The effects of filter paper strip (FPS) storage time and temperature on antibody detection are poorly understood despite widespread use in wildlife research. We collected sera and FPSs from 23 wild turkeys (Meleagris gallopavo) and 20 northern bobwhites (Colinus virginianus) experimentally infected with West Nile virus (WNV) to compare FPS storage methods with WNV plaque reduction neutralization test (PRNT) sensitivity. FPS storage methods included: immediate elution after drying, and storage at −20°C for 3 mo, −20°C for 6 mo, room temperature (RT) for 3 mo, and RT for 6 mo prior to elution. FPS eluates and sera were co-titrated to determine endpoint antibody titers, which were compared between FPS sera eluted immediately and sera, and among FPS eluates that underwent different storage conditions. Antibody titers were ~4-fold less in FPS sera eluted immediately versus sera, and dropped more frequently below PRNT detection threshold in northern bobwhites, which had ~10-fold lower serum antibody titers than wild turkeys. Antibody titers were lower in FPS samples stored at RT and for 6 mo. WNV serologic surveys may result in falsely low seroprevalence estimates if FPSs are stored at RT for ≥ 3 mo before elution. Abstract: Background: Measurements of human growth hormone (hGH) are a prerequisite for identifying a deficiency or excess. Our study is the first to investigate the reliability of a very sensitive assay for the quantification of GH in dried blood spots on filter paper. Objective: Validation of a commercially-available enzyme-linked immunoassay (ELISA) for measuring hGH from filter paper samples of dried blood. Methods: We used an assay system (ELISA, E022, Mediagnost) based on polyclonal rabbit antibodies. Its suitability is ascribable to its very high sensitivity (1.6ng/L) and virtual absence of interfering factors, excepting for a cross-reactivity with high pegvisomant concentrations. Results: hGH was found to be stable in dried blood spots on filter paper (No. 903, Whatman) over eight days at 37°C. Extraction of hGH from filter paper, in comparison to EDTA plasma, was 107% (SD 8.1%; n =6) over a range from 2.4 to 34.5μg/L. Linear regression analysis (n =119) showed a correlation of R 2 =0.97 for the hGH concentration in serum and on filter paper samples. Conclusion: Our findings demonstrate the reliability of measurements of hGH in dried blood spots on filter paper. The advantages of this method are the low sample volume and the easy transport, storage, and handling of samples. Mathematical models have been considered as a robust tool to support biological and medical studies of the coronavirus disease 2019 (COVID-19). This new disease is caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). This paper develop a within-host SARS-CoV-2 dynamics model with logistic growth for healthy epithelial cells, humoral (antibody) immune response and general SARS-CoV-2-target incidence rate. The model is incorporated with four mixed (distributed/discrete) time delays, delay in the formation of latent infected epithelial cells, delay in the formation of active infected epithelial cells, delay in the activation of latent infected epithelial cells, and maturation delay of new SARS-CoV-2 particles. The model is formulated as a system of delay differential equations (DDEs). We establish that the model's solutions are non-negative and ultimately bounded. We deduce that the model has three equilibria and their existence and stability are perfectly determined by two threshold parameters. We prove the global stability of the model's equilibria by utilizing the Lyapunov method and applying the LaSalle's invariance principle. To support and illustrate our theoretical findings we present numerical simulations for the model with a special form of the general incidence rate function. The effect of time delays on the SARS-CoV-2 dynamics is addressed. We observe that increasing time delays values can have the same impact as drug therapies in suppressing viral progression. Fast and precise diagnostic techniques are required for the treatment of many disorders. Biosensors are one of the diagnostic devices that are applicable in biological and medical sciences. Biosensors could be utilized to recognize biological molecules with high sensitivity. Biosensors are consisted of different components and have different types. Each type of biosensor is used in a particular field according to its specific features. Nanobodies are a novel class of antibodies with small size, high affinity, and specificity to their target. The unique properties of nanobodies make them appropriate tools for