The refractive index points to the interplay between light and objects, which is rarely studied down to micronano scale. Herein, we demonstrated a conventional bright-field imaging technique to determine the local refractive index of single particles combined with a series of refractive index standard solutions. This intrinsic optical property is independent with the particle size and surface roughness with a single chemical component. Furthermore, we accurately tuned refractive index of homemade core-shell nanoparticles by adjusting the ratio of core-to-shell geometry. This simple and effective strategy reveals extensive applications in exploring, designing and optimizing the physical and optical characterizations of composite photonic crystals with high precision. It also indicates potentials in the field of reflective displays, optical identification, and encryption.

The refractive index (RI) is a fundamental physical property of materials. Although measurement of the RI of biological nanoparticles (BNPs) in aqueous media is of great importance to basic research and biomedical applications, it is hampered by their tiny size, large intrinsic heterogeneity, and weak scattering. Here, we report the development of a label-free technique that can determine the RI of individual viruses and small extracellular vesicles (sEVs) with high precision and an analysis rate up to 10 000 particles per minute. This was achieved via the combination of high-sensitivity light-scattering detection by nanoflow cytometry (nFCM) and the Mie theory calculation. With the measured RIs for T7 virions, T7 capsids, and sEVs, the concentrations of nucleic acid in viral particles and protein in the lumen of sEVs were estimated. Furthermore, building upon a simplified core-shell model, the RIs of sEVs ranging from 40 to 200 nm were obtained. By using these RIs, a statistically robust size distribution of sEVs was acquired in minutes with accuracy and resolution matched closely with those of cryo-TEM measurements. Our approach could become an important tool in the RI determination of single BNPs.

Silicon (Si) is promising as a field enhancement material because of its high abundance, low toxicity, and high refractive index. The field enhancement effect intensifies light-matter interactions, which improves photocatalysis, solar cell performance, and sensor sensitivity. To manufacture field enhancement materials on a production scale, the fabrication technique must be simple, cost-effective, fast, and highly reproducible and must produce a high enhancement factor (EF). Herein, we report on an economical and efficient fabrication method for a field enhancement substrate consisting of a two-dimensional Si wire array (2D-SiWA). Herein, a novel liquid crystal microarray (LCM) film with optical regulation ability is first constructed by combining liquid crystals (LC) and the highly ordered microporous structure of inverse opal photonic crystals (IOPhCs). The LCM films are fabricated by infiltrating LC molecules into the LC polymer with the structure of IOPhCs, and their properties are very different from those without the LC. Interestingly, the optical property of LCM films can be controlled by changing the orientation of LC molecules, which varies with the interfacial force. In combination with polarization images, spectral reflection peak, circular dichroism spectra, potential difference, and fluorescence images of LCM films, the mechanism of this change is investigated. It is found that the exposed basic group of single-stranded DNA is the key to the change of the optical property of LC microarrays. Meanwhile, the optical signals of LC microarrays based on the PhCs provide a novel LC signal mode for an LC sensing system (microspectral signal mode), and it can be recorded by a fiber-optic spectrometer, which is a great improvement on LC sensing signals. Therefore, the LC microarray sensing signal can be used for accurate analysis of targets by the change of the reflection peak intensity of PhCs. When the LC molecules are induced by different aptamers, the LC microarray sensing interface can be further used for the determination of different targets, such as cocaine and Hg2+. The research on LCM films is of significant value for the development of LC sensing technology and also shows great application prospects in biochemical sensing fields.

Terahertz (THz) spectroscopy is a powerful tool for unambiguously extracting complex-valued material properties (e.g., refractive index, conductivity, etc.) from a wide range of samples, with applications ranging from materials science to biology. However, extracting complex refractive indices from THz time-domain spectroscopy data can prove challenging, especially for multilayer samples. These challenges arise from the large number of transmission-reflection paths the THz pulse can take through the sample layers, leading to unwieldy strings of Fresnel coefficients. This issue has often been addressed using various approximations. However, these approximations are only applicable to specific classes of samples and can give erroneous results when misapplied. An alternative to this approach is to programmatically model all possible paths through the sample. The many paths through the sample layers can be modeled as a tree that branches at every point where the paths diverge, i.e., whenever the pulse can either be transmitted or reflected. This tree can then be used to generate expressions relating the unknown refractive index to the observed time domain data. Here, we provide a freely available open-source package implementing this method as both a MATLAB library and a corresponding graphical user interface, which can also be run without a MATLAB license (https://github.com/YaleTHz/nelly). We have tested this method for a range of samples and compared the results to commonly used approximations to demonstrate its accuracy and wide applicability. Our method consistently gives better agreement than common approximations.

Microlens arrays (MLAs) are acquiring a key role in the micro-optical system, which have been widely applied in the fields of imaging processing, light extraction, biochemical sensing, and display technology. Compared with solid MLAs, liquid MLAs have received extensive attention due to their natural smooth interface and adjustability. However, manufacturing tunable liquid MLAs with ideal structures is still a key challenge for current technologies. In this paper, a novel and simple optofluidic method is demonstrated, enabling the tunable focusing and high-quality imaging of liquid MLAs. Tunable droplets are fabricated and self-assembled into arrays as the MLAs, which can be easily adjusted to focus, form images, and display different focal lengths. Tuning of MLAs' focusing properties (range from 550 to 5370 μm) is demonstrated by changing the refractive index (RI) of the droplets with a fixed size of 200 μm, which can be changed by adjusting the flow rates of the two branch streams. Also, the corresponding numerical apertures of the MLAs range from 0.026 to 0.26. Furthermore, the MLAs' functionality for microparticle imaging applications is also illustrated. Combining the MLAs with a 4× objective, microparticle imaging is magnified two times, and the resolution has also been improved on the original basis. Besides, both the size and RI of the MLAs in an optofluidic chip can be further adjusted to detect samples at different positions. These MLAs have the merits of high optical performance, a simple fabrication procedure, easy integration, and good tunability. Thus, it shows promising opportunities for many applications, such as adaptive imaging and sensing.

