Abstract

The oak lace bug (OLB) Corythucha arcuata (Say, 1832) is an invasive alien species (IAS) that potentially could have many negative impacts on European oak health. Certain measures can be applied to counteract these effects. However, these measures may not be acceptable for forest managers or other stakeholder groups, such as private forest owners, environmental NGOs or the general public. Thereby, we set out to study the perception and knowledge of foresters and other stakeholders on the health status of European oak forests affected by oak lace bug and to investigate what forest health management measures would be acceptable to these target groups. An online survey questionnaire was designed and distributed via social networks, as well as professional networks via e-mails. The survey questionnaire was completed by 2084 respondents from nine European countries: Austria, Croatia, Belgium, France, Hungary, Italy, Romania, Serbia and Slovenia. Even though only a little over 60% of respondents reported they had noticed the discolouration of oak leaves caused by OLB, almost all (93%) considered it to be a problem. As respondents come from a country where C. arcuata is widespread and established, people’s general knowledge and awareness of OLB began to increase. The survey revealed that foresters thought that the insect affected photosynthesis, acorn crop and the aesthetics of the trees, but cannot cause death of trees. However, they assume that the value of the wood would decrease (this fact is also supported by the respondents who are connected to an environmental NGO), but that OLB does not affect property value. However, forest owners claim that the value of the property can be affected and that people would avoid entering the forest. In terms of potential control methods, respondents preferred biological or mechanical measures over chemical ones. We consider this study to be a good basis for further research on the topic of perception, knowledge and attitudes related to OLB since we can expect that the IAS, such as OLB, will certainly spread to European countries that were not included in this survey.

International audience; The objective of this study was to compare the enteric methane emission and milk production parameters in cows in mid to late lactation allowed either a 6-h restricted grazing period as a supplement to one of 2 partial mixed rations (PMR), or receiving a basal PMR in confinement without grazing (PMR-C). The 2 partial mixed rations were one basal (PMR-G) and one with increased inclusion of maize silage (PMR(CS)-G) (42.8 vs 55.4% maize silage). A balanced crossover design with 3 treatments and 3 successive 4-week treatment periods using 24 Holstein-Friesian cows was used. The cows were habituated to the basal PMR and consecutively to 6-h grazing during respectively 3 and 2 weeks preceding the experiment, as they were previously used to confinement. Fresh grass on pasture, water and PMR in the barn were provided ad libitum. Strip grazing was used to provide a daily fresh strip of palatable grass to motivate cows to graze. Cow data including milk yield, concentrate intake and PMR intake were recorded automatically. Fresh grass intake was estimated using the n-alkanes technique by providing C32 through oral boluses for 10 consecutive days. Emissions were measured using 2 GreenFeeds (one in the barn and one on pasture) throughout the day. Analysis was done using a linear mixed model with treatment, period and days in milk as fixed effects and cow as random effect. Results showed that dry matter intake was higher in confined cows (PMR-C) than grazing cows fed the same PMR (PMR-G), but not different from grazing cows fed a PMR with higher inclusion of maize silage (PMR(CS)-G). A significant reduction of absolute methane production (−12%) (P < 0.001) was found in the grazing treatments compared with non-grazing cows. Methane yield (−7% and −10% g/kg dry matter intake (DMI)) and intensity (−8% and −12% g/kg fat and protein corrected milk (FPCM)) were significantly lower (P < 0.001 and P = 0.003 resp.) in the grazing treatments. FPCM was lower in PMR-G compared with confined cows (PMR-C), but not different from PMR(CS)-G. NDF content of the different rations was 341, 378 and 365 g/kg DMI for PMR-C, PMR-G and PMR(CS)-G respectively, and net energy of lactation (NEL) was 6.4, 6.8 and 6.4 MJ/kg DMI respectively. We concluded that grazing combined with an increased maize silage inclusion in the barn PMR (i.e., PMR(CS)-G) resulted in the best trade-off between milk production and methane reduction in cows in mid and late lactation.

Abstract

International audience; Peptide marker identification is an important step in development of a mass spectrometry method for multiple allergen detection, since specificity, robustness and sensitivity of the overall analytical method will depend on the reliability of the proteotypic peptides. As part of the development of a multi-analyte reference method, discovery analysis of two incurred food matrices has been undertaken to select the most reliable peptide markers. Six allergenic ingredients (milk, egg, peanut, soybean, hazelnut, and almond) were incurred into either chocolate or broth powder matrix. Different conditions of protein extraction and purification were tested and the tryptic peptide pools were analysed by untargeted high resolution tandem mass spectrometry and the resulting fragmentation spectra were processed via a commercial software for sequence identification. The analysis performed on incurred foods provides both a prototype effective and straightforward sample preparation protocol and delivers reliable peptides to be included in a standardized selected reaction monitoring method.

Abstract

The widespread use of High-Throughput Sequencing (HTS) for detection of plant viruses and sequencing of plant virus genomes has led to the generation of large amounts of data and of bioinformatics challenges to process them. Many bioinformatics pipelines for virus detection are available, making the choice of a suitable one difficult. A robust benchmarking is needed for the unbiased comparison of the pipelines, but there is currently a lack of reference datasets that could be used for this purpose. We present 7 semi-artificial datasets composed of real RNA-seq datasets from virus-infected plants spiked with artificial virus reads. Each dataset addresses challenges that could prevent virus detection. We also present 3 real datasets showing a challenging virus composition as well as 8 completely artificial datasets to test haplotype reconstruction software. With these datasets that address several diagnostic challenges, we hope to encourage virologists, diagnosticians and bioinformaticians to evaluate and benchmark their pipeline(s).

Abstract

In this paper, for the first time, the in-to-out-body path loss between a capsule antenna placed inside the cows’ rumen and a distant gateway was characterized at 868 MHz. Measurements were conducted on five different fistulated cows in a dairy barn. The in-body antenna gain was then de-embedded from the wireless channel. The difference between free space measurements and in-to-out-body path loss assessment was used to quantity the path loss increase due to the cows’ body. Results have shown an increase of the path loss on average (all cows) by 50.6 dB, with a variation between 43.7 and 55.3 dB. The obtained results were used to calculate the range of a LoRa (Long range) based network accounting for the antenna channel. With an input transmit power of 14 dBm, ranges up to 175 m in indoor and 364 m in outdoor were obtained depending on the used bit rate.

Abstract

In this Letter, for the first time, the in-to-out-body path loss between an antenna placed inside the cows' rumen and a distant gateway was characterised at 433 MHz. Measurements were conducted on seven different fistulated cows using a signal generator and a spectrum analyser. Subsequent measurement of the antenna in free space was used to quantify the path loss increase due to the cow body. Results have shown an increase of the path loss by 45.5 dB on average (all cows), with a variation between 39.7 and 51.1 dB. Also, the measured path loss values as a function of the transmitter-receiver distance in a dairy barn were well fitted by a log-normal path loss model. The obtained models were used to calculate the range of a LoRa (Long range) based network. Ranges up to 100 m were obtained depending on the used transmit power and bit rate.
