive for Covid-19. The literature review confirmed that CT and LUS cannot diagnose this disease, but that both can help in the management and staging of this disease. There is no literature to prove that LUS at the bedside may be beneficial from the view of decreased transmission to other health care workers and bystanders due to reduced transit but comparing the transit pathway and contact leads one to propose that this would be so. Pregnant patients with COVID-19, young children and patients in the reproductive stage would also benefit from LUS since there is no radiation dose and the critical patient in distress will benefit from testing at the bedside.

Abstract

This study evaluated the microtensile bond strength (μTBS) of resin-based composite (RBC) to dentin after different immediate dentin sealing (IDS) strategies and surface-conditioning (SC) methods and on two water storage times. Human molars (n=48) were randomly divided into eight experimental groups involving four different IDS strategies-IDS-1L with one layer of adhesive, IDS-2L with two layers of adhesive, IDS-F with one layer of adhesive and one layer of flowable RBC, and DDS (delayed dentin sealing) with no layer of adhesive (control)-and two different SC methods-SC-P with pumice rubbing and SC-PC with pumice rubbing followed by tribochemical silica coating. The μTBS test was performed after one week and after six months of water storage, being recorded as the "immediate" and "aged" μTBS, respectively. This paper examines the available information on smoking cessation and posttraumatic stress disorder (PTSD). Unfortunately, there are only two available preliminary smoking cessation intervention studies for smokers with PTSD. The paper first reviews the epidemiology of PTSD, as well as smoking rates in the VA and veterans with PTSD. Next, it presents preliminary information regarding the relationship between PTSD symptoms and smoking. Finally, it presents various approaches to smoking cessation for PTSD smokers in the VA, and explores potential fruitful avenues for enhancing smoking cessation rates in this population.

Abstract

In this work we show results of experiments aiming to understand the stability of a Gas Electron Multiplier (GEM) detector. While operating the detector, we monitor temperature, pressure and humidity in order to understand the contribution of each of these parameters to the behaviour of the devise. We also use a Single Wire proportional counter as a reference detector so to keep track of the gain fluctuations given by external influences (pressure, temperature, gas composition, etc.). In this way it is possible to spot fluctuation due to the intrinsic properties of these new detectors. A special effort is devoted to explaining and discussing different experimental drawbacks that appeared on the way in order to set a precedent, and as far as possible to avoid them in future experiences.

Abstract

Epidermal cysts are very common lesions. Here we present the unusual case of an epidermal cyst occurring between the anal sphincters, presenting as a lump in the perineum. This was successfully excised with careful dissection of the intersphincteric plane. To our knowledge this is the only case of its kind reported in the literature.

Abstract

We are reporting the synthesis, the physicochemical, and the hydrogen sorption properties of a novel bcc high entropy alloy Ti0.30V0.25Cr0.10Zr0.10Nb0.25. At room temperature, the alloy rapidly absorbs hydrogen reaching a capacity of 2.0 H/M (3.0 wt.%) and forming a dihydride with fcc structure, as confirmed by both synchrotron X-ray diffraction and neutron diffraction. The absorption Pressure–Composition Isotherms corroborated with synchrotron X-ray diffraction prove that the reaction with hydrogen occurs within two steps, i.e., bcc alloy → bcc monohydride → fcc dihydride. The thermodynamic parameters calculated for the second step transformation evidence the formation of a stable dihydride with ΔHabs = −75 kJ/molH2. The phase transition during hydrogen/deuterium desorption was investigated by in situ synchrotron X-ray and neutron diffraction confirming a reversible reaction with hydrogen. Furthermore, the cycling properties show a decrease of the capacity over the first cycles followed by a stabilization at 2.44 wt.%, whereas the absorption kinetics improve after the first cycle reaching full capacity after only 30 s at room temperature.