Refereed/Peer-reviewed A multifunction, high-sensitivity, and temperature-compensated optical fiber DNA hybridization sensor combining surface plasmon resonance (SPR) and Mach–Zehnder interference (MZI) has been designed and implemented. We demonstrate, for the first time to our knowledge, the dual-parameter measurement of temperature and refractive index (RI) by simultaneously using SPR and MZI in a simple single-mode fiber (SMF)–no-core fiber (NCF)–SMF structure. The experimental results show RI sensitivities of 930 and 1899 nm/RIU and temperature sensitivities of 0.4 and −1.4 nm/°C for the MZI and SPR, respectively. We demonstrate a sensitivity matrix used to simultaneously detect both parameters, solving the problem of temperature interference of RI variation-based biosensors. In addition, the sensor can also distinguish biological binding events by detecting the localized RI changes at the fiber’s surface. We realize label-free sensing of DNA hybridization detection by immobilizing probe DNA (pDNA) onto the fiber as the probe to capture complementary DNA (cDNA). The experimental results show that the sensor can qualitatively detect cDNA after temperature compensation, and the limit of detection (LOD) of the sensor reaches 80 nM. The proposed sensor has advantages of high sensitivity, real time, low cost, temperature compensation, and low detection limit and is suitable for in situ monitoring, high-precision sensing of DNA molecules, and other related fields, such as gene diagnosis, kinship judgment, environmental monitoring, and so on.

Chip-scale SARS-CoV-2 testing was demonstrated using silicon nitride (Si3N4) nanoslot fluidic waveguides to detect a tagged oligonucleotide with a coronavirus DNA sequence. The slot waveguides were fabricated using complementary metal-oxide-semiconductor (CMOS) fabrication processes, including multiscale lithography and selective reactive ion etching (RIE), forming femtoliter fluidic channels. Finite difference method (FDM) simulation was used to calculate the optical field distribution of the waveguide mode when the waveguide sensor was excited by transverse electric (TE) and transverse magnetic (TM) polarized light. For the TE polarization, a strong optical field was created in the slot region and its field intensity was 14× stronger than the evanescent sensing field from the TM polarization. The nanoscale confinement of the optical sensing field significantly enhanced the light-analyte interaction and improved the optical sensitivity. The sensitivity enhancement was experimentally demonstrated by measuring the polarization-dependent fluorescence emission from the tagged oligonucleotide. The photonic chips consisting of femtoliter Si3N4 waveguides provide a low-cost and high throughput platform for real-time virus identification, which is critical for point-of-care (PoC) diagnostic applications.

Aluminum has recently attracted considerable interest as plasmonic material due to its unique optical properties, but most work has been limited to nanostructures. We report here novel SPR biosensing with aluminum thin-films using the standard Kretschmann configuration that has previously been dominated by gold films. Electron-beam physical vapor deposition (EBPVD)-prepared Al films oxidize in air to form a nanofilm of Al(2)O(3,) yielding robust stability for sensing applications in buffered solutions. FDTD simulations revealed a sharp plasmonic dip in the visible range that enables measurement of both angular shift and reflection intensity change at a fixed angle. Bulk and surface tests indicated that Al films exhibited superb sensitivity performance in both categories. Compared to Au, the Al/Al(2)O(3) layer showed a marked effect of suppressing non-specific binding from proteins in human serum. Further characterization indicated that Al film demonstrated a higher sensitivity and a wider working range than Au films when used for SPR imaging analysis. Combined with its economic and manufacturing benefits, the Al thin-film has the potential to become a highly advantageous plasmonic substrate to meet a wide range of biosensing needs in SPR configurations.

Determination of size and refractive index (RI) of dispersed unlabeled subwavelength particles is of growing interest in several fields, including biotechnology, wastewater monitoring, and nanobubble preparations. Conventionally, the size distribution of such samples is determined via the Brownian motion of the particles, but simultaneous determination of their RI remains challenging. This work demonstrates nanoparticle tracking analysis (NTA) in an off-axis digital holographic microscope (DHM) enabling determination of both particle size and RI of individual subwavelength particles from the combined information about size and optical phase shift. The potential of the method to separate particle populations is demonstrated by analyzing a mixture of three types of dielectric particles within a narrow size range, where conventional NTA methods based on Brownian motion alone would fail. Using this approach, the phase shift allowed individual populations of dielectric beads overlapping in either size or RI to be clearly distinguished and quantified with respect to these properties. The method was furthermore applied for analysis of surfactant-stabilized micro- and nanobubbles, with RI lower than that of water. Since bubbles induce a phase shift of opposite sign to that of solid particles, they were easily distinguished from similarly sized solid particles made up of undissolved surfactant. Surprisingly, the dependence of the phase shift on bubble size indicates that only those with 0.15-0.20 μm radius were individual bubbles, whereas larger bubbles were actually clusters of bubbles. This label-free means to quantify multiple parameters of suspended individual submicrometer particles offers a crucial complement to current characterization strategies, suggesting broad applicability for a wide range of nanoparticle systems.

