Microplastics are plastic particles or fibers with a diameter of less than 5 mm, and they widely exist in the environment and pose potential risks to the ecosystem and human health. Microplastics detection can provide basic data for formulating effective environmental protection strategies. In this paper, the physical, chemical and biological detection methods of microplastics are reviewed, and the advantages and disadvantages of different methods are analyzed. China attaches great importance to food security. With the strong promotion of the government, China's grain output has achieved a great feat from "shortage" to "surplus in bumper years" and then to "rising steadily", achieving a record "increase for the tenth consecutive year". From the current balance of food supply and demand in China, the food security situation can be guaranteed on the whole. However, if we analyze the food security in China from the perspective of agricultural sustainable development strategy, there are some difficulties, such as the reduction of cultivated land area, the decline of cultivated land quality, the fragility of agricultural ecological environment, and the transformation of residents' consumption structure. This paper analyzes the difficulties faced by the development of food security in China under the strategy of sustainable agricultural development, and puts forward that to implement the strategy of sustainable agricultural development, it is necessary to realize the coordinated development of food security, science and technology, ecological environment, resources and society. The application of new media technology in rural areas of China provides more convenient conditions for agricultural products publicity, transaction information dissemination and user feedback. In particular, e-commerce platform based on Internet technology provides more convenient sales channels for characteristic agricultural products. Through the research on the application of new media technology in rural areas of Anhui Province, this paper explores the actual effect of new media technology on the development of rural e-commerce. The questionnaire survey is used to understand the development status of new media in rural areas of China. They are; the development of "Internet government affairs" has begun to take shape; the official account of WeChat has shortened the time and space between the production and consumption of agricultural products. Live interaction gives full play to the powerful role of fan economy; digital media has not been widely used. Through the form of on-the-spot visits, this paper expounds the application effect of new media in the agricultural field of Anhui Province from three perspectives of the government, agricultural enterprises and farmers. It analyzes the problems existing in the application of new media in rural e-commerce in Anhui Province. They are; the new media market is still full of problems; The infrastructure construction of rural e-commerce is relatively poor; Farmers lack the guidance of professional talents; farmers' acceptance and learning ability is poor. 60. Analysis on the Influencing Factors and Control Measures of "Non-Grain Conversion" of Cultivated Land in Agricultural Land Transfer.

- Abstract
Through the method of literature analysis, this paper analyzes the present situation of the "non-grain conversion" of cultivated land in the transfer of agricultural land in China, summarizes the influencing factors and consequences of the phenomenon of "non-grain conversion" of cultivated land in the transfer of agricultural land, and finally puts forward some measures. The study found that costs and benefits are the fundamental factors affecting the "non-grain conversion" of cultivated land in agricultural land transfer, and other main factors include government and policy guidance, industrial and commercial capital to the countryside, labor transfer and so on. The "non-grain conversion" of cultivated land in the transfer of agricultural land is a serious threat to China's food security, leading to the decline of the quality of agricultural land and the destruction of the ecological environment, as well as the risk of breach of contract in "industrial and commercial capital to the countryside". Based on the understanding of the influencing factors of non-grain conversion, this paper puts forward the following measures: strengthening the control of the use of cultivated land in agricultural land transfer; implementing accurate subsidies for grain cultivation and ensuring the benefit of grain growers; adopting grain planting incentive mechanism to increase grain enthusiasm; strengthening media publicity and education to enhance food security awareness. Flowering is a prerequisite for apple fruiting, and apple flower buds are mixed buds, that is, the vegetative organs and flower structure exist in the same terminal bud simultaneously, which are formed in the year before flowering and fruiting, mainly including spur terminal buds and axillary buds. The infrequent formation of flower buds during its growth and biennial bearing are closely related to flower bud differentiation. Therefore, this paper reviews the research progress of flower bud differentiation of apple from the morphological differentiation, plant hormones and flowering-related genes, in order to provide a theoretical reference for efficient cultivation and stable yield of apple. 66. Cloing and Bioinformatics Analysis of ndk Gene from Vibrio alginolyticus.