Abstract

BY-COVID Work Package (WP) 2 brings together datasets and catalogues of metadata across domains, assesses data governance and access procedures, and contributes to increase the FAIRness level of different data sources. It aims to align metadata descriptions and other semantic information, first within domains (e.g., biomolecular and imaging, clinical and health, survey, etc), and in a second stage (in alignment with WP3 developments) across domains to propose a reference catalogue with harmonised metadata descriptions, which can facilitate data access and eventually enable data mobilisation for the analysis workflows specified by WP4 . Deliverable 2.2 (D2.2) focuses on data access and transfer across research domains and jurisdictions. Originally intended to address technical, legal, and organisational barriers in data sources, its scope expanded to include all types of data that also link to pathogen variants via SARS-CoV-2 DataHubs (WP1). The objectives of D2.2 are two-fold: ensuring compliance with data privacy principles for controlled access data and proposing strategies to enhance data interoperability. Efforts included developing a survey, conducting ad-hoc meetings with key stakeholders, and aligning with the FAIR data principles. The deliverable contributes to ongoing efforts to facilitate efficient and responsible data sharing, which should enable faster and coordinated responses in future pandemics.

Abstract

The identification of a mutant of C/EBPα (C/EBPαR35A) that greatly accelerates BMT helped to illuminate the mechanism. Thus, incoming C/EBPα binds to PU.1, an obligate partner expressed in B cells, leading to the release of PU.1 from B cell enhancers, chromatin closing and silencing of the B cell program. Released PU.1 redistributes to macrophage enhancers newly occupied by C/EBPα, causing chromatin opening and activation of macrophage genes. All these steps are accelerated by C/EBPαR35A, initiated by its increased affinity for PU.1. Wild-type C/EBPα is methylated by Carm1 at arginine 35 and the enzyme’s perturbations modulate BMT velocity as predicted from the observations with the mutant. Increasing the proportion of unmethylated C/EBPα in granulocyte/macrophage progenitors by inhibiting Carm1 biases the cell’s differentiation toward macrophages, suggesting that cell fate decision velocity and lineage directionality are closely linked processes. Abstract

International audience; Polycrystalline materials exhibit physical properties that are driven by both the interatomic crystallographic structure as well as the nature and density of structural defects. Crystallographic evolutions driven by phase transitions and associated twinning process can be observed in situ in three-dimensional (3D) using monochromatic synchrotron radiation at very high temperatures (over 1000 C). This paper focuses on continuous measurements of the 3D-reciprocal space maps by high-resolution x-ray diffraction as a function of temperature along a phase transition process occurring between 1200 C and room temperature. These high precision measurements allow observing the reciprocal space node splitting and the evolution of the diffuse scattering signal around that node as a function of temperature. The capability of this experimental method is illustrated by direct in situ high temperature measurements of the 3D splitting of a reciprocal space node due to phase transition recorded on dense pure zirconia polycrystals.

Abstract

Coherent X-ray Diffraction Imaging (CDI) [1] allows for the in situ 3D mapping of the structural changes during phasetransitions and chemical reactions, with a fair spatial resolution of the order of magnitude of 10 nm [2]. Using thistechnique in multi-Bragg conditions (Bragg CDI on multiple reflections), it was possible to follow the structural evolution(change in morphology, variation of the displacement and strain fields) as a function of temperature in a 300 nm Ni 3Fenanocrystal. During this volume transition, which happens between 500°C and 600°C, a core-shell structure isprogressively observed with two distinct lattice parameters (difference of 0.39%, corresponding to two different Ni:Feratios), leading to highly strained and coherent domains. A rounding of the particle is also observed, with a disappearingof the initial facets. Static molecular calculations (LAMMPS) of different systems, such as faceted and core-shellnanoparticles, complete this work and allow for the confirmation of a core-shell structure. This study shows the perfectability of modern phase retrieval methods to reconstruct highly strained systems such as core-shell nanoparticles.