Confocal and multiphoton optical imaging techniques have been powerful tools for evaluating the performance of and monitoring experiments within microfluidic devices, but this application suffers from two pitfalls. The first is that obtaining the necessary imaging contrast often requires the introduction of an optical label which can potentially change the behavior of the system. The emerging analytical technique stimulated Raman scattering (SRS) microscopy promises a solution, as it can rapidly measure 3D concentration maps based on vibrational spectra, label-free; however, when using any optical imaging technique, including SRS, there is an additional problem of optical aberration due to refractive index mismatch between the fluid and the device walls. New approaches such as 3D printing are extending the range of materials from which microfluidic devices can be fabricated; thus, the problem of aberration can be obviated simply by selecting a chip material that matches the refractive index of the desired fluid. To demonstrate complete chemical imaging of a geometrically complex device, we first use sacrificial molding of a freeform 3D printed template to create a round-channel, 3D helical micromixer in a low-refractive-index polymer. We then use SRS to image the mixing of aqueous glucose and salt solutions throughout the entire helix volume. This fabrication approach enables truly nonperturbative 3D chemical imaging with low aberration, and the concentration profiles measured within the device agree closely with numerical simulations.

Modifying material properties in simple macromolecules such as polyethylene (PE) is achieved by different connection modes of ethylene monomers resulting in a plurality of possible topologies-from highly linear to dendritic species. However, the challenge still lies within the experimental identification of the topology and conformation of the isolated macromolecules because of their low solubility, which demands methods with specific solvents and high operating temperatures. Additionally, a separation technique has to be coupled to different detection methods to meet the specific demands of the respective characterization goal. In this work, we report a quadruple-detector high temperature size exclusion chromatography (HT-SEC) system which contains online multiangle laser light scattering, dynamic light scattering, differential viscometry, and differential refractometry detectors. Quadruple-detector HT-SEC was successfully applied to explore the full range of physical parameters of various PE samples with different branching topologies ranging from highly linear macromolecules, polymers with moderate level of branching, to highly branched PEs with hyperbranched structure. This method is a useful tool not only to investigate molecular weight, mass distribution, and size but also to enable access to important factors which describe the conformation in dilute solution and branching density.

Refractive index-based sensors offer attractive characteristics as nondestructive and universal detectors for liquid chromatographic separations, but a small dynamic range and sensitivity to minor thermal perturbations limit the utility of commercial RI detectors for many potential applications, especially those requiring the use of gradient elutions. As such, RI detectors find use almost exclusively in sample abundant, isocratic separations when interfaced with high-performance liquid chromatography. Silicon photonic microring resonators are refractive index-sensitive optical devices that feature good sensitivity and tremendous dynamic range. The large dynamic range of microring resonators allows the sensors to function across a wide spectrum of refractive indices, such as that encountered when moving from an aqueous to organic mobile phase during a gradient elution, a key analytical advantage not supported in commercial RI detectors. Microrings are easily configured into sensor arrays, and chip-integrated control microrings enable real-time corrections of thermal drift. Thermal controls allow for analyses at any temperature and, in the absence of rigorous temperature control, obviates extended detector equilibration wait times. Herein, proof of concept isocratic and gradient elution separations were performed using well-characterized model analytes (e.g., caffeine, ibuprofen) in both neat buffer and more complex sample matrices. These experiments demonstrate the ability of microring arrays to perform isocratic and gradient elutions under ambient conditions, avoiding two major limitations of commercial RI-based detectors and maintaining comparable bulk RI sensitivity. Further benefit may be realized in the future through selective surface functionalization to impart degrees of postcolumn (bio)molecular specificity at the detection phase of a separation. The chip-based and microscale nature of microring resonators also make it an attractive potential detection technology that could be integrated within lab-on-a-chip and microfluidic separation devices.

We present a novel spectroscopic quantitative phase imaging technique with a wavelength swept-source, referred to as swept-source diffraction phase microscopy (ssDPM), for quantifying the optical dispersion of microscopic individual samples. Employing the swept-source and the principle of common-path interferometry, ssDPM measures the multispectral full-field quantitative phase imaging and spectroscopic microrefractometry of transparent microscopic samples in the visible spectrum with a wavelength range of 450-750 nm and a spectral resolution of less than 8 nm. With unprecedented precision and sensitivity, we demonstrate the quantitative spectroscopic microrefractometry of individual polystyrene beads, 30% bovine serum albumin solution, and healthy human red blood cells.