diagnostic applications. In this paper, we review biosensors, and their features and roles in medicine. During the last month of 2019, a new Coronavirus from China started to spread all around the world causing a pandemic emergency still ongoing. The outbreak made imperative the need for diagnostic and screening tests that could identify the current and past infection state of an individual. Occupational medicine is facing a very demanding challenge, as the pandemic set off the need to re-evaluate many aspects of workplace safety. A fundamental role has been played by tests used to diagnose COVID-19 and to isolate infected asymptomatic subjects, with a view to the viral evolution and the emerging variants. However, the need for the urgent set-up of new methods for assessing both new and past infections has resulted in a large number of methods, not always comparable with each other, in terms of laboratory techniques, viral antigens used for detection, and class of antibodies detected. These factors make it difficult to understand the serological test results and their possible application. In this paper, we reviewed the types of assays currently available, to address some key aspects that characterize each technique, and might have an impact on results interpretation. The fully synthetic humanized phage antibody library has the advantages including the minimized immunogenicity, which frequently happened in hybridoma cell-based antibody production. In this paper, using the constructed diverse complementarity determining region gene library and the germline gene as the backbone, we constructed eight single-chain antibody libraries and a combinatorial antibody library with a big capacity of 1.41 × 1010. M13EEA helper phage that was engineered from M13KO7 was applied to prepare phage antibody library. The eukaryotic expression of T-cell immune receptor with Ig and ITIM domain (TIGIT) antigen was used as a target antigen for screening. The screening of antigen-specific single-chain Fc-fused protein was performed through evaluation of binding affinity based on ELISA analysis. The IgG antibody was prepared with the screened single-chain protein. Finally, the CB3 antibody was screened out which exhibits the highest binding affinity with TIGIT with the Kd value of 8.155 × 10−10 M. Antibodies to the 65 kD isoform of glutamic acid decarboxylase (GAD65) have been associated with incident Type 2 Diabetes Mellitus, however results are inconsistent. To assess the association between GAD65 antibody positivity and incident Type 2 Diabetes Mellitus in a non-diabetic adult (≥18 years) population, in a systematic review and meta-analysis. A systematic literature search was conducted in Pubmed (MEDLINE) and Embase until January 14th, 2019. Included studies were 1) prospective studies on the association between GAD65 antibodies and incident Type 2 Diabetes Mellitus; 2) in a non-diabetic adult (≥18 years) population. To strengthen the review, unpublished data from 1302 Hoorn Study participants were included. Data extraction and quality assessment were performed independently by two observers. Ten studies were rated for methodological quality and seven were pooled using a random-effects meta-analysis, of which 2 strong, 2 moderate and 3 of low methodological quality. The pooled risk estimate of incident Type 2 Diabetes Mellitus for GAD65 antibody positivity, compared to GAD65 antibody negativity was 3.36 (95% CI: 1.9–5.9). This result was robust to sensitivity analyses. Heterogeneity between studies was significant with I2 statistic of 79% (p < 0.0001). However, excluding one study showed a decrease of I2 to 19% (p < 0.0001), explaining a large part of the heterogeneity. GAD65 antibody positivity was associated with an increased risk of future Type 2 Diabetes Mellitus in adults. • Ten papers on GAD65 antibodies and incident T2DM were identified from the literature. • In a meta-analysis, GAD65 antibody positivity was associated with >3 times increased risk of incident T2DM. • This finding was robust to sensitivity analyses. • One study explained a large part of the observed heterogeneity. Abstract Therapeutic monoclonal antibodies (mAbs) market is strongly contributing to the rising growth of the biotechnology industry. Despite the increasing number of inventions over time, a few therapeutic mAbs are currently marketed. This paper focuses on developing an emerging score to select/rank promising therapeutic mAbs patents, based on a hierarchical decision model using expert's opinion. Six attributes related to each factor concerning patent status, patent owner's profile and mAbs medical relevance