Abstract

International audience; X-ray diffraction tomography is a well-developed technique to study the structure of heterogeneous materials which makes it a tool of choice for the non-destructive investigation of cultural heritage microsamples. Characterizing such complex materials with a high enough spatial resolution requires acquiring large amounts of diffraction images, followed by a complex sequence of data management to localize the crystalline phases in the sample. Here we propose to use multivariate analysis in order to automatically decompose the data in a small set of components, each representing the diffraction pattern of one or a small number of phases. This makes phase identification and quantification of each component much more efficient and leads to a quantitative knowledge of the phase content in each voxel of the tomographic reconstruction. We show that non-negative matrix factorization is very efficient for this purpose, with a computing time well compatible with in-line data analysis in order to assess the quality of measurements during experiments at synchrotron beamlines. Here, we apply the method to the investigation of microsamples from medieval applied brocade decors selected from a wooden statue, and the results are validated a posteriori by comparison with ex situ destructive techniques.

Abstract

The Senegalese sole (Solea senegalensis) is an economically important flatfish species. In this study, a genome draft was analyzed to identify microsatellite (SSR) markers for whole-genome genotyping. A subset of 224 contigs containing SSRs were preselected and validated by using a de novo female hybrid assembly. Overall, the SSR density in the genome was 886.7 markers per megabase of genomic sequences and the dinucleotide motif was the most abundant (52.4%). In silico comparison identified a set of 108 SSRs (with di-, tetra- or pentanucleotide motifs) widely distributed in the genome and suitable for primer design. A total of 106 markers were structured in thirteen multiplex PCR assays (with up to 10-plex) and the amplification conditions were optimized with a high-quality score. Main genetic diversity statistics and genotyping reliability were assessed. A subset of 40 high polymorphic markers were selected to optimize four supermultiplex PCRs (with up to 11-plex) for pedigree analysis. Theoretical exclusion probabilities and real parentage allocation tests using parent-offspring information confirmed their robustness and effectiveness for parental assignment. These new SSR markers were combined with previously published SSRs (in total 229 makers) to construct a new and improved integrated genetic map containing 21 linkage groups that matched with the expected number of chromosomes. Synteny analysis with respect to C. semilaevis provided new clues on chromosome evolution in flatfish and the formation of metacentric and submetacentric chromosomes in Senegalese sole. Abstract

International audience; We show two examples of resonant x-ray scattering experiments performed in situ on the INS2 end-station of the BM32 beamline at ESRF. In the first one the inversion parameter of thin cobalt ferrite films grown on Ag(001) by oxygen-assisted MBE is found. In spinel structure this parameter quantifies the distribution of divalent and trivalent cations on tetrahedral and octahedral sites, and strongly influence the electronic and magnetic properties of the ferrite. By measuring grazing incidence resonant x-ray diffraction intensity versus energy of several spinel peaks at the iron and cobalt K-edges and comparing with theoretical calculations performed with the FDMNES code, we could find that cobalt occupies mostly octahedral sites, resulting in an inversion parameter of 0.88(5). 1 In the second experiment we studied Co-Pt nanoparticles self-organized on an ultrathin aluminum oxide film on Ni3Al(111), a template with a 4.1 nm hexagonal surface lattice. Co and Pt were subsequently deposited on the template layer seeded with a few Pd atoms per unit cell. Bimetallic nanoparticles organize at the corners of the oxide hexagonal surface cell giving sharp peaks in the small angle x-ray scattering pattern. The analysis of the RXD measured at the cobalt K edge on these SAXS features evidences then a core-shell structure. GISAXS of 0.15 nm Co on alumina template (Pd seeded), and RXD of the (10) reflection (Co K edge). These examples demonstrate the capabilities of BM32 for in situ resonant scattering studies. At short term, they will be improved with the installation of a crystal analyser.