- Abstract
[Objectives] The paper was to clone and analyze bioinformatics of ndk gene from Vibrio alginolyticus. [Methods] A pair of specific primers was designed based on the ndk gene sequence of V. alginolyticus HY9901. The full length of ndk gene was amplified by PCR and bio informatics analysis was performed. MEGA 5.0 software was used to construct NDK phylogenetic tree by neighbor-joining method. SWISS-MODEL program was used to obtain the three-dimensional structural model of single subunit from NDK protein. [Results] The ndk gene, molecular structural formula C7CB Hlow N]92 02J4 S7, was 426 bp in total, encoding 141 amino acids, with the theoretical molecular weight of 15.871 99 kD and the theoretical pl value of 5.13. The prediction results of protein subcellular localization, Signal? 5. 0, TMHMM Server 2.0 and SoftBerry-Psite showed that NDK mainly existed in the cytoplasm, and the protein was unstable and hydrophobic. There was neither signal peptide cleavage site, nor transmembrane region and KEGG metabolic pathway. The amino acid sequence had two protein kinase C phosphorylation sites, a casein kinase II phosphorylation site, a N-myristoylation site, three microbody C-terminal target signal sites, and a nucleoside diphosphate kinase active site. Homology analysis showed that the NDK of V. alginolyticus had high homology with that of V. diabolicus, with a similarity of 98.58%. Analysis of the structural functional domain revealed that the protein had one NDK structural functional domain. The prediction results of secondary structure showed that the a-helix, random coil, [3-sheet and extended strand accounted for 53.19%, 28.37%, 7.09% and 11.35%, respectively. Analysis of NDK protein via STRING database demonstrated that the proteins interacting with NDK protein were NrdA, NrdB, GmK, CmK, TmK, PyrG, PyrH, RelA, FolE and SpoT. "Three new developments" is the guiding ideology of Chinese-style high-quality development in the new era. Jujube (Zizyphus jujube Mill.) is a plant of Zizyphus of Rhamnaceae, which originated in China and has been listed as one of the "five fruits" since ancient times. Xinjiang is the most important jujube producing area in China, and its yield has accounted for 50% of the national jujube yield. In the five prefectures and cities of southern Xinjiang, as well as the regimental farms of southern and eastern Xinjiang of Xinjiang Production and Construction Corps, the jujube industry is an advantageous characteristic industry in agriculture. The development prospect of Pennisetum Sinese Roxb. is receiving much concern of domestic researchers. In this paper, literature of Pennisetum Sinese Roxb. was taken as basis, and bibliometric analysis method was used to make statistics of distribution of years, research institutions, authors, and research fields. The results showed that research fields of Pennisetum Sinese Roxb. in China were mainly ecological restoration, accounting for 13.27% of the total literature; the research took on dynamic and diversified cooperation trend. In recent 5 years, the number of papers significantly increased, accounting for 93. 04% of the total literature. 72. Strategies for Prioritizing Ecological Construction with Consideration of Agricultural Industry Development in Northern China's Agropastoral Ecotone: A Perspective of Opportunity Cost.

In northern China's agro-pastoral ecotone, there are conflicts between agricultural industry and ecological construction resource competition, and also problems urgently to be solved for economic growth and sustainable ecological improvement. Taking the development concept of "lucid waters and lush mountains being mountains of gold and silver" as the guiding principle of prioritizing ecological and green development, this paper studied the strategies for prioritizing ecological construction with consideration of economic development of agricultural industry from the perspective of opportunity cost. Through the analysis of opportunity cost, externality, and Pareto optimal theory, and using the calculation method of the unit equivalent factor value of terrestrial ecological service function, this paper calculated the ecological service value of forest, grassland, and farmland. It found that the ecological value of forest and grassland per unit area was greater than the ecological value of agricultural industry. This indicates that the opportunity cost of farmland for ecological construction was less than that of agricultural industry. Based on the theoretical and quantitative analysis, it came up with recommendations for establishing a comprehensive system for prioritizing ecological construction with consideration of agricultural industry development and establishing and improving the incentive mechanism of ecological construction. 73. Spatial Differences in the Development Level of Agricultural Producer Services in China.