were analyzed. The desirability levels of each attribute were also assessed. Our data shows the medical relevance factor as the most important, contributing 50% of the emerging score. Among the attributes, the most important under patent status was proper geographic coverage and wider patent scope; for organization's profile was the preexistence of approved drugs; and for medical relevance, the clinical phase performance. A group of 1053 patents related to therapeutic mAb were scored, and the most promising were concerning combination therapy using immune checkpoint inhibitors. The study has managerial implications related to patent portfolio management and patent valuation, and provides instructions to rank mAbs patents according to the emerging score defined by attribute's importance in order to improve the identification of future innovations pathways. Anaplasma Marginale, a rickettsial gram-negative bacterium, is responsible for causing bovine anaplasmosis which has widely spread among the different parts of the world causing death among cattle. It is mainly spread through the transmission of infected ticks. The manifestation of infected ticks could be controlled to a limited extent. However, subsequently, it is very much necessary for the cattle to develop strong immunity against A. marginale. It could be possible if a subunit vaccine is created against the rickettsial bacteria and introduced into the blood of the cattle for trials. In this paper, we have conducted a study to develop a multi-epitope subunit vaccine against A.marginale by using reverse vaccinology process. Five highly immunogenic outer membrane proteins of A. marginale were selected such as MSP1a, MSP2, Vir B10, OMP1 and OMP11. These proteins bind to the MHC class I molecules of the epithelial cells of mammals and MHC class II molecules of the antigen-presenting cells such as B-cells. The MHC class I molecules present antigens to the cytotoxic t-lymphocytes whereas the MHC class II molecules present antigens to the helper t-lymphocytes thus, initiating a strong immune response among the bovine species. We constructed three vaccine sequences from which two vaccine constructs V2 and V3, were found to be stable with high solubility and negative allergenicity. We also performed molecular docking of the two refined vaccine constructs with Rp-105 (30% resemblance in sequence to TLR4) and TLR-9. The vaccines interacted with the receptors with high binding energy. The best-designed vaccine construct was then reverse transcribed and adapted for the host E.coli K12 strain. The reverse transcribed sequence was later inserted into the pET28a (+) vector for the cloning and expression of the vaccine. The study could be a better initiative for in vitro and in vivo practice in developing an effective vaccine against A. marginale. • The five Outer surface proteins- MSP1a, MSP2, Vir B10, OMP1, and OMP11 were the most antigenic. • The proposed epitopes displayed high antigenic scores, high conservancy and non allergenicity. • Both the modeled vaccine V2 and V3 interacted with the bovine receptors rp-105 (TLR4) and TLR9 with high binding energy. • The designed construct, V3 was adapted for E. coli (K12) for cloning and expression. • Sequence of vaccine constructs showed no similarity with human proteins. In presented paper we have developed new system for cancer theranostics based on vascular endothelial growth factor (VEGF) targeted magnetic nanoparticles. Conjugation of anti-VEGF antibodies with bovine serum albumin coated PEGylated magnetic nanoparticles allows for improved binding with murine breast adenocarcinoma 4T1 cell line and facilitates doxorubicin delivery to tumor cells. It was shown that intravenous injection of doxorubicin loaded VEGF targeted nanoparticles increases median survival rate of mice bearing 4T1 tumors up to 50%. On the other hand magnetic resonance imaging (MRI) of 4T1 tumors 24 h after intravenous injection showed accumulation of nanoparticles in tumors, thus allowing simultaneous cancer therapy and diagnostics. Abstract: This paper provides direct evidence that earthworm immune cells, coelomocytes, are exposed to bio-reactive quantities of metals within 3 days after dermal exposure, and that they respond by upregulating metallothionein (MT) and heat shock protein (HSP70, HSP72) expression. Indirect support for the hypothesis that coelomocytes are capable of trafficking metals was also obtained. Coelomocytes were expelled from adult individuals of Eisenia fetida after 3-day exposure either to metal ions (Zn, Cu, Pb, Cd) or to distilled water (controls) via filter papers. The number