Data processing; DNA sequencing; Genomic analysis Tratamiento de datos; Secuenciación de ADN; Análisis genómico Tractament de dades; Seqüenciació d'ADN; Anàlisi genòmica Methods to reconstruct the mitochondrial DNA (mtDNA) sequence using short-read sequencing come with an inherent bias due to amplification and mapping. They can fail to determine the phase of variants, to capture multiple deletions and to cover the mitochondrial genome evenly. Here we describe a method to target, multiplex and sequence at high coverage full-length human mitochondrial genomes as native single-molecules, utilizing the RNA-guided DNA endonuclease Cas9. Combining Cas9 induced breaks, that define the mtDNA beginning and end of the sequencing reads, as barcodes, we achieve high demultiplexing specificity and delineation of the full-length of the mtDNA, regardless of the structural variant pattern. The long-read sequencing data is analysed with a pipeline where our custom-developed software, baldur, efficiently detects single nucleotide heteroplasmy to below 1%, physically determines phase and can accurately disentangle complex deletions. Our workflow is a tool for studying mtDNA variation and will accelerate mitochondrial research. Here, we describe that IL-6 is necessary for C/EBPα-enhanced reprogramming of B cells into iPSCs but not for B cell to macrophage transdifferentiation. C/EBPα overexpression activates both Il6 and Il6ra genes in B cells and in PSCs. In embryo development, Cebpa is enriched in the trophectoderm of blastocysts together with Il6, while Il6ra is mostly expressed in the inner cell mass (ICM). In addition, Il6 expression in blastocysts requires Cebpa. Blastocysts secrete IL-6 and neutralization of the cytokine delays the morula to blastocyst transition. The observed requirement of C/EBPα-regulated IL-6 signaling for pluripotency during somatic cell reprogramming thus recapitulates a physiologic mechanism in which the trophectoderm acts as niche for the ICM through the secretion of IL-6. Abstract

International audience; A deep understanding of the solid-state phase transition processes of zirconia is a mandatory requirement for the development of new zirconia-based materials useful for many industrial applications. For five decades, the monoclinic ⇔ tetragonal phase transition is described as a martensitic one and it is well known that it is associated with a large unit cell volume variation that promotes the appearance of elastic strains and also microcracking. In the present paper, we study, through in situ high temperature x-ray diffraction experiments, the coupling between strain relaxation and the martensitic phase transition into a pure zirconia bulk polycrystal. Quantitative analysis of the diffraction signal allows us to disentangle, with respect to the temperature variation, the phase transition and the microcracking processes, and we demonstrate that a high temperature postelaboration thermal treatment induces an increase in the stored elastic energy. Finally, we show that in such polycrystals exhibiting a crystal size distribution and in which the crystals are under internal stresses, the tetragonal to monoclinic phase transition process evolves from a first order one to a second order one when the temperature decreases

Abstract

Acetylation; Neurogenesis Acetilación; Neurogénesis Acetilació; Neurogènesi Regulation of histone acetylation dictates patterns of gene expression and hence cell identity. Due to their clinical relevance in cancer biology, understanding how human embryonic stem cells (hESCs) regulate their genomic patterns of histone acetylation is critical, but it remains largely to be investigated. Here, we provide evidence that acetylation of histone H3 lysine-18 (H3K18ac) and lysine-27 (H3K27ac) is only partially established by p300 in stem cells, while it represents the main histone acetyltransferase (HAT) for these marks in somatic cells. Our analysis reveals that whereas p300 marginally associated with H3K18ac and H3K27ac in hESCs, it largely overlapped with these histone marks upon differentiation. Interestingly, we show that H3K18ac is found at “stemness” genes enriched in RNA polymerase III transcription factor C (TFIIIC) in hESCs, whilst lacking p300. Moreover, TFIIIC was also found in the vicinity of genes involved in neuronal biology, although devoid of H3K18ac. Our data suggest a more complex pattern of HATs responsible for histone acetylations in hESCs than previously considered, suggesting a putative role for H3K18ac and TFIIIC in regulating “stemness” genes as well as genes associated with neuronal differentiation of hESCs. The results break ground for possible new paradigms for genome acetylation in hESCs that could lead to new avenues for therapeutic intervention in cancer and developmental diseases. Abstract