Background correction is a very important step that must be performed before peak detection or any quantification procedure. When successful, this step greatly simplifies such procedures and enhances the accuracy of quantification. In the past, much effort has been invested to correct drifting baseline in one-dimensional chromatography. In fast online comprehensive two-dimensional liquid chromatography (LC×LC) coupled with a diode array detector (DAD), the change in the refractive index (RI) of the mobile phase in very fast gradients causes extremely serious baseline disturbances. The method reported here is based on the use of various existing baseline correction methods of one-dimensional (1D) liquid chromatography to correct the two-dimensional (2D) background in LC×LC. When such methods are applied orthogonally to the second dimension (2D), background correction is dramatically improved. The method gives an almost zero mean background level and it provides better background correction than does simple subtraction of a blank. The properties imparted, oftentimes synergistically, by the different components of copolymers and blends account for the widespread use of these in a variety of industrial products. Most often, however, processing and end-use of these materials (especially copolymers) is optimized empirically, due to a lack of understanding of the physicochemical phase-space occupied by the macromolecules. Here, this shortcoming is addressed via a quintuple-detector size-exclusion chromatography (SEC) method consisting of multiangle static light scattering (MALS), quasi-elastic light scattering (QELS), differential viscometry (VISC), ultraviolet absorption spectroscopy (UV), and differential refractometry (DRI) coupled online to the separation method. Applying the SEC/MALS/QELS/VISC/UV/DRI method to the study of a poly(acrylamide-co-N,N-dimethylacrylamide) copolymer in which both monomer functionalities absorb in the same region of the UV spectrum, we demonstrate how to determine the chemical heterogeneity, molar mass averages and distribution, and solution conformation of the copolymer all in a single analysis. Additionally, through the various mutually independent conformational and architectural metrics provided by combining the five detectors, including the fractal dimension (derived from two different detector combinations), two different dimensionless size parameters, the chemical heterogeneity, and the persistence length, it is shown that the monomeric arrangement is more alternating than random at lower molar masses, thus causing the copolymer to adopt a more extended conformation in solution in this molar mass (M) regime. At high M, however, the copolymer is shown to be and to behave more like a random coil homopolymer, after passing through a 250 kg mol(-1)-broad region of intermediate chain flexibility. Thus, the combination of five detectors provides a unique means by which to determine absolute properties of the copolymer, solution-specific physical behavior, and the underlying chemical basis of the latter. The quintuple-detector method was also extended to the study of blends of polyacrylamide and poly(N,N-dimethylacrylamide) homopolymers to quantitate their molar mass, solution conformation, and chemical heterogeneity and to shed light on the breadth of the distributions of the component species. The method presented should be applicable to the study of copolymers and blends in which either one or both component moieties or polymers absorb in the UV region and can be implemented using not only SEC but other size-based separation methods as well.

We have used simultaneous quartz crystal microbalance-dissipation (QCM-D) monitoring and four-detector optical reflectometry to monitor in situ the structural transformation of intact vesicles to a lipid bilayer on a gold surface. The structural transformation of lipid vesicles to a bilayer was achieved by introducing a particular amphipathic, alpha-helical (AH) peptide. The combined experimental apparatus allows us to simultaneously follow the acoustic and optical property changes of the vesicle rupturing process upon interaction with AH peptides. While QCM-D and reflectometry have similar sensitivities in terms of mass and thickness resolution, there are unique advantages in operating these techniques simultaneously on the same substrate. These advantages permit us to (1) follow the complex interaction between AH peptides and intact vesicles with both acoustic and optical mass measurements, (2) calculate the amount of dynamically coupled water during the interaction between AH peptides and intact vesicles, (3) demonstrate that the unexpectedly large increase of both adsorbed mass and the film's energy dissipation is mainly caused by swelling of the vesicles during the binding interaction with AH peptides, and (4) permit us to understand the structural transformation from intact vesicles to a bilayer via the AH peptide interaction by monitoring viscoelastic properties, acoustic mass, optical mass, and thickness changes of both the binding and destabilization processes. From the deduced "hydration signature" we followed the complex transformation of lipid assemblies. On the basis of this information, a mechanism of this structural transformation is proposed that provides new insight into the process of vesicle fusion on solid substrates.

An evaporative light scattering detector (ELSD) coupled with HPLC was used for the first time to characterize membranes. Polydispersed PEG-200, 600, and 1000 were selected as probe molecules to study the dependence of membrane retention on molecular weight via a gradient eluted HPLC separation coupled to ELSD detection. The results show that HPLC/ELSD is a really general and powerful technique to study the nanofiltration (NF) process since it does not require any special properties for the solutes (chromospheres or fluorophores) and possesses the required sensitivity. Especially in solvent resistant NF (SRNF), where a wide range of organic solvents is used, the ELSD detector was not affected by the interaction between solvent and solutes, which is a critical issue compared to other more common detectors.

Whispering gallery mode (WGM) resonators are small, radially symmetric dielectrics that recirculate light through continuous total internal reflection. High-Q resonances are observed that shift in response to changes in surrounding refractive index, leading to many applications in label-free sensing. Surface binding measurements with WGM resonators have demonstrated competitive analytical detection metrics compared to other sensing schemes. Similar figures of merit for detecting bulk refractive index changes, however, have proven more challenging. This has limited their use in applications such as capillary electrophoresis (CE), where their compact footprint and refractive index sensitivity offers advantages in nondestructive, universal detection. Here we couple WGM detection with CE by introducing a modulation scheme to improve detection limits. Phase sensitive WGM (PS-WGM) detection is developed to monitor real-time shifts in the WGM spectrum due to changes in surrounding refractive index. We directly compare phase sensitive detection with spectral measurements normally used to track WGM shifts. We report an improvement in detection limits by almost 300-fold using the PS-WGM method. The integrated CE with PS-WGM approach is demonstrated by detecting the separation of a three-component mixture of cations (Na(+), Li(+), and K(+)).