of coelomocytes was significantly decreased after Cu, Pb, or Cd treatment. Cytospin preparations of coelomocytes were subjected to immunoperoxidase staining with monoclonal antibodies against human heat shock proteins (HSP70 or HSP72), or rabbit polyclonal antibodies raised against metallothionein 2 (w-MT2) of Lumbricus rubellus. Applied antibodies detected the respective proteins of E. fetida and revealed that the expression of HSP70, HSP72 and w-MT2 proteins was either induced or significantly enhanced in coelomocytes from metal-exposed animals. In conclusion, stress protein expression in earthworm coelomocytes may be used as sensitive biomarkers of metal contaminations. Further experimentation is needed for quantitative analysis of kinetics of metal-induced stress protein expression in earthworm coelomocytes. [Copyright &y& Elsevier]

The theory that antibody (Ab) directed against the TSH receptor (TSHR) (TSHRAb) is the causal factor of Graves' disease seems unlikely. Corticosteroids have not had a curative effect on the hyperthyroidism of Graves' disease despite their effectiveness for other autoimmune diseases. Two kinds of TSHRAb, thyroid-stimulating Ab (TSAb) and thyroid-blocking Ab (TBAb), are known as causal factors of hyperthyroidism and hypothyroidism, respectively. Previously, we reported that TSAb may be thyroid stimulating animal IgG-like hormone and TBAb may be the precursor of TSAb. In this paper we suggested that TBAb (precursor) converts to TSAb (active form) via the action of the protease, colloid antigen 2 (CA2). We speculate that the conversion of TBAb to TSAb is controlled by two factors: the protease and an anti-protease Ab. When anti-protease Ab levels are high, the patient exhibits hypothyroidism due to the increase in TBAb levels caused by neutralization of the protease. When anti-protease Ab levels are negative, the patient's hypothyroidism disappeared by the negative serum TBAb due to increased protease. An immunoglobulin G (IgG) with enzyme activity is known as an abzyme, which may be an undeveloped form. IgG with hormone activity may be likewise called an abhormone, which could also be an undeveloped form. The tumor marker CEA is a known member of the IgG supergene family. Many ancestral versions of proteins may have been produced as an IgG form. Possible participation of colloid antigen 2 and abhormone for the etiology of Graves' disease is suggested. The adhesive interactions between molecular recognition units (such as specific peptides and antibodies) and antigens or other receptors on the surfaces of tumors are of great value in the design of targeted nanoparticles and drugs for the detection and treatment of specific cancers. In this paper, we present the results of a combined experimental and theoretical study of the adhesion between Luteinizing Hormone Releasing Hormone (LHRH)/Epherin type A2 (EphA2)–AFM coated tips and LHRH/EphA2 receptors that are overexpressed on the surfaces of human Triple Negative Breast Cancer (TNBC) tissues of different histological grades. Following a histochemical and immuno-histological study of human tissue extracts, the receptor overexpression, and their distributions are characterized using Immunohistochemistry (IHC), Immunofluorescence (IF), and a combination of fluorescence microscopy and confocal microscopy. The adhesion forces between LHRH or EphA2 and human TNBC breast tissues are measured using force microscopy techniques that account for the potential effects of capillary forces due to the presence of water vapor. The corresponding adhesion energies are also determined using adhesion theory. The pull off forces and adhesion energies associated with higher grades of TNBC are shown to be greater than those associated with normal/non-tumorigenic human breast tissues, which were studied as controls. The observed increase in adhesion forces and adhesion energies are also correlated with the increasing incidence of LHRH/EphA2 receptors at higher grades of TNBC. The conformation and orientation of proteins immobilised on synthetic materials determine their ability to bind their antigens and thereby the sensitivity of the microarrays and biosensors employing them. Plasma immersion ion implantation (PIII) of polymers significantly increases both their wettability and protein binding capacity. This paper addresses the hypothesis that a PIII treated polymer surface modifies the native protein conformation less significantly than a more hydrophobic untreated surface and that the differences in surface properties also affect the protein orientation. To prove this, the orientation and conformation of rat