The integration of physical and high-density genetic maps is a very useful approach to achieve chromosome-level genome assemblies. Here, the genome of a male Senegalese sole (Solea senegalensis) was de novo assembled and the contigs were anchored to a high-quality genetic map for chromosome-level scaffolding. Hybrid assembled genome was 609.3 Mb long and contained 3403 contigs with a N50 of 513 kb. The linkage map was constructed using 16,287 informative SNPs derived from ddRAD sequencing in 327 sole individuals from five families. Markers were assigned to 21 linkage groups with an average number of 21.9 markers per megabase. The anchoring of the physical to the genetic map positioned 1563 contigs into 21 pseudo-chromosomes covering 548.6 Mb. Comparison of genetic and physical distances indicated that the average genome-wide recombination rate was 0.23 cM/Mb and the female-to-male ratio 1.49 (female map length: 2,698.4 cM, male: 2,036.6 cM). Genomic recombination landscapes were different between sexes with crossovers mainly concentrated toward the telomeres in males while they were more uniformly distributed in females. A GWAS analysis using seven families identified 30 significant sex-associated SNP markers located in linkage group 18. The follicle-stimulating hormone receptor appeared as the most promising locus associated with sex within a region with very low recombination rates. An incomplete penetrance of sex markers with males as the heterogametic sex was determined. An interspecific comparison with other Pleuronectiformes genomes identified a high sequence similarity between homologous chromosomes, and several chromosomal rearrangements including a lineage-specific Robertsonian fusion in S. senegalensis. Abstract

High entropy alloys belong to a new and promising class of functional materials for solid-state hydrogen storage. In this context, a novel single-phase body centered cubic (bcc) high entropy alloy Ti0.30V0.25Zr0.10Nb0.25Mo0.10 was prepared. The physicochemical and hydrogen sorption properties have been determined by both laboratory and large-scale facilities. This alloy can quickly absorb hydrogen up to 2.0 H/M (2.8 wt.%) at room temperature and forms a face centered cubic (fcc) hydride, as proven by synchrotron X-ray diffraction. The Pressure–Composition Isotherm and in situ neutron diffraction during hydrogen/deuterium desorption reaction suggest that the alloy experiences a reversible single step phase transition (bcc↔fcc). PDF analysis from X-ray total scattering data points out that the hydride phase possesses an average fcc structure with random atoms distribution and small lattice distortion. Despite an initial small fading of the capacity, the alloy withstands 20 absorption/desorption cycling without phase decomposition, as demonstrated by kinetic measurements coupled with X-ray diffraction and microstructural study by SEM-EDS. Moreover, the complete hydrogen absorption occurs in less than 30 s at room temperature and the kinetic improves during cycling.

Abstract

International audience; A complete study on the thermal stability of the crystal structure and electrical properties, as well as on the phase decomposition, of the high-performance thermoelectric colusite Cu26Cr2Ge6S32 is presented. A combination of results from differential scanning calorimetry, thermogravimetric analysis, in situ neutron powder diffraction, and Seebeck coefficient and electrical resistivity temperature cycling experiments show that the high thermoelectric performance colusite Cu26Cr2Ge6S32 is stable at least up to 700 K in a nonoxidative atmosphere. A superficial sulfur loss without phase decomposition is observed above approximate to 760 K, leading to a progressive increase of both electrical resistivity and Seebeck coefficient. The colusite phase, Cu26Cr2Ge6S32, starts to decompose above 830 K into Cu8GeS6 and CuCrS2 phases. This leads to a Ge enrichment of the colusite phase (i.e., decrease of the Cu/Ge ratio). This initial decomposition step is followed by a second one above 880 K, related to the structural modification from the cubic structure of colusite into the monoclinic Cu2GeS3 structure. Above 930 K, the colusite Cu26Cr2Ge6S32 phase is fully decomposed. The influence on the crystal structure of the superficial sulfur loss detected a few dozens of degrees before the colusite Cu26Cr2Ge6S32 phase decomposition is discussed and compared to the results obtained on the colusite Cu26V2Ge6S32 phase, stable at least up to 1000 K. A singular thermal evolution of the unit cell parameter of the latter is observed and discussed. Finally, a discussion on the thermoelectric performance optimization by process-induced structural disordering in these two colusite phases is given.