Due to the differences in regional resources level, economic development level and government support strength, there are clear regional differences in the development of China's agricultural producer service industry. Based on the data of agricultural producer services from 31 provinces in China from 2003 to 2016, the spatial differences in the development level of China's agricultural producer services were measured in this paper using the Theil index. The results showed that there are significant spatial differences in China's agricultural producer services, and regional differences are the main reason for the overall differences. To reveal the influencing factors of the spatial differences in the level of development of China's agricultural producer services, theoretical analysis was conducted in this paper from four aspects: location factors, economic factor, policy factor and technical factors. Using the panel data from 2003 to 2016, empirical research was carried out. It was found that regional economic development level and government support strength are the main factors affecting the differences in the development level of agricultural producer services in China. 74. Competitiveness and Complementarity of Cocoon Silk Trade between China and Southeast Asia.

In the context of the Belt and Road Initiative,it is of great significance to study the trade ties between China and Southeast Asia with cocoon silk as a link. Trend analysis,trade competitiveness index and trade complementarity index were comprehensively used in this paper to study the competitiveness and complementarity of the cocoon silk trade between China and Southeast Asia. The results show that the export trade volume of cocoon silk of China and Southeast Asia is decreasing while fluctuating,but it is difficult for other regions to replace their export trade status in the world market. The development trend of the replacement of the silkworm cocoon trade by the raw silk trade is presented. The cocoon silk trade of China and Southeast Asia is characterized by dislocation competition. China’s raw silk export trade is competitive and has become the leader of the raw silk export camp. In order to promote the economic and financial development of Henan Province,this paper constructs the financial ecological system of Henan Province,evaluates the " quantity" of Henan financial ecology by entropy method-TOPSIS method,and evaluates the " quality" of Henan financial ecology by DEA-Malmquist method. It is found that the " quantity" of financial ecology in Henan Province shows the early advantage,but the power of late advantage is insufficient,and the " quality" of financial ecology in Henan Province is improved obviously,but it only depends on the technological progress,showing a lack of sustained momentum. Based on this,this paper puts forward some suggestions on promoting the development of digital finance,creating a good credit environment and actively preparing for the establishment of a diversified financial system,so as to improve the quality of financial ecology in Henan Province. - Abstract
In order to explore researches about the chromosome karyotype analysis and fluorescence in situ hybridization (FISH) technology in China, using the bibliometric method, taking "fluorescence in situ hybridization (FISH) " and "chromosome" as Key words:, this paper made a statistical analysis on the literature published in China National Knowledge Infrastructure (CNKI) during 2002 -2016. The results indicated that the number of papers published in 2002 was the smallest (37), while the number of papers published in 2012 was the largest (125). In terms of the distribution of organizations of authors, in 1201 papers, 11 organizations published papers ≥15, accounting for 21.65%. In terms of distribution of papers published by different periodicals, 11 periodicals published papers ^10, accounting for 17. 65%. In terms of the papers supported by foundation projects, in all papers searched, 377 papers were supported by foundation projects, accounting for 31.39%. In terms of the distribution of doctoral and master's dissertations, 259 papers were master's dissertations, accounting for 21.57% ; 92 papers were doctoral dissertations, accounting for 7. 66%. - Abstract
[Objectives] To isolate and identify characteristics of the cellulose-degrading bacteria. [Methods] In view of the poor effect of crop straw composting, the strains which can degrade cellulose were isolated and purified from the soil piled with rice straw, and the strains A2 and A5 with high efficiency of cellulose degradation were finally screened through the transparent circle, cellulase activity and filter paper disintegration experiments. [Results] It was confirmed that the strains A2 and A5 could degrade cellulose well. The results of 16s rDNA showed that A2 was Pseudoxanthomonas mexicana and A5 was Bacillus cibi. [ Conclusions] The results of this study are expected to provide high-quality strain resources for the degradation of cellulose in straw, and have important application value for improving the efficiency of straw composting. Based on the panel data of 41 cities in the Yangtze River Delta from 2007 to 2018, this paper empirically tests the impact of digital economy development on urban ecological efficiency in the Yangtze River Delta. The results show that the development level of digital economy in Yangtze River Delta urban agglomeration is fluctuating and rising; the development of digital economy has a significant positive role in promoting the improvement of urban ecological efficiency; there is significant regional heterogeneity in the promotion of ecological efficiency by digital economy, especially in central cities. 80. Study on Top Grafting Technology of Bare Scions of Walnut Trees in Eastern Hebei.