anti-mouse CD34 antibody immobilized on untreated and PIII treated polycarbonate (PC) were investigated using ToF-SIMS and FTIR-ATR spectroscopy. Analysis of the primary structure of anti-CD34 antibody and principal component analysis of ToF-SIMS data were applied to detect the difference in the orientation of the antibody attached to untreated and PIII treated PC. The difference in the antibody conformation was analysed using deconvolution of the Amide I peak (in FTIR-ATR spectra) and curve-fitting. It was found that compared to the PIII treated sample, the antibody immobilized on the untreated PC sample has a secondary structure with a lower fraction of β-sheets and a higher fraction of α-helices and disordered fragments. Also, it was found that anti-CD34 antibody has a higher tendency to occur in the inactive ‘tail-up’ orientation when immobilized on an untreated PC surface than on a PIII treated surface. This paper discusses potential avenues to abate the last bulwark to the HIV-1 eradication i.e. viral reservoirs in the body, consisting mainly of viral DNA hidden, in latent form, into long-lived memory CD4 + T-cells, resulting unaffected by either drugs or immune system. Activation of the innate immune system is necessary for the induction of adaptive immune responses against invading pathogens. In part, this is achieved by recognition of molecules associated with infection by a plethora of pattern recognition receptors comprising Toll-like receptors (TLRs) which are express on numerous cells of immune system such as, dendritic cells and B cells, and are activated by some TLR ligands. TLR-9, localized in endosomal compartment, specifically recognizes unmethylated oligonucleotide sequences containing CpG motifs which are particularly abundant in microbial genome, including HIV-1 DNA. Naïve B cells are activated following antigen binding to the B cell receptor (BCR). The complex antigen/BCR, internalises and synergises with TLR leading to hyper activation of B cells and antibodies production. Cross-link of the BCR with TLR9, followed by upregulation of TLR-9, in response to self DNA-containing antigens, on DNA-specific B lymphocytes lead to the development of DNA-specific autoantibodies. This occur in systemic autoimmune disorders, such as, systemic lupus erythematosus (SLE). Although such DNA-specific B lymphocytes, are usually present in B cell repertoire at low frequency, they normally do not produce autoantibodies, they can be activated by antigen BCR ligation, with plasmacytoid dendritic cells (pDCs) providing helper signals. The hypothesis here derives by insight that stimulation of BCR, by exogenously added HIV-1 DNA, on potentially reactive HIV-1 DNA-specific B cells, from HIV-1 and/or HIV-1/SLE patients, co-cultured with autologous viral DNA-stimulated pDCs, could lead to desirable in vitro production of antibodies direct against HIV-1 DNA by involving BCR/TLR9 pathway. These antibodies, suitably delivered into infected or uninfected CD4 + T-cells, by a carrier, such as, peptide transduction domain of Tat, could selectively recognize viral nucleic acid, rich in CpG motifs, respect to host DNA, both in productively than latently infected T cells. Because HIV-1 DNA into latently infected cells is heavily methylated and deacetylated, co-treatment with a small molecule inhibitor of DNA methylation, such as 5-aza-2′-deoxycytidine (aza-Cdr), and histone deacetylase inhibitors (HDACi), they would favor higher accessibility by antibodies to viral DNA, which is harbored into human chromosomes. The in vitro production of anti-HIV-1 DNA antibodies and their in vivo reintroduction, could find future application as interesting strategy to pave the way to an eventual cure to decrease and/or eliminate viral reservoirs from the body, and providing possible therapeutic applications not only for HIV-1/AIDS, but also for other infectious diseases. The article focuses on papers presented at the October 2003 New England Society of Allergy Fall Meeting in Portsmouth, New Hampshire. The paper "Idiopathic Chronic Urticaria and Angioedema After Vasectomy," by Dr. David H Dreyfus suggests autoantibody production and autoreactive t-cells triggered by vasectomy trigger urticaria through epitope spreading from sperm antigens to other antigens shared with mast cells. The paper "Cow's Milk Protein Induced Anaphylaxis in a Six-Week-Old Male Without a Personal or Family History of Atopy," by Dr. Jordan Scott and Dr. Kelly Stone illustrates that food-induced anaphylaxis can occur even in early infancy despite a negative family history of atopy and a negative personal history of eczema.