Abstract

Brain metastases are the most common tumor of the brain with a dismal prognosis. A fraction of patients with brain metastasis benefit from treatment with immune checkpoint inhibitors (ICI) and the degree and phenotype of the immune cell infiltration has been used to predict response to ICI. However, the anatomical location of brain lesions limits access to tumor material to characterize the immune phenotype. Here, we characterize immune cells present in brain lesions and matched cerebrospinal fluid (CSF) using single-cell RNA sequencing combined with T cell receptor genotyping. Tumor immune infiltration and specifically CD8+ T cell infiltration can be discerned through the analysis of the CSF. Consistently, identical T cell receptor clonotypes are detected in brain lesions and CSF, confirming cell exchange between these compartments. The analysis of immune cells of the CSF can provide a non-invasive alternative to predict the response to ICI, as well as identify the T cell receptor clonotypes present in brain metastasis., The use of CSF for diagnosis of metastatic brain tumors could be of clinical and patient benefit. Here the authors undertake a single-cell RNA analysis of CSF and brain to determine whether the phenotype in the CSF is reflective of the phenotype in the tumor.

Abstract

International audience; Two polychrome sculptures from the late Middle Age were investigated by means of non-invasive techniques with the aim of characterizing sophisticated relief decoration named "applied brocade". Here, we have used portable instrumentation solely in order to perform X-ray powder diffraction and fluorescence measurements at the surface of the sculptures. The results show a reliable localisation and identification of the different materials making up an applied brocade: a filler material, a tin-based foil and a gilding, even in the case of a largely overpainted sculpture. More particularly, a direct identification of the organic material used for the filler as natural beeswax is proposed. Differences between the two investigated artworks have also been identified: (i) in the structure of the calcium sulphate preparation layers (one or double layer of gesso) (ii) in the presence or absence of a priming paint layer under decors (iii) in the choice of the material used to make the gilding: pure gold or probable zwischgold. The characterization of the degradation of this type of decorations was also assessed, namely romarchite (SnO) and cassiterite (SnO 2) were identified and their relative proportion estimated.

Abstract

International audience; We demonstrate that convenient thermal treatment of a specific sapphire vicinal surface can induce the formation of a fully two-dimensional (2D) ordered surface made of a periodic assembly of (006) facets. The similarity between the resulting surface topography and patterns represented in the "Hexagon series" of paintings by Vasarely is really striking! We thus propose to call these surfaces as "nanoscaled Vasarely surfaces". We also show that the self-organisation process, which is driven by the minimisation of the free energy of a closed system, results in a quasi-linear isothermal growth of the facets' surface area over time.

Abstract

International audience; A new quinary multi-principal element alloy Ti0.30V0.25Zr0.10Nb0.25Ta0.10 was prepared by high temperature melting technique and the physicochemical as well as hydrogen sorption properties have been determined. The as-cast alloy crystallizes into a single-phase bcc lattice and can very quickly absorb hydrogen at room temperature forming a fcc hydride phase with capacity of 2 H/M (2.5 wt.%). The absorption/desorption of hydrogen is reversible and occurs within one step, as proven by in situ neutron diffraction for the desorption reaction. The capacity is slightly fading during the first 10 cycles and then stabilizes at around 2.2 wt.% for the next 10 cycles.

Abstract

Intercalation of alkali atoms within the lamellar transition metal dichalcogenides is a possible route toward a new generation of batteries. It is also a way to induce structural phase transitions authorizing the realization of optical and electrical switches in this class of materials. The process of intercalation has been mostly studied in three-dimensional dichalcogenide films. Here, we address the case of a single-layer of molybdenum disulfide (MoS$_2$), deposited on a gold substrate, and intercalated with cesium (Cs) in ultra-clean conditions (ultrahigh vacuum). We show that intercalation decouples MoS$_2$ from its substrate. We reveal electron transfer from Cs to MoS$_2$, relative changes in the energy of the valence band maxima, and electronic disorder induced by structural disorder in the intercalated Cs layer. Besides, we find an abnormal lattice expansion of MoS$_2$, which we relate to immediate vicinity of Cs. Intercalation is thermally activated, and so is the reverse process of de-intercalation. Our work opens the route to a microscopic understanding of a process of relevance in several possible future technologies, and shows a way to manipulate the properties of two-dimensional dichalcogenides by "under-cover" functionalization., Comment: 6 figures

