Neuropathic pain is a debilitating chronic condition provoked by a lesion in the nervous system and it induces functional alterations to the noradrenergic locus coeruleus (LC), affecting distinct dimensions of pain, like sensorial hypersensitivity, pain-induced depression, and anxiety. However, the neurobiological changes induced by nerve damage in the LC remain unclear. Here, we analyzed excitatory and inhibitory inputs to the LC, as well as the possible damage that noradrenergic neurons suffer after the induction of neuropathic pain through chronic constriction injury (CCI). Neuropathic pain was induced in male Sprague-Dawley rats, and the expression of the vesicular glutamate transporter 1 or 2 (VGLUT1 or VGLUT2), vesicular GABA transporter (VGAT), and cleaved caspase-3 (CC3) was analyzed by immunofluorescence 7 (CCI7d) or 28 days after the original lesion (CCI28d). While no significant differences in the density of VGLUT1 puncta were evident, CCI7d induced a significant increase in the perisomatic VGLUT2/VGAT ratio relative to Sham-operated and CCI28d animals. By contrast, when the entire region of LC is evaluated, there was a significant reduction in the density of VGLUT2 puncta in CCI28d animals, without changes in VGLUT2/VGAT ratio relative to the CCI7d animals. Additionally, changes in the noradrenergic soma size, and a lower density of mitochondria and lysosomes were evident in CCI28d animals. Interestingly, enhanced expression of the apoptotic marker CC3 was also evident in the CCI28d rats, mainly co-localizing with glial fibrillary acidic protein but not with any neuronal or noradrenergic marker. Overall, short-term pain appears to lead to an increase of markers of excitatory synapses in the perisomatic region of noradrenergic cells in the LC, an effect that is lost after long-term pain, which appears to activate apoptosis.

[EN] First-episode psychosis (FEP) patients show structural brain abnormalities at the first episode. Whether the cortical changes that follow a FEP are progressive and whether age at onset modulates these changes remains unclear. This is a multicenter MRI study in a deeply phenotyped sample of 74 FEP patients with a wide age range at onset (15-35 years) and 64 neurotypical healthy controls (HC). All participants underwent two MRI scans with a 2-year follow-up interval. We computed the longitudinal percentage of change (PC) for cortical thickness (CT), surface area (CSA) and volume (CV) for frontal, temporal, parietal and occipital lobes. We used general linear models to assess group differences in PC as a function of age at FEP. We conducted post-hoc analyses for metrics where PC differed as a function of age at onset. We found a significant age-by-diagnosis interaction effect for PC of temporal lobe CT (d=0.54; p=002). In a post-hoc-analysis, adolescent-onset (≤19y) FEP showed more severe longitudinal cortical thinning in the temporal lobe than adolescent HC. We did not find this difference in adult-onset FEP compared to adult HC. Our study suggests that, in individuals with psychosis, CT changes that follow the FEP are dependent on the age at first episode, with those with an earlier onset showing more pronounced cortical thinning in the temporal lobe. [ES] El objeto de este trabajo final de grado es realizar un análisis desde el punto de vista estructural de la catedral gótica Notre Dame de Paris. Una vez realizado este análisis estructural y conocido suficientemente el comportamiento de los elementos con capacidad portante, se estudiarán distintas soluciones y se realizarán propuestas con otros materiales. El uso de materiales con propiedades mecánicas completamente diferentes a la piedra permitirá comparar cómo pueden resolverse las distintas soluciones estructurales que conforman la catedral e incluso realizar una propuesta de sustitución de las bóvedas afectadas en el reciente incendio, pudiendo limitarse la intervención a una mera sustitución de los elementos afectados con intención conservacionista o a una propuesta estructural nueva., [EN] The aim of this Final Degree Project is to perform an analysis of the gothic cathedral Notre Dame de Paris from the structural point of view. After performing the structural analysis and knowing sufficiently the behavior of the elements with bearing capacity, different solutions will be studied, and proposals made with other materials. The use of materials with mechanical properties completely different from the stone will allow comparing how the different structural solutions of the cathedral can be solved and even make a proposal to replace the vaults affected in the recent fire, the intervention being limited to a mere substitution of the affected elements with conservation intent or a new structural proposal.