The two-dimensional entropy image segmentation algorithm has a huge amount of computation problems. Based on immune adjustment mechanism, a new image segmentation algorithm is designed to solve the problem in this paper. In this method, vaccine theory is combined with entropy theory to propose the entropy vaccine operator. Entropy vaccine operator is selected as an antibody vaccine. Extract updated vaccine bank by using entropy calculation. In this way can make the formation of optimal segmentation threshold and the update of vaccine library with adaptive, and the select of vaccine more accuracy and more effective. The simulation results indicate that the searching speed of algorithm is accelerated and the adaptability is increased due to entropy genetic operator. Abstract: In this paper we explore the consequences of a heterogeneous immune response in individuals on the evolution of a rapidly mutating virus. We show that several features of the incidence and phylogenetic patterns typical of influenza A may be understood in this framework. In our model, limited diversity and rapid drift of the circulating viral strains result from the interplay of two interacting subpopulations with different types of immune response, narrow or broad, upon infection. The subpopulation with the narrow immune response acts as a reservoir where consecutive mutations escape immunity and can persist. Strains with a number of accumulated mutations escape immunity in the other subpopulation as well, causing larger epidemic peaks in the whole population, and reducing strain diversity. Overall, our model produces a modulation of epidemic peak heights and patterns of antigenic drift consistent with reported observations, suggesting an underlying mechanism for the evolutionary epidemiology of influenza, in particular, and other infectious diseases, more generally. The aim of this paper is to describe the structure, production and function of secretory immunoglobulin A (sIgA) as well as changes of its concentration caused by exercise of various intensity and duration. Immunoglobulin A is the main class of antibodies present in the body secreted fluids such as saliva, tears or mucus from the intestines. It is generally recognized that IgA, due to its dominance in the immune system of mucous membranes, is the first line of defence against harmful environmental factors. The secretion and composition of saliva depends on the activity of the sympathetic and parasympathetic nervous systems. Physical activity, stimulating the autonomous nervous system, may reduce the amount of saliva and/or inhibit its secretion. The relationship between physical activity and the suppression of the immune system is not fully understood, but it is known that moderate intensity exercise can improve immune defences, while extreme effort can reduce them by creating an increased risk of upper respiratory tract inflammation (URTI). In athletes, the lowest risk of upper tract infection was connected with the case of moderate intensity exercise. It is now believed that the relationship between exercise volume and the risk of URTI has the shape of the letter “J”. This means that both too little and too much physical activity may increase the risk of upper respiratory tract infection. Training optimization and correct balance between exercise and rest periods may reduce the risk of adverse changes in the immune system and decrease the frequency of URTI. [ABSTRACT FROM AUTHOR]

Abstract: Based on clonal selection principle and the immunodominance theory, a new immune clustering algorithm, Immunodomaince based Clonal Selection Clustering Algorithm (ICSCA) is proposed in this paper. Firstly, by introducing a new immunodomaince operator to Clonal Selection Algorithm (CSA), the gene of elites in antibody population can be extracted and generalized to ordinary antibodies so as to gain on-line priori knowledge and share information among individuals. Then, one iteration of Fuzzy C-means clustering algorithm (FCM) and adaptive updating mechanism of antibody population are utilized to improve the diversity of antibody population in order to speed up the convergence speed. The proposed method has been extensively compared with FCM, GA-clustering algorithm (GACA) and Clonal Selection Algorithm based FCM (CSAFCM) over a test suit of several real life data sets and synthetic data sets. Experimental results indicate the superiority of the ICSCA over FCM, GAFCM and CSAFCM on clustering accuracy and robustness. [Copyright &y& Elsevier]