Abstract

At the kilo- to megabase pair scales, eukaryotic genomes are partitioned into self-interacting modules or topologically associated domains (TADs) that associate to form nuclear compartments. Here, we combine high-content super-resolution microscopies with state-of-the-art DNA-labeling methods to reveal the variability in the multiscale organization of the Drosophila genome. We find that association frequencies within TADs and between TAD borders are below ~10%, independently of TAD size, epigenetic state, or cell type. Critically, despite this large heterogeneity, we are able to visualize nanometer-sized epigenetic domains at the single-cell level. In addition, absolute contact frequencies within and between TADs are to a large extent defined by genomic distance, higher-order chromosome architecture, and epigenetic identity. We propose that TADs and compartments are organized by multiple, small-frequency, yet specific interactions that are regulated by epigenetics and transcriptional state., Eukaryotic genomes are partitioned into self-interacting modules or topologically associated domains (TADs) that exist at the kilo-megabase scale. Abstract

International audience; Prelithiation of silicon/graphite-based composite anodes is a promising strategy to limit Li-ion battery capacity loss over long cycling. We report on the spontaneous-corrosion-driven-lithiation (SCDL) of lithium metal on the anode surface of a-Si/c-FeSi$_2$/graphite//LiNi$_{0·6}$Mn$_{0·2}$Co$_{0·2}$O$^2$ cells, and compare it to electrochemically-driven-lithiation (EDL). High cyclability performances are achieved, i.e. more than 1760 cycles at 100% depth-of-discharge with less than 11% capacity loss at C/2. To quantify the fate of the lithium supply over prolonged cycling and follow in situ the benefits of prelithiation, we perform simultaneous operando synchrotron wide-angle and small-angle X-ray scattering (WAXS/SAXS). By accessing both the graphite lithiation process (WAXS) and the nanostructural variations of the silicon phase (SAXS), we obtain a complete picture of the lithium repartition during lithiation/delithiation of the prelithiated anode. Results indicate that the additional lithium is stored in the silicon, which acts as a reservoir compensating for lithium trapping. The silicon phase therefore plays an essential role but without the detrimental effect of large volume variations. By design, SCDL leads to a more heterogeneous prelithiation compared to EDL given that only the electrode area directly in contact with the lithium metal is initially prelithiated. However, we show that during cycling the additional lithium is redistributed throughout the electrode by migrating into the silicon and, finally, the two methods yield equivalent systems, with similar electrode morphologies and a much increased cell performance over cycling. Prelithiation could therefore be used for the production of high energy density and highly stable Li-ion batteries, implementing the easily up-scalable SCDL process.

Abstract

Background: Although native to North America, the invasion of the aphid-like grape phylloxera Daktulosphaira vitifoliae across the globe altered the course of grape cultivation. For the past 150 years, viticulture relied on grafting-resistant North American Vitis species as rootstocks, thereby limiting genetic stocks tolerant to other stressors such as pathogens and climate change. Limited understanding of the insect genetics resulted in successive outbreaks across the globe when rootstocks failed. Here we report the 294-Mb genome of D. vitifoliae as a basic tool to understand host plant manipulation, nutritional endosymbiosis, and enhance global viticulture. Results: Using a combination of genome, RNA, and population resequencing, we found grape phylloxera showed high duplication rates since its common ancestor with aphids, but similarity in most metabolic genes, despite lacking obligate nutritional symbioses and feeding from parenchyma. Similarly, no enrichment occurred in development genes in relation to viviparity. However, phylloxera evolved > 2700 unique genes that resemble putative effectors and are active during feeding. Population sequencing revealed the global invasion began from the upper Mississippi River in North America, spread to Europe and from there to the rest of the world. Conclusions: The grape phylloxera genome reveals genetic architecture relative to the evolution of nutritional endosymbiosis, viviparity, and herbivory. The extraordinary expansion in effector genes also suggests novel adaptations to plant feeding and how insects induce complex plant phenotypes, for instance galls.