One major goal in the surface plasmon resonance (SPR) technique is the reliable detection of small molecules as well as low analyte concentrations. This can be achieved by a viable signal amplification strategy. We therefore investigated optimal liposome characteristics for use as a signal enhancement system for SPR sensors, as liposomes excel not only at versatility but also at colloidal stability and ease of functionalization. These characteristics include the encapsulation of high refractive index markers, lipid composition, liposome size, and surface modifications to best match the requirements of the SPR system. Our studies of the binding of biotinylated liposomes to surface-immobilized streptavidin show that the refractive index of the encapsulant has a major influence on the SPR signal and outweighs the influence of the thin lipid bilayer. Thus, the signal amplification properties of liposomes can be adjusted to the respective needs of any analytical task by simply exchanging the encapsulant solution. In this work, a maximum enhancement factor of 23 was achieved by encapsulating a 500 mM sucrose solution. Dose-response studies with and without liposome enhancement revealed an improvement of the limit of detection from 10 nmol L(-1) to 320 pmol L(-1) streptavidin concentration with a much higher sensitivity of 3 mRIU per logarithmic unit of the concentration between 500 pmol L(-1) and 10 nmol L(-1).

We developed a novel miniaturized and multiplexed, on-capillary, refractive index (RI) detector using liquid core optical ring resonators (LCORRs) for future development of capillary electrophoresis (CE) devices. The LCORR employs a glass capillary with a diameter of approximately 100 mum and a wall thickness of a few micrometers. The circular cross section of the capillary forms a ring resonator along which the light circulates in the form of the whispering gallery modes (WGMs). The WGM has an evanescent field extending into the capillary core and responds to the RI change due to the analyte conducted in the capillary, thus permitting label-free measurement. The resonating nature of the WGM enables repetitive light-analyte interaction, significantly enhancing the LCORR sensitivity. This LCORR architecture achieves dual use of the capillary as a sensor head and a CE fluidic channel, allowing for integrated, multiplexed, and noninvasive on-capillary detection at any location along the capillary. In this work, we used electro-osmotic flow and glycerol as a model system to demonstrate the fluid transport capability of the LCORRs. In addition, we performed flow speed measurement on the LCORR to demonstrate its flow analysis capability. Finally, using the LCORR's label-free sensing mechanism, we accurately deduced the analyte concentration in real time at a given point on the capillary. A sensitivity of 20 nm/RIU (refractive index units) was observed, leading to an RI detection limit of 10-6 RIU. The LCORR marries photonic technology with microfluidics and enables rapid on-capillary sample analysis and flow profile monitoring. The investigation in this regard will open a door to novel high-throughput CE devices and lab-on-a-chip sensors in the future.

A simple, stable, ultrasensitive dual-capillary dual-bicell (DCDB) microinterferometic backscattering detection (MIBD) system was developed. In DCDB MIBD, a He-Ne laser beam passes through a half-wave plate onto the cross section of two capillaries, one for reference and another for sensing analyte. The position of the backscattered fringe from each capillary, which are in proximity or essentially identical thermal environments, was detected with matched bicell photodetectors. The configuration was found to effectively compensate for thermal drift, which is normally the major source of noise in refractive index (RI) detection systems. It is shown that passive environmental compensation leads to greatly enhanced signal in nanoscale refractometry preformed by MIBD. An order of magnitude improvement in detection limits over single channel configurations is possible. Performance reaches the 10(-9) RIU level for like solvents in the presence in extremely large thermally induced RI gradients. At this level of detectability, DCDB MIBD could facilitate nanoliter-volume, femtomole-level universal detection in applications ranging from mu-HPLC and on-chip CE to scanning microcalorimetry.

An integrated optical metal clad leaky waveguide (MCLW) sensor device has been developed for the detection of bacteria. This is more sensitive than waveguide sensors currently in use. The MCLW device has been fabricated to extend the evanescent field to provide significant light intensity over the entire volume of the bacteria bound on the chip surface within this field. This in turn increases the interaction of the light with the entire volume of the bacteria. MCLW devices have been used for detecting refractive index changes, scattering, and fluorescence from bacterial spores captured on an immobilized antibody. The detection limit of Bacillus subtilis var. niger bacterial spores using refractive index detection was 8 x10(4) spores/mL. The scattering intensity of the BG spores was found to be three times greater than the scattering intensity generated using surface plasmon resonance. The extended light propagation along the direction of flow for a few millimeters provides an effective interrogation approach to increase the area of detection to detect low concentrations down to 1 x 10(4) spores/mL. The sensor was then optimized by studying the key factors affecting sensor performance including changing the pH of the medium, type of antibody immobilization matrix, sensor surface regeneration approaches, and longevity of the sensor.

The physicochemical properties of nanosized colloidal drug carrier systems are of great influence on drug efficacy. Consequently, a broad spectrum of analytical techniques is applied for comprehensive drug carrier characterization. It is the primary objective of this paper to present asymmetrical flow field-flow fractionation (AF4), coupled online with multiangle light scattering detection, for the characterization of gelatin nanoparticles. Size and size distribution of drug-loaded and unloaded nanoparticles were determined, and data were correlated with results of state-of-the-art methods, such as scanning electron microscopy and photon correlation spectroscopy. Moreover, the AF4 fractionation of gelatin nanoparticulate carriers from a protein model drug is demonstrated for the first time, proposing a feasible way to assess the amount of loaded drug in situ without sample preparation. This hypothesis was set into practice by monitoring the drug loading of nanoparticles with oligonucleotide payloads. In this realm, various fractions of gelatin bulk material were analyzed via AF4 and size-exclusion high-pressure liquid chromatography. Mass distributions and high-molecular-weight fraction ratios of the gelatin samples varied, depending on the separation method applied. In general, the AF4 method demonstrated the ability to comprehensively characterize polymeric gelatin bulk material as well as drug-loaded and unloaded nanoparticles in terms of size, size distribution, molecular weight, and loading efficiency.