- Abstract
[Objectives] To accelerate the improvement of walnut varieties and change the cultivation status cf low yield and poor quality. [Methods] Through experimenting on the optimization technology of walnut trees, this paper studied the suitable top grafting period and scientific top grafting protection measures of walnut trees in Eastern Hebei, and established the top grafting technology system of walnut. Moreover, this paper explored the impact of different time and scientific management and protection measures on the survival rate and growth of bare scion top grafting cf walnut trees. [Results] The grafting time had a great impact on the growth of new shoots, and there were significant differences in the survival rate among different grafting periods. The highest grafting survival rate was 96.97% on April 27 and the lowest grafting survival rate was 66.67% on May 4; the average survival rate was 78.10%. Among them, the survival rate of top grafting was the highest on April 20 and April 27. The growth of new shoots grafted in the early stage was greater than that in the late stage. The growth of new shoots and branches on April 8 and April 20 were the largest, with lengths of 160.3 and 143.7 cm; the thickness was 1.72 and 1.43 cm; the branch lengths were 79.8 and 63.4 cm; the branch thickness was 0.98 and 0.82 cm; the growth index was also the highest on April 8 and April 20 at 10, 20, 30 d after grafting, which was significantly different from other time periods. [Conclusions] Walnut trees with early grafting in Eastern Hebei can quickly restore their crow growth and had higher yield, thus this technology can be widely applied in Eastern Hebei or Yanshan area. Based on the analysis of rice planting and export development in Heilongjiang Province in 2013 - 2021, this paper comprehensively used three export competitiveness analysis methods, namely, trade competitiveness index, revealed comparative advantage (RCA) analysis and international market share analysis, to compare the four major rice exporters in the world and the three major rice export provinces and cities in China. Besides, it evaluated the rice export competitiveness of Heilongjiang Province from multiple perspectives. It analyzed the reasons for low competitiveness in terms of export product structure, export market structure, production and processing technology, and brand building. 87. Establishment of Molecular Biological Method for Identification of Bacteria by 16S rDNA and gyrB Gene.

- Abstract
[Objectives] The paper was to establish a molecular biological method for identification of bacterial strains. [Methods] The thalli of standard bacterial strains existing in the laboratory were collected and genomic DNA was extracted for amplification of 16S rDNA and gyrB gene. The 16S rDNA and gyrB gene sequences were obtained after sequencing. Sequences were aligned and analyzed via EzBioCloud and NCBI database, and phylogenetic trees were constructed to determine the species relationship of strains. Meantime, they were compared with known strains. [Results] This method could identify 5 standard strains accurately to the species level. The 16S rDNA and gyrB gene sequences were aligned and analyzed in EzBioCloud database and NCBI database. The strain with the max score was consistent with the known strain. And the query cover and ident were both above 99%. [Conclusions] The established molecular biological method for identification of bacterial strains by 16S rDNA and gyrB gene has good accuracy, which effectively solves the problem that the laboratory identification of bacteria relies on traditional methods and the accuracy can not be guaranteed, and further improves the identification ability of laborator)' bacterial strains. Taking Yunnan Province as an example, this paper calculated the real integrated land productivity (RILP) of 16 prefectures (cities) in Yunnan Province for the past 11 years (2009-2019), and analyzed its spatial distribution pattern. The research results show that the RILP of the whole Yunnan Province and all prefectures (cities) showed a slow upward trend. Among them, the province's RILP showed the highest increase in 2019 compared with the previous year; the RILP of all prefectures (cities) in Yunnan Province has significant spatial autocorrelation, showing obvious positive spatial agglomeration characteristics. Based on this, this paper proposes to improve the overall RILP in various regions according to local conditions and formulate countermeasures for coordinated development of various prefectures (cities). [Objectives] This study was conducted to gain an in-depth understanding of the research status of fields related to soil aggregates. [ Methods] The academic papers published from 2001 to 2020 in the field of soil aggregates were searched in the CNKI database serving as the statistical source using the subject terms "soil" and "aggregate" under advanced search, and visually analyzed by bibliometric statistical methods, from the perspectives of the annual number of published papers, the number of papers published by authors, and highly cited papers. [ Results] The trend of the number of published papers in the past 20 years was mainly divided into three stages; 2001 -2007, 2008 - 2016, and 2017 -2020. There were 192 first authors in the literature related to soil aggregates. The authors with the top 3 published papers were Wang Erli, 2hang 2hengxiong, Li Yangbing, and Yang Yonghui (tied for third). Three highly cited papers were published in journals related to "ecology", and the top 10 most cited papers focused on the "formation and stability" of soil aggregates and the relationship between aggregates and soil organic matter and biochar. 97. Effect of Magnetic Field Technology on Preservation Quality of Fresh-cut Apple.