In this paper, we synthesize magnetite-based nanocomposites modified by meso-2,3-dymercaptosuccin acid with biologically active immobilized components (normal human immunoglobulin (Ig) and 2-mercaptonycotin acid (2- MNC). Also, we investigate physical-and-chemical properties of magnetic suspensions based on them. We develop the method of chemical surface modification of single-domain magnetite with thiol groups. The obtained nanocomposite is biofunctionalized by normal human immoglobulin by using sulfo-SMCC heterofunctionalized reagent. We immobilize model biologically active compound 2-MNCs on the surface of magnetically sensitive nanocomposite by the reaction of thiols disulfide exchange using dipiridyl-disulfide. By employing XRT methods and Fourier infrared spectroscopy, we prove the presence of disulfide bridges and SH-groups on the modified surface, and determine the quantitative content of thiol groups. Abstract: This paper proposed an improved adaptive artificial immune system (IA-AIS) for complex optimization problems in continuous search space. In this IA-AIS optimization, several operators are improved or revised which aim at faster convergence speed and better optimal solution. Further speaking, cloning and reproduction of each offspring candidate antibody are proportional to the power of its parent affinity from the antigen; while mutation of each offspring candidate antibody is inversely exponentially determined by its parent affinity from the antigen. Also, suppression operator between antibodies is dynamically controlled according to their concentration. In other words, the suppression level is proportional to their Euclidian distance in continuous search space. The effectiveness of these improvements of operators is experimentally verified. Furthermore, comparative investigations are carried out between the proposed IA-AIS optimization and other optimization utilities. Finally a persuasive case about the proportional-integral-differential (PID) controller tuning demonstrates the potential searching capability and practical value of IA-AIS optimization. This paper presents an emergent pattern recognition approach based on the immune network theory and hierarchical clustering algorithms. The immune network allows its components to change and learn patterns by changing the strength of connections between individual components. The presented immune-network-based approach achieves emergent pattern recognition by dynamically generating an internal image for the input data patterns. The members (feature vectors for each data pattern) of the internal image are produced by an immune network model to form a network of antibody memory cells. To classify antibody memory cells to different data patterns, hierarchical clustering algorithms are used to create an antibody memory cell clustering. In addition, evaluation graphs and L method are used to determine the best number of clusters for the antibody memory cell clustering. The presented immune-network-based emergent pattern recognition (INEPR) algorithm can automatically generate an internal image mapping to the input data patterns without the need of specifying the number of patterns in advance. The INEPR algorithm has been tested using a benchmark civil structure. The test results show that the INEPR algorithm is able to recognize new structural damage patterns. [ABSTRACT FROM AUTHOR]

Evaluation of immune status of dogs, in different physiological, against two of the worst diseases of dogs, it is a necessity as long as contamination is a real nest of puppies. In principle, protective immunity correlates with high antibody titres, but any study that has such a topic, establish protective serum antibody levels to specific pathogens. A major difficulty in the comparative analysis of specific immune response obtained in different serological studies is the manner in which antibody titers were determined. Therefore, to facilitate further research on the same theme of this paper, it was decided to use a test semi quantitatively (dot-ELISA) commercially available, approved by many laboratories. The first physiological assessment was undertaken bitches immune response was non-pregnant and lactating female dogs. The dynamics of post-vaccination antibody in this category, vaccinated up to 12 months before serological evaluation, revealed progressive reduction of protective antibody titers to CPV and CDV. The highest antibody titers after vaccination were obtained from nonpregnant bitches and non-tested 1-3 months after manage their vaccine, 60% of those with titers over 1:650 and 1:160 over 20%. The second physiological condition of bitches who did the evaluation of immune response was pregnant bitches. Values post-vaccination antibodies to pregnant bitches, stud previously vaccinated were above the minimum threshold for CDV and CPV protective, 60% of pregnant with titers of over 1:160 and 40% between 1:80 and 1:160 titers. The third physiological state of bitches that did the evaluation of immune response was lactating bitches. Unlike dogs pregnant group, the group postvaccination antibody levels lactating bitches were at the limit of the minimum protective CDV (above 1:80) at 60% of test animals and 40% of bitches were in period lactanţie had valuesgreater than 1:160. If anti-CPV IgG titer percentage bitches with the lower limit of protection was only 20%, the remaining animals with titers of over 1:160. [ABSTRACT FROM AUTHOR]