A simple method for converting local reflectivity changes measured in surface plasmon resonance (SPR) microscopy to effective adlayer thicknesses and absolute surface coverages of adsorbed species is presented. For a range of high-contrast angles near the SPR resonance where the local metal surface's reflectivity changes linearly with angle, the change in reflectivity at fixed angle is proportional to the change in effective refractive index (eta(eff)) near the surface. This change in eta(eff) can be converted to absolute adsorbate coverage using methods developed for quantitative SPR spectroscopy. A measurement of the change in reflectivity due to changes in refractive index of bulk solutions, i.e., percent reflectivity change per refractive index unit (RIU), is the only calibration required. Application of this method is demonstrated for protein adsorption onto protein/DNA arrays on gold from aqueous solution using an SPR microscope operating at 633 nm. A detection limit of 0.072% change in absolute reflectivity is found for simultaneous measurements of all 200 microm x 200 microm areas within the 24-mm(2) light beam with 1-s time averaging. This corresponds to a change in effective refractive index of 1.8 x 10(-5) and a detection limit for protein adsorption of 1.2 ng/cm(2) (approximately 0.5 pg in a 200-microm spot). The linear dynamic range is Deltaeta(eff) = approximately 0.011 RIU or approximately 720 ng/cm(2) of adsorbed protein. Using a nearby spot as a reference channel, one can correct for instrumental drift and changes in refractive index of the solutions in the flow cell.

Fourier transform (FT)-plasmon waveguide resonance (PWR) spectroscopy measures light reflectivity at a waveguide interface as the incident frequency and angle are scanned. Under conditions of total internal reflection, the reflected light intensity is attenuated when the incident frequency and angle satisfy conditions for exciting surface plasmon modes in the metal as well as guided modes within the waveguide. Expanding upon the concept of two-frequency surface plasmon resonance developed by Peterlinz and Georgiadis (Opt. Commun. 1996, 130, 260), the apparent index of refraction and the thickness of a waveguide can be measured precisely and simultaneously by FT-PWR with an average percent relative error of 0.4%. Measuring reflectivity for a range of frequencies extends the analysis to a wide variety of sample compositions and thicknesses since frequencies with the maximum attenuation can be selected to optimize the analysis. Additionally, the ability to measure reflectivity curves with both p- and s-polarized light provides anisotropic indices of refraction. FT-PWR is demonstrated using polystyrene waveguides of varying thickness, and the validity of FT-PWR measurements are verified by comparing the results to data from profilometry and atomic force microscopy (AFM).

A new method is described for the simultaneous determination of absorbance and refractive index of a sample medium. The method is based on measurement of the analyte-modulated modal power distribution (MPD) in a multimode waveguide. In turn, the MPD is quantified by the far-field spatial pattern and intensity of light, i.e., the Fraunhofer diffraction pattern (registered on a CCD camera), that emerges from a multimode optical fiber. Operationally, light that is sent down the fiber interacts with the surrounding analyte-containing medium by means of the evanescent wave at the fiber boundary. The light flux in the propagating beam and the internal reflection angles within the fiber are both affected by optical absorption connected with the analyte and by the refractive index of the analyte-containing medium. In turn, these angles are reflected in the angular divergence of the beam as it leaves the fiber. As a result, the Fraunhofer diffraction pattern of that beam yields two parameters that can, together, be used to deduce refractive index and absorbance. This MPD based detection offers important advantages over traditional evanescent-wave detection strategies which rely on recording only the total transmitted optical power or its lost fraction. First, simultaneous determination of sample refractive index and absorbance is possible at a single probe wavelength. Second, the sensitivity of refractometric and absorption measurements can be controlled simply, either by adjusting the distance between the end face of the fiber and the CCD detector or by monitoring selected modal groups at the fiber output. As a demonstration of these capabilities, several weakly absorbing solutions were examined, with refractive indices in the range from 1.3330 to 1.4553 and with absorption coefficients in the range 0-16 cm-1. The new detection strategy is likely to be important in applications in which sample coloration varies and when it is necessary to compensate for variations in the refractive index of a sample.

Pooled human milk oligosaccharides were fractionated by anion-exchange chromatography on AG 1-X2 and by an improved gel filtration procedure that allowed the separation of large oligosaccharides on Toyopearl HW 40 (S) and Bio-Gel P-6 columns, respectively. The analysis of the resulting nonderivatizated fractions by matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) revealed several neutral and acidic high-molecular-weight oligosaccharides. So far unknown acidic oligosaccharides containing up to 20 monomers were detected in a molecular mass range of 2094-3626 Da. Furthermore, neutral structures containing up to 35 monosaccharides were identified after fractionation on Toyopearl HW 40 (S) and subsequent P-6 fractionation, demonstrating the suitability of the applied method for the preparation of oligosaccharides in this high-molecular-mass range. The composition of the detected oligosaccharides was found to be the same as those previously identified in oligosaccharides of lower masses. However, an enormous structural heterogeneity was observed when acidic and neutral fractions were characterized by high-pH anion-exchange chromatography with pulsed amperometric detection (HPAEC-PAD). From our analysis we may conclude that each molecular mass identified by MALDI-MS corresponds to a variety of isomeric structures. The total number of oligosaccharides occurring in human milk may consequently be much higher than estimated before.

