Dengue viruses ( DENVs; serotypes 1-4) are members of the flavivirus family. The envelope protein ( E) of DENV has been defined as the principal antigenic target in terms of protection and diagnosis. Antibodies that can reliably detect the E surface glycoprotein are necessary for describing and mapping new DENV epitopes as well as for developing more reliable and inexpensive diagnostic assays. In this study, we describe the production and characterization of a monoclonal antibody (mAb) against a recombinant DENV-2 E protein that recognizes a sequential antigen in both native and recombinant form located in domain II of the E protein of all four DENV serotypes. We confirmed that this mAb, C21, recognizes a sequence located in the fusion peptide. In addition, C21 does not have neutralizing activity against DENV-2 in an in vitro system. Furthermore, the C21 mAb is an ideal candidate for the development of research reagents for studying DENV biology because it cross-reacts with the four dengue serotypes. [ABSTRACT FROM AUTHOR]

To evaluate if heterogeneity of tissue cancer antigen 125 ( CA125) expression is present in epithelial serous adenocarcinomas. Furthermore, to investigate whether there is a correlation between levels of CA125 tissue expression, serum level of CA125, stage, and grade. A total of 10 patients diagnosed with serous ovarian adenocarcinomas were included. Preoperative blood samples were collected to determine serum CA125 levels. Tumor tissue from primary surgery was collected and processed for immunohistochemical analyses. CA125 was expressed in varying degrees in tumor tissues from all patients. Mean tissue CA125 expression for each patient ranged from 36% to 98%. Intrapatient variations in tissue expression ranged from 10% to 90% point. No significant correlations between levels of CA125 tissue expression, serum level of CA125, stage, and grade were found. We found that the tissue expression of CA125 is heterogenic. Although most patients had a high mean expression, it covers a large intrapatient variation in expression. This suggests that if using CA125 as a tissue marker and anti- CA125 (oregovomab) as immunotherapy treatment in future studies, it will be necessary to take heterogeneity into consideration and examine a larger number of biopsies. Saadatnia G, Mohamed Z, Ghaffarifar F, Osman E, Kazemi Moghadam Z, Noordin R. Toxoplasma gondii excretory secretory antigenic proteins of diagnostic potential. APMIS 2012; 120: 47-55. Infection with Toxoplasma gondii is widespread and important in humans, especially pregnant women and immunosuppressed patients. A panel of tests is usually required for diagnosis toxoplasmosis. Excretory secretory antigen (ESA) is highly immunogenic, and thus it is a good candidate for investigation into new infection markers. ESA was prepared from tachyzoites of RH strain of T. gondii by mice intraperitoneal infection. Sera were obtained from several categories of individuals who differed in their status of anti- Toxoplasma IgM, IgG and IgG avidity antibodies. The ESA was subjected to SDS-PAGE, two-dimensional gel electrophoresis and Western blot analysis. Antigenic bands of approximate molecular weights of 12, 20 and 30 kDa, when probed with anti-human IgM-HRP and IgA-HRP, showed good potential as infection markers. The highest sensitivity of the bands was 98.7% with combination of IgM and IgA blots with sera of patients with anti- Toxoplasma IgM+ IgG+. The specificities were 84% and 70% with sera from other infections and healthy controls in IgM blots and IgA blots respectively. By mass spectrometry, the 12 kDa protein was identified as thioredoxin. The two top proteins identified for 20 kDa molecule were microneme protein 10 and dense granule protein 7; whereas that for 30 kDa were phosphoglycerate mutase 1 and phosphoglycerate mutase. Sun Q, Yang X, Asim MBR, Jiao F, He X, Zhong B, Li D, Lu S. Different challenge terms determine disease patterns of antigen-induced pulmonary inflammation in E3 rats. APMIS 2011; 119: 229-38. Antigen induced pulmonary inflammation (AIPI) in rats, a classic animal model for asthma, has greatly contributed to the understanding of the disease pathogenesis, especially for the inflammation process. E3 rats are recently used to induce AIPI model for its susceptibility to pulmonary inflammation, but the features of AIPI with different antigen challenge terms on E3 rats require to be elucidated systemically. The aim of this study was to compare AIPI disease patterns in E3 rats with different challenge terms. E3 rats were immunized and challenged with ovalbumin (OVA) for 1, 4, and 8 weeks. Histological methods were used to determine morphological changes in lungs and cell types in bronchoalveolar lavage fluid. Nitric oxide (NO) concentration was assayed by Griess method. IL-4 and TGF-β expression were detected by real-time PCR. ELISA was used for the determination of serum IgE and OVA-specific IgG1. The results showed that all the sensitized E3 rats had a strong influx of eosinophils into the airway. In 1-week challenge group, the rats showed stronger inflammation, such as elevated levels of NO, delayed type hypersensitivity, IL-4 expression, and inflammatory cell infiltration; while in 8-week challenge group, rats manifested significant tissue destruction, accumulation of collagen and mucus production, and higher levels of antibody production, and TGF-β expression. Hence, the detail characterizations of AIPI model challenged for different terms demonstrated that E3 rats challenged with antigen for 1 week are suitable for studying acute pulmonary inflammation; meanwhile, the model established in the rats challenged for 8 weeks is appropriate for understanding pathogenesis of lung remodelling in chronic inflammation. The aim of the study was to establish possible factors which play a role in progression of gastritis to atrophic gastritis in long-term follow-up among the Estonian population, to assess the association between the host immune response and different Helicobacter pylori antigens and autoantigens in relation to the histological parameters of gastritis in the antrum and corpus. ELISA and immunoblot were used for detection of IgG to H. pylori acid glycine-extracted cell surface proteins, CagA protein, and H. pylori HSP60. Anticanalicular autoantibodies (ACAB) in the serum were evaluated according to Faller et al. (1996). Apoptosis was evaluated using the TUNEL method. Study subjects were 1958 persons from an unselected Estonian population, and 70 persons from a sample from Saaremaa Island, who had been investigated over a period of 18 years. Seropositivity for CagA was a sign of gastritis activity [OR=14.8 (4.5–50.3)] and atrophy [OR=7.0 (2.1–23.1)] and might predict development of atrophy, particularly in the corpus [OR=7.1 (1.8–27.7)]. The prevalence of ACAB increased significantly with duration of H. pylori gastritis from 22% in 1985 to 46% in 1997 (p=0.004). Immune response to H. pylori HSP60 indicates chronic inflammation in the antrum (p=0.003). Apoptosis of gastric epithelial cells is largely dependent on grade of activity of gastritis, and, particularly in the antrum, on grade of H. pylori colonization (p=0.01; p=0.02), but is not associated with development of atrophy. The article focuses on diagnosis of Borna disease virus (BDV) infection. It discusses how applied diagnostic systems influence the pathogenicity concepts of BDV, especially if low sensitivity levels obscure the detection of silent carriers. It looks at how the discovery of BDV circulating immune complexes (CIC) and antigen-based EIA systems inevitably implies a shift to the epidemiologically consistent concept about BDV as a fairly prevalent mild pathogen.