La artritis reumatoide es una enfermedad inflamatoria y autoinmune crónica que afecta principalmente a las articulaciones. Se produce una inflamación articular que daña las diferentes estructuras deformándolas, llegando a incapacitar al paciente. Se caracteriza por inflamación de la membrana sinovial con infiltración de células inmunes, degradación del cartílago, con muerte de condrocitos, y destrucción del hueso por desregulación del mecanismo de degradación-reparación. La activación aberrante de las células presentadoras de antígeno frente a antígenos propios modificados inicia una cascada inflamatoria que lleva a la activación linfocitaria contra este antígeno, la proliferación de linfocitos B y la producción de autoanticuerpos, y a la liberación de mediadores proinflamatorios tanto en el foco inflamatorio como en la circulación general. Esto lleva a la hiperactivación osteoclástica e inhibición osteoblástica produciendo la degradación del hueso subcondral. Esta respuesta inflamatoria está mediada por linfocitos Th1, Th2 y Th17, entre otros. No obstante, para compensar este aumento de linfocitos proinflamatorios, el organismo activa la vía resolutiva a través de los linfocitos T reguladores FOXP3+ que fracasan en su intento por paliar la inflamación. Así, el proceso se retroalimenta positivamente hasta la degradación completa de las articulaciones. Hoy en día existen muchos tratamientos para combatir la enfermedad, pero se centran exclusivamente en frenar su avance y paliar los síntomas dolorosos. Además, no se ha conseguido revertir la degradación ya producida. En este sentido, es importante continuar investigando tanto la enfermedad como posibles vías y dianas terapéuticas. Así, en los últimos años han aparecido diversas terapias biológicas capaces de mejorar la enfermedad, pero no son efectivas en todos los casos o pueden acabar fracasando por tolerancia o por incompatibilidad con nuestro propio sistema inmune. La proteína relacionada con la paratohormona (PTHrP) y sus derivados peptídicos han sido estudiados recientemente como tratamiento para las enfermedades óseas. Uno de ellos, osteostatina (PTHrP(107-111)), ha mostrado propiedades antiinflamatorias y antisenescentes en osteoblastos y también antiosteoclastogénicas. El objetivo del presente estudio ha sido profundizar en el conocimiento farmacológico de este péptido, determinando sus efectos en un modelo animal de artritis reumatoide y en células humanas. Se empleó el modelo de artritis inducida por colágeno (CIA) en ratones DBA/1. Después del inicio de la artritis, se administró osteostatina a 80 µg/kg o 120 µg/kg por vía subcutánea durante 13 días. A día 14 se sacrificaron los animales y se les extrajeron los ganglios linfáticos para la caracterización de las poblaciones linfocitarias por citometría de flujo y para ensayos de proliferación celular. Unas patas fueron seccionadas y homogenizadas para determinar mediadores, otras se emplearon en el estudio histológico de la articulación y otras para determinar la densidad ósea mediante rayos X. También se cuantificaron diversos mediadores en el suero de los ratones. Osteostatina fue capaz de reducir los signos clínicos de la artritis, así como los principales mediadores inflamatorios locales como IL-1β, TNFα e IL-6. Además, aumentó la principal citocina antiinflamatoria IL-10. Todo ello acorde con los resultados obtenidos con la citometría de flujo de los linfocitos de ganglio linfático donde se observó un aumento de la población FOXP3+ respecto a la RORγT+. Los cortes histológicos mostraron una reducción de la degradación del cartílago y de la destrucción ósea que se confirmó con el análisis de rayos X. Por otro lado, se separaron monocitos de sangre humana para su posterior diferenciación a osteoclastos y estudiar la capacidad de osteostatina de reducir este proceso. Se utilizaron las concentraciones de 100 nM, 250 nM y 500 nM. Tras 2 o 7 días de diferenciación, se lisaron las células y se midieron distintos genes implicados en la diferenciación de osteoclastos por RT-qPCR. También se separaron los linfocitos, se trataron con osteostatina a 100 nM y 500 nM y se caracterizaron las poblaciones por citometría de flujo. Se observó que osteostatina es capaz de bloquear la diferenciación osteoclástica a partir de monocitos humanos mediante la disminución de la expresión de genes clave como RANK, OSCAR y catepsina K, sobre todo en los primeros estadios de diferenciación. Además, el péptido provocó un aumento de los linfocitos FOXP3+ en comparación con los RORγT en los linfocitos extraídos de sangre humana. Así puede concluirse que osteostatina tiene un perfil inmunomodulador, antiinflamatorio y antirresortivo, demostrado tanto en CIA como en células humanas. Sería interesante seguir profundizando en el estudio de esta molécula para desarrollar posibles tratamientos en enfermedades inflamatorias crónicas articulares como es la artritis reumatoide.

In chronic inflammatory joint diseases, such as rheumatoid arthritis, there is an important bone loss. Parathyroid hormone-related protein (PTHrP) and related peptides have shown osteoinductive properties in bone regeneration models, but there are no data on inflammatory joint destruction. We have investigated whether the PTHrP (107-111) C-terminal peptide (osteostatin) could control the development of collagen-induced arthritis in mice. Administration of osteostatin (80 or 120 &mu, g/kg s.c.) after the onset of disease decreased the severity of arthritis as well as cartilage and bone degradation. This peptide reduced serum IgG2a levels as well as T cell activation, with the downregulation of ROR&gamma, t+CD4+ T cells and upregulation of FoxP3+CD8+ T cells in lymph nodes. The levels of key cytokines, such as interleukin(IL)-1&beta, IL-2, IL-6, IL-17, and tumor necrosis factor-&alpha, in mice paws were decreased by osteostatin treatment, whereas IL-10 was enhanced. Bone protection was related to reductions in receptor activator of nuclear factor-&kappa, B ligand, Dickkopf-related protein 1, and joint osteoclast area. Osteostatin improves arthritis and controls bone loss by inhibiting immune activation, pro-inflammatory cytokines, and osteoclastogenesis. Our results support the interest of osteostatin for the treatment of inflammatory joint conditions.

[EN] Nuclear factor (erythroid-derived 2)-like 2 (Nrf2) is a master regulator of cellular protective processes. Rheumatic diseases are chronic conditions characterized by inflammation, pain, tissue damage and limitations in function. Main examples are rheumatoid arthritis, systemic lupus erythematosus, osteoarthritis and osteoporosis. Their high prevalence constitutes a major health problem with an important social and economic impact. A wide range of evidence indicates that Nrf2 may control different mechanisms involved in the physiopathology of rheumatic conditions. Therefore, the appropriate expression and balance of Nrf2 is necessary for regulation of oxidative stress, inflammation, immune responses, and cartilage and bone metabolism. Numerous studies have demonstrated that Nrf2 deficiency aggravates the disease in experimental models while Nrf2 activation results in immunoregulatory and anti-inflammatory effects. These reports reinforce the increasing interest in the pharmacologic regulation of Nrf2 and its potential applications. Nevertheless, a majority of Nrf2 inducers are electrophilic molecules which may present off-target effects.