Herein, we describe the construction of a novel dual-pad referencing surface plasmon resonance (SPR) sensor utilizing electrolytic grafting of diazonium salts to individually functionalize two gold pads positioned in a single fluidic channel. Using a dove prism, a simple single axis optical train may be employed without compromising the analytical performance. Once functionalized, one pad is used as the analytical sensing pad for detection of molecular interactions while the other serves as the reference pad, compensating for background refractive index fluctuations. The reference pad effectively compensates bulk refractive index changes and temperature variations as well as other nonspecific effects. The sensor was applied to calibration of a pH-responsive polymer layer in the presence of bulk refractive index and temperature variations. Monitoring selective attachment of a protein is also demonstrated. To our knowledge, this is the first implementation of in-channel referencing SPR sensor utilizing diazonium salt-based surface chemistry.

Herein, a nanoporous alumina was fabricated to use as a mold in transforming nanopillar structures onto a thin film polymer by thermal nanoimprint lithography (NIL). The size of the pores was successfully controlled by varying the applied voltages and etching time. These nanoporous structures were transferred to the Cyclo-olefin polymer (COP) film surface from the porous mold by a thermal nanoimprinting process. A plasmonic substrate was fabricated by sputtering a thin layer of gold onto this nanopillar polymer structure, and the refractive index response in a variety of media was evaluated. Finally, the biosensing capacity of this novel plasmonic substrate was verified by analysis of Human immunoglobulin and achieved a minimum detection limit of 1.0 ng/mL. With the advantages of mass production with consistent reproducibility stemming from the nanoimprint fabrication process, our gold-capped polymeric pillars are ready for the transition from academic interest into commercialization systems for practical use in diagnostic applications.

High surface area, sol-gel derived macroporous silica films doped with gold nanoparticles (AuNP) are used as a platform for high-density affinity-based immobilization of functional structure-switching DNA aptamer molecules onto Michelson interferometer long-period grating (LPG) fiber sensors, allowing for label-free detection of small molecular weight analytes such as adenosine triphosphate (ATP). The high surface area afforded by the sol-gel derived material allowed high loading of DNA aptamers, while the inclusion of gold nanoparticles within the silica film provided a high refractive index (RI) overlay, which is required to enhance the sensitivity of the LPG sensor according to our numerical simulations. By using a structure-switching aptamer construct that could release an oligonucleotide upon binding of ATP, the effective change in RI was both enhanced and inverted (i.e., binding of ATP caused a net reduction in molecular weight and refractive index), resulting in a system that prevented signals originating from nonspecific binding. This is the first report on the coupling of aptamers to LPG fiber sensors and the first use of high RI AuNP/silica films as supports to immobilize biomolecules onto the LPG sensor surface. The dual functionality of such films to both improve binding density and LPG sensor cladding refractive index results in a substantial enhancement in the sensitivity of such sensors for small molecule detection.

Gradient ion-exchange chromatography (IEC) was interfaced with a low-angle laser light scattering photometer (LALLS) and ultraviolet (UV) and refractive index (RI) detectors connected in series for on-line determination of the differential refractive index (dn/dc) of proteins and eventually their molecular weights (Mw's). Interfacing of gradient HPLC with a RI detector was made possible by using two isorefractive buffers, which helped generate stable baselines for the LALLS and RI detectors. An optically modified, laser based RI detector was used for determination of dn/dc. On-line determinations of dn/dc required smaller amounts of sample compared to off-line determinations (3-4 mg vs 200 mg). Also, dn/dc could be measured by making one repetitive injection of the sample, if the molar absorptivity of the sample was known. dn/dc values determined on-line were in close agreement with those determined off-line. Additionally, percent recovery of the sample was calculated on-line, and this provided true concentration detected at the UV detector, which was then used for determination of Mw.

A metal-clad waveguide (MCWG) sensor comprised of a nanoporous waveguiding layer on a metal cladding layer is advantageous in sensing of biomolecules because of a high surface area of nanopores and a sharp dip in the reflection spectrum due to characteristics of the MCWG mode. Here, a porous anodic alumina (PAA)/aluminum (Al) film was fabricated on a glass substrate as a MCWG sensor with the Kretschmann geometry, and the sensor response was examined for both colorless bovine serum albumin (BSA) and colored metal complexes by measurements of reflection spectra and Fresnel calculations. The BSA adsorption on the PAA layer induced a parallel redshift of the waveguide coupling dip in the reflection spectrum. The experimental results were well simulated by the five-phase Fresnel calculations which indicated that the redshift of the dip was linearly dependent on the adsorbed amount of BSA. When the response of a MCWG sensor with a PAA layer was compared with that of a MCWG sensor with a nonporous alumina layer, the former showed larger redshift than the latter, due to a large adsorbed amount of BSA in the PAA layer with high surface area. For the adsorption of colored Ru[Bphen(3)](2+) and Fe[Phen(3)](2+), the effect of both the real and imaginary parts of the complex refractive index on the sensor response was examined. As a result, a redshift of the waveguide coupling dip was observed for both metal complexes irrespective of the wavelength region examined; this could be ascribed to the changes in the real part of the refractive index due to the adsorption of metal complexes on the PAA layer. Meanwhile, an increase in the reflectivity was observed when the coupling wavelength was close to that of the absorption bands of the metal complexes; this could be ascribed to the changes in the imaginary part of the refractive index of the PAA layer. Using the sensor response caused by the changes in the imaginary part, absorption spectral profiles of metal complexes could be reproduced.