- Abstract
[Objectives] The paper was to study the effect of magnetic field technology on physicochemical properties of fresh-cut apple during storage. [Methods] Using fresh-cut apples as materials, magnetic field assisted refrigeration and conventional refrigeration control groups were set up, and the preservation effects of magnetic field assisted refrigeration on fresh-cut apples were tested by measuring the changes of microorganisms and physicochemical indicators. In this experiment, the conditions of magnetic field assisted refrigeration group was set as follows: temperature 4 °C, magnetic field intensity 5 mT, static magnetic field, and storage for 6 d. [Results] At 6 d post storage, the weight loss rate, total number of colonies and total number of fungi of fresh-cut apples increased; the i" value and hardness showed a downward trend; the soluble solid content first increased and then decreased; and the total acid decreased first and then increased. The above indicators in magnetic field assisted refrigeration group changed more gently than that in conventional refrigeration group. Compared with conventional assisted refrigeration, the weight loss rate of fresh-cut apples treated by magnetic field combined with 4 9C refrigeration was reduced by 68.00%; the hardness was increased by 12.23%; the total number of colonies and total number of fungi were reduced by 64.15% and 37.50%; and the L" value did not change much. High content of soluble solids and total acid was maintained. [Conclusions] Magnetic field assisted refrigeration delays quality deterioration of fresh-cut apples during storage, prolongs shelflife, and further improves their edible quality and commodity value. 100. Distribution Characteristics of Soil Total Nitrogen in Low-efficiency Forest Land in the Northern Windy Desert Area of Jingbian County.

[Objectives] The paper was to investigate the distribution characteristics of soil total nitrogen in low-efficiency forest land in the northern windy desert area of Jingbian County. [Methods] The distribution of soil total nitrogen in the 0-40 cm soil layer of 5 towns in the northern windy desert area of Jingbian County was studied through field sampling and laboratory detection. [Results] The average soil total nitrogen contents of Hongdunjie Town, Haizetan Town, Huanghaojie Town, Ningtiaoliang Town, Dongkeng Town and windy desert area in the 0-20 cm soil layer were 0.259, 0.224, 0.242, 0. 248, 0.431 and 0.275 g/kg, respectively. The soil total nitrogen content in Dongkeng Town was higher than those in other towns, while there was little difference among other regions. The average total nitrogen contents in the 20 -40 cm soil layer were 0.239, 0.285, 0.113, 0.262, 0.349 and 0.241 g/kg, respectively. The soil total nitrogen content in Huanghaojie Town was slightly lower than those in other towns, while that in Dongkeng town was higher. The variation coefficient of soil total nitrogen content in the survey area was greater than 30%, and there was great difference in spatial distribution. With the increase of soil depth, there was little change in soil total nitrogen content, namely the soil total nitrogen contents in 5 towns and windy desert area were not statistically different in the 0-40 cm soil layer. According to the nutrient grading standard of the second national soil survey, the soil total nitrogen content in the survey area was in the deficiency grade.