Serological diagnosis of Chamydophila pneumoniae is usually undertaken by complement fixation test (CFT) or by microimmunofluorescence (MIF). A number of commercial methods for detecting C. pneumoniae-specific IgG have been developed. The aim of this study was to compare the performance characteristics of six methods for the diagnosis of pneumonia due to C. pneumoniae, including CFT (in house), MIF (Vircell, Spain), and four ELISAs (Medac, Germany; Savyon, Israel; Serion, Germany; and DRG, Germany). ELISA-Medac, ELISA-Savyon, ELISA-DRG and MIF use C. pneumoniae antigens while ELISA-Serion and CFT use Chlamydophila genus-specific antigen. Acute and convalescent samples from 85 pneumonia patients were studied. Using CFT, cases were initially classified as due to Chlamydophila (43 cases); to other agents (23 cases) (influenza A and B, Mycoplasma pneumoniae, respiratory syncytial virus, adenovirus and Legionella pneumophila); or as negative (19 cases). Cases were considered positive if they showed seroconversion, a significant rise in titer or high titer; and were finally classified as positive if they gave a positive result in at least three assays. Sensitivity values ranged from 87% to 97.8%; and specificity from 84.6% to 97.4%. In conclusion, the assays compared appear to be useful tools for the diagnosis of pneumonia due to Chlamydophila. Lohse L, Nielsen J, Kamstrup S, Oleksiewicz MB, Eriksen L. Porcine humoral immune responses to multiple injections of murine monoclonal antibodies. APMIS 2005;113:489–96. In humans and cattle, multiple injections of murine monoclonal antibodies (m-mAbs) induce anti-mouse antibody responses. The objectives of the present study were to investigate whether a similar response could be seen when pigs were subjected to m-mAb therapy, and to study the kinetics of such a response. In two separate animal experiments, long-term treatment was performed with m-mAbs at low-dose levels and therapeutic levels, respectively. Two specific m-mAbs that recognized cognate antigen in the pigs (CD4 and CD8 surface antigens on T-lymphocytes) and two irrelevant control m-mAbs having no cognate antigen in the pigs were used. Enzyme-linked immunosorbent assays (ELISA) were used to quantitate the circulating m-mAbs, as well as the induced pig anti-mouse antibodies (PAMA), in serum samples from m-mAb-treated pigs. As expected, we generally saw vigorous PAMA responses within 10 days after the start of m-mAb treatment with the specific m-mAbs. However, the different mAbs showed striking differences in the kinetics and levels of PAMA responses, differences that might be ascribed to the m-mAb formulation and epitope specificity. In conclusion, treatment of pigs with m-mAbs against T-cell surface antigens induced rapid PAMA responses. This may influence and possibly decrease the effect of the m-mAb treatment by narrowing the time period where m-mAbs can efficiently be used for cell depletion. [ABSTRACT FROM AUTHOR]

The article highlights a study which examined Borna disease virus (BDV) infection in children with psychiatric disorders. The study involved children from Rome, Italy who have either psychiatric diseases or participated in an epidemiological study and their sera were tested for BDV infection by a triple enzyme-linked immunosorbent assay (ELISA). No significant differences were found between the antibodies, antigens and circulating immune complexes (CIC) in the two groups.

The article discusses several research findings on anti-neutrophil cytoplasmic antibody (ANCA). Several research suggested that ANCA could be directed against several neutrophil cytoplasmic antigens. Other studies srutinize the role of the nasal carriage of Staphylococcus aureus but its mechanisms remain unclear. A large number of researchers believed that granulomas are producing not only ANCA but also a number of important disease regulating substances being released locally and systemically.