We present a simple chip-based refractometer with a central organic light-emitting diode (OLED) light source and two opposed organic photovoltaic (OPV) detectors on an internal reflection element (IRE) substrate, creating a true dual-beam sensor platform. For first-generation platforms, we demonstrate the use of a single heterojunction OLED based on electroluminescence from an Alq(3)/TPD heterojunction (tris-(8-hydroxyquinoline)aluminum/N,N'-bis(3-methylphenyl)-N,N'-diphenylbenzidine) and light detection with planar heterojunction pentacene/C(60) OPVs. The sensor utilizes the considerable fraction of emitted light from conventional thin-film OLEDs that is coupled into guided modes in the IRE, instead of into the forward (display) direction. A ray-optics description is used to describe light throughput and efficiency-limiting factors for light coupling from the OLED into the substrate modes, light traversing through the IRE substrate, and light coupling into the OPV detectors. The arrangement of the OLED at the center of the chip provides for two sensing regions: a "sample" channel and a "reference" channel, with detection of light by independent OPV detectors. This configuration allows for normalization of the sensor response against fluctuations in OLED light output, stability, and local fluctuations (temperature) that might influence sensor response. The dual-beam configuration permits significantly enhanced sensitivity to refractive index changes, relative to single-beam protocols, and is easily integrated into a field-portable instrumentation package. Changes in refractive index (DeltaRI) between 10(-2) and 10(-3) RI units could be detected for single beam operation, with sensitivity increased to DeltaRI approximately 10(-4) RI units when the dual-beam configuration is employed.

We describe the synthesis and characterization of degradable nanogels that display bulk erosion under physiologic conditions (pH = 7.4, 37 degrees C). Erodible poly(N-isopropylmethacrylamide) nanogels were synthesized by copolymerization with N,O-(dimethacryloyl) hydroxylamine, a cross-linker previously used in the preparation of nontoxic and biodegradable bulk hydrogels. To monitor particle degradation, we employed multiangle light scattering and differential refractometry detection following asymmetrical flow field-flow fractionation. This approach allowed the detection of changes in nanogel molar mass and topology as a function of both temperature and pH. Particle erosion was evident from both an increase in nanogel swelling and a decrease in scattering intensity as a function of time. Following these analyses, the samples were recovered for subsequent characterization by direct particle tracking, which yields hydrodynamic size measurements and enables number density determination. Additionally, we confirmed the conservation of nanogel stimuli-responsivity through turbidity measurements. Thus, we have demonstrated the synthesis of degradable nanogels that erode under conditions and on time scales that are relevant for many drug delivery applications. The combined separation and light scattering detection method is demonstrated to be a versatile means to monitor erosion and should also find applicability in the characterization of other degradable particle constructs.

Gold nanoparticles bound to substrates exhibit localized surface plasmon resonance (LSPR) in their optical extinction spectra at visible and near-infrared wavelengths. The LSPR wavelength is sensitive to the surrounding refractive index, enabling a simple, label-free immunoassay when capture antibodies are bound to the nanoparticles. Gold bipyramids are nanoparticles with a penta-twinned crystal structure, which have a sharp LSPR because of their high monodispersity. Bipyramid substrates were found to have a refractive index sensitivity ranging from 288 to 381 nm/RIU (-0.62 to -0.68 eV/RIU), increasing with the nanoparticle size and aspect ratio. In an immunoassay, the bipyramid substrates yielded higher sensitivity than nanorods and nanospheres. An immunoassay sensitivity constant which depends on both the optical properties of the nanoparticle and conjugation chemistry was found to be K(LSPR) = 0.01 nm x microm(2) for gold bipyramids.

An optically modified high-performance liquid chromatography refractive index detector was developed to allow complete on-line determinations for biopolymer molecular weights. On-line concentration, refractive index, specific refractive index increment (dn/dc2)mu, and Rayleigh factor were determined under flow injection analysis (FIA) and size exclusion chromatography (SEC) conditions using low-angle laser light scattering, ultraviolet, and modified refractive index detection. This instrumental system is capable of determining absolute on-line molecular weights. The error and time requirements involved in conventional methodologies for proteins have been reduced. Sample quantities have been reduced from 150 to 200 mg, in conventional off-line methods, to less than 2 mg for on-line FIA and 0.5 mg for on-line SEC, if mass absorptivities (a) are known. Otherwise, the determination of a will be the most sample-demanding step, requiring about 3 mg of the pure protein. On-line measurements of (dn/dc2)mu are in good agreement with traditional off-line values established at Donnan equilibrium (usually within 5%). In addition, this technique provides true injected mass as determined by the UV detector, after chromatographic exposure where losses may occur, which is then used in the calculation of biopolymer molecular weight.