In-vitro immunofluorescent assays/imaging are routinely used methods of detecting antigens. The ability to perform ocular angiography to study the choroidal and retinal vasculature in real time provides us with a unique opportunity to perform real time in-vivo immunofluorescent imaging. This unique combination of in-vivo immunofluorescent imaging and live imaging of choroidal and retinal circulation can help detect antigens of infective organisms in-vivo to diagnose causative infective aetiology in cases of choroiditis/retinitis. Abstract: Background: Latent autoimmune diabetes in adults (LADA) represents a subgroup of diabetes mellitus. LADA is characterised by adult-onset diabetes and circulating autoimmune antibodies. LADA patients may need a different therapeutic approach than the usual type 2 diabetes mellitus. When LADA is inadequately diagnosed as type 2 diabetes mellitus, LADA patients will mistakenly be exposed to a high dose of oral glucose lowering drugs and their possible side effects. Aim: To assess which clinical features predict the presence or absence of LADA in patients older than 25 years presenting with hyperglycemia. Methods: A structured Medline and Embase search was conducted. Titles and abstracts were screened using predetermined selection criteria. Critical appraisal was based on standardized validity criteria for diagnostic research. Results: One-hundred and eighty-four papers were retrieved of which after assessment of relevance and validity 2 studies remained for further analysis. One study reported a probability of LADA of 0.99 with one or two out of the following five clinical features: age at onset <50 years; acute symptoms; BMI<25kg/m2; a history of autoimmune disease; a family history positive for diabetes mellitus. The other study reported a probability of LADA of zero with none of the following clinical features and of 0.32 with one out of three: fasting blood glucose≥15mmol/l and/or HbA1c ≥10%; 10% reduction in body weight in the previous 3 months; BMI<25kg/m2. Conclusion: Further testing for LADA by measurement of autoimmune antibodies appears to be unnecessary in the absence of a specific set of clinical features. Before initiating therapy applying the above criteria may help to separate LADA from usual type 2 diabetes. [Copyright &y& Elsevier]

Abstract: This paper aims at developing novel bioactive fibrous mats for protein immobilization and for protein separation/purification. For this purpose, an amphiphilic triblock copolymer, biotinylated poly(ethylene glycol)-b-poly(l-lactide)-b-poly(l-lysine) was co-electrospun together with poly(l-lactide-co-glycolide) into ultrafine fibers ∼2μm in diameter, and a layer of blocking agent was coated on the fiber surfaces to block off possible non-specific binding of proteins. The biotin species retained their ability to specifically recognize and bind streptavidin, and the immobilized streptavidin could further combine with biotinylated antibodies, antigens and other biological moieties. Horseradish peroxidase-labeled streptavidin and fluorescein isothiocyanate-labeled goat globulin were used to detect the immobilizations of streptavidin and rabbit anti-goat IgG(H+L) via enzyme-linked immunoassay and confocal laser scanning microscope, respectively. The immobilized antigen was eluted from the fiber substrate with a glycine/HCl solution and the eluted antigen retained its bioactivity. Therefore, these biotin-carrying composite fibers have a variety of uses, including selective immobilization of functional proteins, antigen/antibody separation and purification, and vaccine preparation. Summary: This paper critically reviews the association of infection by Chlamydophila pneumoniae (C. pneumoniae) and Alzheimer''s disease (AD). The aging population has increased interest in finding the cause of AD, but studies have yielded contradictory results that are likely due to varying diagnostic tools and different uses of diagnostic tests. Knowledge of AD''s characteristics, risk factors, and hypothesized etiologies has expanded since Alois Alzheimer''s initial description of AD. Epidemiologic and projection studies provide incidence estimates of AD through a two-stage method: (1) primary diagnosis of dementia by cognitive testing such as Mini-Mental State Examination (MMSE), and (2) clinical diagnosis of AD through criteria such as National Institute of Neurological and Communicative Diseases and Stroke/Alzheimer''s Disease and Related Disorders Association (NINCDS–ADRDA). Cross-sectional studies yield prevalence estimates of infection by C. pneumoniae by detecting immunoglobulins through laboratory tests such as microimmunofluorescence (MIF). Studies examining the association of C. pneumoniae and AD are limited, but brain autopsy provides information about presence, proximity to areas associated with AD, and bacterial load. Standardization of diagnostic techniques would allow for better comparability of studies, but uncertainty about the best method of diagnosis of infection by C. pneumoniae and AD may call for revised or novel diagnostic tools.
